Abstract:Four previously unreported phenylpropanoid glycosides ( 1 – 4 ), together with four known analogues ( 5 – 8 ), were isolated from the leaves of Illicium dunnianum . The structures of these new compounds were elucidated based on spectroscopic analysis (HR-ESI-MS, NMR, IR, UV) and chemical methods. Abstract:The ethyl acetate extract from leaves of Illicium micranthum Dunn exhibited radical scavenging ability on DPPH radical-scavenging assay. Further phytochemical investigation led to the isolation of a new sesquiterpenoid, illicinolide C (1), and eight known compounds (2-9). The structural elucidations of the compounds were based on spectroscopic methods and by comparison of their spectroscopic data with those reported in the references. Except compound 9, all the other compounds were isolated from I. micranthum Dunn for the first time, while compounds 1 and 2 were new to the Illiciaceae family.

Abstract:Nine new terpenoids (1-3, 5-10), including three cyclolanostane triterpenoids (1-3) and six isopimarane diterpenoids (5-10), along with six known terpenoids (4, 11-15), were isolated from the leaves and twigs of Dysoxylum gotadhora. Compound 1 represents a class of rare natural 21,24-epoxy cyclolanostane-type triterpenoids, and compounds 2 and 3 are the first examples of 21,25-epoxy cyclolanostane-type triterpenoids. Compounds 1, 2, 4, 8, and 15 exhibited noteworthy inhibition of nitric oxide production induced by lipopolysaccharide in RAW 264.7 cells with IC50 values of 25.5, 41.5, 27.4, 14.5, and 45.2 mu M, respectively.

Abstract:A new triterpene glycoside ilexdunnoside A (1) and a new sulfated triterpene derivative ilexdunnoside B (2), together with five known analogues 3-7 were isolated from the roots of Ilex dunniana Levl. The structures were established by NMR spectroscopic analysis and acid hydrolysis. Results of an in vivo study of the biological activity showed that 75% ethanol and n-butanol extracts of the plant displayed anti-inflammatory activities against ear edema in mice, with inhibition rates of 23.5% and 37.5%, respectively, at a dose of 50 mg/kg. Furthermore, Compounds 1, 2 and 3 exhibited moderate indirect inhibitory effects on lipopolysaccharide-induced NO production in BV2 microglial cells in vitro, with IC50 values of 11.60, 12.30 and 9.70 mu M, respectively.

Abstract:Thirty-six compounds were isolated from extract of the stem bark of Illicium burmanicum, including twelve previously undescribed prenylated C6-C3 compounds and a norsesquiterpene lactone: illicidione D (1), illicidione E (2), illicidione F (3), illicidione G (4), (2R,4S,11R)-12-O-ethylillifunone C (5), (2R,4S,11R)-illifunone C-12-O-β-d-glucopyranoside (6), (2R,4S,11R)-2-hydroxyillifunone C (7), 4-epi-2,3-dehydroillifunone C (8), illiburmanone A (9), illiburmanone B (10), illiburmanlactone A (11), (2S,4S,5S,11R)-2,3-dihydroillicione E (12), and illiburmanolside A (13). Their structures were determined based on extensive spectroscopic data analyses, including MS, NMR, and ECD spectra. The anti-inflammatory activity of the isolated compounds (1-36) was evaluated, and compounds 7, 12, 14, and 18 exhibited inhibitory effects in RAW 264.7 cells induced by LPS.

Abstract:The occurrence of inflammation is closely related to the activation of the NLRP3 inflammasome. IL-1β produced during the activation of the NLRP3 inflammasome has strong pro-inflammatory activity and can also promote the release of inflammatory factors by other immune cells, exacerbating inflammatory damage to tissues. Utilizing IL-1β as the detection index to find small-molecule inhibitors targeting NLRP3 from natural products will benefit the search for drugs for inflammation-related diseases. During the exploration of anti-inflammatory active components derived from the flowers of Dolichos lablab L., an ingredient in traditional Chinese medicine with dual applications in both medicinal treatment and dietary consumption, fourteen compounds (1-14), including seven previously unreported ones, named flosdolilabnitrogenousols A-D (1-4) and flosdolilabsaponins A-C (5-7), were found. Their structures were established through extensive NMR spectra determination, HR-ESI-MS analysis, ECD calculations, and chemical reactions. Flosdolilabsaponin A (5) stands out as an exceptionally rare tetracyclic lactone oleane-type saponin. Additionally, the inhibitory activity on IL-1β release of all compounds, without cytotoxicity, was evaluated using BMDMs stimulated with LPS/Nigericin. An Elisa assay revealed that compounds 1, 8, 9, and 11-14 exhibited significant inhibition of IL-1β release at a concentration of 30 μM. Structure-activity relationships were also discussed. This study indicates that D. lablab flowers possess anti-inflammatory activity, which might exert its effect by suppressing the activation of the NLRP3 inflammasome.

Abstract:Three new lignans, melodinins A-C (1-3), together with four known analogues (4-7), were isolated from the stems of Melodinus suaveolens. Their structures including relative configurations were determined by spectroscopic methods, including 1D and 2D NMR, and HRESIMS. Their absolute configurations were unambiguously assigned by electronic circular dichroism (ECD) experiments, especially the application of ECD exciton chirality method. Anti-inflammatory evaluation revealed that compounds 1, 2, 4, and 5 exhibited moderate inhibitory effects on beta-glucuronidase release in rat polymorphonuclear leukocytes (PMNs) induced by platelet-activating factor, with IC50 values ranging from 1.68 to 9.56 mu M.

Abstract:Five novel lignans, namely styraxjaponica A-E ( 1–5 ), together with eight known compounds ( 6–13 ) were isolated from the leaves of Styrax japonicus Siebold & Zucc. Their chemical structures were characterized by extensive analysis of 1D and 2D NMR, UV, IR, HRESIMS spectroscopic analysis as well as by comparison to the literature. The absolute configurations of the new compounds were further determined by quantum chemical electronic circular dichroism (ECD) calculations powered by time-dependent density functional theory (TDDFT). Moreover, the anti-inflammatory effects of compounds 1–5 in lipopolysaccharide (LPS)-induced RAW 264.7 cells were also evaluated by measuring nitric oxide (NO) concentrations. All compounds displayed significant anti-inflammatory activity without affecting cell viability in vitro .

Abstract:Five new neolignans (1-4 and 9), two pairs of neolignan epimers (5-8), and two new aromatic glycosides (10 and 11) have been isolated from the stem bark of Illicium difengpi. Their structures were determined by spectroscopic methods, including 1D and 2D NMR, HRESIMS, CD experiments, and chemical methods. The absolute configurations of the 3,4-diol moiety in 1 and 1,3-diol moiety in 2 were confirmed by Snatzke's method, observing the induced circular dichroism after addition of dimolybdenum tetraacetate in DMSO. Compounds 3, 4, and 11 exhibited moderate anti-inflammatory activities with IC(50) values ranging from 1.62 to 24.4 microM, while compound 3 displayed antioxidant activity with an IC(50) value of 42.3 microM.

Abstract:Six new dihydrobenzofuran lignans, named illiciumlignans A⿿F (compounds 1⿿6), along with 15 known compounds (7⿿21) were isolated from the branches and leaves of Illicium wardii. The structures of 1⿿6 were determined using a combination of 1D and 2D NMR, HR-ESI⿿-MS, and CD spectroscopic data. Illiciumlignan D (4) is the first reported dihydrobenzofuran lignan arabinofuranoside that is derivatized with the arabinofuranose moiety on C-9⿲. Compounds 1⿿21 were evaluated for cytotoxic activity against four human cancer cell lines. Compounds 8, 12 and 20 exhibited significant activity against human cancer cell lines (A549, SKOV3, HepG2 and HCT116), with IC50 values ranging from 2.7 to 14.9μM.

Abstract:Amomum villosum Lour. is a medicinal and edible plant, whose medicinal parts are dried and mature fruits, and its stems and leaves are always treated as waste. HPLC-MS/MS analysis showed that the chemical components contained in the stems/leaves of A. villosum and those in fruits are quite different. To discover potential active ingredients from the stems/leaves of A. villosum, phytochemical evaluation of the stems/leaves of A. villosum was conducted to isolate and identify-four undescribed compounds (1, 2a, 2b, and 3) along with 41 known ones (4a, 4b, 5a, 5b, and 6-42). All isolated compounds were assessed for their anti-inflammatory and antioxidant activities. Among them, compounds 5b, 33, 34, and 38 exhibited anti-inflammatory activity, and compounds 1, 4a, 4b, 6, 7, 15, 33, 35, 37, and 41 showed antioxidant effects. Among them, the new compound 1 showed a significant antioxidant effect via activation of NRF2/HO-1 pathways. Therefore, the leaves and stems of A. villosum may be served as a potential medicine or dietary supplement for preventing and treating diseases resulting from inflammation and oxidative stress.

Abstract:Two new sesquiterpenes (1 and 2), one new alkaloid (4), one new neolignan glycoside (14), together with 19 known compounds were isolated from the branches of Alangium barbatum. Their structures were established by spectroscopic analysis. The absolute configuration of 21 was determined for the first time by single-crystal X-ray diffraction analysis with Cu-K alpha irradiation. Some of isolates were evaluated for their inhibitory effects on NO production in LPS-induced RAW264.7 macrophages. Compounds 1, 3 and 9 at 10 mu M concentrations exhibited mild inhibitory effects on NO production in the range of 25.0 to 32.4. Moreover, these compounds at 25 mu M concentration were also found to mildly suppress NF-kappa B activation in TNF-alpha induced expression of NF-kappa B-Luc in Hela cells with inhibition percent in the range of 15.0 to 25.0%.

The structures of compounds 1–4 were determined by extensive spectroscopic data analysis and comparison with reported references. Compounds 1–4 were evaluated for their inhibitory activity against LPS-induced NO production in RAW264.7 cells. Among them, compounds 1 and 2 showed certain anti-inflammatory activities with IC 50 values of 4.09 ± 0.52 and 6.24 ± 0.64 mM, respectively.

Abstract:Three new labdane‐type diterpenoids, calcaratarin E, villosumtriol, and 12‐epi‐villosumtriol (1‐3) were isolated from the fruits of Amomum villosum, along with seven known diterpenoids (4‐10). Through comprehensive analysis of chemical evidence and spectral data including UV, 1D and 2D NMR, HR‐ESI‐MS, IR, and X‐ray crystallography, the structures of these novel compounds were successfully determined. Additionally, the inhibitory effects of compounds 2‐10 on NO production in lipopolysaccharide (LPS)‐induced RAW264.7 cells were evaluated. Notably, compound 6 exhibited the most significant inhibitory effect with an IC50 value of 1.74 ± 0.69 μM.

Abstract:Licorice, the root and rhizome of Glycyrrhiza uralansis Fisch, is one of the most frequently used Traditional Chinese Medicines in rigorous clinical trials to remove toxins and sputum, and to relieve coughing. However, the aerial parts are not used so widely at present. It has been reported that the aerial parts have many bioactivities such as anti-microbial and anti-HIV activities. In this study, we aimed to discover the bioactive compounds from the leaves of G. uralensis. Four new compounds, licostilbene A-B (1-2) and licofuranol A-B (3-4), together with eight known flavonoids (5-12), were isolated and identified from the leaves of G. uralensis. Their structures were elucidated mainly by the interpretation of high-resolution electrospray mass spectrometry (HR-ESI-MS) and nuclear magnetic resonance (NMR) spectroscopic data. Compared with quercetin, which showed a 50% inhibitory concentration (IC50) value of 4.08 g/mL, compounds 1-9 showed significant anti-inflammatory activities by inhibiting lipopolysaccharide (LPS)-induced nitric oxide (NO) production with IC50 values of 2.60, 2.15, 3.21, 3.25, 2.00, 3.45, 2.53, 3.13 and 3.17 g/mL, respectively. The discovery of these active compounds is important for the prevention and treatment of inflammation.

Abstract:A dibenzylbutane-type lignan (16), along with eight furofuran-type (1-8), five furan-type (9-13), two dibenzylbutane-type (14 and 15), two bibenztetrahydronaphthalene-type lignans (17 and 18), two neolignans (19 and 20), and six phenolic derivatives (21-26) were isolated from an MeOH extract of the stem bark of Albizia julibrissin Durazz. The chemical structures of the obtained compounds were elucidated by nuclear magnetic resonance (NMR) and mass spectrometry (MS) analyses. Of the evaluated compounds, 14 were isolated from A. julibrissin and the Fabaceae family for the first time. Anti-inflammatory effects of the isolated analogs were investigated in terms of the inhibition of the nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated murine RAW264.7 macrophage cells. Ten compounds (10-12, 14, and 17-22) displayed significant dose-dependent inhibitory effects against the NO production, with IC50 values ranging from 5.4 to 19.2 µM. Moreover, eight compounds (1-4, 9, 13, 15, and 16) exhibited moderate inhibitory activities, with IC50 values ranging from 21.0 to 62.5 µM.

Abstract:A new lignan, tirpitzin A (17) together with 20 known compounds (1-16, and 18-21) were isolated from the ethyl acetate soluble fraction of ethanol extract of the aerial parts of Tirpitzia ovoidea. The structure of new compound was elucidated by means of spectroscopic analysis. Of the known compounds, 7-21 were isolated from Linaceae family for the first time. The pharmacological activity of the crude extracts was tested using a mouse inflammation model induced by dimethyl benzene. The results demonstrated that the ethyl acetate soluble fraction had anti-inflammatory activity. Moreover, the cytotoxic and anti-inflammatory activities of some compounds were studied. The new compound 17 showed moderate cytotoxic effect against BxPC-3 cell line (IC50 = 19.51μmol·L-1) and Compound 10 showed significant cytotoxicity against HepG2, HL-60, U87 and BxPC-3 cell lines with IC50 values in the range 4.2-8.3μmol·L-1. Additionally, Compounds 2, 10, 11, and 13 exhibited potent inhibitory effects on LPS-induced nitric oxide production in RAW 264.7 macrophages at the concentration of 50μmol·L-1.
Phenolics and terpenoids with good anti-inflammatory activity from the fruits of Amomum villosum and the anti-inflammatory mechanism of active diterpene

Abstract:The fruits of Amomum villosum are often considered a medicinal and food homologous material and have been found to have therapeutic effects in chronic enteritis, gastroenteritis, and duodenal ulcer. The aim of this study is to discover the anti-inflammatory active ingredients from dried ripe fruits of A. villosum and to elucidate the molecular mechanisms. We verified that the inhibitory activity of the ethyl acetate extract was superior to Dexamethasone (Dex), so we ultimately chose to study the ethyl acetate extract from the fruits of A. villosum. A total of 33 compounds were isolated from its ethyl acetate extract, including nine known diterpenoids (compounds 1-9), twelve known sesquiterpenoids (compounds 10-21), ten known phenolics (compounds 22, 23, 25-29, 31-33) and two new phenolics (24 and 30). On the basis of chemical evidences and spectral data analysis (UV, ECD, Optical rotation data, 1D and 2D-NMR, HR-ESI-MS, NMR chemical shift calculations), the structures of new compounds were elucidated. Among these compounds, isocoronarin D (5) was found to have good anti-inflammatory activity. Further research has found that isocoronarin D can down-regulate the protein levels of COX2 and NOS2, activate Nrf2/Keap1 and suppress NF-κB signaling pathway in LPS-induced RAW264.7 cells. In addition, isocoronarin D inhibited inflammasome assembly during inflammasome activation by hampering the binding of NLRP3 and ASC. Further evidence revealed that isocoronarin D suppressed the assembly of the NLRP3 inflammasome via blocking the formation of ASC specks. From these results, isocoronarin D may be the important bioactive compound of A. villosum and exhibits anti-inflammatory effects by regulating the NF-κB/Nrf2/NLRP3 axis in macrophages.
