Abstract:β - Amino acids are structural motifs widely found in therapeutic natural products, novel biomimetic polymers and peptidomimetics. As a convergent method, the synthesis of stereoenriched β-amino amides through the asymmetric Mannich reaction requires specialized amide substrates or a metal catalyst for enolate formation. By a redesign of the Ugi reaction, a conceptually different solution to prepare chiral β-amino amides was established using ambiphilic ynamides as two-carbon synthons. The modulation of ynamides or oxygen nucleophiles concisely furnished three classes of β-amino amides with generally good efficiency as well as excellent chemo- and stereo-control. The utility is verified in the preparation of over 100 desired products that bear one or two contiguous carbon stereocentres, including those that directly incorporate drug molecules. This advance also provides a synthetic shortcut to other valuable structures. The amino amides could be elaborated into β-amino acids, anti -vicinal diamines, γ-amino alcohols and β-lactams or undergo transamidation with amino acids and amine-containing pharmaceuticals.

Abstract:Stereoselective protonation is a challenge in asymmetric catalysis. The small size and high rate of transfer of protons mean that face-selective delivery to planar intermediates is hard to control, but it can unlock previously obscure asymmetric transformations. Particularly, when coupled with a preceding decarboxylation, enantioselective protonation can convert the abundant acid feedstocks into structurally diverse chiral molecules. Here an anchoring group strategy is demonstrated as a potential alternative and supplement to the conventional structural modification of catalysts by creating additional catalyst–substrate interactions. We show that a tailored benzamide group in aminomalonic acids can help build a coordinated network of non-covalent interactions, including hydrogen bonds, π – π interactions and dispersion forces, with a chiral acid catalyst. This allows enantioselective decarboxylative protonation to give α-amino acids. The malonate-based synthesis introduces side chains via a facile substitution of aminomalonic esters and thus can access structurally and functionally diverse amino acids.

Abstract:A novel copper-catalyzed cross-dehydrogenative-coupling (CDC) process of arylglycine derivatives with N-heteroarenes for the straightforward synthesis of α-aryl-α-heteroaryl α-amino acid scaffolds has been successfully developed. This protocol exhibits a broad substrate scope with good functional group compatibility by utilizing air as the sole oxidant. Abstract:β-Amino acids are frequently found as important components in numerous biologically active molecules, drugs and natural products. In particular, they are broadly utilized in the construction of bioactive peptides and peptidomimetics, thanks to their increased metabolic stability. Despite the number of methodologies established for the preparation of β-amino acid derivatives, the majority of these methods require metal-mediated multistep manipulations of prefunctionalized substrates. Here we disclose a metal-free, energy-transfer enabled highly regioselective intermolecular aminocarboxylation reaction for the single-step installation of both amine and ester functionalities into alkenes or (hetero)arenes. A bifunctional oxime oxalate ester was developed to simultaneously generate C-centred ester and N-centred iminyl radicals. This mild method features a remarkably broad substrate scope (up to 140 examples) and excellent tolerance of sensitive functional groups, and substrates that range from the simplest ethylene to complex (hetero)arenes can participate in the reaction, thus offering a general and practical access to β-amino acid derivatives.

Abstract:Unnatural chiral α-tertiary amino acids containing two different carbon-based substituents at the α-carbon centre are widespread in biologically active molecules. This sterically rigid scaffold is becoming a growing research interest in drug discovery. However, a robust protocol for chiral α-tertiary amino acid synthesis remains scarce due to the challenge of stereoselectively constructing sterically encumbered tetrasubstituted stereogenic carbon centres. Herein we report a cobalt-catalysed enantioselective aza-Barbier reaction of ketimines with various unactivated alkyl halides, including alkyl iodides, alkyl bromides and alkyl chlorides, enabling the formation of chiral α-tertiary amino esters with a high level of enantioselectivity and excellent functional group tolerance. Primary, secondary and tertiary organoelectrophiles are all tolerated in this asymmetric reductive addition protocol, which provides a complementary method for the well-exploited enantioselective nucleophilic addition with moisture- and air-sensitive organometallic reagents. Moreover, the three-component transformation of α-ketoester, amine and alkyl halide represents a formal asymmetric deoxygenative alkylamination of the carbonyl group.

Abstract:The development of methods for the assembly of secondary α-alkyl amines remains a central challenge to chemical synthesis because of their critical importance in modulating the physical properties of biologically active molecules. Despite decades of intensive research, chemists still rely on selective N-alkylation and carbonyl reductive amination to make most amine products. Here we report the further evolution of a carbonyl alkylative amination process that, for the first time, brings together primary amines, aldehydes and alkyl iodides in a visible-light-mediated multicomponent coupling reaction for the synthesis of a wide range of α-branched secondary alkylamines. In addition to exploring the tolerance and limitations in each reaction component, we also report preliminary applications to the telescoped synthesis of α-branched N-heterocycles and an N-alkylation protocol that is selective for primary over cyclic secondary amines. Our data support a mechanism involving addition of an alkyl radical to an uncharged alkyl imine which, to the best of our knowledge, has not previously been described. We believe that this method will enable practitioners of synthetic chemistry in academic and industrial settings to approach the synthesis of these important molecules in a manner that is streamlined compared to established approaches.

Abstract:The stable α-boryl aldehydes are important amphoteric synthetic intermediates that act as linchpins in organic synthesis. However, current approaches to these important building blocks typically rely on oxidative processes, which might lead to issues such as lack of product diversity and functional group compatibility. Here, we report the design and utilization of 2-(BMIDA) acrylaldehyde as a suitable coupling partner toward photoredox-catalyzed intermolecular radical addition reaction for the rapid synthesis of functionalized α-boryl aldehydes. With a commercially available photocatalyst and readily accessible alkyltrifluoroborates as radical precursors, a wide range of α-boryl aldehydes were obtained in up to 90% yield. Preliminary mechanistic studies suggested that the reaction might proceed through intermolecular radical addition and radical-polar crossover processes. The synthetic potential of the method was further demonstrated by scale-up reactions, several synthetic transformations, and derivatization of bioactive molecules. Moreover, by simply switching the radical precursors to α-amino acids, diverse 4-boryl 2,3-dihydropyrroles could be accessed via the intermolecular radical addition/cyclization sequence. Furthermore, the MIDA boronate-bearing six- or seven-membered heterocycles could also be readily obtained when the corresponding β- or γ-amino acid was used. This strategy provides a facile, robust, and diversity-oriented synthetic approach to functionalized α-boryl aldehydes or borylated heterocycles.

Abstract:Efficient and modular synthesis of structurally diverse 1,4-diketones from readily available building blocks represents an essential but challenging task in organic chemistry. Herein, we report a multi-component, regioselective bis-acylation of olefins by merging NHC organocatalysis and photoredox catalysis. With this protocol, a broad range of 1,4-diketones could be rapidly assembled using bench-stable feedstock materials. The robustness of this method was further evaluated by sensitivity screening, and good reproductivity was observed. Moreover, the diketone products could be readily converted into functionalized heterocycles, such as multi-substituted furan, pyrrole, and pyridazine. Mechanistic investigations shed light on the NHC and photoredox dual catalytic radical reaction mechanism.

Abstract:The synthesis of aryl amines from 3-alkynyl-2-pyrones and various amines is described. Mechanistically, the aryl amines are proposed to arise from the 3-alkynyl-2-pyrone substrates through their selective opening in a 1,6-fashion by secondary amines followed by decarboxylation and an unexpected rearrangement. The proposed mechanism is supported by quantum chemical transition-state calculations, which are consistent with the regiochemical outcome. The scope of this transformation spans a variety of 3-alkynyl-2-pyrones and a range of secondary amines. The influence of the secondary amine coupling partners on reaction efficiency was elucidated through data-driven modeling as well as scope exploration. These latter studies revealed that the steric bulk of the secondary amine coupling partner under the reaction conditions serves as a strong indicator of overall reaction efficiency.

Abstract:The β‐lactam scaffolds are the prevalent structural units in antibiotics and natural products. Herein, three‐component asymmetric Kinugasa/aldol and Kinugasa/Mannich cascade reactions have been developed for constructing α‐quaternary chiral β‐lactams. This strategy enables sequential assembly of alkynes, nitrones and aldehydes (or imines) and creates three contiguous stereogenic centers, controlled by copper(I) catalysts and chiral bis(oxazolidine) ligands. The developed protocol features high enantioselectivity, good reaction efficiency, and mild operational conditions representing a practical and sustainable synthetic tool for the construction of functionalized chiral β‐lactams.

Abstract:Primary γ-Amino acids are essential structural scaffolds with biologically actives in pharmaceutical compounds. However, their preparation from readily available substrates remains challenging. Here, we report a direct approach to achieve the energy transfer-driven carboimination of alkenes for synthesis of a diverse collection of valuable γ-amino acids. Additionally, a variety of γ-lactam derivatives are produced in only two steps from the simple substrates. Furthermore, this transformation features mild reaction conditions, operational simplicity, and excellent functional group tolerance.

Abstract:The development of modular, chemically diverse and regioselective reactions for expanding the cycloaddition reaction toolbox hinges heavily on a strict functional 1,3-dipole and dipolarophile pairing exploration. In this study, we disclosed a step-economic, nitrite-triggered cycloaddition reaction between bromide and alkyne/alkene for modular assembly of isoxazoles (or isoxazolines). This process underwent an in-situ Ac-L-methionine (S11)-catalyzed sulfur ylide generation, nitrite-triggered the generation of a sulfonium masked 1,3-dipole, as well as subsequent cascade cycloaddition with alkyne (or alkene) by the simultaneous releasing of Ac-L-methionine (S11). With intrinsic advantages of high regio-selectivity, excellent efficiency, wide functionality tolerance, biocompatible condition and operational simplicity, this reaction is robust for the direct diversification of pharmaceutics and bio-relevant motifs. Moreover, this intermolecular cycloaddition reaction could be well expanded to the intramolecular cycloaddition for fused-ring isoxazoles and proximity sulfide induced cycloaddition for the specific methionine modification.

Abstract:A modular, atroposelective total synthesis of micitide 982 (1) is reported. The feature of this report is the gram-scale C-H biarylation of N-phthaloyl-L-alanine followed by atroposelective Larock macrocyclization. This modular approach allowed the construction of a highly strained atrop-Tyr-Trp cross-linkage with unprecedented atropisomerism, as well as the first total synthesis of micitide 982 (1).

Abstract:A new general de novo synthesis of pharmaceutically important alpha-aryl beta-perfluoroalkyl ketones has been disclosed. Compared with trifluoromethylation-initiated radical 1,2-aryl migration of alpha,alpha-diaryl allylic alcohols, this protocol employs a new strategy of biomimetic carbene catalysis to assemble alkene, aldehyde and perfluoroalkyl reagents, providing access to products with excellent flexibility of the aryl unit and perfluoroalkyl group. This method also demonstrates excellent functional group compatibility, including some Grignard reagent sensitive groups.

Abstract:Amino acid surfactants have gained significant importance in overcoming the limitations of conventional surfactants, notably, their low biocompatibility and biodegradability. However, the current amino acid surfactants lack multifunctional properties due to the nonreactivity of their aliphatic chains, necessitating the development of a new type of amino acid surfactant. A novel melanin-like amino acid surfactant and a biomimetic synthesis route were devised by mimicking the biosynthesis of melanin. Renewable natural polyphenol compounds with catechol moieties were utilized as building blocks for the hydrophobic group. In a proof-of-concept experiment, ethyl protocatechuate was oxidized to o-quinone and subsequently covalently linked to the amino group of lysine via Michael addition. The chemical structure was verified using liquid chromatography-tandem mass spectroscopy. The melanin-like amino acid surfactant exhibited excellent surface-active properties, with a critical micelle concentration of 1.59 mN m-1. Furthermore, it demonstrated remarkable emulsifying, foaming, solubilizing, dispersing, and wetting capabilities. Notably, it also possessed multifunctionality, including antibacterial activity, antioxidant activity, robustness, and mildness. These outstanding properties indicate significant potential for various applications. This strategy offers innovative insights and a versatile, modular toolbox for synthesizing multifunctional amino acid surfactants that mimic melanin. The approach allows for the easy interchange of o-quinone building blocks, which is akin to snap jewelry.

Abstract:Unnatural amino acids (UAAs) have shown great potential to enhance the pharmacological properties of peptides and represent a key motif in a wide range of biologically relevant natural products. Herein, we disclose a protocol that facilitates the asymmetric synthesis of UAAs from widely abundant amines through photoredox-mediated α-amino C–H bond activation. The platform utilizes either the strongly oxidizing organic acridinium or decatungstate as a photocatalyst to generate α-amino radicals from a variety of amines, which are then effectively coupled with a chiral glyoxylate-derived N-sulfinyl imine. The disclosed platform provides a general entry to various decorated unnatural α-amino acid derivatives, accommodating a diverse array of functional groups.

Abstract:Polyfunctionalized arenes are privileged structural motifs in both academic and industrial chemistry. Conventional methods for accessing this class of chemicals usually involve stepwise modification of phenyl rings, often necessitating expensive noble metal catalysts and suffering from low reactivity and selectivity when introducing multiple functionalities. We herein report dehydrogenative synthesis of N -functionalized 2-aminophenols from cyclohexanones and amines. The developed reaction system enables incorporating amino and hydroxyl groups into aromatic rings in a one-shot fashion, which simplifies polyfunctionalized 2-aminophenol synthesis by circumventing issues associated with traditional arene modifications. The wide substrate scope and excellent functional group tolerance are exemplified by late-stage modification of complex natural products and pharmaceuticals that are unattainable by existing methods. This dehydrogenative protocol benefits from using 2,2,6,6-tetramethylpiperidine 1-oxyl (TEMPO) as oxidant that offers interesting chemo- and regio-selective oxidation processes. More notably, the essential role of in situ generated water is disclosed, which protects aliphatic amine moieties from overoxidation via hydrogen bond–enabled interaction.
