CRISPR/Cas9 System Mediated SIRT7 Gene Knockout Promotes Melanogenesis by MITF via MAPKS and BMP Activation in Melanoma Cells

Abstract:Mitogen-activated protein kinase (MAPK) and bone morphogenetic proteins (BMP) have been known as the crucial targets for the modulation of microphthalmia-associated transcription factor related to melanin production. However, the detailed mechanisms by which SIRT7 mediates melanogenesis remain obscure. The aim of this study was to elucidate the role of SIRT7 through MAPK and BMP pathways in melanogenesis. The SIRT7 gene in the SIRT7 gene knockout B16F1 melanoma (KO) cells were established by CRISPR/Cas9 system. The base indel sequencing results of SIRT7 KO cells were analyzed using the Ice analysis online program. Besides, the virtual translation and protein modeling in normal and KO cells were predicted using Expasy and Swiss protein modeling, respectively. The melanin amount was quantified by the measurement of fluorescence intensity caused by the oxidative reaction of hydrogen peroxide and melanin. Western blot and immunofluorescence staining assay were used to analyze the expression level of proteins related to MAPK and BMP signaling pathways. The SIRT7 gene in KO cells was analyzed to have a single base "T" insertion near 3 bases upstream to the protospacer adjacent motif site. In virtual transcription and protein modeling, SIRT7 represented the premature termination and the removal of the active site of Zn 2+- binding site owing to the replacement of Cys with Val. The melanin amount in KO cells was increased by 294% compared with that in normal cells. The KO cells displayed the up-regulation of p38 and BMP6 but the down-regulation of p-ERK and BMP4. Above results suggest that SIRT7 could be used as a potential target for the treatment of melanoma diseases.
