Abstract:The wide use in health care of intravenous bags and tubing sets for patient-side fluid delivery set the stage for transferring the disposability of filtration bag systems, single-use systems, to the biopharma markets. Single-use technologies used for separation, clarification, purification and sterilizing processes are key operations to achieving production efficiencies and cost reductions. This chapter discusses membrane filtration, depth filtration and viral clearance operations that utilize single-use technology devices. Sterilizing-grade and bacterial retentive filters are polymeric membranes typically having rigid pore structure through the cross-sectional matrix. Cell harvesting technologies such as conventional centrifugation or stacked disc depth filtration are difficult to implement as single-use systems. Single-use bags used for storage of liquids were the first application in the move toward single-use technology in the biopharma market. Single-use bioreactors and fermenters are replacing the traditional stainless steel hardware in the biopharma processing operation with a growing preference, and in the cell expansion seed train area of production.
