Abstract:Strychnine is an indole alkaloid consisted of highly conjugated seven rings, which is known as a target molecule for many organic chemists from its isolation. In this review, four recent synthetic studies of strychnine were described, which were reported after the review by Overmann et al. in 2012.

Abstract:Known for the complexity of its structure, strychnine remains to be one of the challenging targets of total synthesis. Generations of chemists have contributed to the total synthesis of strychnine since Woodward's groundbreaking synthesis. Today, new strategies and tactics are still emerging for the optimization of the route of strychnine synthesis. In this review, I discuss the documents about strychnine synthesis in the past five years with the aim of demonstrating the trend of the strategy and tactics of total synthesis.

Abstract:The first asymmetric total syntheses of the fused-pyrrole alkaloids strychnuxinal and strychnuxin have been achieved in 6 and 7 steps, respectively, starting from commercially available (±)-4-chlorostyrene oxide. Key steps in the synthetic route include a regioselective epoxide opening, a reductive etherification sequence to form the central 1,4-oxazine ring, and a late-stage phenol synthesis using a mild palladium-catalyzed coupling reaction. Notably, the optimized synthetic sequence presented avoids the use of traditional protecting groups. Total synthesis of these two structurally related natural products confirmed both their constitution (via NMR and X-ray crystallography) and their absolute configuration (via optical rotation). Abstract:The Pictet-Spenglerase strictosidine synthase (STR) has been characterized as the central enzyme in the biosynthesis of around 2000 monoterpenoid indole alkaloids in plants. In the light of a high therapeutic value and huge scaffold diversity these alkaloids represent, STR as an enzyme has attracted great attentions in recent years, intending to be utilized in the formation of new interesting alkaloids with unusual substitution pattern or even with novel scaffolds. For outlining the application potential that STR possesses, together with insight into the reaction mechanism catalyzed by STR, strategies and methods for exploring the applicability of STR have been updated in this article by taking R. serpentina STR (RS-STR) and C. roseus. STR (CR-STR) as representative models, followed by introducing the latest released complex structures of RS-STR with new substrates. Examples provided here, including substrate scaffold tailoring, X-ray crystal complex structure comparison, protein engineering and biosynthetic pathway reprogramming, pave the way to finally construct novel alkaloids libraries by chemo-enzymatic approaches.

Abstract:Strychnine is synthesized via a longest linear sequence of six steps from commercially available starting materials. Key steps include a base-mediated intramolecular Diels–Alder reaction of a tryptamine-derived Zincke aldehyde, a Ru-catalyzed trans-hydrosilylation of 1,4-butynediol, and a tandem Brook rearrangement/intramolecular conjugate addition reaction that affords the Wieland–Gumlich aldehyde.

Abstract:A bioinspired photo-Fries/imine capture cascade reaction was developed in continuous-flow mode, which facilitated the rapid construction of a series of diversely functionalized 2,7-heterocycle-fused tetrahydrocarbazoles, the ubiquitous core structures embedded in strychnos and akuammiline-type monoterpene indole alkaloids. The synthetic utility of this novel method has been preliminarily explored by the first total synthesis of (+)-alsmaphorazine C and formal synthesis of (+)-strictamine in a concise and efficient manner.

Abstract:It was developed a concise synthetic route resulting in the first syntheses of bis-Strychnos alkaloids (-)-sungucine, (-)-isosungucine, and (-)-strychnogucine B from commercially available (-)-strychnine. Employing a highly convergent synthetic strategy, it was demonstrated that both Strychnos monomers could be efficiently prepared from commercially available (-)-strychnine. The venerable Mannich reaction was enlisted to join the two Strychnos monomers in a biomimetic fashion. Subsequent epimerization and olefin isomerization yielded (-)-strychnogucine B. Functional group manipulation transformed (-)-strychnogucine B into (-)-sungucine and (-)-isosungucine. Computational chemistry was employed to rationalize the regiochemical course of key steps en route to the bis-Strychnos targets.

Abstract:A diastereoselective approach for the total synthesis of an unusual atropisomer of the Schisandra triterpenoid (+/-)-schiglautoneA is described. The efficient synthetic strategy features three key transformations: 1) two sequential titanium(III)-catalyzed radical cyclization/homologation reactions to construct the trans-fused [6,7] bicycle as well as install the quaternary carbons at C10 and C14 with the desired stereo-chemistry; 2) a Claisen rearrangement followed by a ring-closing metathesis to forge the strained nine-membered ring; and 3) a substrate-controlled Michael addition to enable the introduction of the C17 side-chain with good diastereoselectivity.

Abstract:In the work reported herein, the concise and enantioselective total synthesis of the Schizozygine alkaloid (-)-strempeliopine was developed. This synthetic strategy featured the palladium-catalyzed decarboxylative asymmetric allylic alkylation of N-benzoyl lactam to set up the absolute configuration at the C20 position, a highly diastereoselective one-pot Bischler-Napieralski/lactamization and iminium reduction sequence for the construction of the pentacyclic core structure, and the late-stage dearomative addition of indole, leading to the otherwise difficult-to-achieve hexacyclic indoline framework with complete control of four neighbouring stereocenters.

Abstract:The Strychnos alkaloids have long been regarded as landmark targets for chemical synthesis due to their captivating architectures and notorious biological properties. The family has fascinated synthetic and medicinal chemists for almost 100 years, since the decades-long debate over the structure of the 'flagship' member strychnine, and its first total synthesis by Woodward. Featuring a dense array of carbo- and heterocyclic rings, and numerous stereochemical challenges, these molecules have inspired many synthetic strategies. In spite of this rich history, the design of approaches that can access multiple family members, in an asymmetric, concise, and atom-economical fashion, remains a significant challenge. Here we show that thiophene S,S-dioxides (TDOs) offer a modular and highly concise entry to the Strychnos natural products. We show how the rapid assembly of a tryptamine-tethered TDO enables the synthesis of family member akuammicine in just three steps from tryptamine, with minimal waste generation. We further demonstrate exceptional levels of stereocontrol in unprecedented asymmetric cycloadditions of chiral thiophene S,S-dioxides; these afford tricyclic indolines that are of interest for medicinal chemistry applications, and also enable highly concise, stereoselective and scalable syntheses of the Strychnos alkaloids by either intra- or intermolecular asymmetric cycloadditions. Our results highlight the potential of TDOs as important building blocks in asymmetric cycloaddition chemistry for the streamlined synthesis of polycyclic organic frameworks; in the case of the Strychnos alkaloids, this means that both natural products and analogues that have previously been inaccessible can now be prepared in a handful of transformations.

Abstract:Strychnine is a natural product that, through isolation, structural elucidation and synthetic efforts, shaped the field of organic chemistry. Currently, strychnine is used as a pesticide to control rodents 1 because of its potent neurotoxicity 2,3 . The polycyclic architecture of strychnine has inspired chemists to develop new synthetic transformations and strategies to access this molecular scaffold 4 , yet it is still unknown how plants create this complex structure. Here we report the biosynthetic pathway of strychnine, along with the related molecules brucine and diaboline. Moreover, we successfully recapitulate strychnine, brucine and diaboline biosynthesis in Nicotiana benthamiana from an upstream intermediate, thus demonstrating that this complex, pharmacologically active class of compounds can now be harnessed through metabolic engineering approaches.
