Antioxidant activity of Hibiscetin and Hibiscitrin: insight from DFT, NCI, and QTAIM

Abstract:The present work focuses on the structural and antioxidant ability of the naturally occurring flavonol Hibiscetin and its glycoside Hibiscitrin investigated in gas, water, and benzene media using M06-2X functional level of density functional theory with 6–311 + G ( d , p ) basis set. Relaxed potential energy scans of the hydroxyl groups and the quantum theory of atoms in molecules are used to study the strength of intramolecular hydrogen bonding and non-covalent interactions, which are critical for the antioxidant action of polyphenols. Thermodynamic factors associated with polyphenols' three primary anti-radical mechanisms have been investigated. Both molecules were found to exhibit the same trend in their reaction enthalpies. The bond dissociation enthalpies of Hibiscetin are lower than Hibiscitrin. However, the difference is not well pronounced. Their comparable reactivity is confirmed by the examination of Frontier molecular orbitals (FMO) and global descriptive characteristics. According to the study, radical attacks prefer to occur at the 8-OH position. The FMO and electrostatic potential map confirm the location of the active site.
