Abstract:Heat is a cardinal feature of inflammation, yet its impacts on immune cells remain uncertain. We show that moderate-grade fever temperatures (39°C) increased murine CD4 T cell metabolism, proliferation, and inflammatory effector activity while decreasing regulatory T cell suppressive capacity. However, heat-exposed T helper 1 (T H 1) cells selectively developed mitochondrial stress and DNA damage that activated Trp53 and stimulator of interferon genes pathways. Although many T H 1 cells subjected to such temperatures died, surviving T H 1 cells exhibited increased mitochondrial mass and enhanced activity. Electron transport chain complex 1 (ETC1) was rapidly impaired under fever-range temperatures, a phenomenon that was specifically detrimental to T H 1 cells. T H 1 cells with elevated DNA damage and ETC1 signatures were also detected in human chronic inflammation. Thus, fever-relevant temperatures disrupt ETC1 to selectively drive apoptosis or adaptation of T H 1 cells to maintain genomic integrity and enhance effector functions.

Hyperthermia Differentially Regulates TLR4 and TLR2-mediated Innate Immune Response

Abstract:Fever influences multiple parameters of the immune response. However, the mechanisms by which fever manipulates immune response remain undefined. Here we present the evidences that fever range hyperthermia differentially regulates immune response to lipopolysaccharide (LPS) and lipoteichoic acids (LTA) through modulating Toll-like receptor (TLR) signaling. Pretreatment with 39.5 degrees C temperature enhanced LPS, but not LTA, induced NF-kappaB activation and TNF-alpha, IL-6 production in human macrophages. Consistently, expression of TLR4, but not TLR2, was up-regulated by 39.5 degrees C treatment. The increase in LPS-induced cytokine production was inhibited by TLR4-blocking antibody, indicating the enhancement of LPS-induced cytokine production by 39.5 degrees C pretreatment was TLR4-dependent. Pretreatment of mice with 39.5 degrees C temperature also enhanced LPS, but not LTA, induced TNF-alpha and IL-6 production in vivo. These results support the concept that fever range hyperthermia might activate innate immune response by promoting TLR4 expression and signaling, providing a possible mechanistic explanation for the function of fever in regulating innate immune responses.

Abstract:Abstract Fever is a common sign of clinical disease but its biological purpose is unclear. Several cytokines mediate fever as well as the regulation of immune responses, and it was previously shown that murine thymocyte proliferative responses to IL-1 and IL-2 are exquisitely temperature-sensitive. The present studies were performed to determine whether such a high degree of temperature sensitivity was also a property of the primary in vitro immune responses of mature, murine splenic T cells. Using a physiological range of temperatures spanning those of normal skin (29 degrees C) to those of febrile core tissues (39 degrees C), primary murine spleen cell proliferative responses to either Con A, solid-phase anti-CD3 antibody, specific Ag, or allogenic cells were all found to display a high degree of temperature sensitivity. A similar pattern of strong temperature sensitivity was found for the capacity of primary MLR cultures to generate allospecific effector CTL. All of these forms of T-cell temperature sensitivity are expressed both in terms of the absolute degree of the response and in terms of its kinetics, the warmer temperatures up to 35 degrees C to 37 degrees C giving larger responses sooner than the lower physiological temperatures of 29 degrees C to 33 degrees C, at which these responses are markedly inhibited relative to those at 37 degrees C. In contrast, the effector cytolytic activity of a previously formed population of CTL is remarkably insensitive to the same temperature changes that strongly regulate the generation of those effector cells. The temperatures that usually define fever are those appropriate to the core tissues of a febrile animal, yet temperature changes in this range (37 degrees C-39 degrees C) had little stimulatory effect upon the T-cell responses studied, and were, in some cases, actually inhibitory. Temperature changes appropriate to the peripheral tissue temperatures in an animal becoming febrile (29 degrees C-37 degrees C) appear to be the ones that strongly regulate mature primary T-cell responses. These results suggest that one purpose of either fever or local tissue inflammation may be to temporarily ablate the cooler portions of normal thermal gradients, thereby selectively amplifying the emergence of T-cell immunity in peripheral tissues.

Abstract:We have examined the possibility that hyperthermia, such as that occurring during fever, may benefit the immune response. The effect of temperature on the in vitro immune response of unprimed murine spleen cells against the antigen sheep erythrocytes was tested. Hyperthermia potently augmented the plaque-forming cell response. Temperature- sensitive events occurred early in the culture period. Subsets of lymphocytes were independently assessed for effects of temperature on their activation and function. We showed that the beneficial effect of elevated temperature on the plaque-forming cell response probably occurs during the priming stage of T helper cells, and neither improves the delivery of help or the activation of B cells, nor impairs suppressor T cell generation or function. We propose that this powerful immunopotentiating effect of hyperthermia may account for the selective value of the fever response. This suggests taht the monokine interleukin 1, which is the endogenous mediator of fever, may promote immune responses both through a direct action on lymphocytes, and indirectly by an action on the central nervous system resulting in fever.

Abstract:Key PointsThe hallmark fever response during infection and disease has been maintained throughout hundreds of millions of years in endothermic (warm-blooded) and ectothermic (cold-blooded) species.Febrile temperatures boost the probability of an effective immune response by stimulating both the innate and the adaptive arms of the immune system.The pyrogenic cytokine interleukin-6 (IL-6) contributes to two phases of the febrile response: it elevates the core body temperature via thermoregulatory autonomic mechanisms, and it also serves as a thermally sensitive effector molecule that amplifies lymphocyte trafficking into lymphoid organs.There is emerging evidence that adrenergic signalling pathways associated with thermogenesis can greatly influence immune cell function.Thus, the thermal element of fever serves as a systemic alert system that broadly promotes immune surveillance in the setting of infection and disease.

Abstract:Fever improves survival and shortens disease duration in microbial infections. However, the mechanisms of these beneficial responses still remain elusive. Toll-like receptors (TLRs) play important roles in sensing microbes invading and therefore we hypothesized that fever range temperature may enhance responsiveness of dendritic cells (DCs) to lipopolysaccharide (LPS) by promoting TLR4 expression and signaling. In this study, we found that pretreatment of DCs with 39.5 degrees C temperature can up-regulate TLR4 expression in DCs and enhances LPS-induced DC production of interleukins (IL) IL-6, IL-10 and IL-12 but not tumor necrosis factor alpha (TNF-alpha). Blockade of the autocrine action of IL-10 could increase LPS-induced TNF-alpha and IL-12 production in DCs. Further experiments confirmed that TLR4 ligation activates extracellular signal-regulated kinase (ERK), p38, and nuclear factor-kappaB pathways more potently in DCs pretreated with 39.5 degrees C. We conclude that fever range temperature can promote TLR4 expression and signaling in DCs, leading to enhancement of immune responses to inflammatory stimuli. These results might reveal a possible mechanistic explanation for the significance of fever in activating innate immune responses.

Abstract:Summary Fever is a hallmark symptom of disease across the animal kingdom. Yet, despite the evidence linking temperature fluctuation and immune response, much remains to be discovered about the molecular mechanisms governing these interactions. In patients with rheumatoid arthritis, for instance, it is clinically accepted that joint temperature can predict disease progression. But it was only recently demonstrated that the mitochondria of stimulated T cells can rise to an extreme 50°C, potentially indicating a cellular source of these localized ‘fevers’. A challenge to dissecting these mechanisms is a bidirectional interplay between temperature and immunity. Heat shock response is found in virtually all organisms, activating protective pathways when cells are exposed to elevated temperatures. However, the temperature threshold that activates these pathways can vary within the same organism, with human immune cells, in particular, demonstrating differential sensitivity to heat. Such inter-cellular variation may be clinically relevant given the small but significant temperature differences seen between tissues, ages, and sexes. Greater understanding of how such small temperature perturbations mediate immune responses may provide new explanations for persistent questions in disease such as sex disparity in disease prevalence. Notably, the prevalence and severity of many maladies are rising with climate change, suggesting temperature fluctuations can interact with disease on multiple levels. As global temperatures are rising, and our body temperatures are falling, questions regarding temperature–immune interactions are increasingly critical. Here, we review this aspect of environmental interplay to better understand temperature’s role in immune variation and subsequent risk of disease.
Inositol-Requiring Enzyme 1 Pathway and Autophagy Drive Sequential Response of Endothelial Cells to Febrile Range Hyperthermia

Abstract:Context: Fever is an evolutionarily conserved and adaptive response during infections. However, prolonged fever has numerous systemic metabolic and functional side effects. In the heart, prolonged fever associated with infection is linked to fatal adverse effects, particularly involving impaired coronary circulation. Yet, the direct relationship between elevated temperature and coronary microcirculation dysfunction, remains to be fully demonstrated. In this study, we aimed to explore the specific responses of human coronary artery endothelial cells (HCAECs) to fever-range hyperthermia. Method: HCAECs were cultured at either 37°C or 40°C for up to 24 hours. Transcriptomic and proteomic profiles were obtained through microarray and mass spectrometry after 6, 12, and 24 hours of exposure. Key signaling pathways, upstream regulators, and candidate mechanisms were identified and validated at the mRNA and protein levels using mechanistic approaches. Results: Prolonged hyperthermia compromised HCAEC function, evidenced by cell detachment, loss of junctions, and increased permeability. HCAECs rapidly activated the unfolded protein response (UPR), including IRE1α activation and XBP1 splicing. Additionally, autophagic flux was significantly elevated, participating in the degradation of junction proteins. Pharmacological inhibition of IRE1α reduced the autophagic burden, protected against cell detachment, and preserved junction integrity over time. Conclusion: Our findings reveal that in response to fever-range hyperthermia, the IRE1α-autophagy axis regulates the survival and function of coronary endothelial cells. This mechanism could play a key role in modulating endothelial responses during infection and contribute to the pathological outcomes of fever. Furthermore, it may be relevant in local inflammatory conditions with elevated temperatures.

Abstract:Fever, an elevated thermoregulatory “set-point,” occurs in vertebrates from fishes through mammals in response to infection with appropriate pathogens. The long phylogenetic history of fever supports the hypothesis that fever has an adaptive or beneficial role (i.e., fever is a component of the host's immunological defenses). Besides providing insight into determining the role of fever in disease, ectothermic vertebrates have also served as excellent animal models to specifically answer many questions relating to fever and disease. For example, survival studies using goldfishes, lizards, and newborn mammals which are also virtually ectothermic) have shown that an elevated body temperature increases the survival rate of the infected organism. Furthermore, by using ectotherms it has been possible to demonstrate that the reduction in serum iron which accompanies most infections is not directly attributable to an elevation in body temperature. Nevertheless, a rise in body temperature, coupled with a fall in serum iron, appears to constitute a coordinated host defense mechanism in response to at least some bacterial infections in reptiles and mammals, and perhaps other groups of vertebrates.

Abstract:PML/RARα fusion protein is the oncogenic driver in acute promyelocytic leukemia (APL). While most APL cases are cured by PML/RARα targeting therapy, relapse and resistance can occur due to drug-resistant mutations. Here we report that thermal stress destabilizes PML/RARα protein, including clinically identified drug-resistant mutants. AML1/ETO and TEL/AML1 oncofusions show similar heat shock susceptibility. Mechanistically, mild hyperthermia stimulates aggregation of PML/RARα in complex with nuclear receptor corepressors leading to ubiquitin mediated degradation via SIAH2 E3 ligase. Hyperthermia and arsenic therapy destabilize P/R via distinct mechanisms and are synergistic in primary patient samples and in vivo including 3 refractory APL cases. Collectively, our results suggest that by taking advantage of a biophysical vulnerability of PML/RARα, thermal therapy may improve prognosis in drug-resistant or otherwise refractory APL. These findings serve as a paradigm for therapeutic targeting of fusion oncoprotein associated cancers by hyperthermia. Statement of Significance: Hyperthermia destabilizes oncofusion proteins including PML/RARα and acts synergistically with standard arsenic therapy in relapsed and refractory acute promyelocytic leukemia. Changes in Body Surface Temperature Play an Underappreciated Role in the Avian Immune Response

Abstract:Fever and hypothermia are well-characterized components of systemic inflammation. However, our knowledge of the mechanisms underlying such changes in body temperature is largely limited to rodent models and other mammalian species. In mammals, high dosages of an inflammatory agent (e.g., lipopolysaccharide [LPS]) typically leads to hypothermia (decrease in body temperature below normothermic levels), which is largely driven by a reduction in thermogenesis and not changes in peripheral vasomotion (i.e., changes in blood vessel tone). In birds, however, hypothermia occurs frequently, even at lower dosages, but the thermoeffector mechanisms associated with the response remain unknown. We immune challenged zebra finches (<i>Taeniopygia guttata</i>) with LPS, monitored changes in subcutaneous temperature and energy balance (i.e., body mass, food intake), and assessed surface temperatures of and heat loss across the eye region, bill, and legs. We hypothesized that if birds employ thermoregulatory mechanisms similar to those of similarly sized mammals, LPS-injected individuals would reduce subcutaneous body temperature and maintain constant surface temperatures compared with saline-injected individuals. Instead, LPS-injected individuals showed a slight elevation in body temperature, and this response coincided with a reduction in peripheral heat loss, particularly across the legs, as opposed to changes in energy balance. However, we note that our interpretations should be taken with caution owing to small sample sizes within each treatment. We suggest that peripheral vasomotion, allowing for heat retention, is an underappreciated component of the sickness-induced thermoregulatory response of small birds.

Body temperature changes during simulated bacterial infection in a songbird: fever at night and hypothermia during the day

Abstract:Although fever (a closely regulated increase in body temperature in response to infection) typically is beneficial, it is energetically costly and may induce detrimentally high body temperatures. This can increase the susceptibility to energetic bottlenecks and risks of overheating in some organisms. Accordingly, it could be particularly interesting to study fever in small birds, which have comparatively high metabolic rates and high, variable body temperatures. We therefore investigated two aspects of fever and other sickness behaviours (circadian variation, dose dependence) in a small songbird, the zebra finch. We injected lipopolysaccharide (LPS) at the beginning of either the day or the night, and subsequently monitored body temperature, body mass change and food intake for the duration of the response. We found pronounced circadian variation in the body temperature response to LPS injection, manifested by (dose-dependent) hypothermia during the day but fever at night. This resulted in body temperature during the peak response being relatively similar during the day and night. Day-to-night differences might be explained in the context of circadian variation in body temperature: songbirds have a high daytime body temperature that is augmented by substantial heat production peaks during activity. This might require a trade-off between the benefit of fever and the risk of overheating. In contrast, at night, when body temperature is typically lower and less variable, fever can be used to mitigate infection. We suggest that the change in body temperature during infection in small songbirds is context dependent and regulated to promote survival according to individual demands at the time of infection.

Abstract:Aim Animals exhibit physiological changes designed to eliminate the perceived danger, provoking similar symptoms of fever. However, a high‐grade fever indicates poor clinical outcomes. Caspase11 (Casp11) is involved in many inflammatory diseases. Whether Casp11 leads to fever remains unclear. In this study, we investigate the role of the preoptic area of the hypothalamus (PO/AH) microglia Casp11 in fever. Methods We perform experiments using a rat model of LPS‐induced fever. We measure body temperature and explore the functions of peripheral macrophages and PO/AH microglia in fever signaling by ELISA, immunohistochemistry, immunofluorescence, flow cytometry, macrophage depletion, protein blotting, and RNA‐seq. Then, the effects of macrophages on microglia in a hyperthermic environment are observed in vitro. Finally, adeno‐associated viruses are used to knockdown or overexpress microglia Casp11 in PO/AH to determine the role of Casp11 in fever. Results We find peripheral macrophages and PO/AH microglia play important roles in the process of fever, which is proved by macrophage and microglia depletion. By RNA‐seq analysis, we find Casp11 expression in PO/AH is significantly increased during fever. Co‐culture and conditioned‐culture simulate the induction of microglia Casp11 activation by macrophages in a non‐contact manner. Microglia Casp11 knockdown decreases body temperature, pyrogenic factors, and inflammasome, and vice versa. Conclusion We report that Casp11 drives fever. Mechanistically, peripheral macrophages transmit immune signals via cytokines to microglia in PO/AH, which activate the Casp11 non‐canonical inflammasome. Our findings identify a novel player, the microglia Casp11, in the control of fever, providing an explanation for the transmission and amplification of fever immune signaling.

Abstract:Abstract In vitro responses of human peripheral blood lymphocytes (PBL) were found to be markedly enhanced by culture at 40°C rather than at the conventional temperature of 37°C. We studied proliferative responses of lymphocytes by activation by phytohemagglutinin (PHA) concanavalin A (Con A), pokeweed mitogen (PWM), and allogeneic lymphocytes in mixed lymphocyte culture (MLC) and found enhancement of DNA synthesis at the higher temperature. Cytotoxic T cell responses to allogeneic cells were also enhanced when MLC was done at 40°C. These enhanced immune responses appear to be due in part to increased numbers of participating cells. If in vitro lymphocyte responses correlate with in vivo responses, then fever associated with infection or tumor may be beneficial whereas that associated with autoimmune disorders may have a detrimental effect.

High body temperature increases gut microbiota-dependent host resistance to influenza A virus and SARS-CoV-2 infection

Abstract:Fever is a common symptom of influenza and coronavirus disease 2019 (COVID-19), yet its physiological role in host resistance to viral infection remains less clear. Here, we demonstrate that exposure of mice to the high ambient temperature of 36 °C increases host resistance to viral pathogens including influenza virus and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). High heat-exposed mice increase basal body temperature over 38 °C to enable more bile acids production in a gut microbiota-dependent manner. The gut microbiota-derived deoxycholic acid (DCA) and its plasma membrane-bound receptor Takeda G-protein-coupled receptor 5 (TGR5) signaling increase host resistance to influenza virus infection by suppressing virus replication and neutrophil-dependent tissue damage. Furthermore, the DCA and its nuclear farnesoid X receptor (FXR) agonist protect Syrian hamsters from lethal SARS-CoV-2 infection. Moreover, we demonstrate that certain bile acids are reduced in the plasma of COVID-19 patients who develop moderate I/II disease compared with the minor severity of illness group. These findings implicate a mechanism by which virus-induced high fever increases host resistance to influenza virus and SARS-CoV-2 in a gut microbiota-dependent manner.

Abstract:Fever raises body temperature (T ) from ∼37°C to beyond 38°C to combat pathogens. While generally well tolerated below 40°C, fevers can induce seizures in 2-5-year-old neurotypical children. This study investigates how neuronal activity is maintained during fever-range temperatures. Recordings of layer (L) 4-evoked spiking in L2/3 mouse somatosensory cortex show that excitatory pyramidal neurons (PNs) may remain inactive, stay active, cease activity, or initiate activity as temperature rises from 30°C (standard in electrophysiology studies) to 36°C (normal T ) and then to 39°C (fever-range). Similar proportions of neurons cease or initiate spiking. Thus, “STAY” PNs, which remain active across temperatures changes, are crucial for maintaining stable spiking activity. STAY PNs are more prevalent at younger postnatal ages. To sustain spiking during temperature increases, STAY PNs adjust their depolarization levels to match the spike threshold while remaining temperature-insensitive in input resistance. In the striatum, STAY medium-spiny neurons are likely dopamine (D)2-type receptor-expressing and share similar characteristics with STAY PNs. Intracellular blockade of the thermosensitive channel , but not , significantly decreased the population of STAY PNs and reduced spiking at 39°C. Therefore, function may be critical for maintaining cortical activity during fever.

Abstract:A spontaneously occurring temperature increase in solid tumors has been reported sporadically, but is largely overlooked in terms of cancer biology. Here we show that temperature is increased in tumors of patients with pancreatic ductal adenocarcinoma (PDAC) and explore how this could affect therapy response. By mimicking this observation in PDAC cell lines, we demonstrate that through adaptive changes in lipid metabolism, the temperature increase found in human PDAC confers protection to lipid peroxidation and contributes to gemcitabine resistance. Consistent with the recently uncovered role of p38 MAPK in ferroptotic cell death, we find that the reduction in lipid peroxidation potential following adaptation to tumoral temperature allows for p38 MAPK inhibition, conferring chemoresistance. As an increase in tumoral temperature is observed in several other tumor types, our findings warrant taking tumoral temperature into account in subsequent studies related to ferroptosis and therapy resistance. More broadly, our findings indicate that tumoral temperature affects cancer biology.
Fever-range hyperthermia promotes cGAS-STING pathway and synergizes DMXAA-induced antiviral immunity

Abstract:Background: Fever-range hyperthermia or fever-range temperature (hereafter FRT) improves survival and shortens disease duration in microbial infections. However, the mechanisms of these beneficial effects still remain elusive. We hypothesized that FRT might enhance cell responsiveness to infections by promoting cGAS-STING signaling to cause enhanced production of IFN-β. Objective: To investigate the effect fever-range hyperthermia on cGAS-STING pathway. Methods: RAW 264.7 and cGAS-/- RAW 264.7 cells, stimulated with 5μg/ml herring testis DNA (htDNA), were heated to 39.5°C and analyzed for the expression of cGAS, STING, IFN-β, and the synthesis of cGAMP and IRF3 phosphorylation. In vivo, wild type C57BL/6J mice were subjected to whole body hyperthermia (WBH) at 39.5°C. The mice were then challenged with influenza virus and analyzed for antiviral response in term of IFN-β expression, body weight and survival. Results: We found that 39.5°C FRT upregulated the expression of cGAS and STING, and induced the synthesis of cGAMP and production of IFN-β in htDNA-transfected RAW 264.7 cells more potently as compared to 37°C. Moreover, FRT+DMXAA-treated cells were better protected from vesicular stomatitis virus (VSV)-induced cytotoxicity in vitro in contrast to the nonprotected control (no FRT and DMXAA) or DMXAA treatment alone. In vivo, FRT at 39.5°C, co-administered with DMXAA, significantly induced the expression of IFN-β, showed reduced weight loss mice and exhibited 25% more survival over the course of 14 days as compared to DMXAA treated mice 37°C. Conclusion: We conclude that fever-range hyperthermia promotes cGAS-STING pathway to cause increased expression of IFN-β and mediate its antiviral effects.

Abstract:Cytoadhesion of P. falciparum‐infected red blood cells to endothelial cells leads to significant upregulation of genes associated with immune response, nucleosome assembly, NF‐kappa B signaling, and angiogenesis in these cells. Increasing the temperature to 40°C (fever) led to further upregulation of many genes, particularly those involved in cytokine production and angiogenesis. Malaria remains a significant global health problem, mainly due to Plasmodium falciparum, which is responsible for most fatal infections. Infected red blood cells (iRBCs) evade spleen clearance by adhering to endothelial cells (ECs), triggering capillary blockage, inflammation, endothelial dysfunction and altered vascular permeability, prompting an endothelial transcriptional response. The iRBCIT4var04/HBEC‐5i model, where iRBCs present IT4var04 (VAR2CSA) on their surface, was used to analyze the effects of iRBC binding on ECs at different temperature (37°C vs. 40°C). Binding of non‐infected RBCs (niRBCs) and fever alone altered the expression of hundreds of genes in ECs. Comparing the expression profile of HBEC‐5i cells cultured either in the presence of iRBCs or in the presence of niRBCs revealed significant upregulation of genes linked to immune response, nucleosome assembly, NF‐kappa B signaling, angiogenesis, and antiviral immune response/interferon‐alpha/beta signaling. Raising the temperature to 40°C, simulating fever, led to further upregulation of many genes, particularly those involved in cytokine production and angiogenesis. In summary, the presence of iRBCs stimulates ECs, activating several immunological pathways and affecting antiviral (−parasitic) mechanisms and angiogenesis. Our data uncovered the induction of the interferon‐alpha/beta signaling pathway in ECs in response to iRBCs.

Abstract:Fever is a complex physiological response enhancing immune surveillance during infection and inflammation. Fever-range whole-body hyperthermia (WBH) treatment can experimentally mimic the febrile condition in mice. Here, we describe a protocol for the treatment of mice with WBH and normothermia. We describe the isolation of T cells from mouse spleen followed by the evaluation of T-cell adhesion and transmigration. This animal model can be applied to studying the dysfunction of the immune system induced by fever. For complete details on the use and execution of this protocol, please refer to Lin et al. (2019).
