Genetics and epigenetics of the SLC6A4 gene in depression

Abstract:Depression is a complex and multifactorial psychiatric disorder manifesting itself individually. Its occurrence acts through the interaction of individual genetic background and stress events experienced by each person throughout life. The genetic factors involved in the etiology of depression are not yet fully elucidated, however, several genes operate in different pathways related to its development. Furthermore, these genetic factors are influenced by environmental factors due to epigenetic modifications. The serotonin transporter gene (SLC6A4) is one of the most studied genes associated with depression. The genetic polymorphisms present in this gene are directly related to gene transcription level. In addition, epigenetic mechanisms such as DNA methylation, histone modifications, and noncoding RNA interact and influence SLC6A4 expression. Therefore, the purpose of the chapter is to demonstrate the association of the genetic background, the stress events, and changes in the epigenetic profiles in SLC6A4, for the contribution to depression development.
