Abstract:The incidence of invasive aspergillosis (IA) , which is commonly caused by Aspergillus fumigatus, has increased recently in immunocompromised patients and has become the common cause of death in these patients. Antifungal resistance is one of the reasons for treatment failure. Since the first itraconazole-resistant A. fumigatus was reported in 1997, the reports on clinical strains of triazole-resistant A. fumigatus have increased, as well as studies on the resistant mechanisms. In this paper, the known molecular mechanisms of antifungal resistance in Aspergillus, especially in A. fumigatus, are reviewed.
Dectin-1 Mrna Expression in Lung of Different Immune Status Mice Infected with Aspergillus Fumigatus

Abstract:Objective To study dectin-1 mRNA expression in lung of different immune status mice infected with Aspergillusfumigatus, and to explore the influence of immunosuppressive agent on the expression of dectin-1 and its relationship with the progression of disease. Methods Mice were divided into four groups which were normal control, immuncompromised, immuncompromised with A.fumigatus inoculation and immuncompetent with A.fumigatus inoculation groups. We explored the kinetic mRNA expression of dectin-1 in lung of different groups by real-time quantitative PCR. Pulmonary fungal burden assessment was performed to reflect the progressing of disease during the experimental time course. Results On day 3 after inoculation, pulmonary fungal burden of the immuncompromised mice was higher than that of the immuncompetent group. On day 1 and 3, dectin-I mRNA expression in lung of the immuncompromised group was much lower than that of the normal control. On day 3, dectin-1 mRNA expression in lung of the immuncompetent mice infected with A.fumigatus was much higher than that of the immuncompromised with A.fumigatus inoculation and the normal control groups ( P < 0.05 ). Conclusion During the infection, expression of dectin-1 in lung of the immuncompetent group was strikingly increased, which may play an important role on the defence to A.fumigatus invasion. Cyclophosphamide inhibited the expression of dectin-1 in lung of mice which may be one of the mechanisms of the invasive pulmonary aspergillosis development induced by eyclophosphamide. Abstract:Objective To investigate the effects of immunosuppressive agent on mRNA expression of anti-aspergillus infection-associated receptors on alveolar macrophage.Methods Thirty Kunming mice were randomly divided into two groups:normal control group(n=6)and immunocompromise group(150 mg/kg cyclophosphamide,intraperitoneal injection,n=24).Six mice were randomly taken to collect alveolar macrophage by bronchoalveolar lavage from immunocompromise group at time of 4,8,12 and 24 hours after immunosuppression.Mice in normal control group were sacrificed to collect alveolar macrophage after receiving normal saline intraperitoneal injection.Reverse transcription-polymerase chain reaction was used for determining the mRNA expression of Toll-like receptor 2(TLR2),TLR4 and dendritic cell-associated C-type lectin 1(Dectine-1).Results TLR2 mRNA expression decreased signiffieantly four hours after cyclophosphamide intraperitoneal injection,and decreased continually to 24 hours(all P＜0.01).The decrease of TLR4 mRNA expression appeared eight hours after immunosuppression,and decreased continually to 24 hours(all P＜0.0 1).There was not significant difference on Dectine-1 mRNA expression between normal control group and immunocompromise group.Conclusions The results suggest that cyclophosphamide could down-regulate the mRNA expression of TLR2 and TLR4 on alveolar maerophage.
Clinical Analysis on Ninty-Six Cases of Secondary Aspergillosis in COPD Patients

Abstract:Objective To discuss the clinical features and control measure of respiratory infection associated with aspergillosis secondary to chronic obstructive pulmonary disease.Method 96 cases of COPD patients with aspergillus in their sputum culture,admitted into the First Affiliated Hospital of Zhejiang University from June 2007 to June 2008,were collected and their clinical manifestations,treatment and treatment outcome were analyzed.Result Among the 96 cases of COPD patients with aspergillosis,9 cases patients died after treatment,87 cases improved including 45 cases treated with non-antifungal agents,38 cases with fluconazde and 13 cases with the addition of Voriconazole.Conclusion COPD patients with aspergillosis were increasing in recent years.The risk factors of aspergillus infection includes repeatedly long-term hospitalization,advance in age,irrational antibiotics and hormones application,as well as latrogenic and invasive operation.Early diagnosis,early treatment and rational precautionary can be a key to reduce the occurrence of respiratory infection by aspergillus.
Cyclophosphamide Reduces Dectin-1 Expression in the Lungs of Naive and Aspergillus Fumigatus-Infected Mice

Abstract:Aspergillus fumigatus is an important opportunistic fungal pathogen. Patients treated with chemotherapeutic agent such as cyclophosphamide are susceptible to invasive pulmonary aspergillosis. Dectin-1 is a pattern recognition receptor critically involved in immune responses to A. fumigatus. Therefore, we tested whether cyclophosphamide treatment could cause alterations in dectin-1 expression in the lung, which could contribute to invasive pulmonary Aspergillus infections in patients. We established a murine A. fumigatus infectious model to investigate the kinetics of dectin-1 expression in lung tissues in the presence or absence of cyclophosphamide treatment. During infection, dectin-1 expression was strikingly increased in immunocompetent mice infected with A. fumigatus as compared to those in a non-infected control group. In vitro macrophages stimulated with heat-inactivated A. fumigatus conidia expressed a significantly elevated level of dectin-1. Infected mice treated with cyclophosphamide showed decreased levels of dectin-1 and a higher fungal burden in the lung than the infected mice without cyclophosphamide treatment. These results suggest that dectin-1 is involved in host defense against A. fumigatus infection and that suppression of dectin-1 expression caused by cyclophosphamide may contribute to susceptibility to infections caused by this fungus in the immunocompromised host.</.
Efficacy of the Combination of Voriconazole and Caspofungin in Experimental Pulmonary Aspergillosis by Different Aspergillus Species

Abstract:Objectives Invasive pulmonary aspergillosis (IPA) caused by Aspergillus fumigatus , Aspergillus flavus , or Aspergillus niger is associated with high mortality. We evaluated the efficacy and compared the therapeutic effect differences of voriconazole (VRC) in combination with caspofungin (CAS) in transiently neutropenic rats infected by A. fumigatus, A. flavus , or A. niger. Methods Treatment groups consisted of VRC (10 mg/kg q12 h) monotherapy, CAS (1 mg/kg/day) monotherapy, combination of VRC (10 mg/kg q12 h) + CAS (1 mg/kg/day), and no drug for 10 consecutive days. The efficacy and the difference in the treatments were evaluated through prolongation of survival, reduction in serum galactomannan levels and residual fungal burden, and histological studies. Results For all the strains, the combination of VRC and CAS led to significant prolongation in survival ( P < 0.05) and reduction in residual fungal burden ( P < 0.05) compared with CAS alone, and decrease in serum galactomannan levels ( P < 0.05) compared with either agent alone. The survival in the combined therapy groups was significantly improved compared to VRC monotherapy for the strains of A. flavus and A. niger ( P < 0.05), but no significant difference for the strains of A. fumigatus ( P > 0.05). Conclusions Combination of VRC and CAS was synergistic in IPA by A. flavus and A. nige r, but small efficacy benefits in IPA by A. fumigatus .
Change of the concentration of IFN-γand infectious Aspergillus organisms in lung in the model of invasive pulmonary aspergillosis

Abstract:Objective To establish the model of invasive pulmonary aspergillosis (IPA) and assay the influence on the host defense against Aspergillus infection when immunity was suppressed in mice.Methods Immunocompromised mice were made by treatment with cyclophosphamide administered intraperitoneally (i.p.).Suspension of conidia was applied to the nostrils of mice to make the model of IPA.Lungs were harvested and homogenized.Portions of homogenates were cultured to determine the number of CFU.IFN-γ in bronchoalveolar lavage fluid was determined by a cytokine-specific ELISA kit.Reverse transcription PCR (RT-PCR) analysis was done to determine the mRNA of IFN-γ in lung cells.Mortality of different rice was calculated.Results Compared with immunocompetent mice,the immunocompromised mice demonstrated a high mortality and had significantly higher concentration of infectious Aspergillus organisms in their lung tissues.In accordance with the increase of Aspergillus organisms,the levels of IFN-γ in lung tissues got higher.Lung sections from immunocompromised mice revealed patterns of lesions characterized by signs of bronchial wall damage,peribronchial necrosis,and the presence of numerous infiltrating inflammatory cells.Conidia and hyphae were seen in these mice.In contrast,these features were not observed in immunocompetent mice whose lungs were characterized by few inflammatory cells infiltration,and few fungal growth after inoculation.Conclusion The levels of IFN-γ in lung tissues are related to the infectious Aspergillus organisms.The immunity and T cells play a major role in host defense against Aspergillus infection.
[Differentially expressed genes in immunosuppressive and healthy mice after intranasal inoculation of Aspergillus fumigatus]

Abstract:Objective: To screen out the differential expression genes between the immune suppressive host and healthy host after intranasal inoculation of Aspergillus fumigatus (A fumigatus). Methods: Six immune suppressive mice and 6 healthy mice were inoculated with A, fumigatus intranasally. Three days after the inoculation the mice were killed and autopsied. The total RNA of the lungs was isolated. Gene chip technology was used to examine the differentially expressed genes. Results: 66 differentially expressed genes were screened out of the 4098 genes. In the experimental group the genes encoding CD22, CD53, and 24p3 and the genes related to engulfment and digestion of macrophage or monocytes were up-regulated. The genes encoding surfactant associated protein C, immunoglobulin lambda chain and the genes related to pulmonary metabolism were down-regulated. The genes encoding IL18, CD8, and CD28 were not differentially expressed. Conclusion: The up-regulation of CD22 gene may be one of the ways by which the host defense is inhibited. CD53 may take part in the regulation of engulfment of phagocytes. 24p3 protein, an acute stress protein, protects the host from excessive inflammatory response.
Vitamin D Deficiency Causes Defective Resistance to Aspergillus Fumigatus in Mice Via Aggravated and Sustained Inflammation

Abstract:BACKGROUND: Vitamin D plays an important role in pulmonary resistance and immunity, and its deficiency has been linked to various respiratory infections. Little is known about the effect of vitamin D deficiency on host pulmonary defense to Aspergillus fumigatus (A. fumigatus). METHODS: Mice raised on vitamin D sufficient or deficient diets were infected intratracheally with A. fumigatus conidia. Mortality, fungal growth, weight loss and lung histology were monitored. Alveolar macrophages (AMs) were stimulated with A. fumigatus conidia in vitro. The kinetics of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6), chemokines (CXCL1, CCL3), and pattern recognition receptors (Toll-like receptor [TLR] 2, TLR 4 and dectin-1) expression in the lungs and AMs were measured. RESULTS: Upon A. fumigatus infection, vitamin D deficient mice showed higher mortality, greater fungal load, and more weight loss than its sufficient counterparts. Vitamin D deficient mice demonstrated aggravated and prolonged histological evidence of lung inflammation as well as enhanced BAL cell counts, dominated by neutrophils after A. fumigatus inoculation. Increased basal levels of pro-inflammatory cytokines in the lungs and AMs from naïve vitamin D deficient mice were observed. Upon A. fumigatus exposure, vitamin D deficiency led to enhanced and sustained expression of TNF-α, IL-1β, IL-6, CXCL1 and CCL3 both in vivo and in vitro. Up-regulation of TLR2, TLR4 and dectin-1was observed in the lungs and AMs from vitamin D deficient mice both at baseline and after A. fumigatus exposure. CONCLUSIONS: Vitamin D deficiency causes defective pulmonary resistance to A. fumigatus in mice, possibly by the enhanced basal expression of pattern recognition receptors and pro-inflammatory cytokines, which induced excessive inflammatory response in response to A. fumigatus challenge.
Improved Clinical Outcomes in Chinese Renal Allograft Recipients Receiving Lower Dose Immunosuppressants

Abstract:Background. The application of potent immunosuppressants has decreased the incidence of acute rejection and increased short- and long-term graft survival; however, these drugs cause a variety of complications. In China, many transplant centers have adopted the immunosuppressive protocols based on the white population, neglecting the differences between the races. The purpose of this study was to explore a suitable immunosuppressive regimen for Chinese renal allograft recipients.Methods. Two hundred cadaveric renal allograft recipients who underwent transplantation between July 1999 and October 2001 were observed. Before October 2000, 104 recipients received the conventional dose of immunosuppressants; thereafter, 96 recipients received lower dose treatment. Doses of immunosuppressive agents, the incidence of acute rejection and pulmonary infection, and patient and graft survival rates were compared between the two groups.Results. Doses of mycophenolate mofetil (MMF) and cyclosporine A (CsA) administered in the conventional dose group were significantly higher than in the lower dose group at 3 months posttransplant, as was prednisone at 6 months posttransplant. The incidence of acute rejection and subclinical rejection that was biopsy-proven or diagnosed by clinical manifestations was 17.3% and 19.8%, respectively, in the conventional dose group and the lower dose group within the first 6 months, and no significant difference was noted (P=0.55). The incidence of pulmonary infection, especially severe infection, was much higher in the conventional treatment group (40.1% and 26.9%, respectively) than that in the lower dose group (11.5% and 5.2%, respectively), and the differences were statistically significant (P<0.001). The corresponding 1-year survival rate of patients was 87.4% and 97.9% (P<0.01), and that of renal grafts was 85.5% and 96.9% (P<0.01), for patients receiving conventional dose and lower dose immunosuppressive drugs, respectively. The rate of death with a functioning allograft caused by infection in the conventional dose group was significantly higher than that in the lower dose group (12.5% vs. 0%, P<0.01).Conclusions. The regimen of lower dose MMF, CsA, and prednisone in combination can significantly reduce the incidence of pulmonary infection, especially severe pulmonary infection, without increasing the incidence and severity of allograft rejection.

Abstract:Immunodeficiency is a state of compromised immune mechanisms that leads to increased vulnerability to opportunistic infections. [ 1 ] Opportunistic infections in immunocompromised patients can present a diagnostic challenge. Nocardia and Aspergillus fumigatus are opportunistic pathogens and easily overlooked owing to their rarity, leading to delayed diagnosis and treatment. In patients with connective tissue diseases, steroids use may lead to an increased risk of opportunistic infections with a poor prognosis. Pneumonia in immunocompromised individuals often has high incidence and mortality rates. However, co-infection with Nocardia and A fumigatus has rarely been reported. We report a case of pneumonia with Nocardia and A fumigatus successfully treated.

Abstract:Objective To study the influence on host defense against Aspergillus infection when immunized with Aspergillns-pulsed DC in mice.Methods DC were generated from mice bone marrow cells and pulsed with the antigen of Aspergillus fumigatus.Then DC were transfered into mice via the lateral tail vein.Immunocompromised mice were made by treatment with cyclophosphamide administered intraperitoueally(i.p.).Suspension of conidia was applied to the nostrils of mice to develop the model of invnsive pulmonary aspergillosis (IPA).Lungs were harvested and homogenized.Portions of homogenates were cultured to determine the number of CFU.IFN-γin lung homogenates was determined by a cytokine-specific ELISA kit.Reverse transcription PCR(RT-PCR)analysis was used to determine the mRNA of IFN-γin spleen cells.Results Compared with naive DC and heat-inactivated Aspergillus fumigatus(HAF)immunized mice,the mice immunized with Aspergillus-pulsed DC demonstrated prolonged survival time,had significantly lower concentration of infectious Aspergillus organisms in their lung tissues,and had higher levels of IFN-γin spleen cells and lung tissues(3.60±1.57 mg/ml vs 1.10±0.42 ng/ml, 1.35±0.47 ng/ml).Lung sections from HAIr and na(?)ve DC-treated mice revealed patterns of lesions characterized by bronchial wall damage, peribronchial necrosis,and the presence of numerous intimating inflammatory cells,Conidia and hyphae were seen in these mice.In contrast, these features were not observed in HAF-pulsed DC treated mice whose lung were characterized by few inflammatory ceils infiltration,and no evidence of fungal growth after inoculation.Conclusion Transfered into mice,Aspergillus-pulsed DC can induce Thl response to the fungus and enhance their host defense against Aspergillus infection.
Role of excessive inflammatory response in death of immunosuppressed invasive pulmonary aspergillosis mice

Abstract:Objective Immunosuppressed mice were injected with suspension of Aspergillus fumigates spore by tracheotomy to establish invasive pulmonary aspergillosis (IPA) animal models and pathological changes of lung tissue were dynamically observed in order to provide a basis for the IPA mouse studies and learn more about IPA disease development.Methods C57BL/6J mice were intraperitoneally injected with cyclophosphamide 250 mg/kg twice,building immune suppression.Under anesthesia,the neck of mice was cut a 0.3 cm incision,the trachea was isolated and inoculated with Aspergillus fumigatus.The mice general state,survival,pulmonary fungal load,gross and pathological changes in lung tissue were dynamically observed.Results The mortality rate on the fourth day after Aspergillus fumigatus infection in mice was 100％.The content of Aspergillus spores in lungs on the first day was the highest,and then decreased.The gross observation showed lung tissue bleeding,edema,and white fungus foci,and the degree gradually increased.Pathological results showed alveolar inflammatory cells,including lymphocytes infiltration,heavy bleeding and necrosis increased with time,the normal alveolar structure gradually disappeared.Conclusions By intraperitoneal injection of cyclophosphamide,mice are immunosuppressed,and mouse model of Aspergillus infection is successfully established by tracheotomy,excessive inflammatory response is an important reason of death.
Clinical and Microbiological Characteristics of Aspergillosis at a Chinese Tertiary Teaching Hospital

Abstract:Background: Aspergillus spp. infection in immunocompromised patients results in increasing morbidity and mortality. This study investigated clinical and microbiological characteristics of aspergillosis in a Chinese tertiary teaching hospital. Methods: A total of 114 patients with aspergillosis were included over a 5-year period at Ruijin Hospital. In sum, 114 Aspergillus strains were isolated and identified at species level using matrix-assisted laser desorption ionization time-of-flight mass spectrometry, confirmed by ITS gene region and β-tubulin (BenA) gene sequencing. Sensititre YeastOne was used in vitro to test susceptibility to antifungal drugs: amphotericin B, itraconazole, voriconazole, posaconazole, isavuconazole, micafungin, anidulafungin, and caspofungin. Results: The median age of the patients was 61 (19) years, men accounted for 53.5% (n=61) of the sample, about 64% were immunocompromised, and 36% had underlying diseases. Pulmonary diseases accounted for 27.2%. Aspergillus isolates were mainly isolated from sputum (n=42, 36.8%). Antifungal therapy was administered to 106 (93.0%) patients and voriconazole (n=76, 66.7%) was the most frequently used as empirical therapy. Aspergillus fumigatus (n=69, 60.5%) was the most common species. There was a 73.7% concordance between MALDI-TOF MS and molecular identification. All Aspergillus isolates showed good susceptibility to anidulafungin and caspofungin. Conclusion: Immunocompromised patients are an at-risk population for aspergillosis, and voriconazole was used as empirical therapy in Ruijin Hospital, China. A. fumigatus was the predominant Aspergillus species causing aspergillosis, and A. flavus - as non-A. fumigatus species are increasing - the second-leading cause of aspergillosis. Anidulafungin and caspofungin were the most active in vitro against the Aspergillus isolates tested. The MALDI-TOF MS method showed good accuracy for identification of common Aspergillus spp. In vitro antifungal-susceptibility testing is crucially important for decisions on effective therapy with aspergillosis.
Transient Overexpression of Gamma Interferon Promotes Aspergillus Clearance in Invasive Pulmonary Aspergillosis

Abstract:Cytokines are critical molecules necessary for normal lung pathogen host defences. Gamma interferon (IFN-gamma) and T1-phenotype immune responses are important components of host defence against Aspergillus. Therefore, we hypothesized that transient overexpression of IFN-gamma within the lung could augment host immunity against Aspergillus. Here it was showed that intranasal administration of 5 x 10(7) colony-forming units (CFU) of Aspergillus fumigatus (Af ) induced the expression of IFN-gamma. Mice were intranasally (i.n) administrated with 5 x 10(8) PFU of a recombinant adenovirus vector containing the murine IFN-gamma cDNA (AdmIFN-gamma), and challenged 24 h later with Af. We observed that i.n. administration of AdmIFN-gamma resulted in about a fourfold increase in levels of IFN-gamma and IL-12 within the lung, about a 75% reduction in lung fungal contents at day 2 and a more than threefold higher survival rate in the AdmIFN-gamma-treated group compared to the controls (P < 0.01). This protection effect was not found when AdmIFN-gamma was i.p. administrated. Alveolar macrophages and lung leucocytes isolated from i.n. AdmIFN-gamma-treated animals displayed enhanced killing of intracellular Aspergillus organisms ex vivo. These results demonstrate that transient overexpression of IFN-gamma could augment host defence against Aspergillus.
Innate and Adaptive Immune Responses Induced by Aspergillus Fumigatus Conidia and Hyphae

Abstract:Previous research indicated that hyphae of Aspergillus fumigatus ( A. fumigatus ) rather than conidia could successfully build a pulmonary aspergillosis model in immunocompetent mice. In this study, we compared the immune responses induced by hyphae and conidia to explore the possible mechanism of this striking phenomenon. Herein, a novel method was designed and adopted to quantify hyphal fragments. Murine macrophages RAW264.7 and human peripheral blood mononuclear cells were stimulated by A. fumigatus hyphae and conidia in vitro, respectively, and then immunological reactions were measured. Male C57BL/6 mice were challenged with conidia and hyphae through intratracheal inoculation. Dynamic conditions of mice were recorded, and RNA-seq measured corresponding immune responses. The results of the study confirmed that hyphae could induce more intensive inflammation than conidia in vitro and in vivo. However, macrophages revealed a higher production of ROS and M1 polarisation in response to conidia stimuli. Additionally, conidia could promote Th1 cell differentiation, while hyphae could increase the CD4/CD8 ratio. RNA-seq validated the fact that those multiple immunologically relevant pathways were more strongly activated by hyphae than conidia, which also promoted Th2 cell differentiation and suppressed Th1 signalling. Both hyphae and conidia could activate Th17 signalling. In general, conidia and hyphae induced distinctly different host immune responses, and the immune responses induced by conidia played a better protective effect. Therefore, the unique function of hyphae in the spread and infection of Aspergillus should be emphasised, and more research is required to clarify the underlying mechanisms for better understanding and management of aspergillosis.
Glucocorticoids Suppress NF-κB-Mediated Neutrophil Control of Aspergillus fumigatus Hyphal Growth

Abstract:Glucocorticoids are a major class of therapeutic anti-inflammatory and immunosuppressive drugs prescribed to patients with inflammatory diseases, to avoid transplant rejection, and as part of cancer chemotherapy. However, exposure to these drugs increases the risk of opportunistic infections such as with the fungus Aspergillus fumigatus, which causes mortality in >50% of infected patients. The mechanisms by which glucocorticoids increase susceptibility to A. fumigatus are poorly understood. In this article, we used a zebrafish larva Aspergillus infection model to identify innate immune mechanisms altered by glucocorticoid treatment. Infected larvae exposed to dexamethasone succumb to infection at a significantly higher rate than control larvae. However, both macrophages and neutrophils are still recruited to the site of infection, and dexamethasone treatment does not significantly affect fungal spore killing. Instead, the primary effect of dexamethasone manifests later in infection with treated larvae exhibiting increased invasive hyphal growth. In line with this, dexamethasone predominantly inhibits neutrophil function rather than macrophage function. Dexamethasone-induced mortality also depends on the glucocorticoid receptor. Dexamethasone partially suppresses NF-κB activation at the infection site by inducing the transcription of IκB via the glucocorticoid receptor. Independent CRISPR/Cas9 targeting of IKKγ to prevent NF-κB activation also increases invasive A. fumigatus growth and larval mortality. However, dexamethasone treatment of IKKγ crispant larvae further increases invasive hyphal growth and host mortality, suggesting that dexamethasone may suppress other pathways in addition to NF-κB to promote host susceptibility. Collectively, we find that dexamethasone acts through the glucocorticoid receptor to suppress NF-κB-mediated neutrophil control of A. fumigatus hyphae in zebrafish larvae.

Abstract:Aim:Aspergillus fumigatus is one of the most common opportunistic fungi that can cause invasive infection. To profile the kinetic variation of immune cells and cytokines after exposure to A. fumigatus thoroughly, we established a pulmonary A. fumigatus infection model in temporarily immunosuppressed mice. Materials & methods: Systematic and kinetic studies of different immune cells and cytokines were performed. Results: We observed that the granulocytes and macrophages recruited to the site of infection played an important role in the infectious phase. There was a significant increase in the cytokines IFN-gamma, IL-6, TNF-alpha as well as the chemokines CXCL1, MIP-1 alpha, MIP-2 and CCL5 after infection. IL-10 was found to participate in balancing the anti-inflammatory response in the recovery phases. The immune response mediated by T cells was mainly presented by the Th1-type on day 7 after exposure with a high proportion of IFN-gamma(+) CD4(+) T cells and CD4(+)CD44(high)CD62L(low) effector T cells. Conclusion: These kinetic parameters of the immune response might provide diagnostic clues for A. fumigatus infection.
Adoptive Immunity Mediated by HLA-A*0201 Restricted Asp F16 Peptides-Specific CD8+ T Cells Against Aspergillus Fumigatus Infection

Abstract:Aspergillus fumigatus (A. fumigatus) is the most common pathogen of invasive aspergillosis (IA), a life-threatening infection in immunocompromised patients. Recent findings revealed that CD8+ T cells can mediate cytotoxic activity against A. fumigatus. Here, we bioinformatically identified three HLA-A*0201-restricted peptides from Asp f16, an A. fumigatus antigen which was previously shown to be involved in T cell immunity. Our immunological results demonstrated that these peptides can potently induce cytotoxic T lymphocyte (CTL) response in CD8+ T cells, thus, damaging the conidia and hyphae of A. fumigatus. Moreover, the Asp f16 peptides can also raise Th1 cell-like response, as measured by interferon-γ (IFN-γ) enzyme-linked immunosorbent spot (ELISPOT). Furthermore, we established an invasive pulmonary aspergillosis model in HLA-A*0201 transgenic mice. Adoptive transfer of Asp f16 peptides-specific CTL significantly extended the overall survival time in the A. fumigatus-infected immunocompromised mice. In conclusion, our results demonstrate that the Asp f16 peptides might provide immunity against invasive A. fumigatus infection.
