Abstract:Objective: Lead exposure is one of major public health problems. The aim of this study was to examine in vitro the effect of lead on lymphocyte intracellular Ca2+ ([Ca2+]i). Methods: Lymphocytes were separated from mice spleen and treated with three doses of PbCl2 (1, 10 and 100 mu mol/L), [Ca2+]i was measured in a Thermo Labsystems Fluoroskan Ascent after loading the cells with Fura2-AM probe. Results: The lymphocyte [Ca2+]i in 10 and 100 mu mol/L PbCl2 groups increased to the highest level after 10 min of lead exposure (P<0.05). After I h of lead exposure, however, lymphocyte [Ca2+]i levels were not statistically different from that in control group cells (P>0.05). Removal of Ca2+ from external solution did not significantly affect the PbCl2-induced lymphocyte [Ca2+]i, PbCl2 increased the [Ca2+]i under both normal Ca2+ and Ca2+-free conditions. With pretreatment of calmodulin (CaM) antagonist W7, lymphocyte [Ca2+]i levels were still high, but [Ca2+]i levels were not as high as those in the absence of W7. Conclusion: When the lymphocytes were exposed to PbCl2 ranging from 1 to 100 mu mol/L, the lymphocyte [Ca2+]i level was increased, but the increase appeared reversible CaM may play a role in the process of the effect of lead on the lymphocyte [Ca2+]i. Study on the Neurotoxic Effects of Low-Level Lead Exposure in Rats

Abstract:Objective: To investigate effects of developmental lead exposure on nitric oxide synthase (NOS) activity in different brain regions and on N-methyl-D-aspartate (NMDA) receptor mRNA expression in the hippocampus of rats. On the basis of these observations, we explored possible mechanisms by which lead, exposure leads to impaired learning and memorizing abilities in children. Methods: A series of rat animal models exposed to low levels of lead during the developing period was established (drinking water containing 0.025%, 0.05% and 0.075% lead acetate). NOS activities in the hippocampus, the cerebral cortex, the cerebellum and the brain stem were determined with fluorescence measurement and levels of mRNA expression of the NMDA receptor 2A (NR2A) subunit and NMDA receptor 2B (NR2B) subunit in the rat hippocampus were measured with Retro-translation (RT-PCR). Results: There were no differences in the body weight of rat pups between any of the groups at any given time ( P >0.05). The blood lead level of Pb-exposed rat pups showed a systematic pattern of change: at 14 d of age, it was lower than that at 7 d of age, then rising to the peak level at 21 d and finally falling to lower levels at 28 d. The hippocampal NOS activities of lead-exposed groups were all lower than that of the control group on the 21 st and 28th day ( P <0.01). NOS activities in the cerebellum of lead-exposed groups were all lower than that of the control group on the 21st, and 28th day ( P <0.001) and the NOS activity of the 0.025% group was significantly lower than that of the 0.05% groups on the 28th day ( P <0.05). NOS activity in the cerebral cortex of the 0.075% group was significantly lower than that of the control, 0.025% and 0.05% groups on the four day spans ( P <0.001). There was no significant difference of NOS activity in the brain stem between any lead-exposed group and the control group on the four day spans. In the 0.05% and the 0.075% groups the level of NR2A mRNA expression was higher than that in the control group at 7 d and 14 d of age ( P <0.05). In the 0.025% group, the level of NR2A was found to be higher than that in the control group at 7 d of age only ( P <0.05). No significant differences were found for the levels of NR2B mRNA expression between any of the groups at any given time. Conclusions: NOS activity in the hippocampus, the cerebral cortex and the cerebellum are inhibited by lead exposure. The degree of the inhibitory effect depends on the time span of exposure and the lead concentration. Developmental low-level lead exposure was found to raise the level of NR2A mRNA expression in the hippocampus of rats. Developmental low-level lead exposure does not affect the level of NR2B mRNA expression in the hippocampus.
The Role of Modified Citrus Pectin As an Effective Chelator of Lead in Children Hospitalized with Toxic Lead Levels.

Abstract:Context Lead toxicity is an ongoing concern worldwide, and children, the most vulnerable to the long-lasting effects of lead exposure, are in urgent need of a safe and effective heavy metal chelating agent to overcome the heavy metals and lead exposure challenges they face day to day.Objective This clinical study was performed to determine if the oral administration of modified citrus pectin (MCP) is effective at lowering lead toxicity in the blood of children between the ages of 5 and 12 years.Method Hospitalized children with a blood serum level greater than 20 mu g/dL, as measured by graphite furnace atomic absorption spectrometry (GRAS), who had not received any form of chelating and/or detoxification medication for 3 months prior were given 15 g of MCP (PectaSol) in 3 divided dosages a day. Blood serum and 24-hour urine excretion collection GFAAS analysis were performed on day 0, day 14, day 21, and day 28.Result This study showed a dramatic decrease in blood serum levels of lead (P=.0016; 161% average change) and a dramatic increase in 24-hour urine collection (P=.0007; 132% average change).Conclusion The need for a gentle, safe heavy metal chelating agent, especially for children with high environmental chronic exposure, is great. The dramatic results and no observed adverse effects in this pilot study along with previous reports of the safe and effective use of MCP in adults indicate that MCP could be such an agent. Abstract:To investigate the effects of chronic lead exposure on expression of autophagy-associated proteins in rat hippocampus.SD rats were randomly divided into three groups: control group was given distilled water, lead-exposed groups were given 0.5 g/L (low-dose) or 2.0 g/L(high-dose) lead acetate solution in drinking water. The rat pups started to drink the lead content water until 60 d maturity. The lead contents in blood and brain samples were analyzed by graphite furnace atomic absorption spectrophotometry. The expressions of Beclin 1, LC3, LAMP2 and cathepsin B proteins were detected by Western blot and immunohistochemistry.Compared with control group, the contents of lead were significantly higher in blood and hippocampus samples in chronic lead-exposed rats (P<0.01). Western blot showed that the expression of Beclin 1 and LC3-II/LC3-I increased significantly in high dose lead-exposed group compared with control group (P<0.05 or P<0.001). The confocal laser immunostaining results demonstrated that increased immunofluorescence staining of cathepsin B in hippocampal neurons compared with control animals.The disturbance of autophagy-lysosome signaling molecules might be partially contribute to neurotoxicity of chronic lead exposure.
Chronic Lead Exposure Results in Auditory Deficits and Disruption of Hair Cells in Postweaning Rats

Abstract:Objective: The effects of lead exposure on cognitive function have been studied intensively over the past decade, but less attention has focused on its impact on auditory function. This study is aimed at investigating the effect of lead on the cochlea and the molecular mechanisms responsible for its actions. Methods: 0.2% lead acetate was administered to rats in drinking water for 30, 60, and 90 days. Brainstem auditory evoked responses (ABR) were recorded, and morphological changes in the hair cells were observed. We also measured glutathione (GSH) and malondialdehyde (MDA) concentrations and antioxidant enzyme activities such as catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and glutathione reductase (GR) activities in the cochlea. Results: Lead exposure increased the ABR threshold and slightly prolonged the latencies of wave II and wave IV in rats. Abnormally shaped hair cells and loss of hair cells were found in the cochlea basilar membrane, together with degenerative changes in spiral ganglion neurons following lead exposure. The activities of some antioxidant enzymes were also reduced in association with upregulation of MDA expression. These effects may be caused by impaired catalytic function of the enzymes as a result of lead interaction. Conclusion: The antioxidant system of the cochlea in the immature rat brain is highly vulnerable to developmental lead exposure. Oxidative stress may therefore represent a possible mechanism for lead-induced auditory deficits.

Abstract:Toxicity of lead on male reproductive functions has raised wide public concern as environmental lead contamination remains common worldwide. Conflicting and controversial data are available regarding effects of lead on male fertility. More importantly, our knowledge on effects of lead on sperm DNA integrity is significantly limited. Thus, further studies should focus on this issue. In the current study, adult male mice were exposed to a series of lead acetate concentrations in drinking water for six weeks. Following administration, lead levels in blood, testicles, and epididymis were measured, and potential changes in morphology of testis and epididymis due to lead exposure were identified. We also analyzed sperm parameters, including sperm density, viability, motility, and morphology, to evaluate quality of sperm collected from epididymis. Especially, hypothetical influence of lead on sperm DNA integrity was also evaluated by terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick end labeling, alkaline comet assay, and sperm chromatin structure assay. Lead exposure possibly exerted no effect on growth of mice because these animals acquired similar body weight gain during the experimental period. However, high lead concentrations (0.5% and 1%) in drinking water affected sperm motility and increased percentage of spermatozoa with abnormal morphology. In groups treated with 0.25%, 0.5%, and 1% lead acetate, percentages of sperm cells showing DNA breaks and chromatin structure damage significantly increased. Altogether, lead exposure not only exhibits adverse effects on sperm physiological parameters, but also impairs DNA structure and integrity. These effects may lead to significant decline in male fertility.
From Molecular to Functional Effects of Different Environmental Lead Exposure Paradigms

Abstract:Lead is a heavy metal whose widespread use has resulted in environmental contamination and significant health problems, particularly if the exposure occurs during developmental stages. It is a cumulative toxicant that affects multiple systems of the body, including the cardiovascular and nervous systems. Chronic lead exposure has been defined as a cause of behavioral changes, inflammation, hypertension, and autonomic dysfunction. However, different environmental lead exposure paradigms can occur, and the different effects of these have not been described in a broad comparative study. In the present study, rats of both sexes were exposed to water containing lead acetate (0.2% w/v), from the fetal period until adulthood. Developmental Pb-exposed (DevPb) pups were exposed to lead until 12 weeks of age (n = 13); intermittent Pb exposure (IntPb) pups drank leaded water until 12 weeks of age, tap water until 20 weeks, and leaded water for a second time from 20 to 28 weeks of age (n = 14); and the permanent (PerPb) exposure group were exposed to lead until 28 weeks of age (n = 14). A control group (without exposure, Ctrl), matched in age and sex was used. After exposure protocols, at 28 weeks of age, behavioral tests were performed for assessment of anxiety (elevated plus maze test), locomotor activity (open-field test), and memory (novel object recognition test). Metabolic parameters were evaluated for 24 h, and the acute experiment was carried out. Blood pressure (BP), electrocardiogram, and heart (HR) and respiratory (RF) rates were recorded. Baroreflex gain, chemoreflex sensitivity, and sympathovagal balance were calculated. Immunohistochemistry protocol for NeuN, Syn, Iba-1, and GFAP staining was performed. All Pb-exposed groups showed hypertension, concomitant with a decrease in baroreflex gain and chemoreceptor hypersensitivity, without significant changes in HR and RF. Long-term memory impairment associated with reactive astrogliosis and microgliosis in the dentate gyrus of the hippocampus, indicating the presence of neuroinflammation, was also observed. However, these alterations seemed to reverse after lead abstinence for a certain period (DevPb) and were enhanced when a second exposure occurred (IntPb), along with a synaptic loss. These results suggest that the duration of Pb exposure is more relevant than the timing of exposure, since the PerPb group presented more pronounced effects and a significant increase in the LF and HF bands and anxiety levels. In summary, this is the first study with the characterization and comparison of physiological, autonomic, behavioral, and molecular changes caused by different low-level environmental lead exposures, from the fetal period to adulthood, where the duration of exposure was the main factor for stronger adverse effects. These kinds of studies are of immense importance, showing the importance of the surrounding environment in health from childhood until adulthood, leading to the creation of new policies for toxicant usage control.
Evaluating the Genotoxic Effects of Workers Exposed to Lead Using Micronucleus Assay, Comet Assay and TCR Gene Mutation Test.

Abstract:To evaluate the genotoxic effects of lead (Pb) exposure, 25 workers in a workplace producing storage battery were monitored for three genetic end-points using micronucleus (MN) assay, comet assay and TCR gene mutation test. Twenty-five controls were matched with workers according to age, gender and smoking. The air Pb concentration in the workplace was 1.26 mg/m(3). All subjects were measured for Pb concentration of blood by atom absorption spectrophotometry. The mean Pb concentration of blood in workers (0.32 mg/l) was significantly higher than that in controls (0.02 mg/l). The results of MN test showed that the mean micronuclei rate (MNR) and mean micronucleated cells rate (MCR) in workers were 9.04+/-1.51 per thousand and 7.76+/-1.23 per thousand, respectively, which were significantly higher than those (2.36+/-0.42 per thousand and 1.92+/-0.31 per thousand) in controls (P<0.01). It was found in the comet assay that the mean tail length (MTL) of 25 workers and 25 controls were 2.42+/-0.09 and 1.02+/-0.08 microm, respectively, there was significant difference between workers and controls for MTL (P<0.01), also the difference of the mean tail moment (MTM) between workers (0.85+/-0.05) and controls (0.30+/-0.09) was very significant (P<0.01). However, in TCR gene mutation assay Mfs-TCR of workers and controls were 1.69+/-0.15 x 10(-4) and 1.74+/-0.17 x 10(-4), respectively, there was no significant difference between workers and controls (P>0.05). The results of our study indicated that the genetic damage was detectable in 25 workers occupationally exposed to lead.
Effect of Lead Exposure on the Accumulation of Copper and Iron in Central Nervous System of Rats

Abstract:Objective: To investigate the dynamic changes of copper and iron contents in brain tissue, body fluids and barriers of rats exposed to lead at different periods in order to provide a theoretical basis for the study of the mechanism of lead nerve injury. Methods: Sixty-four healthy adult SPF male SD rats were randomly divided into control group and lead exposure group, after one week of adaptive feeding, rats in the lead exposure group were treated with 250 mg/L lead acetate, and rats in control group were treated with ordinary drinking water, the experimental period was 12 weeks. After exposure for 3, 6, 9 and 12 weeks, the samples including blood, choroid plexus, cerebrospinal fluid, cortex, hippocampus, striatum, hypothalamus, amygdala, substantia nigra and cerebellum were obtained. Lead, copper and iron content in all kinds of samples were detected by Inductively Coupled Plasma Mass Spectrometry(ICP-MS). The measurement data were presented as Mean±SD, Comparison of metal contents in different tissues of rats at different time analyzed using repeated measurement analysis of variance, Two-variable correlation analysis using Spearman correlation test.The relationship between lead exposure experiod and copper and iron in samples was studied by using trend test. Results: After 12 weeks of lead exposure compared with the control group, lead contents in cortex, hippocampus, striatum, hypothalamus, amygdala, substantia nigra and cerebellum of rats were 2.21, 2.44, 2.95, 3.53, 4.01, 1.85 and 2.86 folds of control group, and the differences were statistically significant(P<0.05). At the same time, lead content in blood, cerebrospinal fluid,choroid plexus, brain microvessels and bones increased. The increase rate in the amygdala and cerebrospinal fluid ranked first among brain tissue or barrier,which were 4.01 and 3.0 folds respectively. Compared with the control group, Compared with the control group, copper content in cortex,hippocampus, striatum, hypothalamus,amygdala, cerebellum,blood,cerebrospinal fluid,choroid plexus and cerebral microvasculature showed an increasing trend among rats following 3,6,9,12 weeks of lead exposure. Copper content change in the striatum was highest among all brain tissue. The increase rate of copper content in the striatum was at the top among brain tissues. After 12 weeks of lead exposure,copper content in brain microvessels was 4.98 folds higher than that of the control group (P<0.05). After lead exposure at different periods,the iron content in the cortex, hippocampus, striatum,cerebrospinal fluid,choroid plexus and brain microvessels of experimental rats all increased(P<0.05). And the iron increase rate in the hypothalamus or cerebrospinal fluid increase ranked first among brain tissues or body fluid the most obviously. Conclusion: With the increase of exposure time, lead exposure can changes in the contents of copper and iron in different brain tissues,body fluids and barriers in rats,among which, the contents of copper and iron in the amygdala,cerebrospinal fluid and brain microvessels increase significantly. This may be related to nerve damage from lead exposure.
Influence of montmorillonite nanocomposite on tissue levels of lead and blood biochemical parameters in pigs exposed to lead.

Abstract:Eighteen （Duroc × Landrace × Yorkshire） castrated boars,55 days of age and weighing approximately 19 kg,were used to investigate the effects of montmorillonite nanocomposite on tissue levels of lead as well as changes in blood biochemical parameters in pigs fed lead contaminated diets.The barrows were randomly assigned to one of three treatments （n=6） including a control which were fed a basal diet based on corn and soybean meal.The remaining two groups were fed the basal diet supplemented with 10 mg/kg lead either with or without 0.5% montmorillonite nanocomposite for 50 days.Pigs treated with lead had significantly increased levels of lead in their tissues.Lead accumulated in all tissues of the body with the highest accumulation found in the hair.When montmorillonite nanocomposite was given along with lead,tissue concentrations of lead did not differ from the control.Fecal excretion of lead increased significantly in lead-exposed pigs treated with montmorillonite nanocomposite.A progressive and appreciable accumulation of lead was seen in blood with a concomitant increase in zinc protoporphyrin levels during the course of treatment.However,zinc protoporphyrin levels did not differ from the control when montmorillonite nanocomposite was administered along with lead.The activity of δ-amino levulinic acid dehydratase in blood was significantly increased in lead treated pigs compared with the control.However,in pigs treated with lead and montmorillonite nanocomposite in combination,the activity of δ-amino levulinic acid dehydratase was similar to control pigs.Our results indicate that montmorillonite nanocomposite treatment of pigs exposed to lead resulted in reduced lead concentrations in body tissues.It appears that montmorillonite nanocomposite functions by increasing fecal excretion of lead.
Rat Testis Damage Caused by Lead Sulfide Nanoparticles After Oral Exposure

Abstract:Lead sulfide nanoparticals (PbS NPs) is an important semiconductor material due to its unique physical and chemical properties, but its potential health hazard to reproductive system is not clear. In the current study, we systematically explored the reproductive toxicity of PbS NPs in rats by measuring the body weight and testicular coefficient, testing serum testosterone levels, and studying the sperm survival rate and sperm abnormality rate. Furthermore, in order to study the toxic mechanism we performed lead contents measurements in testis, and investigated the pathology in testis. Our results confirmed that PbS NPs showed high reproductive toxicity due to PbS NPs in rats' testicular tissue by the establishment of PbS NPs chronic exposure model.

Early-life exposure to lead changes cardiac development and compromises long-term cardiac function

Abstract:Lead (Pb) is widely used in industrial and daily-use consumer products. Early-life exposure may increase the risk of lead-related heart problems in childhood. However, the effects of early-life lead exposure on fetal heart development and long-term cardiac outcomes are unknown. In this study, pregnant ICR mice were exposed to lead acetate trihydrate (50 mg/kg/d) via oral gavage from gestation day 1.5 until offspring weaning. Thereafter, the second hit model was established, two groups of offspring (4 weeks old) were either administered sterile saline or Angiotensin II (Ang II) for 4 weeks until euthanasia. We investigated lead-induced offspring heart damage from embryonic period to adulthood by echocardiographic analysis, pathological H&E staining, and ultrastructural examination, as well as mitochondrial function detection. The results showed early-life lead exposure predisposed offspring mice to decreased ejection fraction, increased left ventricular volume, accompanied by hypertrophy and dilation, cardiomyocyte sarcomere dysplasia, abnormal mitochondrial structure, mitochondrial dysfunction, and decreased expression of key sarcomeric and mitochondrial genes, rendering them more susceptible to cardiac hypertrophy, vascular wall thickening, cardiac fibrosis, apoptosis, and heart failure induced by Ang II infusion. This study elucidates early-life low dose lead exposure compromises cardiac development and exacerbates second hit-induced cardiac pathological responses in adulthood, which furnishes crucial scientific evidence pertaining to the cardiac toxicity and risk evaluation associated with early-life exposure to lead.

Abstract:Lead exposure has been implicated in the aetiopathogenesis of male infertility via an oxidative stress-sensitive pathway. Conversely, acetate has been shown to confer cellular protection by improving the antioxidant defense mechanism. Yet, the effect of acetate on lead-induced testicular toxicity, viz., dysregulation of testicular steroidogenesis and spermatogenesis, has not been reported. The present study probed the influence of acetate on lead-induced dysregulation of testicular steroidogenesis and spermatogenesis. In our study, a reduction in body weight gain and testicular weight was identified in lead-exposed rats. While histopathological results established distortion of testicular histoarchitecture, reduced germ cell count, and suppressed spermatogenesis, biochemical studies confirmed that lead-deregulated testicular steroidogenesis was associated with reduced circulating gonadotropin-releasing hormone and gonadotropins, as well as down-regulated testicular 3β-HSD and 17β-HSD activities. These findings were accompanied by increased testicular malondialdehyde, TNF-α, IL-1β, and IL-6, and reduced glutathione, thiol and non-thiol protein levels, total antioxidant capacity, superoxide dismutase, and catalase activities. In addition, lead exposure increased NFkB and Bax levels, as well as caspase 3 activity, but reduced Bcl-2 levels. However, co-administration of acetate ameliorated lead-induced alterations. Collectively, acetate attenuated lead-induced dysregulation of testicular steroidogenesis and spermatogenesis by targeting oxidative stress, NFkB-mediated inflammation, and caspase 3-driven apoptosis. Impact of Quercetin and Omega-3 Fatty-Acid on Lead-Induced Alterations in Reproductive Parameters in Male Wistar Rats

Abstract:Exposure to environmental toxins like Lead has been associated with male infertility. Whether or not potent antioxidants like omega 3 Fatty acids and/or quercetin could reduce the effect of lead on male sexual functions deserve scientific investigation. Thirty-five (35) male albino Wistar rats were assigned into 7 groups: Group 1 (normal control), Group 2 (sham-control1), Group 3 (sham-control 2), Group 4 (Lead group), Group 5 (Lead + Omega-3), Group 6 (Lead + Quercetin) and Group 7 (Lead + Omega-3 + Quercetin). Lead was given orally at 20mg/kg bwt, quercetin 20mg/kg bwt s.c, Omega-3 14.29mg/kg bwt orally. The animals all had free access to rat food and water for 56 days. After which they were sacrificed, and semen and blood samples were collected for assay. The results showed no significant difference between normal control group and sham controls. Sperm function parameters (sperm count, motile, viable and normal sperms) in the lead treated group was significantly reduced compared to the control. Omega 3 and/or quercetin administration reversed the reductions in sperm function parameters to near control levels. Hormone levels (LH and Testosterone) were significantly reduced in lead group compared to the normal control but were reversed after quercetin and omega 3 treatment. However, there was no significant difference in the FSH level among experimental groups. Conclusively, omega 3 and/or quercetin ameliorates the harmful effects of lead on reproductive parameters by improving sperm functions (total sperm count, viability, motility, and morphology). A combination of both quercetin and omega 3 provided better ameliorative effect than either omega 3 or quercetin.
[Effects of Lead Exposure on 18 Elements in Blood and Excretions in Rats].

Abstract:OBJECTIVE:To investigate the effects of lead exposure on lead and other metal elements contents in rats.METHODS:SD rats were randomly divided into control group and several experiment groups of different doses. The rats were exposed to lead acetate through intragastric administration every other day for 5 times. The whole blood, urine and feces of all the rats were collected. The concentrations of lead and 18 metal elements in these samples were determined by inductively coupled plasma mass spectrometry (ICP-MS) and inductively coupled plasma atom emission spectrometry (ICP-AES).RESULTS:No significant difference among the groups was found for body weight and organ-body ratios of the rats after lead exposure (P>0.05). With the increase of exposure dose, lead content in blood, total lead in urine and feces tended to increase, while the total lead in urine no longer increased in the high dose group. Significant differences among the groups (P<0.05) were observed for the sodium, magnesium, potassium, strontium, antimony, thallium and bismuth contents in the whole blood, the potassium, iron and antimony contents in the urine, and the calcium, iron, zinc, copper, thallium, bismuth and rare earth elements contents in the feces.CONCLUSION:The effect of lead on the metabolism of divalent metal ions, namely calcium, magnesium, iron, zinc, copper and strontium ion, may be due to the competition of lead with the ions for common delivery carrier. Lead exposure induces the excretion of light rare earth elements and toxic elements (thallium and bismuth), and changes the antimony, sodium and potassium contents in rats. But there is no effect of lead on molybdenum and cadmium in rats.

Abstract:In the current study, we investigated the effect of lead chloride (PbCl2) administration (50 and 100 ppm) on organ and body weight as well as its bioaccumulation during pregnancy and the postnatal period in mice. We showed that lead has no effect on the body weight of mice. However, spleen weight is affected by the two doses of PbCl2 while liver and kidney weights are altered only by the 100-ppm dose. Inductively coupled plasma atomic emission spectrometry (ICP-AES) analysis showed that lead accumulates in the blood, spleen, and thymus. Both doses of PbCl2 significantly reduced splenocyte and thymocyte cell counts after stimulation with lipopolysaccharide (LPS) and phytohemagglutinin A (PHA), respectively. On the other hand, we showed that the levels of Th1 cytokines (interleukin-2 (IL-2), interferon gamma (IFN-γ)), and tumor necrosis factor alpha (TNF-α) were reduced in the serum of mice treated with PbCl2 in a dose-dependent manner, as measured by ELISA. The levels of interleukin-4 (IL-4) and interleukin-10 (IL-10) were very low in untreated mice and were also reduced by treatment with PbCl2. The levels of IL-2, IFN-γ, IL-4, IL-10, and TNF-α secretion differentially decreased in LPS-stimulated splenocytes in lead-treated mice. Using PHA-stimulated thymocytes, we observed a reduction in the levels of IL-2, IL-4, IL-10, and TNF-α in the PbCl2-treated groups. However, IFN-γ concentration in the supernatant of these cells was not decreased when mice were treated with 50 ppm of lead.

Abstract:The study aims to evaluate effects of chronic low-dose coexposure to lead (Pb), cadmium (Cd) and manganese (Mn) on hepatorenal toxicity and oxidative stress. Young male Wistar rats were treated with Pb acetate (1.4 mg/kg BW), Cd chloride (0.01 mg/kg BW), Mn chloride (0.14 mg/kg BW) and their combination (Pb + Cd + Mn) by oral gavage, for 15 weeks. Liver enzymes, albumin (Alb), globulin (Glb), total protein, creatinine, urea and electrolyte concentrations were measured in the serum. Hepatic and renal malondialdehyde (MDA), glutathione peroxidase-1 (GPx1) and metallothionein-1 (MT1) concentrations were measured by enzyme immunoassay technique. Chronic exposure to the metals significantly (p < .05) increased serum Glb concentration and decreased Alb/Glb ratio, compared to the controls. Serum creatinine concentration significantly (p < .05) decreased in the Pb, Cd and Pb + Cd + Mn groups, but elevated in the Mn group. Hepatic MDAs rose significantly (p < .05) in the Pb group, while hepatic GPx1 activities increased significantly (p < .05) in the Cd, Mn and Pb + Cd + Mn groups. Hepatic and renal MT1 concentration decreased (p < .05) in the Mn group only. Biochemical alterations were confirmed by light microscopy of the liver and kidneys, which showed degenerative changes. It is concluded that prolonged coexposure to environmentally relevant levels of Pb, Cd and Mn impairs liver and kidney functions via the induction of oxidative stress, and it underlines the importance of studying toxicants in combination.
