Abstract:Avian coccidiosis is one of the major parasitic diseases in the poultry industry. The infection is caused by Eimeria species, and its treatment relies mainly on the administration of anticoccidial drugs, which can result in drug resistance and side effects. The recent trends in avian coccidiosis treatment is directed to the development of a new therapy using herbal compounds. S-Methylcysteine (SMC) is considered one of the organosulfur compounds in garlic that showed promising activity in the treatment of different pathological conditions via a wide range of anti-inflammatory and antioxidant mechanisms. In this study, the anticoccidial activity of SMC was investigated in Eimeria tenella -infected chickens compared to diclazuril as a widely used anticoccidial drug. In this regard, 14-day-old broilers were divided into six groups ( n = 18). The first group (G1) was the healthy control group, while the second group (G2) was the non-infected SMC group treated at a dose of 50 mg/kg b.w. (high dose). Moreover, the third group (G3) was the positive control group (infected and non-treated). The fourth group (G4) was the infected group treated with SMC of 25 mg/kg b.w. (low dose), while the fifth group (G5) was the infected group treated with SMC of 50 mg/kg b.w. (high dose). Conversely, the sixth group (G6) was the diclazuril-treated group. The anticoccidial effects of SMC and diclazuril were evaluated by counting oocysts and recording the body weight gain, feed conversion ratio, clinical signs, lesions, and mortality rate. Interestingly, SMC showed potent anticoccidial activity, which was exemplified by reduction of oocyst count. Furthermore, the biochemical, antioxidant, and anti-inflammatory parameters in the cecal tissues were restored toward their control levels in G4, G5, and G6. Histopathological observation of cecal tissues was consistent with the aforementioned results revealing the ameliorative effect of SMC against E. tenella infection. This study concluded novel findings in relation to the anticoccidial role of SMC as a plant-based compound against the E. tenella -induced coccidiosis in broiler chickens combined with its antioxidative and anti-inflammatory properties. Further studies for exploring the mechanistic pathways involved in this activity and the potential benefits from its use in association with conventional anticoccidial drugs are warranted.

Descriptive Analysis of Fear and Distrust in Early Phases of GSD Projects

Abstract:When globally distributed software development (GSD) is understood as knowledge intensive collaborative work, many of the reasons for problems encountered in GSD projects can be traced back to social conditions framing the collaboration between people at onsite and offsite. A total of 59 interviews were conducted in 8 GSD projects of two major software companies located in Finland. As a result of categorization of problems related to group relations in GSD projects, the lack of trust between onsite and offsite and fears of losing jobs at onsite was found as major problems in the early phases of the projects. Our analysis suggests that poorly communicated reasons GSD can cause severe problems in collaboration between people by laying the ground for fears and for distrust between sites. The study contributes to the GSD research by creating a novel empirical description of the linkages between fear and distrust in GSD.
Use of quantum methods for a consistent approach to combustion modelling: hydrocarbon bond dissociation energies.

Abstract:An attempt has been made to use modern quantum methods to codify the data base concerning bond dissociation energies in hydrocarbons. Calculations have been performed using two hybrid DFT methods, the well-known B3LYP formalism and a newly developed alternative named KMLYP. CBS-Q has also been employed where possible. The combination of experimental uncertainty and theoretical limitations is less than completely satisfactory. However, within uncertainties that translate to a factor of two at 1500 K, many transferable quantities are elucidated. A hybrid method has been developed for the correction of DFT calculations using group additivity. Given that the philosophy behind this work is the understanding that all data bases must be optimised for specific applications, so that avoidance of large errors is more important than absolute precision, the results appear to be quite useful. We are particularly encouraged by the performance of the KMLYP method, given its ease of application to molecules of practical interest.
Efficacy of immunization compared to an anticoccidial drug combination in the management of challenged coccidiosis in Japanese quail

Abstract:This study was carried out to compare the efficacy of immunization, by a low-dose of live sporulated oocysts of different Eimeria species separately, with the efficacy of amprolium plus sulphaquinoxaline in the management of challenged coccidiosis in Japanese quail. Dropping samples were collected and sent to the laboratory for isolation and identification of Eimeria species. Three Eimeria species were isolated and identified as E. bateri, E. uzura, and E. tsunodai. Single oocyst isolation and propagation were done successfully for each species. For the experimental trial, Japanese quails were divided into 11 groups of thirty birds each and given different treatments. The assessment of each treatment relied on clinical signs, mortality, lesion score, oocyst output, weight gain, feed conversion ratio, and hematological parameters. The results revealed that immunization, with any isolated species, gave the best results regarding all tested parameters. Thus, we concluded that immunization by a low-dose of live sporulated oocysts was better compared to amprolium plus sulphaquinoxaline in the management of coccidiosis in Japanese quail.
Studies of resistance to anticoccidials in Eimeria field isolates and pure Eimeria strains

Abstract:Ten Eimeria field isolates from North Germany were studied in battery tests for sensitivity to selected anticoccidials. A high percentage of the Eimeria field isolates (9 out of 10) showed resistance to anticoccidials, mostly multiple resistance. Partial or complete resistance to maduramicin was found in 7 field isolates, to monensin in 6, to salinomycin in 5, to nicarbazin in 8, to halofuginone in 7, to robenidine and toltrazuril in 1, and to diclazuril in 2 field isolates. Multiple resistance had developed in 7 of the 10 isolates. Cross-resistance between maduramicin, monensin, and salinomycin occurred in 5 Eimeria isolates. One isolate showed cross-resistance between diclazuril and toltrazuril. From the resistant isolates 15 pure E. acerculina and 5 pure E. brunetti strains were obtained by single oocyst infections. Seven of the E. acerculina and 4 of the E. brunetti strains showed resistance or partial resistance that was also present in the original isolate. Ten of 11 resistant strains were multiply resistant.

Anticoccidial Effect of Iberogol and Sulfaclozine on Broilers Experimentally Infected with Eimeria tenella

Abstract:Background: Coccidiosis is a common infection among birds that is caused by Eimeria spp. (Protozoa, Phylum Apicomplexa). Eimeria is transmitted fecal-orally and replicated in the intestines. Coccidiosis causes economic losses and increases birds' susceptibility to other diseases. Methods: In this study, the broilers were experimentally infected by Eimeria tenella and then treated with Sulfaclozine and Iberogol. Birds were randomly divided into 5 groups (21 birds each); I: negative control, II: positive control, III: treated by Iberogol, IV: treated by Sulfaclozine, and V: treated by Iberogol+Sulfaclozine. Twenty-one-day-old chickens were infected by 10000 oocysts/ ml/chicken of E. tenella. Treatments were started 1 day after oocyst observation in thirtyday- old chickens. Treatments continued for 3 days, stopped for 2 days, and again continued for the next 2 days. Results: The results were compared for OPG (oocyst per gram), AW (average weight), FCR (feed conversion ratio), and mortality rate among different groups. According to statistical analysis, the worst results were observed in the Iberogol group (III) among other treatment groups (IV and V), and the most effective treatment was reported in the Iberogol+Sulfaclozine group (V). Conclusion: Complex herbal medications, such as Iberogol, can have different activities, but in coccidiosis, its antioxidant and anti-inflammatory effects are more than the anticoccidial effect. Therefore, Iberogol can be used with Sulfaclozine to increase its efficiency.
Anticoccidial Effect of Halofuginone Hydrobromide Against Eimeria Tenella with Associated Histology

Abstract:Halofuginone (stenorol) has been used as an effective anticoccidial reagent for decades but very little is known about its mode of action. In this study, chickens were inoculated with Eimeria tenella oocysts on 14-day-old and medicated with halofuginone at days 0, 1, 2, 3, 4, 5 and 6 post inoculations (groups 0, 1, 2, 3, 4, 5 and 6, respectively). Chickens in group 7 were taken as challenge-unmedicated control and in group 8 unchallenged-unmedicated control. The survival rate, body weight gains (BWG), oocysts production, cecal scores, bloody diarrhea and histological examinations were analyzed to evaluate the anticoccidial efficacy of halofuginone and to initially elucidate its mechanisms. Results showed that halofuginone which acted as a coccidiostatic can significantly enhance the BWG, and decrease both the oocyst shedding and cecal destruction caused by E. tenella infection. The histological slide examination noted that halofuginone was effective when provided 0-2 days post inoculation but only partially effective when applied 3-7 days post infection. The second-generation schizonts treated with halofuginone appeared vacuolated and degenerated. It is concluded that halofuginone can inhibit the parasite's invasion of host cecal hypothetical cell at the early stages of life cycle and later disturb the parasite's development by vacuolation of the schizonts. The resulting abnormal schizonts could not divide into schizoites and were eventually eliminated by the host's immune response.

Abstract:he present study was adopted to evaluate the efficacy of Garlic, Myrrh and Nigella sativa as anticoccidial agents in the ration of broilers against experimental coccidiosis . The parameters used are clinical signs, mortality, live body weight, weight gain, feed consumption, feed conversion ratio, feed efficiency, oocyst output and caecal lesion scoring . One hundred and twenty five Hubbard chicks of one-day old coccidia free were used in this experiment. Chicks were randomly divided into five groups (25 chicks per group). The groups were separately kept according to thetreatment in wire floored. Chickens were given Garlic (Allium sativum) at dose level of 5% of the ration. Myrrh (Commiphora molmol) at rate of 200 ppm and Nigella sativa 5% of freshly minced seeds were added to the ration. All additives were given continousely for 42 days beginning from the 1st day of age .At the age of 14 days; birds were infected experimentally with 80.000 sporulated oocyst/ bird directly into crop using field strain from Eimeria tenella. Faecal samples were examined for oocyst weekly post experimental infection .The obtained results elicited that , all additive obscure the clinical signs, reduced the mortalities ,improved live body weight, weight gain, feed consumption ,feed conversion ratio and feed efficiency as well as decreased shedding of oocyst and reduced caecal lesion scoring as compared with infected untreated group. Garlic was found to be superior in action and responsiveness than Myrrh and Nigella sativa . It could be concluded that theses additives posses an anticoccidial activity and may be valuable for broiler chickens

Abstract:Rumex nervosus leaves (RNL) and Cinnamomum verum bark (CNB), phytogenic herbs, have received much attention in recent years for their antimicrobial properties; however, there is limited knowledge about their potential anticoccidial functions. The prophylactic effects of RNL and CNB were compared with salinomycin, a standard synthetic anticoccidial agent, in broilers experimentally infected with Eimeria tenella ( E. tenella ). One-day-old broiler chicks ( n = 225) were randomly divided into nine groups. Birds were either fed a basal diet containing 1, 3, or 5 g RNL or 2, 4, or 6 g CNB/kg feed, or treated with salinomycin within the basal diet, or the infected (IUT) or non-infected (UUT) groups were fed a basal diet only. Birds infected ( n = 25 bird/group) with 40,000 sporulated E. tenella oocysts/bird at d 21 except UUT. Bodweight gain (BWG) and feed conversion ratio (FCR) were significantly ( p < .05) lower in IUT compared to UUT. On day 7 post-infection (DPI), birds treated with RNL, CNB, or salinomycin had fewer lesions in the caeca and a lower oocyst value, and a higher oocyst reduction rate in the faeces than birds in the IUT. Although RNL was not able to reduce weight loss caused by coccidiosis, CNB at 6 g improved FCR and production efficiency index (PEI) at 7 DPI compared to the infected groups. In conclusion, RNL at 5 g and CNB at 6 g have moderate anti-coccidial activity and could be used to treat poultry coccidiosis in the field. However, more research is needed to identify active ingredients that make it effective compared to commercially available drugs. Phytogenic feed additives prevented weight loss and caecum pathology in broiler chickens at risk of coccidiosis. Rumex nervosus leaves and Cinnamomum verum bark had similar effects to the coccidiostat salinomycin at high doses. Traditional medicinal plants are potential alternatives to pharmaceutical coccidiostats to promote the health and growth of broiler chickens.

Antioxidant properties of D-limonene and its nanoemulsion form enhance its anticoccidial efficiency in experimentally infected broilers with Eimeria tenella: an in vitro and in vivo study

Abstract:The excessive use of conventional medications to treat coccidiosis has led to concerns regarding drug residues in tissues and the emergence of multidrug resistance. Essential oils with anti-inflammatory and antioxidant activities may also have anticoccidial effects. The present study investigated the efficacy of D-limonene and its nanoemulsion form against Eimeria tenella in chickens. An in vitro study was conducted to evaluate the sporulation inhibitory effects of D-limonene on Eimeria tenella oocysts. Five D-limonene concentrations (0.625, 1.25, 2.5, 5, and 10% v/v) were tested alongside positive (10% formalin) and negative (2.5% potassium dichromate) controls. Each ELISA plate well was inoculated with 1200 unsporulated oocysts and incubated at 30 °C for 24, 48, and 72 h. Subsequently, samples were microscopically examined to assess sporulation inhibition and calculate the percentage of sporulated oocysts. For the in vivo study, 125 eight-day-old broiler chicks were divided into five groups of 25 birds each. The control negative group remained uninfected and untreated. The control positive group was challenged with 5 × 104 sporulated Eimeria tenella oocysts. The diclazuril group received 0.2 mg/kg diclazuril in their diet two days prior to, and until 10 days post infection. The D-limonene (DL) and D-limonene nanoemulsion (DLN) groups were challenged with 5 × 104 sporulated E. tenella oocysts at 18 days of age and administered 150 mg/L of their respective treatments in drinking water from day eight until the end of the experiment. Results from the in vitro study demonstrated that D-limonene suppressed oocyst sporulation by 50.83% at its highest concentration of 10%. In the in vivo study, both DL and DLN treated groups exhibited a significant reduction in oocyst output per gram of feces (OPG), along with increased body weight and decreased parasite stages in the cecal tissue. Furthermore, these treatments were associated with elevated levels of antioxidant enzymes such as glutathione peroxidase (GPX), catalase (CAT), and superoxide dismutase (SOD), accompanied by a decrease in malondialdehyde (MDA) and nitric oxide (NO) levels. Particularly, DLN treatment remarkably increased the number of goblet cells. In conclusion, D-limonene and its nanoemulsion represent promising alternatives for managing coccidiosis in poultry. They not only effectively control parasites but also promote intestinal health and boost antioxidant defenses.
In vivo and in vitro anticoccidial efficacy of Astragalus membranaceus against Eimeria papillata infection

Abstract:Coccidiosis is a parasitic disease of wild and domestic animals caused by Eimeria spp. Currently, several drugs are available for the control of this disease but resistance has been confirmed for all them. There is an urgent need, therefore, for the identification of new compounds as alternative treatments to control coccidiosis. Astragalus membranaceus proven to have anti-inflammatory, anti-oxidant, immunomodulating and anticancer activities. The present study was therefore undertaken to assess the in vivo and the in vitro anticoccidial activity of Astragalus membranaceus root (AMR). Mice were divided into five groups, with the first left non-infected and the second, third, fourth and fifth groups being infected with 1×103 sporulated oocysts of E. papillata. The third, fourth and fifth groups also received, respectively, an oral dose of 10, 25 and 50 mg/kg AMR suspended in physiological saline daily for five consecutive days. 50 mg/kg, was the most effective dose, inducing a significant reduction in the number of oocysts output in mice faeces (by about 57%), accompanied by a significant decrease in the number of parasitic stages in jejunal sections. Moreover, the treatment with AMR increased the number of goblet cells and upregulated the expression of its specific gene, MUC2. In addition, our study proved that AMR reduced oxidative damage since levels of TBARS decreased (indicating reduced lipid peroxidation), levels of glutathione (GSH) and glutathione peroxidase (GPX) increased, and the mRNA level of iNOS was downregulated. Also, AMR treatment revealed anti-apoptotic activity as it was able to regulate the gene expression of Bcl-2 in the jejunum of E. papillata infected mice. Finally, the in vitro study revealed that AMR significantly inhabited the oocyst sporulation in a dose dependent manner. Overall, therefore, our results indicate that AMR exhibits significant in vivo and in vitro anticoccidial effects.

Antimicrobial Efficacy of Cinnamon Essential Oil against Avian Pathogenic Escherichia coli from Poultry

Abstract:Colibacillosis, caused by E. coli, is responsible for economic losses in the poultry industry due to mortality, decreased production, and the cost of antibiotic treatments. Prevention of colibacillosis is based on improved biosecurity measures and the use of the vaccine performed with O78 E. coli strains, which is responsible for most cases of colibacillosis. Recently, there has been increased interest in other infection control methods, such as the use of natural compounds. The aim of this study was to evaluate the antimicrobial efficacy of cinnamon essential oil (CEO) against E. coli strains isolated from poultry. The MIC50 and MIC90 of CEO were determined by testing 117 strains belonging to serogroups O78, O2, O128, O139, isolated from laying hens (91 strains), broilers (10 strains), and turkeys (16 strains). The bacterial strains were tested at cell densities of 108 and 106 CFU/mL. At the cell density of 108 CFU/mL, MIC50 and MIC90 were 0.4 and 0.5 μL/mL for most of the tested strains, while they corresponded to 0.5 μL/mL for all strains isolated from broilers and for strains belonging to serogroup O139. At the cell density of 106 CFU/mL, MIC50 and MIC90 were 0.3 and 0.4 μL/mL, regardless of bird species of origin and for strains belonging to serogroups O78 and O2. In addition, a concentration of 0.04 μL/mL of CEO corresponded both to MIC50 and MIC90 for strains belonging to serogroups O139 and O128. Based on these results, cinnamon essential oil showed an effective antibacterial activity against E. coli strains from poultry and could find field application for the prevention of colibacillosis.

The Effect of Supplementation with Weizmannia coagulans Strain SANK70258 to Coccidia-Infected Broilers Is Similar to That of a Coccidiostat Administration

Abstract:To determine whether it could also improve the production performance of Eimeria-infected broilers, Weizmannia coagulans strain SANK70258 (WC) supplementation was compared with coccidiostat lasalocid-A sodium (AM) administration. First, to determine the optimum WC dose, newly hatched broiler chick groups (n = 10) were untreated or consecutively given WC (0.005%, 0.01%, 0.03%, and 0.1%) and AM until slaughter (31 days of age). At day 21, all chicks were infected with coccidia. From the economical and practical viewpoints, 0.03% WC supplementation was the best dose. Second, newly hatched broiler chick groups (n = 10) were untreated or given 0.03% WC and AM. Each group was run in triplicate. At day 21, two chicks/pen with the farthest body weights as per the group’s mean body weight were spared, and the remaining inoculated with coccidia. At days 42 and 49, the WC and AM groups had significantly greater body weights and daily weight gains. Intestinal lesion scores were lower in 29-day-old AM and WC. Oocyst numbers were lower in 29- and 49-day-old AM and WC, but only 29- and 49-day-old AM had higher Escherichia coli levels. To conclude, although WC and AM induced similar growth performance in coccidium-infected chicks, unlike AM, the E. coli levels did not increase with WC.

Abstract:Background: Coccidiosis is one of the most economically significant poultry diseases worldwide, caused by the pathogenic Eimeria species, and is characterized by decreased weight gain (WG) and failure to grow due to malabsorption, low feed conversion rate, bloody diarrhea, and dehydration. Aim: This study investigated the effectiveness of licorice root extract (LRE) in controlling cecal coccidiosis to determine whether its combination with maduramicin could help alleviate the pathological, biochemical, and histopathological effects of cecal coccidiosis in Sasso broiler chicks. Methods: A total of 125 one-day-old Sasso broiler chicks were categorized into five equal groups (n = 25), each consisting of five replicates (n = 5 per replicate). G1-LE received a basal diet supplemented with LRE (3 g/kg); G2-ME received a basal diet containing maduramycin (0.5 g/kg); and G3-LME received a basal diet containing LRE and maduramicin together with the same rates. G4-E (positive control) and G5-N (negative control) received no additives in their feed. Birds in groups (G1-4) were challenged on day 14 of the experiment by orally intercropping a 1 ml suspension of Eimeria tenella sporulated oocysts. Results: Groups of birds fed on LRE and maduramicin separately or together appeared to be in good condition where no deaths or clinical abnormalities were observed, based on the analysis of clinicopathological examination. Compared with the G4-E positive control, the dropping scoring and oocyst shedding of groups G1-LE, G2-ME, and G3-LME along the 10th-day post-challenge (dpc), as well as macroscopic and microscopic lesions scoring at the 7th dpc, was considerably lower. The dual supplementation use of LRE and maduramicin in G3-LME's reduced the harmful effects of coccidian, which appeared only as a mononuclear cellular infiltration and a small number of oocysts invading the intestinal glands. Molecular docking revealed that LRE and maduramicin interacted with E. tenella DNA polymerase, E. tenella apical membrane antigen 1, and microneme protein binding sites resulting in reduced E. tenella replication and invasion. Conclusion: The inclusion of LRE and maduramicin, individually or in combination, in the diet might effectively mitigate the detrimental effects of coccidiosis.
Antimicrobial Efficacy of Indolicidin Against Multi-Drug Resistant Enteroaggregative Escherichia coli in a Galleria mellonella Model

Abstract:Antimicrobial resistance against enteroaggregative Escherichia coli (EAEC), an emerging food-borne pathogen, has been observed in an increasing trend recently. In the recent wake of antimicrobial resistance, alternate strategies especially, cationic antimicrobial peptides (AMPs) have attracted considerable attention to source antimicrobial technology solutions. This study evaluated the in vitro antimicrobial efficacy of Indolicidin against multi-drug resistant enteroaggregative Escherichia coli (MDR-EAEC) strains and further to assess its in vivo antimicrobial efficacy in Galleria mellonella larval model. The minimum inhibitory concentration (MIC; 32 μM) and minimum bactericidal concentration (MBC; 64 μM) of Indolicidin against MDR-EAEC was determined by micro broth dilution method. Indolicidin was also tested for its stability (high-end temperatures, physiological concentration of salts and proteases); safety (sheep RBCs; HEp-2 and RAW 264.7 cell lines); effect on beneficial microflora (Lactobacillus rhamnosus and Lactobacillus acidophilus) and its mode of action (flow cytometry; nitrocefin and ONPG uptake). In vitro time-kill kinetic assay of MDR-EAEC treated with Indolicidin was performed. Further, survival rate, MDR-EAEC count, melanization rate, hemocyte enumeration, cytotoxicity assay and histopathological examination were carried out in G. mellonella model to assess in vivo antimicrobial efficacy of Indolicidin against MDR-EAEC strains. Indolicidin was tested stable at high temperatures (70°C; 90°C), physiological concentration of cationic salts (NaCl; MgCl2) and proteases, except for trypsin and tested safe with sheep RBCs and cell lines (RAW 264.7; HEp-2) at MIC (1X and 2X); the beneficial flora was not inhibited. Indolicidin exhibited outer membrane permeabilization in a concentration- and time-dependent manner. In vitro time-kill assay revealed concentration-cum-time dependent clearance of MDR-EAEC in Indolicidin-treated groups at 120 min, while, in G. mellonella, the infected group treated with Indolicidin revealed an increased survival rate, immunomodulatory effect, reduced MDR-EAEC counts and were tested safe to the larval cells which was concurred histopathologically. To conclude, the results suggests Indolicidin as an effective antimicrobial candidate against MDR-EAEC and we recommend its further investigation in appropriate animal models (mice/piglets) before its application in the target host.

Lasalocid and monensin in finishing diets for early weaned lambs with naturally occurring coccidiosis

Abstract:Early weaned male crossbred lambs (n = 90) with an average weight of 14 kg were used in 6 groups to evaluate the efficacy of monensin (22 mg/g of feed) and lasalocid (12.5, 25, 50, and 100 mg/kg of feed) on naturally occurring coccidiosis. Average 24-hour oocyst output at the start of the experiment ranged from 5.7 x 10(6) to 156.7 x 10(6) for monensin-treated and 100-mg lasalocid-treated groups, respectively. Monensin and lasalocid were effective against Eimeria ninakohlyakimovae of E ahsata. To day 103, body weight gains were similar for the 4 groups fed lasalocid (336 g/day) and averaged 6% higher (P greater than 0.05) than did gains in control and monensin-treated lambs. Feed efficacy averaged 9% higher (P less than 0.05) for lambs fed 25, 50, and 100 mg of lasalocid/kg of feed and 7% higher (P greater than 0.05) for lambs fed monensin and 12.5 mg of lasalocid/kg of feed. There were no treatment effects on digestibility, although lasalocid tended to increase (P greater than 0.05) crude protein digestibility (values were higher on day 76 than on day 28). Lasalocid and monensin were potent manipulators of rumen fermentation. Increasing the amount of lasalocid in the feed was associated with successive increases (P less than 0.05) in molar proportions of propionic acid and decreases (P less than 0.05) in acetic acid. Lasalocid and monensin controlled naturally occurring coccidiosis and improved performance in early weaned lambs (45 days old) under feedlot conditions.

Abstract:Several studies show significant correlations of blood variables, weight, abnormal electrocardiograms, and blood pressure with the incidence of coronary heart disease, but the quantification of fitness and its correlation with coronary heart disease is more difficult. In this study, approximately 3,000 men, average age of 44.6 years, were examined for blood and lipid profile, pulmonary function, percent body fat determination, and Balke maximal performance treadmill stress testing with multilead ECG monitoring. Five age-adjusted cardiorespiratory fitness categories were determined from treadmill times. A consistent inverse relationship among physical fitness categories and resting heart rate, body weight, percent body fat, serum levels of cholesterol and triglycerides, glucose, and systolic blood pressure, was observed. Forced vital capacity was directly related to levels of fitness. These results, although cross-sectional, imply that physical fitness is related to lower coronary risk factors.

Abstract:Increasing resistance of Eimeria species to anticoccidial medications is an issue in the broiler chicken industry. Using drug-sensitive strains in live-coccidiosis vaccines has been shown to improve anticoccidial effectiveness in US-based broiler production. In Canada, litter is removed between flocks, which differ from the US industry practice. Thus, we investigated the use of drug-sensitive vaccine strains in a Canadian broiler production facility with suspected anticoccidial resistance. Weekly fecal samples were collected from flocks before, during, and after vaccine seeding to determine oocyst shedding patterns; following the vaccine seeding, OPG counts from similar aged birds were lower than flocks before live-coccidiosis vaccine use. Eimeria species isolates, collected before and after vaccine seeding, were used in 2 anticoccidial sensitivity tests to evaluate their susceptibility to commercially available anticoccidial medications; a low-dose challenge to define parasite replication, and a high-dose challenge to monitor broiler performance. In both experiments, isolates collected after seeding were more susceptible to almost every anticoccidial medication evaluated compared with the isolates collected before seeding. These results demonstrate an improvement in sensitivity to many anticoccidials after the use of live-coccidiosis vaccines at this facility. However, the regulated removal of litter at the end of each flock required under Canadian broiler chicken production management rules could limit the establishment of vaccine-strain Eimeria species in broiler facilities and could shorten the longevity of improved drug sensitivity observed in this study.

Abstract:BackgroundCoccidiosis is a prevalent problem in chicken production. Dietary addition of coccidiostats and vaccination are two approaches used to suppress coccidia in the practical production. Methionine (Met) is usually the first limiting amino acid that plays important roles in protein metabolism and immune functions in chickens. The present study is aimed to investigate whether increasing dietary Met levels will improve the anticoccidial effects in broilers medicated or vaccinated against coccidia under Eimeria (E.) tenella-challenged condition. Two thousand male Partridge Shank broiler chicks were obtained from a hatchery. After hatch, birds were weighed, color-marked and allocated equally into two anticoccidial treatments, namely medicated and vaccinated groups. Chicks were either fed, from 1 d of age, diets containing coccidiostat (narasin) or diets without the coccidiostat but were inoculated with an anticoccidial vaccine at 3 d of age. At 22 d of age, 1080 chicks among them were randomly allocated evenly into 6 groups under a 2 × 3 treatment with 2 anticoccidial programs and 3 dietary methionine (Met) levels. Chicks medicated or vaccinated against coccidia were fed diets containing 0.45%, 0.56% or 0.68% of Met from 22 to 42 d of age. All chicks were orally introduced with an amount of 5 × 104 sporulated oocysts of E. tenella at 24 d of age. The growth performance, serum anti-oxidative indexes, intestinal morphology, cecal lesion scores, fecal oocyst counts and immune parameters were measured.ResultsThe results showed increasing dietary Met level from 0.45% to 0.56% and 0.68% improved weight gain and feed conversion of broilers medicated against coccidia. In contrast, higher dietary levels of Met did not improve growth performance of the vaccinated chickens. Higher Met levels helped the medicated chickens resist E. tenella infection, as indicated by improved intestinal morphology and immune functions as well as decreased cecal lesion and fecal oocyst counts.ConclusionsAnticoccidial vaccination is a better strategy for controlling coccidiosis than feeding narasin, due to not only greater growth performance, but also the lower Met supplementation. Furthermore, higher dietary Met levels improved growth performance of chickens medicated rather than vaccinated against coccidia under E. tenella-challenged condition.
