Abstract:Peptide cyclization improves conformational rigidity, providing favorable pharmacological properties, such as proteolytic resistance, target specificity, and membrane permeability. Thus, many synthetic and biosynthetic peptide circularization strategies have been developed. PatG and related natural macrocyclases process diverse peptide sequences, generating millions of cyclic derivatives. However, the application of these cyclases is limited by low yields and the potential presence of unwanted...

What problem does this paper attempt to address?
This paper aims to solve the problems of low yield and potential by - products encountered in the peptide cyclization process. Specifically, the researchers designed a covalently fused G - type macrocyclase. By covalently linking the substrate to the N - terminus of the enzyme, the efficiency, yield and accuracy of peptide cyclization were improved. In addition, they also developed an orthogonal control mechanism to achieve precise synthesis in Escherichia coli. This method makes it possible to construct a library containing 4.8 million cyclic peptide derivatives, among which an estimated 2.6 million different cyclic peptides were produced with higher yield and fidelity. ### Main problems solved by the paper 1. **Increasing the yield of peptide cyclization**: Traditional peptide cyclization methods often have low yields, leading to limited practical applications. In this paper, the yield of peptide cyclization was significantly increased by designing a covalently fused G - type macrocyclase. 2. **Reducing the generation of by - products**: In traditional methods, the generation of unwanted intermediates or by - products is a common problem. In this paper, the generation of these by - products was reduced by optimizing the substrate design and the orthogonal control mechanism. 3. **Increasing the fidelity of peptide cyclization**: In the peptide cyclization process, the selectivity of the substrate and the accuracy of the reaction are key factors. In this paper, the fidelity of peptide cyclization was increased by the covalent fusion strategy and the orthogonal control mechanism, ensuring the purity of the target product. ### Main methods and results - **Covalent fusion strategy**: Covalently link the substrate to the N - terminus of the G - type macrocyclase to form a fusion protein. This design enables the substrate to be more effectively recognized and processed by the enzyme, thereby increasing the cyclization efficiency. - **Orthogonal control mechanism**: By introducing an orthogonal control mechanism, such as using the RSI - TruD enzyme to convert the C - terminal cysteine residue into a thiazole ring, the peptide cyclization reaction was further promoted. - **Large - scale library construction**: Using the above methods, a library containing 4.8 million cyclic peptide derivatives was successfully constructed, and sufficient materials were isolated from it for spectroscopic analysis. ### Conclusion The method proposed in this paper not only significantly increases the yield and fidelity of peptide cyclization, but also provides a simple and efficient method for synthesizing cyclic peptides and their libraries. This provides new tools and ideas for applications in the fields of drug discovery and biotechnology.
