Traceability Evaluation of Wild and Cultivated Cordyceps sinensis by Elemental Analysis and GasBench II Coupled to Stable Isotope Ratio Mass Spectrometry

Abstract:Cordyceps sinensis ( C. sinensis ) is highly prone to food fraud in the Chinese market. This study aimed to evaluate wild and cultivated C. sinensis by elemental analysis coupled to stable isotope ratio mass spectrometry (EA-IRMS) and GasBench II coupled to isotope ratio mass spectrometry (GB-IRMS). The orthogonal partial least-squares discriminant analysis (OPLS-DA) model was established to obtain the various significant markers of stable isotope ratios in both the wild and cultivated C. sinensis samples. The δ 2 H, δ 18 O, and δ 15 N values were identified as the significant stable isotope markers in the fresh C. sinensis samples, which could be potentially used to discriminate wild C. sinensis origin traceability. Significantly, the averages of δ 2 H values of fresh C. sinensis water decreased sequentially based on the different origins. The δ 18 O and δ 15 N values were discriminated between the wild and cultivated C. sinensis samples. Lower δ 18 O values and higher δ 15 N values in the C. sinensis were found in the cultivated samples (18.99 ‰ and 4.80 ‰, respectively), whereas higher δ 18 O values and lower δ 15 N values were observed in the wild samples (25.72 ‰ and 2.29 ‰, respectively). Hence, EA/GB-IRMS coupled with OPLS-DA could be optimal strategy for systematic evaluation of traceability of the wild and cultivated C. sinensis in the future.
