Design of peptide-based foldamers for inhibiting the Hdm2-p53 interaction using backbone modification: a molecular dynamics study

Abstract:Protein, as the main cause of cell interactions, can be the key to solving the riddle of cancer and many other diseases. Disorders in protein–protein interactions (PPIs) are often the main cause of several diseases, making them attractive as drug targets. The Hdm2-p53 interaction is an attractive therapeutic target in oncology. In normal cells, p53 activity levels are controlled by negative regulators such as Human double minute 2 (Hdm2). Overexpression of Hdm2 in cancer cells prevents p53 from exerting its normal function. Disruption of the Hdm2-p53 interaction to maintain p53 function is considered a therapeutic approach to cancer treatment. Peptidomimetics have emerged as promising candidates for the treatment of various diseases by modulating PPIs. Herein, an investigation of three classes of unnatural backbone units in the folding behavior of peptides based on the p53 sequence has been reported. Among the unnatural oligomers in the molecular dynamics (MD) simulation study, helix mimetics containing the C α -Me-Leu residue and α-amino isobutyric acid (Aib) have a higher helical content than the natural p53 peptide. Therefore, they can be suitable inhibitors for Hdm2-P53 interaction.
