Abstract:Litter nutrient dynamics contribute significantly to biogeochemical cycling in forest ecosystems. We examined how site environment and initial substrate quality influence decomposition and nitrogen (N) dynamics of multiple litter types. A 2.5-year decomposition study was installed in the Oregon Coast Range and West Cascades using 15 N-labeled litter from Acer macrophyllum , Picea sitchensis , and Pseudotsuga menziesii . Mass loss for leaf litter was similar between the two sites, while root and twig litter exhibited greater mass loss in the Coast Range. Mass loss was greatest from leaves and roots, and species differences in mass loss were more prominent in the Coast Range. All litter types and species mineralized N early in the decomposition process; only A. macrophyllum leaves exhibited a net N immobilization phase. There were no site differences with respect to litter N dynamics despite differences in site N availability, and litter N mineralization patterns were species-specific. For multiple litter × species combinations, the difference between gross and net N mineralization was significant, and gross mineralization was 7–20 % greater than net mineralization. The mineralization results suggest that initial litter chemistry may be an important driver of litter N dynamics. Our study demonstrates that greater amounts of N are cycling through these systems than may be quantified by only measuring net mineralization and challenges current leaf-based biogeochemical theory regarding patterns of N immobilization and mineralization.
Influence of storage on the characteristics of nitrifying sludge

Abstract:The preservation characteristics of nitrifying sludge under different storage methods were investigated. Under starvation conditions in the natural environment, compared to 4°C, -20°C and -20°C with 15% glycerol, the decay rate of nitrifying activity was 0.012 d-1, 0.021 d-1, 0.019 d-1 and 0.013 d-1, and the half-life was 1.71, 1.09, 1.22 and 1.62 months, respectively. After 5 months' storage, the remaining relative activity and relative biomass were 16.0%, 13.6%, 20.8%, 20.4% and 41.5%, 47.6%, 52.2%, 53.6%, respectively. During storage, the heme c and ATP contents declined, and the SV also decreased with the decrease of activity. When storage time was extended, at low temperature, filamentous bacteria dominated in the sludge, the biomass structure became integrated, and the nitrifying sludge changed to black gradually. Under natural conditions, filamentous bacteria decreased, the sludge structure became loose, and the nitrifying sludge turned black rapidly.
Microbial Biomass Dynamics in Recently Air-Dried and Rewetted Soils Compared to Others Stored Air-Dry for Up to 103 Years

Abstract:Our aim was to compare the soil microbial biomass concentration and its activity (measured as CO2–C evolved) following the rewetting and aerobic incubation of soils which have previously been stored air-dry for different periods. Some of the soils have been stored in the Rothamsted sample archive for 103 years, others were comparable freshly sampled soils following air-drying and rewetting and other soils were stored air-dry for 2 years then rewetted for the work described here. Following air-drying, soil ATP concentrations were variable in recently air-dried soil, comprising about 10–35% of the initial ATP concentrations in fresh soil. Following rewetting, the percentage recovery of ATP increased in all soils by 7 days, then declined to between 73% and 87% of the original ATP concentration in the air-dried soils by day 12. Storage of air-dried soils decreased the ability of the microbial biomass to restore its ATP concentrations. For example, the ATP concentration in a soil sampled from stubbed (i.e. tree seedling, saplings and bushes cut frequently to ground level) grassland of the Broadbalk continuous wheat experiment at Rothamsted then air-dried for 2 years was only about 14% of that in the fresh soil at 2 days after rewetting. In other soils from the Hoosfield Barley Experiment, also at Rothamsted, previously given NPK or FYM since 1852, and sampled then stored air-dry for between 13 and 83 years, from 52% to 57% of the ATP in the comparable fresh soils was measured at two days after rewetting. The soil ATP concentration then changed little more up to 12 days. One of the most interesting findings was that while the microbial biomass ATP concentration in the above NPK soils only ranged from about 2 to 4μmol ATPg−1 biomass C, in the FYM soil the microbial biomass ATP concentrations (range 11.5–13.6μmol ATPg−1 biomass C) were the same as we repeatedly measure in fresh moist aerobic soil. We do not yet know the reasons for this. More than twice as much CO2–C was evolved from the long-term stored soils than from freshly sampled ones. However, the specific respiration of the microbial biomass did not change much after the first 12 years of storage, indicating that loss of viability mainly occurred in the earlier years.
Influence of Storage Methods on the Vitality and Growth Rate of Macrofungi

Abstract:The work contains a comparative analysis of methods for storing pure cultures of macrofungi. The study used 20 species of macrofungi from various taxonomic and ecological-trophic groups. Storage was carried out using five methods: serial subculturing, storage under a layer of distilled water and three cryopreservation protocols: a protocol using blocks of agar medium, a "perlite protocol," and a "grain protocol." For the selected cryostorage methods, various cryoprotective compounds (glycerol, trehalose) were used. Radial growth rate was used as a criterion for the state of crops. The values of the radial growth rate obtained immediately after isolation of the pure culture were chosen as the control. It has been shown that the most favorable for preserving the physiological activity of cultures are the storage method under a layer of distilled water, "perlite" and "grain" cryopreservation protocols.

Abstract:In model experiments under forest ecosystem conditions, samples of partially decomposed forest litter and soil from a 0–10 cm layer containing 14C-labeled organic matter were selected. The label was introduced in the form of low molecular weight water-soluble organic substances of individual nature – glycine, uracil and glucose together with unlabelled plant litter in nylon bags. The experiment was carried out in the late autumn period on the territory of the Malinsky forestry, 20 km south-west of Moscow, on a medium loamy sod-podzolic soil. For the study, samples were taken from two selection periods – 40 days after labelling and after two years. The selected samples were extracted and separated into soil organic matter fractions containing 14C in their composition, followed by radiometry. The results showed differences in the transformation and incorporation of radiocarbon of the studied products – glycine, uracil, and glucose into different fractions of soil organic matter of low molecular weight water-soluble substances, depending on their nature.

Abstract:Biological and chemical stabilization of organic C was assessed in soils sampled from the long-term experiments at Rothamsted (UK), representing a wide range of carbon inputs and managements by extracting labile, non-humified organic matter (NH) and humic substances (HS). Four sequentially extracted humic substances fractions of soil organic matter (SOM) were extracted and characterized before and after a 215-day laboratory incubation at 25 degrees C from two arable soils, a woodland soil and all occasionally stubbed soil. The fractions corresponded to biochemically stabilised SOM extracted in 0.5 M NaOH (free fulvic acids (FA) and humic acids (HA)) and chemically plus biochemically stabilised SOM extracted from the residue with 0.1 M Na4P2O7 Plus 0.1 M NaOH (bound FA and HA). Our aim was to investigate the effects of chemical and biochemical stabilization on carbon sequestration. The non-humic to humic (NH/H) C ratio separated the soils into two distinct groups: arable soils (unless fertilised with farmyard manure) had an NH/H C ratio between 1.05 and 0.71, about twice that of the other soils (0.51-0.26). During incubation a slow, but detectable, decrease in the NH/H C ratio occurred in soils of C input equivalent or lower to 4 Mg ha(-1) y(-1), whereas the ratio remained practically constant in the other soils. Before incubation the free to bound humic C ratio increased linearly (R-2 = 0.91) with C inputs in the soils from the Broadbalk experiment and decreased during incubation, showing that biochemical stabilization is less effective than chemical stabilization in preserving humic C. Changes in delta C-13 and delta N-15 after incubation were confined to the free FA fractions. The delta C-13 of free FA increased by 1.48 and 0.80 parts per thousand, respectively, in the stubbed and woodland soils, indicating a progressive biological transformation. On the contrary, a decrease was observed for the bound FA of both soils. Concomitantly, a Delta delta N-15 of up to +3.52 parts per thousand was measured after incubation in the free FA fraction and a -2.58 Delta delta N-15 in the bound FA. These changes, which occurred during soil incubation in the absence of C inputs, indicate that free FA fractions were utilised by soil microorganisms, and bound FA were decomposed and replaced, in part, by newly synthesized FA.The (13)CPMAS-TOSS NMR spectra of free HA extracted before and after 215 days of incubation were mostly unchanged. Litter Decomposition and Soluble Carbon, Nitrogen, and Phosphorus Release in a Forest Ecosystem

Abstract:Litter is an important source of easily mineralizable C, N, and P for microbial metabolism in forest ecosystems; however, its decomposition is dependent upon a variety of biotic and abiotic factors, including litter chemical composition and plant specie, soil properties, and climate. We investigated C, N, and P mineralization patterns of pine litter, oak and a mixture of various species commonly found in wetland landscape position. Litter species were incubated (alone and with soils) under laboratory conditions in the dark for 120 days. Samples were leached weekly and the leachates were analyzed for pH, E4:E6 ratio, dissolved organic carbon (DOC), total N, NO3, NH4, soluble reactive P, and total P. CO2 effluxes during the 120-d incubation period were measure using NaOH traps. Carbon loss was calculated as the sum of DOC and CO2 effluxes. Results indicated that patterns of C and N release varied with litter species and soil type. Mix species treatment resulted in larger DOC and N pulses compared to pine and oak treatments. The majority of the DOC, N, and P leached was retained by the soils. When litters were added to the soils, a greater proportion of the C was lost as CO2, while litter incubated alone lost more C as DOC. This result demonstrated the importance of the soil microbial community affecting the patterns of litter mineralization. Total N concentration and C:N ratio of the litter species were significantly correlated to C loss.
Impacts of sample handling and storage conditions on archiving physiologically active soil microbial communities

Abstract:Soil microbial communities are fundamental to ecosystem processes and plant growth, yet community composition is seasonally and successionally dynamic, which interferes with long-term iterative experimentation of plant-microbe interactions. We explore how soil sample handling (e.g. filtering) and sample storage conditions impact the ability to revive the original, physiologically active, soil microbial community. We obtained soil from agricultural fields in Montana and Oklahoma, USA and samples were sieved to 2 mm or filtered to 45 µm. Sieved and filtered soil samples were archived at -20°C or -80°C for 50 days and revived for 2 or 7 days. We extracted DNA and the more transient RNA pools from control and treatment samples and characterized microbial communities using 16S amplicon sequencing. Filtration and storage treatments significantly altered soil microbial communities, impacting both species richness and community composition. Storing sieved soil at -20°C did not alter species richness and resulted in the least disruption to the microbial community composition in comparison to nonarchived controls as characterized by RNA pools from soils of both sites. Filtration significantly altered composition but not species richness. Archiving sieved soil at -20°C could allow for long-term and repeated experimentation on preserved physiologically active microbial communities.
The effect of litter quality and soil faunal composition on organic matter dynamics in post-mining soil: A laboratory study

Abstract:Laboratory microcosms were constructed consisting of mineral soil (alkaline clay spoil substrate from brown-coal mine tailings) and two types of litter: (1) taken from an unreclaimed site with naturally developed vegetation (mostly Salix caprea) or (2) collected from an alder plantation (a mixture of Alnus glutinosa and A. incana). Microcosms with both kinds of litter were inoculated with five different types of decomposer community: (1) microorganisms-only, (2) microorganisms+mesofauna, (3) microorganisms+mesofauna+litter feeding macro-arthropods (millipedes and diptern larvae), (4) microorganisms+mesofauna+litter dwelling earthworms (Dendrobaena octaedra and Dendrodrilus rubidus) and (5) microorganisms+mesofauna+Lumbricus rubellus. Microorganisms and animals used for microcosms inoculation were collected in the same sites as the litter used in the microcosms. Four replicates for each of 10 treatments were incubated in the dark at 15°C for 90 days.The presence of macrofauna resulted in more extensive litter removal from the litter layer in microcosms derived from the reclaimed site, while in microcosms derived from the unreclaimed site, litter removal increased significantly only in the earthworm treatment. Litter removal was correlated with the accumulation of carbon in the mineral layer of the unreclaimed site microcosms (r=0.802), while for the reclaimed site material it was correlated with C loss from the microcosms (r=0.704). Removal of litter from the litter layer and accumulation of C and N in the mineral layer increased microbial respiration and biomass measured as total PLFA.

Evaluation of Sample Preservation Methods for Poultry Manure

Abstract:When poultry manure is collected but cannot be analyzed immediately, a method for storing the manure is needed to ensure accurate subsequent analyses. This study has 3 objectives: (1) to investigate effects of 4 poultry manure sample preservation methods (refrigeration, freezing, acidification, and freeze-drying) on the compositional characteristics of poultry manure; (2) to determine compositional differences in fresh manure with manure samples at 1, 2, and 3 d of accumulation under bird cages; and (3) to assess the influence of 14-d freezing storage on the composition of manure when later exposed to 25 degrees C for 7 d as compared with fresh manure. All manure samples were collected from a layer house. Analyses performed on the manure samples included total Kjeldahl nitrogen, uric acid nitrogen, ammonia nitrogen, and urea nitrogen. In experiment 1, the storage methods most similar to fresh manure, in order of preference, were freezing, freeze-drying, acidification, and refrigeration. Thoroughly mixing manure samples and compressing them to 2 to 3 mm is important for the freezing and freeze-dried samples. In general, refrigeration was found unacceptable for nitrogen analyses. A significant effect (P < 0.0001) of time for refrigeration was found on uric acid nitrogen and ammonia nitrogen. In experiment 2, the total Kjeldahl nitrogen and uric acid nitrogen were significantly lower (P < 0.05) for 1, 2, and 3 d of accumulation compared with fresh manure. Manure after 1, 2, and 3 d of accumulation had similar nitrogen compositions. The results from experiment 3 show that nitrogen components from fresh manure samples and thawed samples from 14 d of freezing are similar at 7 d but high variability of nitrogen compositions during intermediate times from 0 to 7 d prevents the recommendation of freezing manure for use in subsequent experiments and warrants future experimentation. In conclusion, fresh poultry manure can be frozen for accurate subsequent nitrogen compositional analyses but this same frozen manure may not be a reliable substitute for fresh manure if a subsequent experiment is performed.
Changes in chemical properties and microbial communities' composition of a forest litter-based biofertilizer produced through aerated solid-state culture under different oxygen conditions

Abstract:Fermented forest litter (FFL) is a bioproduct used as biofertilizer for several decades in Eastern Asia and Latin America. It is locally handcrafted by farmers in anaerobic conditions by fermenting forest litter added with agricultural by-products such as whey, cereal bran, and molasses. The aim of this study was to characterize the FFL process and product through gas and liquid chromatography analyses. It also provides some highlights on the influence of O2 on this solid-state culture. Under anoxic condition, a maximum CO2 production rate (CDPR) of 0.41 mL/h∙g dry matter (dm) was reached after 8 days. The main volatile organic compounds (VOCs) were ethanol and ethyl acetate, with a production rate profile similar to CDPR. After 21 days of culture, no residual sucrose nor lactose was detected. Lactic and acetic acids reached 58.8 mg/g dm and 10.2 mg/g dm, respectively, ensuring the acidification of the matrix to a final pH of 4.72. A metabarcoding analysis revealed that heterolactic acid bacteria (Lentilactobacillus, Leuconostoc), homolactic acid bacteria (Lactococcus), and yeasts (Saccharomyces, Clavispora) were predominant. Predicted genes in the microbiome confirmed the potential link between detected bacteria and acids and VOCs produced. When O2 was fed to the cultures, final pH reached values up to 8.5. No significant amounts of lactic nor acetic acid were found. In addition, a strong shift in microbial communities was observed, with a predominance of Proteobacteria and molds, among which are potential pathogens like Fusarium species. This suggests that particular care must be brought to maintain anoxic conditions throughout the process.
Changes in litter and nitrogen deposition differentially alters forest soil organic matter biogeochemistry

Abstract:Global climate change has altered forest productivity across the globe. Although extreme drought and temperature have lowered forest productivity in some regions, increased productivity has been observed in many forests over the last few decades due to increases in atmospheric carbon dioxide, temperature, and nitrogen (N) deposition. These factors can lead to changes in both the quantity and quality of litterfall and root inputs to soil. Additionally, few studies have investigated multifactorial treatments (e.g., detrital changes and N deposition), on soil Carbon (C) sequestration. To investigate the long-term compositional changes to soil organic matter (SOM) in response to litter and N inputs, soil samples were collected from the University of Michigan Biological Station (UMBS) Detrital Input and Removal Treatment (DIRT) site after 15 years of experimental treatments. The samples were characterized using elemental analysis, targeted SOM compound analyses, nuclear magnetic resonance spectroscopy and microbial biomass and community composition measurements. The exclusions of C inputs (litter and/or roots) resulted in molecular-level biogeochemical changes, however, the soils at UMBS are seemingly more resistant to losses in soil C compared to other DIRT studies. Although the addition treatments (Double Litter, Double Wood, Double Litter + N, and N) led to increased soil C concentrations at UMBS, we found evidence for enhanced SOM decomposition occurring with litter additions. Notably, the observations made from the concurrent treatment of Double Litter + N demonstrated that the responses of individual treatments are not representative of simultaneous applications. Collectively, these results demonstrate that soil C biogeochemistry is sensitive to fluctuations in C and N deposition and overall, these processes are dictated by site-specific ecological properties.

Abstract:Abstract The availability of bioresources is a precondition for life science research, medical applications, and diagnostics, but requires a dedicated quality management to guarantee reliable and safe storage. Anecdotal reports of bacterial isolates and sample contamination indicate that organisms may persist in liquid nitrogen (LN) storage tanks. To evaluate the safety status of cryocollections, we systematically screened organisms in the LN phase and in ice layers covering inner surfaces of storage tanks maintained in different biobanking facilities. We applied a culture-independent approach combining cell detection by epifluorescence microscopy with the amplification of group-specific marker genes and high-throughput sequencing of bacterial ribosomal genes. In the LN phase, neither cells nor bacterial 16S rRNA gene copy numbers were detectable (detection limit, 10 2 cells per ml, 10 3 gene copies per ml). In several cases, small numbers of bacteria of up to 10 4 cells per ml and up to 10 6 gene copies per ml, as well as Mycoplasma , or fungi were detected in the ice phase formed underneath the lids or accumulated at the bottom. The bacteria most likely originated from the stored materials themselves ( Elizabethingia , Janthibacterium ), the technical environment ( Pseudomonas , Acinetobacter , Methylobacterium ), or the human microbiome ( Bacteroides , Streptococcus , Staphylococcus ). In single cases, bacteria, Mycoplasma , fungi, and human cells were detected in the debris at the bottom of the storage tanks. In conclusion, the limited microbial load of the ice phase and in the debris of storage tanks can be effectively avoided by minimizing ice formation and by employing hermetically sealed sample containers.

Litter Decomposition and Nutrient Dynamics in Fire-Affected Larch Forests in the Russian Far East

Abstract:Russian boreal forests hold a considerable carbon (C) stock and are subjected to frequent surface fires that unbalance C storage and ecosystem function. Although postfire ecological changes aboveground are well understood, biological C flows (e.g., decomposition in the postfire period) remain unclear. We present the results of a long-term field litterbag experiment on needle litter decomposition in typical Larix gmelinii boreal forests in the Russian Far East. For 3 years, we measured mass loss, C and nitrogen (N) concentrations, lignin and manganese dynamics, respiration intensity and enzyme activity in decaying needles, and environmental conditions (temperature and litter moisture). The decomposition rate at 850 days was 0.435 and 0.213 yr−1 in a control forest and in a forest 15 years after a surface fire, respectively. Early stages of needle decay did not differ among sites, whereas decomposition slowed in later stages in burned forest relative to the control (p < 0.01). This was supported by hampered respiration, slow lignin accumulation in decaying needles, and low peroxidase activity in burned forest. We found no direct N release, and decaying litter immobilization was more pronounced in the control forest. In the later stages, we revealed restrained mass loss and associated C release from larch litter in burned forest. Slow and delayed N release may alter organic matter accumulation, the N cycle, and regeneration of the fire-disturbed larch ecosystem. Our investigations highlight hampered C flow from aboveground litter to soil humus even decades after surface fire in a larch ecosystem. Given the climate-induced increase of fire activity, C retained in the litter layer represents a pool that is more vulnerable to the next fire event.

Abstract:Climate and litter chemistry are major factors influencing litter decay, a process mediated by microbes, such as fungi, nitrogen-fixing bacteria and ammonia-oxidizing bacteria. Increasing atmospheric CO2 concentrations can decrease nitrogen (N) and increase condensed tannin (CT) content in foliar litter, reducing litter quality and slowing decomposition. We hypothesized that reduced litter quality inhibits microbes and is the mechanism causing decomposition to slow. Litterbags of Douglas-fir needles and poplar leaves with a range of N (0.61-1.57%) and CT (2.1-29.1%) treatment and natural acid unhydrolyzable residue (35.3-41.5%) concentrations were placed along climatic gradients in mature Douglas-fir stands of coastal British Columbia rainshadow forests. The structure (diversity, richness and evenness) and composition of microbial communities were analyzed using DGGE profiles of 18S, NifH-universal and AmoA PCR amplicons in foliar litter after 7, 12, 24 and 43 months of decay. High CT and low N concentrations in leaf litter were associated with changes in microbial community composition, especially fungi. Contrary to our hypothesis, high CT and low N treatments did not inhibit microbial colonization or diversity. The joint effects of air temperature and soil moisture on microbial community composition at our sites were more important than the effects of initial litter chemistry.

Abstract:A significant amount of logging residue is produced during roundwood harvesting. Logs are often left in forest sites due to, for example, ecological or logistical aspects. Taking into account the fact that the number of studies focused on changes in the properties of the residue is very low, it was decided to conduct research on the effect of a three-year storage period in forest stands on the chemical properties and energy potential of the wood. The research design allowed for the determination of changes during up to three years of storage. The performed analysis showed a highly negative impact on the characteristics of the material. These adverse effects were probably caused by the activity of microorganisms such as fungi and bacteria, as evidenced by the increased concentrations of ergosterol and low-molecular-weight organic acids. Moreover, it was found that wood stored for three years was characterized by a lower cellulose content; an increased percentage of lignin; a reduced content of sterols (desmosterol, stigmasterol, lanosterol and β-sitosterol) and phenolic acids; and reduced antioxidant activity, as determined with the ABTS+ method. Storing logs also led to a reduction in the energy potential of the residues, as shown by a reduction in net and gross calorific value and an increased ash content.

Abstract:Cookstove emissions are a significant source of indoor air pollution in developing countries and rural communities world-wide. Considering that many research sites for evaluating cookstove emissions and interventions are remote and require potentially lengthy periods of particulate matter (PM) filter sample storage in sub-optimal conditions (e.g., lack of cold storage), an important question is whether samples collected in the field are stable over time. To investigate this, red oak was burned in a natural-draft stove, and fine PM (PM2.5) was collected on polytetrafluoroethylene filters. Filters were stored at either ambient temperature or more optimal conditions (-20 degrees C or -80 degrees C) for up to 3 months and extracted. The effects of storage temperature and length on stability were evaluated for measurements of extractable organic matter (EOM), PM2.5, and polycyclic aromatic compound (PAC) levels in the filter extracts. A parallel, controlled laboratory condition was also evaluated to further explore sources of variability. In general, PM2.5 and EOM in both simulated field and laboratory samples were similar regardless of the storage condition or duration. The extracts were also analyzed by gas chromatography to quantify 22 PACs and determine similarities and/or differences between the conditions. PAC levels were a more sensitive stability measure in differentiating between storage conditions. The findings suggest that measurements are relatively consistent across storage duration/temperatures for filter samples with relatively low EOM levels. This study aims to inform protocols and filter storage procedures for exposure and intervention research conducted in low- and middle-income countries where studies may be budget- and infrastructure-limited.
Contrasting Effects of Forest Type and Stand Age on Soil Microbial Activities: An Analysis of Local Scale Variability

Abstract:Understanding the functioning of different forest ecosystems is important due to their key role in strategies for climate change mitigation, especially through soil C sequestration. In controlled laboratory conditions, we conducted a preliminary study on six different forest soils (two coniferous, two deciduous, and two mixed sites comprising trees of different ages) collected from the same region. The aim was to explore any differences and assess seasonal changes in soil microbial parameters (basal respiration BR, microbial biomass Cmic, metabolic quotient qCO2, dehydrogenase activity DHA, and Cmic:Corg ratio). Indicator- and forest-specific seasonality was assessed. In addition to litter input, soil parameters (pH, nutrient content, texture and moisture) strongly regulated the analyzed microbial indicators. PCA analysis indicated similarity between mature mixed and deciduous forests. Among annual mean values, high Cmic and DHA with simultaneously low qCO2 suggest that the mature deciduous stand was the most sustainable in microbial activities among the investigated forest soils. Research on the interrelationship between soil parameters and forest types with different tree ages needs to be continued and extended to analyze a greater number of forest and soil types.

Transport of Carbon and Nitrogen Between Litter and Soil Organic Matter in a Northern Hardwood Forest

Abstract:We used sugar maple litter double-labeled with 13C and 15N to quantify fluxes of carbon (C) and nitrogen (N) between litter and soil in a northern hardwood forest and the retention of litter C and N in soil. Two cohorts of litter were compared, one in which the label was preferentially incorporated into non-structural tissue and the other structural tissue. Loss of 13C from this litter generally followed dry mass and total C loss whereas loss of 15N (20–30% in 1 year) was accompanied by large increases of total N content of this decaying litter (26–32%). Enrichment of 13C and 15N was detected in soil down to 10–15 cm depth. After 6 months of decay (November–May) 36–43% of the 13C released from the litter was recovered in the soil, with no differences between the structural and non-structural labeled litter. By October the percentage recovery of litter 13C in soil was much lower (16%). The C released from litter and remaining in soil organic matter (SOM) after 1 year represented over 30 g C m−2 y−1 of SOM accumulation. Recovery of litter 15N in soil was much higher than for C (over 90%) and in May 15N was mostly in organic horizons whereas by October it was mostly in 0–10 cm mineral soil. A small proportion of this N was recovered as inorganic N (2–6%). Recovery of 15N in microbial biomass was higher in May (13–15%) than in October (about 5%). The C:N ratio of the SOM and microbial biomass derived from the labeled litter was much higher for the structural than the non-structural litter and for the forest floor than mineral SOM, illustrating the interactive role of substrates and microbial activity in regulating the C:N stoichiometry of forest SOM formation. These results for a forest ecosystem long exposed to chronically high atmospheric N deposition (ca. 10 kg N ha−1 y−1) suggest possible mechanisms of N retention in soil: increased organic N leaching from fresh litter and reduced fungal transport of N from soil to decaying litter may promote N stabilization in mineral SOM even at a relatively low C:N ratio.
