High-performance thin-layer chromatography method development and validation for quantification of naringin in different extracts of Citrus sinensis L. and its antioxidant activity

Abstract:Citrus sinensis (Linn.), also known as sweet orange, is a conventional culinary herb and has been used in different indigenous medicines, especially in Ayurveda owing to its multifaceted pharmacological profile. A simple, specific, accurate and precise high-performance thin-layer chromatography (HPTLC) method for the quantitative determination and validation of naringin in different extracts of C. sinensis was developed for the first time. The mobile phase of glacial acetic acid‒chloroform‒water‒methanol (1:3:0.1:0.5, V/V ) was used for attaining good separation. Densitometric determination was carried out at 296 nm. The calibration curves were reckoned to be linear in the range between 400 and 2000 ng per spot. During the analysis, the ethanolic extract of C. sinensis showed higher content of naringin than dimethyl sulphoxide (DMSO), acetone, and dimethylformamide (DMF) extracts. Further, the antioxidant potential of different extracts of C. sinensis were assessed with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. The developed method of HPTLC was validated for specificity, linearity, accuracy, and precision. The ethanolic extract has unveiled significant antioxidant activity with a percentage inhibition of 70.45%.
