IL-11 increases levels and activates circulating fibrocytes in different pulmonary hypertension animal models.

Abstract:Fibrocytes are described as circulating bone-marrow-derived cells with the ability to adapt to myofibroblast-like phenotype contributing to pulmonary artery remodeling and pulmonary hypertension (PH). Recent works indicate that the IL-11 system promotes lung fibrosis and PH, but the mechanism is poorly understood. The aim of this study is to analyze the role of IL-11 in fibrocytes in vitro and in vivo. Human fibrocytes were isolated from whole blood and stimulated with IL-11, sIL-11Rα or its combination. Fibrocyte-to-myofibroblast transition and fibrocyte adhesion to pulmonary artery endothelial cells (HPAECs) were assessed. The contribution of IL-11 to circulating fibrocytes was evaluated in three animal models. Transgenic Tie2-GFP mice (Tie2-GFP) were subjected to daily injections of rmIL-11. Sprague-Dawley rats and Tie2-GFP mice received an installation of monocrotaline or bleomycin respectively. IL-11 siRNA or non-targeting siRNA were administered to the rats and mice. The circulating fibrocytes were measured by flow cytometry. IL-11 and the combination with sIL-11Rα induce the transformation of circulating fibrocytes to myofibroblasts and participate in their adhesion to the HPAECs. Moreover, IL-11 mice infusion increased the number of circulating fibrocytes with parallel increased levels of CXCL12. In rats or mice treated with monocrotaline or bleomycin, there was an increase in the number of circulating fibrocytes and the levels of CXCL12. Animals transfected with siRNA-IL-11 decreased circulating fibrocytes in both animal models. In conclusion, IL-11 promotes an increase of circulating fibrocytes, their adhesion to HPAECs and phenotypic switching to myofibroblast.
