Abstract:Understanding the molecular mechanisms influencing barley grain quality is essential to develop varieties with optimal requirements for the food, feed, brewing and distilling industries. Over the years, genetic technologies such as QTL, association analyses and gene mapping allowed the identification of many loci influencing barley grain quality aspects. More recently, the development and optimisation of CRISPR/Cas gene editing technology for targeted mutagenesis has transformed genetic manipulation of crop species, including barley. The relative assembly simplicity of CRISPR/Cas components and higher targeting efficiency compared to other nucleases are key factors determining the widespread use and success of this technology. In this review, we describe studies that have applied CRISPR/Cas9 technology to improve barley grain quality and how recent advances in plant expression systems, delivery of CRISPR/Cas components and other molecular tools are contributing to the expansion of this gene editing toolbox.
