Abstract:For more than three decades, the field of bioelectrochemistry has provided novel insights into the catalytic mechanisms of enzymes, the principles that govern biological electron transfer, and has elucidated the basic principles for bioelectrocatalytic systems. Progress in biochemistry, bionanotechnology, and our ever increasing ability to control the chemistry and structure of electrode surfaces has enabled the study of ever more complex systems with bioelectrochemistry. This feature article highlights developments over the last decade, where supramolecular approaches have been employed to develop electrode assemblies that increase enzyme loading on the electrode or create more biocompatible environments for membrane enzymes. Two approaches are particularly highlighted: the use of layer-by-layer assembly, and the modification of electrodes with planar lipid membranes.

Abstract:Since the first demonstration of synthetic host/guest chemistry, the area of supramolecular chemistry has rapidly expanded and become highly influential in the areas of biomedicine, energy materials, catalysis and sensing. In recent years, the use of host/guest chemistry in combination with functional inorganic nanoparticles has resulted in many new applications such as drug delivery, bioimaging and sensing. In this review, we have summarized some of the recent progress that has been made towards the development of organic/inorganic supramolecular nano-systems, which incorporate host/guest chemistry. Initially, we introduce the common receptors and guest molecules that are used in this area, then we discuss the strategies that have been used to construct the supramolecular-based nano-systems using these receptors and guest molecules (host/guest chemistry). With the foundations to these systems discussed, we then highlight the applications of these unique supramolecular systems in the fields of drug delivery, biological imaging, and sensing. Overall, this review is intended to provide a guide on how to combine functional nanomaterials with host/guest chemistry to form supramolecular-based constructs for biological applications. We hope that reading this review will encourage others to explore this rich and exciting area and develop more new systems that could be important to the fields of supramolecular chemistry, materials science, and biomedicine.

Abstract:CONSPECTUS: Host-guest chemistry can greatly improve the selectivity of biomolecule-ligand binding on account of recognition-directed interactions. In addition, functional structures and the actuation of supramolecular assemblies in molecular systems can be controlled efficiently through various host-guest chemistry. Together, these highly selective, strong yet dynamic interactions can be exploited as an alternative methodology for applications in the field of programmable and controllable engineering of supramolecular soft materials through the reversible binding between complementary components. Many processes in living systems such as biotransformation, transportation of matter, and energy transduction begin with interfacial molecular recognition, which is greatly influenced by various external stimuli at biointerfaces. Detailed investigations about the molecular recognition at interfaces can result in a better understanding of life science, and further guide us in developing new biomaterials and medicines. In order to mimic complicated molecular-recognition systems observed in nature that adapt to changes in their environment, combining host-guest chemistry and surface science is critical for fabricating the next generation of multifunctional biointerfaces with efficient stimuli-responsiveness and good biocompatibility. In this Account, we will summarize some recent progress on multifunctional stimuli-responsive biointerfaces and biosurfaces fabricated by cyclodextrin- or cucurbituril-based host-guest chemistry and highlight their potential applications including drug delivery, bioelectrocatalysis, and reversible adsorption and resistance of peptides, proteins, and cells. In addition, these biointerfaces and biosurfaces demonstrate efficient response toward various external stimuli, such as UV light, pH, redox chemistry, and competitive guests. All of these external stimuli can aid in mimicking the biological stimuli evident in complex biological environments. We begin by reviewing the current state of stimuli-responsive supramolecular assemblies formed by host-guest interactions, discussing how to transfer host-guest chemistry from solution onto surfaces required for fabricating multifunctional biosurfaces and biointerfaces. Then, we present different stimuli-responsive biosurfaces and biointerfaces, which have been prepared through a combination of cyclodextrin- or cucurbituril-based host-guest chemistry and various surface technologies such as self-assembled monolayers or layer-by-layer assembly. Moreover, we discuss the applications of these biointerfaces and biosurfaces in the fields of drug release, reversible adsorption and release of some organic molecules, peptides, proteins, and cells, and photoswitchable bioelectrocatalysis. In addition, we summarize the merits and current limitations of these methods for fabricating multifunctional stimuli-responsive biointerfaces in a dynamic noncovalent manner. Finally, we present possible strategies for future designs of stimuli-responsive multifunctional biointerfaces and biosurfaces by combining host-guest chemistry with surface science, which will lead to further critical development of supramolecular chemistry at interfaces.

Abstract:Supramolecular chemistry has emerged as an important technique for the formation of biomaterials, including nano- and microparticles and hydrogels. One specific class of supramolecular chemistry is the direct association of guest-host pairs, which involves host macrocycles such as cyclodextrins and cucurbit[n]urils and a wide range of guest molecules, where association is typically driven by molecule size and hydrophobicity. These systems are of particular interest in the biomedical field due to their dynamic nature, chemical diversity, relative ease of synthesis, and ability to interact with biological or synthetic molecules. In this review, we discuss aspects of polymeric material assembly mediated by guest-host interactions, including the fundamentals of assembly into functional biomedical materials. Additionally, applications of biomaterials that utilize guest-host interactions are discussed with a focus on injectable material formulations, the sequestration and delivery of encapsulated cargo (i.e., drugs, biomolecules), and the investigation of cell-material interactions (i.e., adhesion, differentiation, and delivery). While methodologies for guest-host mediated assembly and biological interaction have rapidly evolved in recent years, they remain far from realizing their full potential in the biomaterials field.

Immobilization of molecular catalysts on electrode surfaces using host-guest interactions

Abstract:Anchoring molecular catalysts on electrode surfaces combines the high selectivity and activity of molecular systems with the practicality of heterogeneous systems. Molecular catalysts, however, are far less stable than traditional heterogeneous electrocatalysts, and therefore a method to easily replace anchored molecular catalysts that have degraded could make such electrosynthetic systems more attractive. Here we applied a non-covalent 'click' chemistry approach to reversibly bind molecular electrocatalysts to electrode surfaces through host-guest complexation with surface-anchored cyclodextrins. The host-guest interaction is remarkably strong and enables the flow of electrons between the electrode and the guest catalyst. Electrosynthesis in both organic and aqueous media was demonstrated on metal oxide electrodes, with stability on the order of hours. The catalytic surfaces can be recycled by controlled release of the guest from the host cavities and the readsorption of fresh guest.

Abstract:Supramolecular chemistry is based on intermolecular bonds, where substances dynamically bind together through noncovalent interactions. These dynamic forces allow the macrocyclic molecules and guest molecules to form stable assemblies, with high stability under physiological conditions, making them suitable for in vivo drug delivery. These dynamic noncovalent bonds are easily influenced by external stimuli such as light, heat, pH, and oxidation; thus, the assemblies induced by supramolecular interactions exhibit high diversity and flexibility in response to external stimuli, providing an effective method for simulating natural and physiological processes. The host-guest interactions induced self-assemblies have been applied across multiple dimensions, ranging from the molecular level to the cellular level, for detoxification, targeted drug delivery, and therapeutic studies. At the molecular level, macrocyclic molecules can encapsulate toxic substances from the bloodstream, serving as a solution for emergency detoxification. At the nanoscale level, host-guest interactions can induce the formation of multiple nanostructures including nanomicelles, nanocapsules, nanovesicles, and nanoparticles. The host-guest interactions can enhance the stability of nanostructures and impart them with stimuli sensitivity, which is highly significant in specific microenvironments like tumors. Nanostructures induced by the host-guest interactions possess optimized drug release profiles and pharmacokinetic features, thereby enhancing the therapeutic efficacy while mitigating side effects. At the microscale level, the host-guest interactions can induce the formation of various microassemblies including hydrogels, microfibers, and microtube aggregates. Moreover, microassemblies show superior potential in morphology transformation for controlling cell activity and diseases. Additionally, at the level of biological components, host-guest interactions can induce the assembly of peptides and organelles within cells and having the cell-cell or cell-particle assemblies as hitchhikers at the cellular level. Therefore, this Account aims to summarize the applications of host-guest interactions induced self-assemblies at various levels and the latest research in supramolecular self-assembly, with a particular focus on the progress in our research group. We hope that this account not only reveals the applications of therapeutic supramolecular self-assemblies but also provides new insights into the design of smart drug delivery systems.

Abstract:Supramolecular polymers constructed by orthogonal self-assembly based on host-guest and metal-ligand interactions are attracting increasing attention currently because of their interesting properties and potential applications. Host-guest interactions impart these polymers with good selectivity and convenient enviro-responsiveness, and metal-ligand interactions endow them with various coordination geometries, strong yet tunable coordination binding abilities, as well as magnetic, redox, photophysical, and electrochromic properties. Therefore, supramolecular polymers constructed by orthogonal host-guest and metal-ligand interactions have wide applications in the fields of soft matter, fluorescence sensing, heterocatalysis, electronics, gas storage, etc. In this critical review, we will address the recent development of supramolecular polymeric systems involving metal-ligand interactions and host-guest molecular recognition. Specifically, we classify the related supramolecular polymers depending on the types of macrocyclic hosts, and highlight their intriguing properties originating from the elegant combination of host-guest complexation and metal centers.

Abstract:Electrochemical stimuli have attracted much attention in recent years as they are simple, clean and can be widely applied in biological systems and material science. As one type of common guest molecules, ferrocene and its derivatives have been well studied with different host molecules, mainly including cyclodextrins, cucurbiturils, pillararenes and calixarenes. This article generally summarizes the recent work on the host-guest interactions between ferrocene derivatives and their host molecules, as well as various supramolecular systems based on these interactions. Abstract:Enzyme cascade reactions play significant roles in bioelectrochemical processes because they permit more complex reactions. Co-immobilization of multienzyme on the electrode could help to facilitate substrate/intermediate transfer among different enzymes and electron transfer from enzyme active sites to the electrode with high stability and retrievability. Different co-immobilization strategies to construct multienzyme bioelectrodes have been widely reported, however, up to now, they have barely been reviewed. In this review, we focus on recent state-of-the-art techniques for constructing co-immobilized multienzyme electrodes including random and positional co-immobilization. Particular attention is given to strategies such as multienzyme complex and surface display. Cofactor co-immobilization on the electrode is also crucial for the enhancement of catalytic reaction and electron transfer, yet, few studies have been reported. The up-to-date advances in bioelectrochemical applications of multienzyme bioelectrodes are also presented. Finally, key challenges and future perspectives are discussed.

Abstract:Despite the fact that noncovalent bonding interactions are ubiquitous, it is primarily those interactions, which are amenable to spectroscopic analysis, that have been well investigated and applied in chemical engineering. New principles and techniques for characterizing noncovalent interactions are required to gain insight into their detailed nature and explore their potential applications. Here we introduce the practice of analytical noncovalent electrochemistry for probing such interactions. The strengths of noncovalent interactions can be determined more accurately by electrochemical means than by relying on spectroscopic measurements. Specifically, electrochemical analyses are capable of recording/identifying minor signals, leading to the discovery of an unexpected 2:1 host–guest complex. Moreover, the proposed technique is capable of probing multiple properties and facilitates the design and screening of active complexes as catalysts. We also demonstrate achieving a high energy density of 495 Wh kg−1 in rechargeable batteries. The analytical procedure provides a fresh perspective for supramolecular science and takes noncovalent chemistry closer to practical applications. Quantifying the strength of noncovalent interactions in supramolecular host–guest systems is key to guiding molecular design for a desired application. Now, a quantitative relationship between noncovalent interactions and electrochemistry is established that provides a new dimension for investigations into noncovalent interactions and enables the control of electrochemical properties in battery engineering.

Abstract:Supramolecular chemistry focuses on the study of species joined by non-covalent interactions, and therefore on dynamic and relatively ill-defined structures. Despite being a well-developed field, it has to face important challenges when dealing with the selective recognition of biomolecules in highly competitive biomimetic media. However, supramolecular interactions reside at the core of chemical biology systems, since many processes in nature are governed by weak, non-covalent, strongly dynamic contacts. Therefore, there is a natural connection between these two research fields, which are not frequently related or share interests. In this feature article, I will highlight our most recent results in the molecular recognition of biologically relevant species, following different conceptual approaches from the most conventional design of elaborated receptors to the less popular dynamic combinatorial chemistry methodology. Selected illustrative examples from other groups will be also included. The discussion has been focused mainly on systems with potential biomedical applications.

Abstract:Using individual molecules as conducting bridges for electrons offers opportunities when investigating quantum phenomena that are not readily accessible from experiments involving ensembles of molecules. The probing of single molecules has led, over the past few decades, to the rise of molecular electronics. Although single-supermolecule electronics is an emerging field, it is not yet a well-defined area of molecular electronics. There is little doubt, however, that single-supermolecule electronics is poised to have an impact on molecular electronics for the simple reason that non-covalent interactions between molecular components in complexes have a profound effect on electron conductivities. In this Review, we survey this emerging field from the standpoint of non-covalent interactions in mechanically interlocked molecules, as well as in supermolecules, and discuss the (super)structure–property relationship of four different interactions associated with (supra)molecular junctions. They are host–guest interactions, hydrogen bonding, π–π interactions, and non-covalent interactions present in mechanically interlocked molecules. We focus our attention on providing a supramolecular-level understanding of charge transport behaviour associated with each interaction, as well as demonstrating the theoretical background and experimental readiness of single-supermolecule electronics for potential applications, such as nucleic acid and peptide sequencing, and the design and production of quantum interference devices, random-access memories and integrated devices.

Abstract:This chapter reviews different procedures for the electrochemical functionalization of nanostructured carbon materials and their relevance for the immobilization of bioelements. Special attention is paid to discuss about the methodologies used in achieving desired changes in surface chemistry of carbon material and how this property has a great influence in the interactions with the bioelement and consequently in the electron transfer process. Key concepts related to the electrochemical functionalization methods allow to understand the experimental approaches to control the functionalization and the scalability and reproducibility of bioelectrode synthesis. Some examples of different electrochemical functionalization strategies for enzyme immobilization are presented that provide evidence of the interest of these procedures in order to obtain better electron transfer between the bioelement and the nanostructured carbon electrode.

Abstract:Supramolecular systems feature dynamic, reversible and stimuli-responsive characteristics, which are not easily achieved by molecular entities. The last decade has witnessed tremendous advances in the investigations of supramolecular systems for various bioapplications, which include drug delivery, anticancer therapy, antibacterial therapy, photodynamic therapy, photothermal therapy, combination therapy, antidotes for residual drugs or toxins, and bioimaging and biosensing. Host-guest chemistry has played a key role in the development of such bioactive supramolecular systems, and natural macrocycles (such as cyclodextrins), synthetic macrocycles (such as calixarenes, cucurbit[ n ]urils, and pillararenes), and porous framework polymers (such as supramolecular organic frameworks and flexible organic frameworks) have been most successfully used as hosts to build different kinds of host-guest systems for attaining designed biofunctions. The self-assembly of rationally designed amphiphilic molecules, macromolecules and polymers represent another important approach for the construction of supramolecular architectures with advanced biofunctions. In this review, we summarize the important contributions made by Chinese researchers in this field, with emphasis on those reported in the past five years.

Abstract:Supramolecular chemistry involving macrocyclic hosts is a highly interdisciplinary and fast-growing research field in chemistry, biochemistry, and materials science. Host-guest based supramolecular assemblies, as constructed through non-covalent interactions, are highly dynamic in nature, and can be tuned easily using their responses to various external stimuli, providing a convenient approach to achieve excellent functional materials. Macrocyclic hosts, particularly cyclodextrins, cucurbit[n]urils, and calix[n]arenes, which have unique features like possessing hydrophobic cavities of different sizes, along with hydrophilic external surfaces, which are also amenable towards easy derivatizations, are versatile cavitands or host molecules to encapsulate diverse guest molecules to form stable host-guest complexes with many unique structures and properties. Interestingly, host-guest complexes possessing amphiphilic properties can easily lead to the formation of various advanced supramolecular assemblies, like pseudorotaxanes, rotaxanes, polyrotaxanes, supramolecular polymers, micelles, vesicles, supramolecular nanostructures, and so on. Moreover, these supramolecular assemblies, with varied morphologies and responsiveness towards external stimuli, have immense potential for applications in nanotechnology, materials science, biosensors, drug delivery, analytical chemistry and biomedical sciences. In this perspective, we present a stimulating overview, discussing simple host-guest systems to complex supramolecular assemblies in a systematic manner, aiming to encourage future researchers in this fascinating area of supramolecular chemistry to develop advanced supramolecular materials with superior functionalities, for their deployment in diverse applied areas.

Abstract:Electrochemical biosensors have potential applications for agriculture, food safety, environmental monitoring, sports medicine, biomedicine, and other fields. One of the primary challenges in this field is the immobilization of biomolecular probes atop a solid substrate material with adequate stability, storage lifetime, and reproducibility. This review summarizes the current state of the art for covalent bonding of biomolecules onto solid substrate materials. Early research focused on the use of Au electrodes, with immobilization of biomolecules through ω-functionalized Au-thiol self-assembled monolayers (SAMs), but stability is usually inadequate due to the weak Au-S bond strength. Other noble substrates such as C, Pt, and Si have also been studied. While their nobility has the advantage of ensuring biocompatibility, it also has the disadvantage of making them relatively unreactive towards covalent bond formation. With the exception of Sn-doped In2O3 (indium tin oxide, ITO), most metal oxides are not electrically conductive enough for use within electrochemical biosensors. Recent research has focused on transition metal dichalcogenides (TMDs) such as MoS2 and on electrically conductive polymers such as polyaniline, polypyrrole, and polythiophene. In addition, the deposition of functionalized thin films from aryldiazonium cations has attracted significant attention as a substrate-independent method for biofunctionalization.
Interfacial Bioelectrochemistry: Fabrication, Properties and Applications of Functional Nanostructured Biointerfaces

Abstract:Bioelectrochemistry is a new interdisciplinary field, which combines biotechnology with electrochemical science, while the emergence of nanotechnology is opening new horizons for the investigation of this field. Nanostructures (such as nanoparticles, nanotubes) have similar dimensions to those of biomolecules (such as proteins or DNA). The combination of nanostructures with biomolecules yields functional nanostructured biointerfaces with synergetic properties and functions. The recent surge of research interest in the bioelectrochemical field is focused on the advanced design and preparation of such potential nanostructured biointerfaces, as well as the fundamental understanding of the interfacial bioelectrochemical processes within these systems. This review describes recent advances in the development of nanostructured biointerfaces based on the nanostructure-biomolecule hybrid systems in the bioelectrochemical field. To begin with, a wide range of nanostructures, which derive from metal, semiconductor, carbon, and so on, are discussed, involving their synthesis, properties, and conjugation with biomolecules, respectively. Sequentially, the fabrication, properties and characterization of functional nanostructured biointerfaces on electrode surfaces are discussed. Particular emphasis is directed to the discussion and exploration of interfacial bioelectrochemical theories of these architectures, which are of fundamental importance to the unraveling of key biological processes and the comprehensive acquaintance of electron-transfer processes. In addition, the wide applications of nanostructured biointerfaces in the bioelectrochemical field are sketched, and prospects and further developments in this exciting interdisciplinary field are also suggested.
Exploiting the Mechanical Bond Effect for Enhanced Molecular Recognition and Sensing

Abstract:The ubiquity of charged species in biological and industrial processes has resulted in ever‐increasing interest in their selective recognition, detection and environmental remediation. Building on the established coordination chemistry principles of the chelate and macrocyclic effects, and host preorganization, supramolecular chemists seek to construct specific three‐dimensional binding cavities reminiscent of biotic systems to enhance host‐guest binding affinity and selectivity. Mechanically interlocked molecules (MIMs) present a wholly unique platform for synthetic host design, wherein topologies afforded by the mechanical bond enable the decoration of three‐dimensional cavities for non‐covalent interactions with a range of target guest geometries. Notably, MIM host systems exhibit mechanical bond effect augmented affinities and selectivities for a variety of charged guest species, compared to non‐interlocked acyclic and macrocycle host analogues. Furthermore, the modular nature of MIM synthesis facilitates incorporation of optical and electrochemical reporter groups, enabling fabrication of highly sensitive and specific molecular sensors. This review discusses the development of recognition and sensing MIMs, from the first reports in the late 20th century through to the present day, delineating how their topologically preorganized and dynamic host cavities enhance charged guest recognition and sensing, demonstrating the mechanical bond effect as a potent tool in future chemosensing materials. Abstract:Supramolecular non‐covalent interaction is the main base of living functions of creatures. It has become a unique strategy for constructing functional materials. The self‐assembled principle of supramolecular hydrogels, and related outstanding bio‐applications for drug delivery, implantable devices and substitutions, tissue engineering, micro‐fluidic devices, and biosensors are summarized by inspiration from humans, animals, and plants. Nature offers a wealth of opportunities to solve scientific and technological issues based on its unique structures and function. The dynamic non‐covalent interaction is considered to be the main base of living functions of creatures including humans, animals, and plants. Supramolecular hydrogels formed by non‐covalent bonding interactions has become a unique platform for constructing promising materials for medicine, energy, electronic, and biological substitute. In this review, the self‐assemble principle of supramolecular hydrogels is summarized. Next, the stimulation of external environment that triggers the assembly or disassembly of supramolecular hydrogels are recapitulated, including temperature, mechanics, light, pH, ions, etc. The main applications of bioinspired supramolecular hydrogels in terms of bionic objects including humans, animals, and plants are also described. Although so many efforts are done for revealing the synergized mechanism of the function and non‐covalent interactions on the supramolecular hydrogel, the complexity and variability between stimulus and non‐covalent bonding in the supramolecular system still require impeccable theories. As an outlook, the bioinspired supramolecular hydrogel is just beginning to exhibit its great potential in human life, offering significant opportunities in drug delivery and screening, implantable devices and substitutions, tissue engineering, micro‐fluidic devices, and biosensors.
