Abstract:Aims: Hydroalcoholic T. dodoneifolius, B.pinnatum and T. catappa leaf extracts were investigated for their acute toxicity and anxiolytic activities. Study Design: Acute toxicity (LD50) was determined using the limit dose acute and Anxiolytic activities were assessed by open-field field behavioural testing in mice. Place and Duration of Study: The study took place in the Neurobehavioural room of Department of Pharmacology & Therapeutics, Faculty of Pharmacy, Ahmadu Bello University, Zaria in late December 2022. Methodology: Groups of mice (n=6; equal sexes) were each exposed to the open-field paradigm (OFT) following 1 hour of single oral administration of distilled water, extracts (125, 250, and 500 mg/kg) and diazepam (0. 5 mg/kg) using the behavioural indices of mean centre zone time (M%CZT), centre zone re-entry (MCZR) and defeaction/urination frequency (MD/UF). Results: Acute toxicity testing shows that the three extracts are safe with LD50 of 5g/kg. Behavioural results indicate, compared to the distilled water treatment (M%CZT, 6.39±1.53; MCZR, 4.67±1.15, & MU/UF, 4.83±0.75), single acute T. dodoneifolius (M%CZT, 7.50±1.73, 13.72±2.43, & 20.94±3.91*; MCZR, 7.33±0.88, 9.50±0.76, & 11.50±1.6*; MD/UF, 4.17±0.48, 3.17±0.54, & 1.83±0.60*), and B.pinnatum (M%CZT, 9.72±1.91, 9.78±1.32, & 19.67±2.01*; MCZR, 6.00±1.15, 10.50±0.76, & 12.50±1.72*; MD/UF, 3.17±0.79, 2.33±0.42, & 1.33±0.49*) extract treatments demonstrated consistent dose-dependency in their anxiolytic activity across the three parameters with T. catappa extract (M%CZT, 6.95±1.27, 12.17±2.01, & 16.84±1.49*; MCZR, 8.17±1.60, 10.17±1.67, & 10.83±1.96; MD/UF, 2.83±0.60, 2.00±0.73, & 3.67±0.42) exhibiting dose-pendent anxiolytic effect only on the M%CZT. However, at the highest dose level of 500 mg/kg, all plant extract treatments caused anxiolytic effects that are comparable with those of 0.5 mg/kg diazepam treatments (M%CZT, 22.61±1.31*; MCZR, 12.17±1.66*; & MD/UF, 2.62±0.21*). Conclusion: These findings justify the traditional use of these medicinal plant extracts in the remediation of nervous and related disorders.

Abstract:Background: The present study investigated the efficacy of Conyza bonariensis, Commiphora africana, Senna obtusifolia, Warburgia ugandensis, Vernonia glabra, and Zanthoxylum usambarense against Bitis arietans venom (BAV), Naja ashei venom (NAV), and Naja subfulva venom (NSV). Methods: 40 extracts and fractions were prepared using n-hexane, dichloromethane, ethyl acetate, and methanol. In vitro efficacy against snake venom phospholipase A 2 (svPLA 2 ) was determined in 96-well microtiter and agarose-egg yolk coagulation assays. in vivo efficacy against venom-induced cytotoxicity was determined using Artemia salina . Two commercial antivenoms were used for comparison. Results: The 96-well microtiter assay revealed poor svPLA 2 inhibition of BAV by antivenom (range: 20.76% ± 13.29% to 51.29% ± 3.26%) but strong inhibition (>90%) by dichloromethane and hexane fractions of C. africana , hexane and ethyl acetate extracts and fraction of W. ugandensis , dichloromethane fraction of V. glabra , and the methanol extract of S. obtusifolia . The methanol extract and fraction of C. africana , and the hexane extract of Z. usambarense strongly inhibited (>90%) svPLA 2 activity in NAV. The hexane and ethyl acetate fractions of V. glabra and the dichloromethane, ethyl acetate, and methanol extracts of C. africana strongly inhibited (>90%) svPLA 2 in NSV. The agarose egg yolk coagulation assay showed significant inhibition of BAV by the dichloromethane fraction of C. africana (EC 50 = 3.51 ± 2.58 μg/mL), significant inhibition of NAV by the methanol fraction of C. africana (EC 50 = 7.35 ± 1.800 μg/mL), and significant inhibition of NSV by the hexane extract of V. glabra (EC 50 = 7.94 ± 1.50 μg/mL). All antivenoms were non-cytotoxic in A. salina but the methanol extract of C. africana and the hexane extracts of V. glabra and Z. usambarense were cytotoxic. The dichloromethane fraction of C. africana significantly neutralized BAV-induced cytotoxicity , the methanol fraction and extract of C. africana neutralized NAV-induced cytotoxicity, while the ethyl acetate extract of V. glabra significantly neutralized NSV-induced cytotoxicity. Glycosides, flavonoids, phenolics, and tannins were identified in the non-cytotoxic extracts/fractions. Conclusion: These findings validate the local use of C. africana and V. glabra in snakebite but not C. bonariensis, S. obtusifolia, W. ugandensis , and Z. usambarense. Further work is needed to isolate pure compounds from the effective plants and identify their mechanisms of action.

Safety Evaluation of a Triterpenoid-Rich Extract from Bamboo Shavings.

Abstract:Triterpenoids, which may have significant application to the development of natural medicines and functional foods as biological active components, are widely distributed throughout the plant kingdom. This paper evaluated the safety of a triterpenoid-rich extract of bamboo shavings (EBS) systematically. (i) Acute toxicity test: The oral maximum tolerated dose of EBS was more than 10 g/kg body weight both in rats and in mice, due to the absence of toxicity according to the criteria of acute toxic classifications. (ii) Mutagenicity test: It had no mutagenicity judged by negative experimental results of Ames test, mouse bone marrow cell micronucleus test and mouse sperm abnormality test. (iii) 30 days feeding study: No abnormal symptoms and clinical signs or deaths had been found in rats in each group during the test. No significant difference had been found in body weight, food consumption and food availability of rats in each test group (P>0.05). In addition, no significant differences were found in each hematology value, clinical chemistry value and organ/body weight ratio, either (P>0.05). No abnormality of any organ was found during histopathological examination. It can be concluded that the extract of bamboo shavings is of low toxicity and support the use of EBS for various foods.
Acute and Subacute Toxicity Studies of Ethanol, Chloroform extracts and Flower oil of Senecio edgeworthii hook plant

Abstract:This study investigated the acute and subacute oral toxicity of SE- Oil, SE-Alcohol and SE-Chloro extracts of Senecio edgeworthii (SE) in a rodent for evaluating its safety profile. Senecio edgeworthii (SE) plant for the acute oral toxicity (300 to 5000mg/kg) and subacute oral toxicity SE- Oil (SE-Oil 50, 200, 500mg/kg) and SE-Chloro (SEC 100, 500, 1000mg/kg) and SE-Alcohol (SEA 100, 500, 1000mg/kg) studies were administered orally according to the guidelines 423 and 407 of OECD, respectively. In the acute oral toxicity study, doses are administered by 300mg/kg, 1000mg/kg, 2000mg/kg and 5000mg/kg (OECD guideline no. 423). SEA and SEC do not show toxicity up to 5000mg/kg; SE- Oil shows toxicity at a dose of 2500mg/kg. In the subacute toxicity study, the daily oral treatment with extracts of SEA and SEC 100, 500, 1000mg/kg, SE-Oil 50, 250, 500mg/kg of extracts for 28 days did not produce any death or hazard. Likewise, SEA and SEC 100, 500, 1000mg/kg, SE-Oil 50, 250mg/kg of Senecio edgeworthii hook no significant changes were recorded in food intake, body weight gain, the biochemical and haematological parameter of control and treated rats. SE-Oil 500mg/kg of significant changes were recorded in food intake and body weight gains are decreased compared to the control group and SE-Oil 500mg/kg there was a significant (P < 0.05) decrease in RBC counts in the treated animals compared to that of the control group counts in the treated animals and induction (ALT, AST, and alkaline phosphatase and Total protein). SE- Oil 500mg/kg of extract treaded group saw mild liver pathological findings characterized by abnormal hepatic configuration was observed with treated animals. The present result from the acute and subacute toxicity study Senecio edgeworthii hook alcohol and chloroform extract is considered relatively safe on acute and subacute oral exposure. Senecio edgeworthii hook flower hexane extract shows acute toxicity at dose 2500mg/kg and in subacute toxicity study, higher dose 500mg/kg.

Abstract:The medicinal plants find wide applications in pharmaceutical, cosmetic, agricultural and food industry. Trema orientalis has emerged as a good source of phytomedicine. T. orientalis has beneficial therapeutic properties, and indicates that it has potential as an effective herbal remedy for several diseases. The present study was undertaken to evaluate the acute and sub-acute toxicity of methanol powder extract of aerial part of Trema orientalis. There were no major changes in the body weight of animal treated with plant extract. The biochemical parameters like ALP, SGPT, SGOT, total protein, globulin, albumin and bilirubin were within the normal limit in the plant extract treated groups of animals. The present study shows that the methanolic extract of Trema orientalis is safe in lower dose for pharmaceutical analysis. Abstract:Ethnopharmacological relevance: Myrianthus arboreus P. Beauv (Cecropiaceae) is a medicinal plant distributed in forests and damp places of tropical Africa. Its leaves are widely used as food and/or for the treatment of various ailments including dysmenorrhoea, female infertility, tumors and diarrhea. However, to the best of our knowledge, no safety assessment of this plant has been reported yet. Aim of study: The present study aimed at evaluating the safety of the aqueous extract of leaves of Myrianthus arboreus (MAA) in Wistar rats through an acute and sub-acute oral administration. Material and methods: In acute oral toxicity, the test was performed according to the Organization for Economic Cooperation and Development (OECD) guidelines Nr. 423 (acute toxicity class method, ATC) with slight modifications. Female Wistar rats were orally treated with the aqueous extract of M. arboreus at the doses of 2000 and 5000mg/kg. In sub-acute toxicity study, using the OECD guidelines Nr. 407, the extract was administered by gavage at the doses of 20, 110 and 200mg/kg/day for 28 consecutive days. Results: A single oral administration of 2000 or 5000mg/kg of the extract induced neither mortality nor exterior signs of toxicity indicating a LD50 >5000mg/kg. In sub-acute study, the extract decreased triglycerides, total cholesterol/high density lipoproteins ratio and atherogenic index of plasma in both sexes at all tested doses. Alanine transaminase decreased in both sexes at 200mg/kg and serum creatinine levels decreased at all tested doses in females. Moreover, significant increases in ovarian and uterine wet weights, red blood cell count, hematocrit, mean corpuscular hemoglobin and hemoglobin were observed at 200mg/kg in females. In males, this extract decreased white blood cell count, lymphocytes and relative weight of seminal vesicles and ventral prostate at 200mg/kg. Conclusion: The aqueous extract of Myrianthus arboreus leaves was non-toxic in acute administration and exhibited a relatively low toxicity potential on accessory sex organs in both sexes, and leukocytes in males following the repeated 28-days oral administration of the dose 200mg/kg.
Acute and Sub-Acute Toxicity Evaluation of the Crude Methanolic Extract of Justicia schimperiana Leaf in Wistar Albino Rats

Abstract:Purpose: This study evaluates the acute and sub-acute toxicity of 80% methanolic extracts of the leaves of Justicia schimperiana in Wistar albino rat models. Methods: Dried powdered leaves of Justicia schimperiana were macerated in 80% methanol. The experiment was conducted in accordance with the Organization for Economic Co-operation and Development guideline 423 for acute and 407 for sub-acute toxicity testing. A single dose of 5000 mg/kg extract was orally administered to three female rats for the acute toxicity study. The plant extract was administered orally for 28 days in daily dosages of 250, 500, and 1000 mg/kg for the sub-acute study. Animals in a control group were given distilled water. A total of 40 rats (5 rats/group/sex) were used for the sub-acute toxicity testing. Daily food intake and weekly body weight measurements were done. The rats were sacrificed at the end of the 28-day treatment period for hematological, biochemical, and histopathological tests. One-way analysis of variance and Kruskal-Wallis tests were employed for the analysis. Results: The single-dose oral administration of the plant resulted in no deaths or serious morbidity. The median lethal dose was >5000 mg/kg. The 28-day oral treatment of the plant extract had no significant effect on general behavior, food intake, organ weight, biochemical parameters, or the majority of the hematological markers, with the exception of the decrease in hemoglobin and hematocrit in the 1000 mg/kg extract-treated groups compared to the controls. Both sexes experienced significant weight increases at all dosage levels. With the exception of minor alterations in a few of the organs, no significant histological change was identified. Conclusion: It is concluded that the single-dose and repeated-dose 28-day oral administration of the methanolic leaf extract of Justicia schimperiana is relatively safe.
The investigation on an ethnic medicinal plant of Elsholtiza bodinieri Vaniot: Chemical constituents, acute, 28-day subacute and 90-day subchronic toxicity evaluation

Abstract:Ethnopharmacological significance: Elsholtiza bodinieri Vaniot, belonging to the family Lamiaceae, has important medicinal value in Yunnan province of China. Traditionally, its aerial parts have been used as an ethnomedicine to treat diaphoresis, headache, fever, cough, pharyngitis, dyspepsia, and hepatitis. However, the safety assessment of E. bodinieri is still unexplored. Aim of the study: This study aimed to investigate the phytochemical constituents of the hot water extract from E. bodinieri (HEEB) and evaluate the 14-day acute, 28-day subacute and 90-day subchronic toxicity by oral administration in Sprague-Dawley (SD) rats. Materials and methods: The chemical constituents of HEEB were analyzed by UHPLC-ESI-HRMS/MS. Firstly, SD rats were chosen for a single oral administration of the maximum dose of 5000 mg/kg to evaluate toxicity. Subsequently, consecutive 28-day subacute and 90-day subchronic toxicity assessments of HEEB were conducted on Sprague-Dawley (SD) rats through repeated doses of 2500, 1250, 625, and 312.5 mg/kg for the former, and 1500, 1000, and 500 mg/kg for the latter. For toxicity evaluation, hematology and serum biochemical indicators were determined, and major organs of the rats were collected to calculate organ coefficients. Additionally, hematoxylin-eosin (H&E) staining was performed on the collected tissues to assess histopathological changes induced by repeated oral administration of HEEB. Results: A total of 23 compounds were identified by UHPLC-ESI-HRMS/MS analysis. Acute toxicity assessment revealed that oral administration of HEEB did not induce mortality and unnormal behavior changes in female rats over a 14-day period, suggesting that the approximate lethal dose (ALD) was higher than 5000 mg/kg. In consecutive 28-day and 90-day toxicity evaluations, HEEB doses of 2500 mg/kg and 1500 mg/kg resulted in hepatic and kidney tissue damage in both female and male rats, which was verified by the increased levels of AST, ALT, BUN, Na+, and Cl-. Conclusions: After the acute, 28-day subacute and 90-day subchronic toxicity evaluation, the No Observed Adverse Effect Level (NOAEL) was determined as 1000 mg/kg/day. These findings not only provided a safety information for its medicinal and edible application, but also promoted the further comprehensive development of this plant.
In vitro antibacterial and antioxidant activity of flavonoids from the roots of Tephrosia vogelii: a combined experimental and computational study

Abstract:Tephrosia vogelii is a traditional medicinal plant used to treat hypertension, diarrhea and urinary disorders. Silica gel chromatographic separation of CH2Cl2/MeOH (1:1) roots extract of T. vogelii afforded seven compounds namely; β-sitosterol (1a), stigmasterol (1b), 6a, 12a-dehydro-deguelin (2), tephrosin (3), maackiain (4), obovatin (5) and 6-oxo, 6a, 12a-dehydro-deguelin (6). GC-MS analysis of essential oils from the root of T. vogelii displayed a total of 17 compounds of which cis-nerolidol (41.7 %) and cadinol (19.7 %) were the major constituents. CH2Cl2/MeOH (1:1) extract, MeOH extract, maackiain (4) and obovatin (5) showed moderate inhibitory activity against Pseudomonas aeruginosa with MIC value of 0.5, 0.66, 0.83 and 0.83 mg/mL, respectively, compared to ciprofloxacin (MIC of 0.078 μg/mL). 6a, 12a-dihydro-deguelin (2), and 6-oxo, 6a, 12a-dehydro-deguelin (6) displayed significant activity against S. epidermis with MIC values of 0.66 mg/mL. Tephrosin (3) and maackiain (4) also showed moderate antibacterial activity against Staphylococcus aureus and Proteus mirabilis with MIC values of 0.83 and 0.5 mg/mL, respectively, compared to ciprofloxacin (0.312 μg/mL). The radical scavenging activity results indicated that tephrosin (3), obovatin (5) and 6-oxo, 6a, 12a-dehydro-deguelin (6) showed potent DPPH scavenging activity with IC50 values of 10.97, 10.43 and 10.73 μg/mL, respectively, compared to ascorbic acid (IC50 of 5.83 μg/mL). The docking prediction results revealed that 6a, 12a-dehydro-deguelin (2) displayed the best binding energy of -8.1 kcal/mol towards pyruvate kinase of S. aureus (PDB ID: 3T07) and -7.9 kcal/mol towards P. mirabilis urease (PDB ID: 1E9Y) and DNA gyrase B of Escherichia coli (PDB: 4F86) receptors compared to ciprofloxacin (-7.2 to -8.0 kcal/mol). Maackiain (4) and obovatin (5) displayed the minimum binding energy of -7.9 and -8.2 kcal/mol towards the LasR protein of P. aeruginosa (PDB: ID 2UV) and S. epidermidis FtsZ (PDB: ID 4M8I), respectively. The SwissADME drug-likeness and Pro Tox II toxicity prediction results indicated that compounds (2-6) obeyed Lipinski's rule of five with 0 violations and none of them were found to be hepatotoxic, mutagenic, and cytotoxic, respectively. The in vitro assessment results supported by the in silico analysis revealed that crude extracts and isolated compounds showed promising antibacterial and antioxidant activity, which proves the therapeutic potential of the roots of T. vogelii.
Acute and subacute toxicity of aqueous extract of the tuber of Kedrostis africana (L.) Cogn in Wistar rats

Abstract:Kedrostis africana (L.) Cogn (Cucurbitaceae) is used in South African traditional medicine and pharmacopoeia as an emetic, purgative and diuretic, and it is used against dropsy in the management of obesity. Aim of the study In this study, acute and subacute toxicity of aqueous extract of K. africanatuber was evaluated in male and female Wistar rats in order to assess its safety profile. Materials and methods In acute toxicity, the effects of a single oral dose (2,000 and 5,000 mg/kg) of aqueous extract was determined in both sexes. General behavior, adverse effects and mortality were determined for 3 h and then periodically for 14 days. The subchronic toxicity test was performed in rats. The effects of the extract in daily single oral administration at the doses of 200, 400 and 600 mg/kg for 28 days were determined. Food and water intakes were monitored daily while body weight was monitored on a weekly bases. Hematological, biochemical and organ parameters were determined at the end of the 28-day administration. Results In the acute study, a single administration of the aqueous extract at the doses of 2,000 and 5,000 mg/kg did not induce mortality. Thus, the LD50 of the aqueous extract of K. africana (AEKA) has been estimated to be higher than 5,000 mg/kg. In the subchronic study, daily oral administration of the AEKA did not result in death of the rats or significant changes in hematological or biochemical parameters at the highest dose of 600 mg/kg. No alteration was observed in body weight, food and water intake. Liver, kidney and heart histopathology did not reveal morphological alteration. Conclusions The results showed that the aqueous tuber extract of K. africana did not cause any death, nor did it cause abnormalities in necropsy and histopathology findings. There were no acute or subchronic toxicity observed, and this indicates that the plant extract could be considered safe for oral medication.
Organ wide toxicological assessment of common edible herbs and their mixtures as used in home remedies

Abstract:The use of home remedies for medicinal purposes, most of which are edible plants has continued to be a practice in many homes. However, there has been an increasing report of chronic use with lethal effect. Among the commonly used herbal/ medicinal plants were ginger, garlic and lemon. These were seen to be prevalent across continents with brewing and crude extraction being the most means of consumption. This study investigated the organ wide toxicity of this extract following chronic consumption of crude extract. Twenty-five albino Wister rats, five in each group were used for this experiment. Each animal received 0.5ml/kg body weight of either ginger extract, garlic extract, lemon juice, or a mixture of equal volumes of all three extract (v/v) respectively twice daily for seven (7) days. Statistics were represented as ±SE; P≤0.05 was considered significant. Previous studies have shown that moderate consumption of these medicinal plants were beneficial and have shown no deleterious effect. This study observed no change in the weight of the experimental animals. The weight of the animals continued to increase except for the group that received lemon and the mixture, but these were not significant. It was observed that chronic consumption induced organ wide toxicity to include the liver, kidney, intestinal epithelium, stomach, and pancreas. These were shown to alter tissue architecture and the cell morphology. Packed cell volume was reduced in the lemon and the group that received a combination of all extracts (p=o.03). Blood differentials showed changes in levels. An elevated basophil level was observed in ginger and garlic (p<0.0001; p=0.0006). Monocyte levels increased progressively across each group when compared to the control with the most elevated level seen in the group that received the mixture (p<0.0001). Lymphocyte count was reduced across all the groups that received the extract except for animals that received ginger. This study suggests the application of caution among users of these medicinal plants and continues to draw attention to the need for harmonization and standardization of safe use doses.
Phytochemical composition and antioxidant activities of some wild edible plants locally consumed by rural communities in northern Uganda

Abstract:Background: Acalypha rhomboidea, Asystacia gangetica, Crassocephalum sacrobasis, Crotalaria ochroleuca, Heterosis rotundifolia, Hibiscus cannabinus, Hibiscus sp., Hibiscus surratensis, Ipomoea eriocarpa, Maerua angolensis, Senna obtusifolia and Vigna membranacea are among the common wild edible plants in the Acholi sub-region, northern Uganda. This study evaluated the phytochemical constituents and antioxidant potential of the plants. Methods: Fresh leaves collected from each plant species were air-dried under shade. The phytochemical contents of the ethanol and petroleum ether extracts were determined using standard protocols. The antioxidant content of the methanolic extracts was assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Results: Preliminary phytochemical analyses indicated the presence of tannins, reducing compounds, alkaloids, flavonoids, flavons aglycones, flavanosides, anthracenosides, anthocyanosides, volatile oils, coumarins, steroid glycosides, sterols and triterpenes. However, the extracts did not contain any emodols and saponins. The results of the quantitative phytochemical analysis showed that the contents of different phytochemicals detected varied significantly (p < 0.05) among the selected plants. The amount of tannins in mg/g (gallic acid equivalent) of dry weight varied from 3.90 ± 0.16 in C. ochroleuca to 10.41 ± 0.78 in I. eriocarpa, total flavonoids in RE, mg/g dry matter from 4.07 ± 0.11 in I. eriocarpa to 14.94 ± 0.08 in S. obtusifolia. Total alkaloids in mg/100 g ranged from 1.59 ± 0.30 in I. eriocarpa to 6.37 ± 0.24 in Hibiscus sp. Total phenolic content in GAE, mg/g dry matter ranged from 13.39 ± 0.26 in A. rhomboidea to 64.25 ± 0.54 in I. eriocarpa. The in vitro antioxidant assays revealed substantial free radical scavenging activity in all the plants. Antioxidant activity expressed as IC50 (ppm) ranged from 13.39 for A. rhomboidea to 64.84 for I. eriocarpa, compared to 12.82 for ascorbic acid standard. The total phenolic compounds and total tannins had significant and positive correlations with DPPH free radical scavenging activity. Conclusion: The findings of this study provide evidence that the species are good natural sources of phytochemicals and antioxidants, whose regular consumption could provide human health benefits by protecting against oxidative stress related diseases. Further research is needed on the structural characterization of the phytochemicals, profiling the plant extracts with high antioxidant activity and determining the antimicrobial activities.

Abstract:Abstract Background Tuberculosis (TB) is one of the leading causes of death globally, and the rise in drug-resistant forms of TB has become a significant threat. Subsequently, it is crucial to explore new, effective and safe anti-TB agents. This study aimed at conducting phytochemical screening, antimycobacterial activity, and acute toxicity of the selected plant species’ crude extracts to assess their toxicological potentials and efficacies against TB. Methods The aqueous and methanol/dichloromethane (DCM) (1:1) extracts of each selected plant species were subjected to phytochemical screening and antimycobacterial activity using microplate alamar blue assay. For acute toxicity, a single dose (2000 mg/kg) of the aqueous extracts was orally administered to each animal following the Organization for Economic Cooperation and Development (OECD) guidelines No. 425 and then observed for 14 days. The animals were closely observed on the general behavior and clinical signs of toxicity, and body weights were recorded. After the termination of the experiment, hematological, biochemical, and histopathological analyses were performed. Results The extracts contained alkaloids, flavonoids, tannins, saponins, steroids, terpenoids, resins, cardiac glycosides, phenolic compounds, and coumarins. Aqueous extracts showed moderate to weak activity against the susceptible (H 37 Rv) M. tuberculosis strain and weak activity against the MDR-TB strain with Minimum Inhibitory Concentrations (MIC μg/mL) ranging from 293.0–2344.0 and 1172.0–4688.0, respectively. Methanol/DCM extracts showed significant to moderate activity against the susceptible TB strain and moderate to weak activity against the MDR-TB strain with MIC (μg/mL) ranging from 98.0–586.0 and 293.0–781.0, respectively. One mortality was recorded from the A. coriaria treated group following the acute toxicity tests, but the LD 50 of all the extracts was estimated to be above 2000 mg/kg. Histopathological analyses did not show any significant lesions in the examined organs except those from the A. coriaria treated group. Conclusion Phytochemical screening of the extracts revealed the presence of alkaloids, tannins, saponins, flavonoids, steroids, terpenoids, resins, cardiac glycosides, phenolic compounds, and coumarins. All the methanol/DCM extracts of the plant species studied have promising antimycobacterial activity. The selected plant extracts studied exhibited low acute toxicity levels except for A. coriaria and could be safe for formulations into herbal products.

Abstract:Plants create secondary metabolites which may or may not have biological activities and are not always benign. Therefore, it is essential to assess both their bioactivities and safety. The objective of this research is to assess the in vitro toxicity and phytochemistry of extracts from Bauhinia galpinii, Combretum caffrum , and Maytenus undata extracts. In the past, it has been demonstrated that these plants possess potent antifungal activities against mycotoxigenic fungal strains. The MTT assay was used to evaluate the cytotoxicity of methanolic leaf extracts of selected plants on the viability of the African Green monkey kidney and the human colorectal adenocarcinoma cells. Furthermore, the ability of these plants to cause genotoxicity in Vero cells was investigated using the micronucleus test. The identity and quantity of the phytocompounds present in the plant extracts were analyzed using liquid-chromatography time-of-flight mass spectrometry (LC-ToF-MS) analysis. B. galpinii, C. caffrum , and M. undata extracts were cytotoxic to Vero cells at 500 μg/ml. All the selected medicinal plant extracts demonstrated dose-dependent cytotoxicity against both Caco-2 and Vero cells. B. galpinii, M. obtusifolia , and M. undata extracts were all genotoxic towards the Vero cells at 500 μg/ml, while Combretum caffrum showed genotoxicity at 250 μg/ml. Phenolic glycosides, coumarins, flavonoids, and hydrolysable tannins were identified as major phytochemicals in the secondary metabolite profiling. Given the results, caution in terms of quantities consumed should be exercised when these plants are used in traditional medicine.

Abstract:Ficus thonningii (Blume) is a medicinal plant whose biological activity has been demonstrated in the management of peptic ulcers. Unfortunately, little scientific data exists on its toxicity. This is why we studied the acute and subacute oral toxicity profile of the hydro-ethanolic extract of the bark of F. thonningii and the systemic exposure of the bioactive components of the plant.Acute toxicity was assessed by administering a single dose of 2000 mg/kg to 5-week-old rats and observed for 14 days. The subacute toxicity study was performed in 6-week-old rats. Animals were orally treated with a daily dose of 125 mg/kg, 250 mg/kg, 500 mg/kg extract for 28 days. A satellite group of animals received 500 mg/kg per day for 28 days and after discontinuation of treatment they were observed for 14 days. Hematological and blood biochemical parameters, as well as kidney and liver histology, were recorded at the end of each experiment. For acute treatment, administration of a dose of 2000 mg/kg did not induce any critical behavioral changes or deaths. Following subacute dosing, biochemical analysis revealed a slight elevation of liver parameters in the 500mg/kg hydroethanolic extract, while no significant increase was observed for kidney parameters. This study showed that daily administration of Ficus thonningii (Blume) for 28 days resulted in the potential to reverse impairment of liver function.
Antisecretory and Spasmolytic Activities of Aqueous and Ethanolic Stem Bark Extracts of Nauclea diderrichii in Wistar Rats

Abstract:Background. Diarrheal diseases are a major cause of morbidity and mortality throughout the world and particularly in developing countries. Nauclea diderrichii is a plant used in traditional medicine in the treatment of anemia, fever, gastric ulcer, malaria, abdominal pain, skin infections, and diarrhea. The present work is aimed at evaluating the antisecretory and spasmolytic activities of aqueous and ethanolic stem bark extracts of Nauclea diderrichii in Wistar rats. Methods. The effect of aqueous and ethanolic extracts of Nauclea diderrichii was tested at doses of 100, 200, and 300 mg/kg on castor oil-induced secretory diarrhea, misoprostol-induced fluid accumulation, and the effect of pretreatment with yohimbine and glibenclamide. They were also tested on normal motility and castor oil- and carbachol-induced hypermotility. Results. The results showed that the aqueous and ethanolic extracts of Nauclea diderrichii significantly ( p < 0.001 ) inhibited castor oil-induced secretory diarrhea at all the doses. Both extracts significantly ( p < 0.001 ) inhibit fluid accumulation induced by misoprostol. The pretreatment with glibenclamide reduced the antidiarrheal activity of aqueous extract of Nauclea diderrichii. The pretreatment with yohimbine did not alter the effect of the aqueous extract of Nauclea diderrichii. On intestine transit as on castor oil- and carbachol-induced motility, the aqueous and ethanolic extracts at doses of 100 and 200 mg/kg reduced significantly ( p < 0.05 , p < 0.01 , and p < 0.001 ) the travelled distance by charcoal and peristaltic index. Conclusions. The study demonstrated that the aqueous and ethanolic extracts of Nauclea diderrichii possess antisecretory and antispasmolytic properties hence its use in traditional medicine against diarrhea.

Abstract:Ethnopharmacological relevance: Mucuna pruriens L is a wild and cultivated leguminous plant which have been used as an aphrodisiac, diuretic, nerve tonic, and antiarthritic agent. Aim: To evaluate the toxicity, antinociceptive, and anti-inflammatory activities of M. pruriens (EEMP) ethanol extract in experimental models. Methods: M. pruriens dried leaves were extracted using aqueous ethanol (30:70). Tests for acute and subacute toxicity were conducted on rats and mice. Mice were used in hotplate, acetic acid, and formalin models to test the antinociceptive activity of EEMP. The anti-inflammatory properties of EEMP (25, 100, and 400 mg/kg) were assessed egg albumin, carrageenan, and formalin-induced oedema models. The study examined the anti-inflammatory mechanism of EEMP (25-400 mg/kg) in rats with an air pouch caused by carrageenan. Air pouch exudates were tested for total leucocytes and differential cell counts, TNF-α, IL-6, myeloperoxidase activity, malondialdehyde, nitrites, and reduced glutathione (GSH). Results: The acute oral toxic dose of EEMP is greater than 2000 mg/kg. There were no significant behavioral, hematological or biochemical alterations seen after 14-days repeated administration of EEMP (200, 400 and 800 mg/kg) in mice. The EEMP demonstrated significant antinociceptive activity in hotplate, acetic acid and formalin-induced nociception in mice. The EEMP significantly and dose dependently reduced paw oedema at 2, 4 and 96 h in the egg-albumin, carrageenan- and formalin-induced paw oedema, respectively. Exudates volume, inflammatory cell counts, TNF-α, IL-6, myeloperoxidase, malondialdehyde and nitrites were significantly reduced, while GSH increased in carrageenan-air pouch of EEMP-treated rats. Conclusion: Mucuna pruriens leaves ethanol extract demonstrated good safety profile as well as antinociceptive and anti-inflammatory activity through mechanisms related to inhibition of oxidative stress and pro-inflammatory cytokines as well as lysosomal membrane stability.
Evaluation of the Antibacterial and Antibiofilm Effects of Ethyl Acetate Root Extracts from Vernonia adoensis (Asteraceae) against Pseudomonas aeruginosa

Abstract:There is an increase in mortality and morbidity in the health facilities due to nosocomial infections caused by multidrug-resistant nosocomial bacteria; hence, there is a need for new antibacterial agents. Vernonia adoensis has been found to possess medicinal value. Plant phytochemicals may have antimicrobial activity against some resistant pathogens. The antibacterial efficacy of root extracts against Staphylococcus aureus and Pseudomonas aeruginosa was investigated using the microbroth dilution method. All extracts from the roots had an inhibitory effect on the growth of both bacteria, with the most susceptible being P. aeruginosa. The most potent extract was the ethyl acetate extract which caused a percentage inhibition of 86% against P. aeruginosa. The toxicity of the extract was determined on sheep erythrocytes, and its effect on membrane integrity was determined by quantifying the amount of protein and nucleic acid leakage from the bacteria. The lowest concentration of extract used, which was 100 µg/ml, did not cause haemolysis of the erythrocytes, while at 1 mg/ml of the extract, 21% haemolysis was observed. The ethyl acetate extract caused membrane impairment of P. aeruginosa, leading to protein leakage. The effect of the extract on the biofilms of P. aeruginosa was determined in 96-microwell plates using crystal violet. In the concentration range of 0-100 µg/ml, the extract inhibited the formation of biofilms and decreased the attachment efficiency. The phytochemical constituents of the extract were determined using gas chromatography-mass spectrometry. Results of analysis showed the presence of 3-methylene-15-methoxy pentadecanol, 2-acetyl-6-(t-butyl)-4-methylphenol, 2-(2,2,3,3-tetrafluoropropanoyl) cyclohexane-1,4-dione, E,E,Z-1,3,12-nonadecatriene-5,14-diol, and stigmasta-5,22-dien-3-ol. Fractionation and purification will elucidate the potential antimicrobial compounds which are present in the roots of V. adoensis.

Abstract:Ethnopharmacological relevance: Blighia sapida, commonly known as Ackee, is a plant native to West Africa, with great cultural and therapeutic value, particularly in Western Nigeria. Traditionally, Blighia sapida capsule is used in western Nigeria to treat ecthyma in sheep and goats by heating it in hot ash. This process causes the capsule to release a liquid, which is then directly applied to the entire affected area of the skin. However, there is limited information available on its phyto-constituents and medicinal effects. Aim of the study: This work examined the bioactive constituents, acute toxicity, and sub-acute toxicity of aqueous and ethanolic extracts of Blighia sapida capsule. Materials and methods: Extraction of phytochemical constituents was carried out with distilled water and ethanol and was concentrated at 40 °C. The phytochemical constituents were determined using a variant 3800/4000 gas chromatography-mass spectrometry (GC-MS) machine. Lorke's method was employed to determine the acute toxicity of the aqueous and ethanolic extracts of Blighia sapida capsule. Results: The GC-MS analysis revealed 15 bioactive compounds in both extracts, with kaempferol being the most abundant. Notable pharmacologically active compounds included pyrrolidin-2-ylmethanol, rutin, quinoline, apigenin, and naringenin. The study observed distinctive differences in aqueous and ethanolic extracts compound weights and peak areas. Acute toxicity study depicts that the lethal dose of aqueous and ethanolic extracts of Blighia sapida capsule is above 5000 mg/kg as no mortality was recorded in the oral administration of 10, 100, 1000, 1600, 2900, and 5000 mg/kg of aqueous and ethanolic extracts. Sub-acute toxicity results indicated no significant adverse effects on kidney and liver function, although some variations in biochemical parameters were observed. Histological analysis showed normal renal and hepatic architecture in treated animals. Conclusion: This study demonstrated that aqueous and ethanolic extracts of Blighia sapida capsule exhibited no acute toxicity and minimal sub-acute toxicity, suggesting they are safe for consumption at the tested doses.
Vernonia amygdalina: a comprehensive review of the nutritional makeup, traditional medicinal use, and pharmacology of isolated phytochemicals and compounds

Abstract:Vernonia amygdalina is a perennial shrub that belongs to the family Asteraceae. The herb is an indigenous African plant that grows in most parts of sub-Saharan Africa. It is probably the most used medicinal plant in the genus Vernonia. Previous studies on the traditional medicinal value, nutritional composition, classes of phytochemical or compound isolation, and evaluation of their pharmacology activity are numerous. This provokes us to review and provide up-to-date evidence-based information on the study plant. A systematic online search using the databases of Google Scholar, PubMed, Science Direct, Wiley, Elsevier and Sci-Hub was carefully applied, using some important key words to get appropriate information. The leafy part of Vernonia amygdalina contributes greatly to the nutritional requirements for human health and to food security since it contains enough concentrations of proximate composition, minerals, and vitamins. The plant parts are used in traditional medicine for many human and animal healthcare purposes, including diarrhea, diabetes, wound healing, tonsillitis, evil eye, retained placenta, headache, eye disease, intestinal parasite, bloating, hepatitis, toothache, anthrax, malaria, urine retention, gastritis, stomach disorders, and snake bites. The chemical analysis revealed the presence of flavonoids, alkaloids, saponins, tannins, triterpenoids, sesquiterpene lactones, steroids, cardiac glycosides, oxalates, phytates, cyanogenic glycosides, and phenols. Additionally, various compounds such as vernolide, luteolin, vernodalol, vernoamyoside A, vernoamyoside B, isorhamnetin, glucuronolactone, and 1-Heneicosenol O-β-D-glucopyranoside were isolated. Some of the isolated compounds pharmacological activity was evaluated against some diseases and showed antioxidant, antibacterial, antifungal, antihelmintic, anticancer, and anti-inflammatory potencies. Thus, the review provides comprehensive information about ethnomedicinal value, nutritional composition, isolated classes of phytochemicals, and compounds, including an evaluation of the pharmacological activity of the isolated compounds of Vernonia amygdalina . A review with this much information could be extremely valuable for future research on developing innovative nutraceutical products.
