Abstract:Subclinical, low-grade, inflammation is one of the main pathophysiological mechanisms underlying the majority of chronic and non-communicable diseases. Several methodological approaches have been applied for the assessment of the anti-inflammatory properties of nutrition, however, their impact in human body remains uncertain, because of the fact that the majority of the studies reporting anti-inflammatory effect of dietary patterns, have been performed under laboratory settings and/or in animal models. Thus, the extrapolation of these results to humans is risky. It is therefore obvious that the development of an inflammatory model in humans, by which we could induce inflammatory responses to humans in a regulated, specific, and non-harmful way, could greatly facilitate the estimation of the anti-inflammatory properties of diet in a more physiological way and mechanistically relevant way. We believe that exercise-induced muscle damage (EIMD) could serve as such a model, either in studies investigating the homeostatic responses of individuals under inflammatory stimuli or for the estimation of the anti-inflammatory or pro-inflammatory potential of dietary patterns, foods, supplements, nutrients, or phytochemicals. Thus, in this review we discuss the possibility of exercise-induced muscle damage being an inflammation model suitable for the assessment of the anti-inflammatory properties of diet in humans.

Abstract:<p>A wide array of molecular pathways has been investigated during the past decade in order to understand the mechanisms by which the practice of physical exercise promotes neuroprotection and reduces the risk of developing communicable and non-communicable chronic diseases. While a single session of physical exercise may induce a transient imbalance of cell homeostasis, repeated physical exercise sessions will improve immunosurveillance and immunocompetence. Additionally, immune cells from the central nervous system will acquire an anti-inflammatory phenotype, protecting central functions from age-induced cognitive decline. This review highlights the exercise-induced anti-inflammatory effect on the prevention or treatment of common chronic clinical and experimental settings. It also suggests the use of pterins in biological fluids as sensitive biomarkers to follow the anti-inflammatory effect of physical exercise.</p>

Abstract:Exhaustive exercise induces systemic inflammatory responses, which are associated with exercise-induced tissue/organ damage, but the sources and triggers are not fully understood. Herein, the basics of inflammatory mediator cytokines and research findings on the effects of exercise on systemic inflammation are introduced. Subsequently, the association between inflammatory responses and tissue damage is examined in exercised and overloaded skeletal muscle and other internal organs. Furthermore, an overview of the interactions between oxidative stress and inflammatory mediator cytokines is provided. Particularly, the transcriptional regulation of redox signaling and pro-inflammatory cytokines is described, as the activation of the master regulatory factor nuclear factor (erythroid-derived 2)-like 2 (Nrf2) is involved directly or indirectly in controlling pro-inflammatory genes and antioxidant enzymes expression, whilst nuclear factor-kappa B (NF-κB) regulates the pro-inflammatory gene expression. Additionally, preventive countermeasures against the pathogenesis along with the possibility of interventions such as direct and indirect antioxidants and anti-inflammatory agents are described. The aim of this review is to give an overview of studies on the systematic inflammatory responses to exercise, including our own group as well as others. Moreover, the challenges and future directions in understanding the role of exercise and functional foods in relation to inflammation and oxidative stress are discussed.

Abstract:Background This study investigated selected inflammatory responses to acute and chronic exercise in individuals with inflammatory bowel disease (IBD). Methods A systematic review and meta-analysis was conducted on all relevant exercise-based intervention publications with IBD participants. The study included articles that utilised a broad range of acute and chronic exercise interventions, with inflammatory biomarkers measured and symptoms documented, both pre- and post-exercise for those with IBD. The search was limited to studies published in English, the use of human participants, and primary studies, with no restrictions on date of publication or participant’s age. Articles were retrieved through the electronic databases: PubMed, SPORTDiscus, and Scopus. This study adhered to Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines. Results Six inflammatory markers were included in the meta-analysis which consisted of five studies. Exercise interventions resulted in no significant difference in IL-6 ( SMD = -0.09; 95% CI = -0.49, 0.30; P = 0.64), TNF-α ( SMD = 0.08; 95% CI = -0.31, 0.48; P = 0.68), CRP ( SMD = -0.04; 95% CI = -0.58, 0.50; P = 0.89), IL-17 ( SMD = 0.15; 95% CI = -0.45, 0.76; P = 0.62), leukocytes ( SMD = 0.40; 95% CI = -0.53, 1.33; P = 0.40) or lymphocytes ( SMD = 0.32; 95% CI = -0.33, 0.97; P = 0.33), thus, indicating exercise may have no effect on inflammatory markers in IBD. Bowel symptoms improved following regular moderate exercise that incorporated stress management. Conclusion Heterogeneity among the identified literature may have led to exercise interventions being ineffective in reducing inflammation. Although the limited number of eligible studies may reduce the reliability of results, it emphasises the need for additional research in this domain. Importantly, no adverse symptomatic responses to exercise indicate that exercise is safe for IBD patients.

Abstract:Key PointsPhysical inactivity increases the risk of type 2 diabetes, cardiovascular diseases, chronic obstructive pulmonary disease, colon cancer, breast cancer, dementia and depression.Physical inactivity leads to the accumulation of visceral fat and consequently the activation of a network of inflammatory pathways. Chronic inflammation promotes the development of insulin resistance, atherosclerosis, neurodegeneration and tumour growth, and subsequently the development of a number of diseases associated with physical inactivity.The protective effect of exercise against chronic inflammation-associated diseases may, to some extent, be ascribed to an anti-inflammatory effect of regular exercise. The anti-inflammatory effect of regular exercise may be mediated by a reduction in visceral fat mass (with a subsequent decreased release of adipokines from adipose tissue) and/or by the induction of an anti-inflammatory environment with each bout of exercise.Possible mechanisms by which exercise exerts its anti-inflammatory effect include: release of interleukin-6 (IL-6) into the circulation from contracting muscle fibres and subsequent increases in circulating levels of IL-10 and IL-1 receptor antagonist; increased circulating numbers of IL-10-secreting regulatory T cells; downregulation of Toll-like receptor expression on monocytes and inhibition of downstream responses (such as pro-inflammatory cytokine production, antigen presentation and co-stimulatory molecule expression); reduction in the circulating numbers of pro-inflammatory monocytes; and inhibition of monocyte and/or macrophage infiltration into adipose tissue.Although regular moderate exercise is associated with a reduced incidence of infection compared with a completely sedentary state, the long hours of hard training undertaken by elite athletes appear to make these individuals more susceptible to infections. Abstract:INTRODUCTION & AIMS Inflammatory bowel disease (IBD) and irritable bowel syndrome (IBS), are characterised by intestinal inflammation that presents severe symptoms and continued complications throughout the lifespan. These gastrointestinal (GI) disorders have a growing prevalence worldwide with no known cure. Management strategies often require a ‘trial and error’ process before an effective pharmacal/s is established; the successful interim varies considerably with prevalent adverse side-effects and gradual decline in the effectiveness of medication. Accordingly, alternative mechanisms to complement management options, reduce side-effects, and improve well-being warrant investigation. Physical activity is known to be beneficial for a variety of inflammatory conditions; the advantageous effects are associated with the exercise-induced inflammatory response directly from active muscle tissue. Despite this knowledge, minimal studies have attempted to quantify the inflammatory effect of exercise. This study investigates the inflammatory response of acute exercise. METHODS Participants were allocated into one of two groups: GI disorders (IBD & IBS; n = 11) or apparently healthy control (AHC; n = 11). All participants completed three sessions: 1) initial familiarisation: questionnaires, anthropometry measures, DEXA scan, and a graded exercise test; 2) protocol 1: moderate-intensity continuous exercise; and 3) protocol 2: alternating high- and low-intensity exercise. Participants were randomly allocated to the acute exercise protocols. Each protocol had a fasted pre-exercise venous blood sample, 35-minutes cycle, fasted blood sample immediately post-, 2hrs post-, and 24hr post-exercise. RESULTS Significant differences were observed between groups in body mass index, waist and hip circumstances, percentage of fat mass, and VO2peak (p < .05). Analyses of venous blood samples will be presented. CONCLUSION With no known cure, lifestyle management is important for those with GI disorders. Exercise hosts several benefits; however, given the inflammatory nature of the condition, the acute responses of exercise are important for clinicians working with IBD/IBS patients.

Abstract:PURPOSE: To explore the effects of exercise preconditioning on serum free mtDNA and mitochondria-associated immune proteins in skeletal muscles under acute centrifugation injury. METHODS: Eight-week-old C57BL male mice were randomly divided into three groups according to their body weight. Each group consisted of 6 mice (NC, RC, RE). Exercise pre-adaptation program: take a non-slope 20 m/min treadmill training for 40 min/d, training for 6 weeks. Acute Sports Injury Program: In the 6th week of exercise training in RE group, 3 days of 10 m/min and 5 minutes of adaptive training without gradients on the RC group were started in the RC group; all mice in the RC and RE groups took 16 m/min,-16 ° Downhill running, training 90 min. After 16 hours of injury training, the mice were sacrificed, and one side of the gastrocnemius muscle was fixed with paraformaldehyde and awaited for HE staining. The tissue samples were detected by PCR, RT-PCR, Western Blotting, and ELISA. RESULTS:1) Serum free mtDNA content in RC group was significantly higher than NC and RE group (P < 0.05) 2) IL6 levels in mouse serum were significantly lower in NC group than in RC and RE group (P < 0.05, P < 0.01); The group was significantly higher than the NC group and RE group (P < 0.05, P < 0.01); 3) The results of STING mRNA detection showed that NC group was higher than RE (P < 0.05) There was no significant difference between RC group and RC group, but RC group was significantly higher than that of RE group (P < 0.01). The results of mRNA expression tests of other molecules in STING pathway showed that only IRF3 expression was significantly lower in RE group than in NC group. (P < 0.05);4) The expression of PINK1 in skeletal muscle was highest in the RC group and significantly higher than that in the RE group (P < 0.01). Parkin was the highest in NC group in skeletal muscle, (P < 0.01). CONCLUSIONS: (1) Increase in serum free mtDNA content caused by acute centrifugation injury exercise is not a cause of inflammatory reaction to skeletal muscle injury, because acute centrifugation injury inhibits the transcription of mitochondria-associated immune proteins in skeletal muscle, and cGAS-STING-TBK1-IRF3 - The IFN-β signaling pathway is not activated. (2) Acute centrifugal injury exercise and exercise preconditioning did not affect PINK1/Parkin-mediated mitochondrial autophagy.
Involvement of neutrophils and macrophages in exhaustive exercise-induced liver, kidney, heart, and lung injuries

Abstract:Moderate exercise is effective for maintaining or improving overall health. However, excessive exercise that exhausts the adaptive reserve of the body or its ability to positively respond to training stimuli can induce tissue damage and dysfunction of multiple organs and systems. Tissue injury, inflammation, and oxidative stress are reportedly induced in the skeletal muscles, liver, and kidneys after exercise. However, the precise mechanisms underlying acute tissue injury after intense exercise have not yet been fully elucidated. Studies using various experimental models of acute tissue injury, other than intense exercise, have demonstrated infiltration of inflammatory cells, including neutrophils and macrophages. These cells infiltrate injured tissues and induce inflammatory and oxidative stress responses by producing inflammatory cytokines and reactive oxygen species, thereby exacerbating tissue injury. In addition to the activation of blood neutrophils and increase in their levels during and/or after prolonged or intense exercise, chemokines that contribute to leukocyte migration are released, facilitating the migration of neutrophils and monocytes into tissues. Therefore, neutrophils and macrophages, activated by exhaustive exercise, may infiltrate tissues and contribute to exhaustive exercise-induced tissue injury. Recently, the contributions of neutrophils and macrophages to various tissue injuries caused by exhaustive exercise have been reported. In this review, we summarize the involvement of neutrophils and monocytes/macrophages in exhaustive exercise-induced non-skeletal muscle tissue injury. In addition, we present novel data demonstrating the contribution of neutrophils and macrophages to exhaustive exercise-induced cardiac and pulmonary injuries. Our study findings and the evidence presented in this review suggest that neutrophils and macrophages may play pivotal roles in exhaustive exercise-induced tissue injuries.

Abstract:Muscle pain affects approximately 11-24% of the global population. Several studies have shown that exercise is a non-pharmacological therapy to pain control. It has been suggested that the endocannabinoid system is involved in this antinociceptive effect. However, the participation of this pathway is unclear. The present study aimed to investigate whether spinal cannabinoid CB2 receptors participate in the exercise-induced antinociception. The inflammatory muscle pain model was induced by the intramuscular injection of carrageenan. Tactile allodynia and thermal hyperalgesia were determined with the von Frey filaments and hot-plate tests. C57BL/6J female mice underwent a swimming training protocol that lasted 3 weeks. This protocol of exercise reduced carrageenan-induced tactile allodynia and thermal hyperalgesia and this effect was prevented by the cannabinoid CB2 receptors inverse agonist AM630 and potentiated by MAFP (inhibitor of the enzyme that metabolizes endocannabinoids) and minocycline (microglia inhibitor). In addition, exercise increased the endocannabinoid anandamide levels and cannabinoid CB2 receptors expression whereas it reduced Iba1 (microglial marker) protein expression as well as pro-inflammatory cytokines (TNF-α and IL-1β) in the spinal cord of mice with inflammatory muscle pain. Swimming training also reduced muscle temperature of carrageen-treated animals. The present study suggests that activation of spinal cannabinoid CB2 receptors and reduction of activated microglia are involved in exercise-induced antinociception.
Interleukin‐10 responses from acute exercise in healthy subjects: A systematic review

Abstract:PURPOSE: Interleukin 10 (IL-10) is a cytokine that plays a critical role with potent anti-inflammatory properties when produced during exercise, limiting host immune response to pathogens and preventing tissue damage. The purpose of this systematic review was to assess the response of IL-10 after acute exercise session in healthy adults.METHODS: Databases of Ovid Medline (1978-2016), CINAHL (1998-2016), EMBASE (2003-2016), SportDiscus (1990-2016), and Web of Science library (1990-2016) were carefully screened. Clinical trials comparing exercise types in healthy individuals were included for pooled analysis. The trials of exercise were methodologically appraised by PEDro Scale.RESULTS: Twelve randomized controlled and crossover trials containing 176 individuals were identified for inclusion. The Kruskal-Wallis test showed no significant differences between type of exercise and the corresponding values in IL-10 [X2(4) = 2.878; p = 0.449]. The duration of exercise was significantly correlated with increase in IL-10 changes (Pearson's r = 1.00, 95%CI: 0.015-0.042, p < 0.0001) indicating that 48% of the variation in IL-10 levels can be explained by the duration of the exercise performed. In addition, despite a linear increase, we did not find a significant correlation with the intensity of exercise and IL-10 changes (Pearson's r = 0.218, 95%CI: -0.554-0.042, p < 0.035).CONCLUSION: Overall, the duration of the exercise is the single most important factor determining the magnitude of the exercise-induced increase of plasma IL-10.

Abstract:Abstract The physiological effects of physical exercise are ubiquitously reported as beneficial to the cardiovascular and musculoskeletal systems. Exercise is widely promoted by medical professionals to aid both physical and emotional wellbeing; however, mechanisms through which this is achieved are less well understood. Despite numerous beneficial attributes, certain types of exercise can inflict significant significant physiological stress. Several studies document a key relationship between exercise and immune activation. Activation of the innate immune system occurs in response to exercise and it is proposed this is largely mediated by cytokine signalling. Cytokines are typically classified according to their inflammatory properties and evidence has shown that cytokines expressed in response to exercise are diverse and may act to propagate, modulate or mitigate inflammation in musculoskeletal health. The review summarizes the existing literature on the relationship between exercise and the immune system with emphasis on how exercise-induced cytokine expression modulates inflammation and the immune response.
Human inflammatory and resolving lipid mediator responses to resistance exercise and ibuprofen treatment

Abstract:Classical proinflammatory eicosanoids, and more recently discovered lipid mediators with anti-inflammatory and proresolving bioactivity, exert a complex role in the initiation, control, and resolution of inflammation. Using a targeted lipidomics approach, we investigated circulating lipid mediator responses to resistance exercise and treatment with the NSAID ibuprofen. Human subjects undertook a single bout of unaccustomed resistance exercise (80% of one repetition maximum) following oral ingestion of ibuprofen (400 mg) or placebo control. Venous blood was collected during early recovery (0-3 h and 24 h postexercise), and serum lipid mediator composition was analyzed by LC-MS-based targeted lipidomics. Postexercise recovery was characterized by elevated levels of cyclooxygenase (COX)-1 and 2-derived prostanoids (TXB2, PGE2, PGD2, PGF2α, and PGI2), lipooxygenase (5-LOX, 12-LOX, and 15-LOX)-derived hydroxyeicosatetraenoic acids (HETEs), and leukotrienes (e.g., LTB4), and epoxygenase (CYP)-derived epoxy/dihydroxy eicosatrienoic acids (EpETrEs/DiHETrEs). Additionally, we detected elevated levels of bioactive lipid mediators with anti-inflammatory and proresolving properties, including arachidonic acid-derived lipoxins (LXA4 and LXB4), and the EPA (E-series) and DHA (D-series)-derived resolvins (RvD1 and RvE1), and protectins (PD1 isomer 10S, 17S-diHDoHE). Ibuprofen treatment blocked exercise-induced increases in COX-1 and COX-2-derived prostanoids but also resulted in off-target reductions in leukotriene biosynthesis, and a diminished proresolving lipid mediator response. CYP pathway product metabolism was also altered by ibuprofen treatment, as indicated by elevated postexercise serum 5,6-DiHETrE and 8,9-DiHETrE only in those receiving ibuprofen. These findings characterize the blood inflammatory lipid mediator response to unaccustomed resistance exercise in humans and show that acute proinflammatory signals are mechanistically linked to the induction of a biological active inflammatory resolution program, regulated by proresolving lipid mediators during postexercise recovery.

Abstract:Exercise-induced muscle damage (EIMD) results in transient muscle inflammation, strength loss, muscle soreness and may cause subsequent exercise avoidance. Omega-3 (n-3) supplementation may minimise EIMD via its anti-inflammatory properties, however, its efficacy remains unclear.

Abstract:Skeletal muscle makes up almost half the body weight of heathy individuals and is involved in several vital functions, including breathing, thermogenesis, metabolism, and locomotion. Skeletal muscle exhibits enormous plasticity with its capacity to adapt to stimuli such as changes in mechanical loading, nutritional interventions, or environmental factors (oxidative stress, inflammation, and endocrine changes). Satellite cells and timely recruited inflammatory cells are key actors in muscle homeostasis, injury, and repair processes. Conversely, uncontrolled recruitment of inflammatory cells or chronic inflammatory processes leads to muscle atrophy, fibrosis and, ultimately, impairment of muscle function. Muscle atrophy and loss of function are reported to occur either in physiological situations such as aging, cast immobilization, and prolonged bed rest, as well as in many pathological situations, including cancers, muscular dystrophies, and several other chronic illnesses. In this review, we highlight recent discoveries with respect to the molecular mechanisms leading to muscle atrophy caused by modified mechanical loading, aging, and diseases. We also summarize current perspectives suggesting that the inflammatory process in muscle homeostasis and repair is a double-edged sword. Lastly, we review recent therapeutic approaches for treating muscle wasting disorders, with a focus on the RANK/RANKL/OPG pathway and its involvement in muscle inflammation, protection and regeneration processes.

Effects of Curcumin on Inflammatory Response During Exercise-Induced Muscle Damage (Literature Review)

Abstract:This study aimed to evaluate the effect of curcumin on the inflammatory response during exercise-induced muscle damage (EIMD) is a literature review. The method applied to this article was a comprehensive strategy such as searching for articles in research journal databases. The databases were taken from Pubmed/MEDLINE, Scopus, Web of Science, and Embase. The keywords were curcumin, inflammation, Exercise-Induced Muscle Damage, Sport, Exercise, and Healthy Lifestyle. There were 30 articles obtained, and 10 articles were analyzed through the purpose, suitability of the topic, sample size, research protocol, and the results of each article. The results of this study explain that curcumin was able to provide anti-inflammatory effects by reducing pro-inflammatory cytokines such as IL-6, IL-8, TNF-α. Curcumin can also reduce muscle pain intensity, decrease CK activity, and increase ROM. The curcumin dose (>180 mg/day) showed to reduce various inflammatory responses due to EIMD. Therefore, it is recommended that curcumin be used in individuals who carry out physical activities, leading to muscle damage and inflammation.

Abstract:Responding to tissue injury, skeletal muscles undergo the tissue destruction and reconstruction accompanied with inflammation. The immune system recognizes the molecules released from or exposed on the damaged tissue. In the local minor tissue damage, tissue-resident macrophages sequester pro-inflammatory debris to prevent initiation of inflammation. In most cases of the skeletal muscle injury, however, a cascade of inflammation will be initiated through activation of local macrophages and mast cells and recruitment of immune cells from blood circulation to the injured site by recongnization of damage-associated molecular patterns (DAMPs) and activated complement system. During the inflammation, macrophages and neutrophils scavenge the tissue debris to release inflammatory cytokines and the latter stimulates myoblast fusion and vascularization to promote injured muscle repair. On the other hand, an abundance of released inflammatory cytokines and chemokines causes the profound hyper-inflammation and mobilization of immune cells to trigger a vicious cycle and lead to the cytokine storm. The cytokine storm results in the elevation of cytolytic and cytotoxic molecules and reactive oxygen species (ROS) in the damaged muscle to aggravates the tissue injury, including the healthy bystander tissue. Severe inflammation in the skeletal muscle can lead to rhabdomyolysis and cause sepsis-like systemic inflammation response syndrome (SIRS) and remote organ damage. Therefore, understanding more details on the involvement of inflammatory factors and immune cells in the skeletal muscle damage and repair can provide the new precise therapeutic strategies, including attenuation of the muscle damage and promotion of the muscle repair.

Abstract:In some asthmatic subjects exercise is followed by a bronchospastic response that generally lasts 30-60 minutes and regresses spontaneously. The initiating stimulus is thought to be the cooling and/or dehydration of airways caused by hyperpnea. The mechanisms leading from thermodynamic changes to airway narrowing are object of controversy. Two hypotheses have been proposed, one suggesting that an increase in the osmolarity of airway lining fluid following water loss causes mediator release, and the other suggesting that an excessive vasodilation during airway rewarming causes vascular engorgement, thus reducing airway caliber. Other controversial issues in exercise-induced asthma are the role of airway inflammation and the question whether a late-phase response may occur. That inflammatory cells and mediators play a role is suggested by increased numbers of eosinophil and epithelial cells in bronchoalveolar lavage and by the efficacy of cromolyn and leukotriene antagonists in preventing EIA. The existence of late-phase response to exercise is questionable because exercise does not cause an increase in airway responsiveness, which usually accompanies late-phase responses to allergens. Furthermore, data of bronchoalveolar lavage and bronchial biopsy suggest that the delayed broncho-constriction that may be observed after exercise reflects airway instability in subjects with more prominent eosinophilic inflammation of airways and is not specific to exercise.

Abstract:Dyslipidemia and inflammation are frequently found in some diseases, such as obesity, type 2 diabetes mellitus, and cancer cachexia. Recent literature has identified that lipids have a pivotal role in the activation of inflammatory pathways, increasing the production of inflammatory cytokines, mainly tumor necrosis factor alpha, interleukin 6 and 1β. On the other hand, cytokines can promote disruption of lipid metabolism, in special cholesterol reverse transport, which is linked to development of atherosclerosis. With this in mind, acute and chronic exercise trainings have been pointed as important tools to counteract both dyslipidemia symptoms and systemic inflammation. Moreover, physical activity has been recommended in the prevention/treatment of the above mentioned outcomes by important health organizations around the world, mainly because it costs less and generates fewer side effects than isolated medicine. Despite the well-documented capacity of acute and chronic exercise training to counteract sustained disease-related immunometabolism, we have chosen to take a look from a current perspective in molecular pathways and in the field of epidemiology. The aim of the present review was therefore to discuss the results of dyslipidemia and inflammatory conditions with acute and chronic exercise training, which underlies the field of molecular pathways and epidemiology. The mechanisms underlying the response to the treatment are considered.
