Abstract:Synthesis and application of azacycloalk-1-enefused oxazol-3-ium salts are summarized. The microreview covers all synthetic methods for obtaining 6,7-dihydro-5 H -pyrrolo[2,1- b ]oxazol-4-ium and 5,6,7,8-tetrahydrooxazolo[3,2- a ]-pyridin-4-ium derivatives that have been classified in two distinct categories: cyclization leading to the formation of azacycloalk-1-ene ring and cyclization leading to the formation of oxazole ring. Application of these azacycloalk-1-enefused oxazol-3-ium salts has been discussed.

Abstract:Background: Although the chemistry of heteroaromatic monoamino azoles has been surveyed more than once in the last decade, the chemistry of the di- and triaminoazoles has not been reviewed. In this article we will survey the synthesis, chemistry and utility of the diaminoazoles. In this review, the chemistry of the diaminoazoles as well as their most important utilities will be surveyed. Objective: The review focuses on recent progress in diaminoazoles (i.e. diaminopyrazoles, diaminoimidazoles, diaminotriazoles and diaminothiazole) with especial references to diaminopyrazoles. The synthesis as well as pharmaceutical utilities are reported. There are several methods for synthesis of diaminopyrazoles. 3,5- Diaminopyrazole and its derivatives are prepared through the reaction of malononitrile or arylhydrazononitrile with hydrazine derivatives. 3,4-Diaminopyrazoles are prepared via nitration of 3-aminopyrazole with subsequent reduction of the produced compound. The diaminopyrazoles have several applications in cosmetic and pharmaceutical industries. They also have useful utilities as a constituent in oxidative hair dyes. Conclusion: We managed to report the common methods for the synthesis of diaminoazoles with especial reference to aminopyrazoles that are prepared through the reaction of malononitrile or hydrazononitriles with hydrazine derivatives. Some important applications that include pharmaceutical utilities such as hair dye constituents are reported.

Abstract:The focus of this review is on the synthetic routes available for different types of pyrazoloquinoline derivatives. There are three types of synthetic methods: i) from pyrazole derivatives; ii) from quinoline derivatives; and iii) miscellaneous methods. The position of the linkage between pyrazole and quinoline rings determines the seven isomers of pyrazoloquinolines. The purpose of this review is to provide a guide for both synthetic and medicinal chemists to discover and design new pyrazoloquinolines for medical purposes.

Abstract:Aryl diazonium salts are versatile building blocks in organic synthesis. In light of the ever-increasing importance of aryl diazonium salts spanning most disciplines of the chemical sciences, we review the recent development of aryl diazonium chemistry over the past seven years (2013-2020). Special emphasis is put on various new transformations involving the generation of radical intermediates via thermal, photochemical, and electrochemical means. Recent advances in the development of transition metal-catalyzed reactions using aryl diazonium salts are also reviewed. Together, these newly developed transformations significantly expand the synthetic chemist's repertoire of aromatic carbon-carbon and carbon-heteroatom bond forming methods using aryl diazonium precursors, providing powerful tools for the synthesis and modification of complex molecular scaffolds.

Abstract:Quinoline and pyrimidine moieties are ubiquitous components in both natural and synthetic compounds, showcasing diverse applications. The fusion of these well‐known structures into hybrid molecules has garnered attention due to their intriguing biological properties. Particularly in the field of medicinal chemistry, numerous studies in the last decade have focused on pyrimido[5,4‐c]quinoline ring systems (PyQs5,4‐c). This review elucidates the synthesis of PyQs5,4‐c and their derivatives using 3,4‐difunctionalized quinoline as a key starting material. The preparation of PyQs5,4‐c involves a series of chemical transformations, including the Friedländer, Ullmann andBiginelli reaction, Vilsmeier‐Haack formylation, Suzuki coupling, and a one‐pot three‐component reaction. These synthetic routes not only offer access to diverse PyQs5,4‐c derivatives but also contribute to the expanding arsenal of methods in the synthesis of biologically relevant compounds.

Abstract:This paper uses basic-catalysed method to convert 1H-pyrazole-4-carboxamides into tri-fused heterocyclic systems containing two kinds of pyrazolotriazolopyrimidine framework. The synthesis of compounds 5-6 and reactions of them with some electrophiles have been investigated. Preliminary bioassay study showed that some compounds displayed high activity.

Abstract:Chromones, six-membered oxygen heterocycles, and pyrazoles, five-membered two-adjacent-nitrogen-containing heterocycles, represent two important classes of biologically active compounds. Certain derivatives of these scaffolds play an important role in medicinal chemistry and have been extensively used as versatile building blocks in organic synthesis. In this context, we will discuss the most relevant advances on the chemistry that involves both chromone and pyrazole rings. The methods reviewed include the synthesis of chromone-pyrazole dyads, synthesis of chromone-pyrazole-fused compounds, and chromones as starting materials in the synthesis of 3(5)-(2-hydroxyaryl)pyrazoles, among others. This review will cover the literature on the chromone and pyrazole dual chemistry and their outcomes in the 21st century.

Abstract:The bicyclic 1-aza-2-azoniaallenium salt intermediates, generated from the azoester species upon treatment with a Lewis acid, have been demonstrated to participate in Huisgen-type cycloaddition with nitriles to result in the formation of fused 6,7,8,9-tetrahydro-5 H-[1,2,4]triazolo[1,5- d][1,4]diazepinium salts. This transformation is interpreted as a regular [3++2] cycloaddition between intermediates as the reactive 1,3-monopole reactants and nitriles as the nucleophilic reagents followed by spontaneous [1,2]-cationic rearrangement. The azoester precursors were easily accessible via oxidation of the corresponding hydrazones using hypervalent iodine oxidant PhI(OAc)2 under mild conditions. The [1,2,4]triazolodiazepine compounds represent a class of N-containing biologically important heterocycles with a new type of scaffold.

Abstract:Heteroarene 1, n-zwitterions are powerful and versatile building blocks in the construction of heterocycles and have received increasing attention in recent years. In particular, pyridinium and quinolinium 1,4-zwitterions have been widely studied and used in a variety of cyclization reactions due to their air stability, ease of use, and high efficiency. Sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions, types of emerging heteroatom-containing synthons, have attracted much attention from chemists. These 1,4-zwitterions, which contain multiple reaction sites, have been successfully used in the synthesis of three- to eight-membered cyclic compounds over the last decade. In this review, we present the exciting progress made in the field of cyclization reactions of sulfur- and nitrogen-based pyridinium and quinolinium 1,4-zwitterions. Moreover, the mechanistic insights, the transition states, some synthetic applications, and the challenges and opportunities are also discussed. We hope to provide an overview for synthetic chemists who are interested in the heterocycle synthesis from cyclization reaction with pyridinium and quinolinium 1,4-zwitterions pyridinium and quinolinium 1,4-zwitterions.

Abstract:Nitrogen- and sulfur-containing heterocyclic compounds widely existed in bioactive natural products and drug molecules with unique bioactivity. The synthesis of nitrogen-and sulfur-containing hybrids had always been a hot topic in the field of synthetic chemistry, and chemists had been committed to developing convenient and efficient strategies to synthesize these compounds. Pyridinium zwitterions with a unique reactivity played a significant role in the construction of nitrogen- and sulfur-containing heterocyclic compounds. In recent years, pyridinium zwitterions had been extensively used in cyclization reactions for the synthesis of novel heterocyclic compounds. This review summarized the recent advances of pyridinium 1,n-zwitterions (n = 4 and 5) participating in cyclization reactions and highlighted the applications of pyridinium 1, n-zwitterions (n = 4 and 5) in the efficient construction of heterocycles.

Abstract:Aziridines are the saturated three-membered cyclic amines that constitute an important group of synthetic intermediates. These could act as a precursor for diverse organic compounds owing to the reactivity due to the ring strain associated with them. The outstanding property of aziridines is their high reactivity towards various nucleophilic and electrophilic reagents to acquire more stable ring-opened or ring-expanded amines that could be obtained from the release of strain energy intrinsic in a small ring. As such, aziridines could be used in the synthesis of 4-7 membered heterocycles of biological and industrial significance, such as azetidines, imidazoles, thiazoles, pyrazines, pyrimidines, benzothiazines, benzodiazepines, etc. Earlier synthesis of aziridines was considered a laborious task due to their instability. However, various synthetic approaches leading to the formation of aziridines are now available in the literature. Recently, green, cost-effective and approaches based on simpler work-up for these reactions have attracted researcher's attention. This review article deals with synthetic routes of aziridines and aziridine applications in organic synthesis.

Abstract:This feature article aims to review the evolution and recent advances in different protocols for synthesizing fused hybrids of N‐heterocycles, specifically [1,2,3]triazolo[1,5‐a] quinoline. Approaches to synthesize this unit will be discussed, including dehydrogenative cyclization, N−N oxidative coupling, Dieckmann condensation, intramolecular Heck reactions, (3+2) cycloaddition, Ullmann‐type coupling, and direct intramolecular arylation reactions. This report outlines the evolution and recent progress about the different protocols to synthesize the N‐heterocycles fused hybrids, specifically [1,2,3]triazolo[1,5‐a]quinoline. This review encompasses a broad range of approaches, describing several reactions for obtaining this since, such as dehydrogenative cyclization, oxidative N−N coupling, Dieckmann condensation, intramolecular Heck, (3+2)‐cycloaddition, Ullman‐type coupling and direct intramolecular arylation reactions. We divided this review in three section based in the starting materials to synthesize the target [1,2,3]triazolo[1,5‐a]quinolines. Starting materials containing quinoline or triazole units previously formed, as well as starting materials which both quinoline and triazole units are formed in situ. Different methods of obtaining are described, such as metal‐free or catalyzed conditions, azide‐free, using conventional heating or alternative energy sources, such as electrochemical and photochemical methods. Mechanistic insights underlying the reported reactions were also described in this comprehensive review.

Abstract:5,6,7,8-tetrahydro-4H-pyrazolo[1,5-a] diazepine carboxylates are valuable scaffolds for drug design and medicinal chemistry. In this paper we disclose a short scalable synthesis and chemical reactivity patterns of the pyrazolo-diazepine pharmacophore. Commercial and cheaply available Methyl pyrazole 3,5-dicarboxylate was alkylated with 3-bromo-N-Boc propyl amine and the resulting derivative underwent concomitant cyclization upon deprotection of the Boc protecting group to yield the pyrazolo-diazepine skeleton. Selective reduction of the lactam was accomplished cleanly using borane and the resulting amine was pro-tected using a tert-butyloxycarbonyl protecting group. The free N-terminal of the diazepine underwent smooth Buchwald and Chan arylations among various standard chemistry applications examined on this pharmacophore.

Abstract:1-Alkyl-N-Boc-5-formylpyrazol-4-amines react with malononitrile and cyanoacetamide to form 5-amino-1H-pyrazolo[4,3-b]pyridine nitriles and amides, respectively. Cyclocondensation of the obtained derivatives with chloroacetaldehyde, bromotrifluoroacetone, or ethyl bromopyruvate leads to the formation of imidazo[1,2-a]pyrazolo[3,4-e]pyridines or to pyrazolo[3′,4′,5,6]pyrido[2,3-d]pyrimidine derivatives in cyclocondensation with orthoesters, ethyl oxalyl chloride, or carbonyldiimidazole.

Abstract:The method for the preparation of 3,6-disubstituted 1,2,4-triazines based on condensation reaction between easily available α-imino esters and isonitrosoacetophenone hydrazones was reported. The significant differences in reaction conditions, ratio of products, and yields between the developed method and the earlier reported approach were demonstrated. The corresponding 2,5-disubstituted pyridines were synthesized from the prepared 1,2,4-triazines, and their photophysical properties were studied. Studies of the photo-physical properties revealed low and moderate luminescence quantum yields, and negligible solvatochromic behavior in case of 4-methoxyphenylpyridine derivative due to the role of donating methoxy group, however, with a low linearity of a Lippert–Mataga plot. Nevertheless, 2,5-disubstituted pyridines are of interest due to simple protocols of synthesis, moderate photophysical properties, and potential applicability in different scientific and industrial areas.
