Abstract:Extracellular polymeric substances (EPS) have a significant effect on the dewaterability of waste-activated sludge (WAS); however, the role of polarity distribution (hydrophilic/hydrophobic characteristics) of EPS in sludge dewatering remains unclear. In this study, the relationship between the characteristics of EPS hydrophilic/hydrophobic fractions (composition and molecular structure) and sludge physicochemical properties of 8 different sludge samples was investigated, and a structural equation model was constructed to describe the potential mechanisms of key EPS parameters on sludge dewatering. Experimental results showed that the variation in sludge EPS polarity led to significant changes in the surface hydrophilicity/hydrophobicity and structure compactness, which affected the sludge dewaterability. Specifically, the increase in the content of tyrosine-like proteins, especially those related to hydrophobic acid fraction (HPO-A), played a key role in increasing the surface hydrophobicity of sludge and promoting its dewaterability. In contrast, humic acids in hydrophilic EPS (HPI) could interact with hydrophobic proteins to form dense biopolymers (characterized by high nonprotonated nitrogen (N nonpr ) content), resulting in a more stable and compact structure of the EPS matrix (characterized by low random coil content) and deterioration of sludge dewaterability. This study provides a new perspective for the exploration of the relationship between hydrophilic/hydrophobic biopolymers and sludge dewaterability.

Abstract:Extracellular polymeric substances (EPS) are known to crucially affect the properties and performance of activated sludge, but the detailed influential mechanisms and the pertinence to specific compositional, structural properties of EPS are still elusive. Such knowledge gaps have severely limited our ability in optimizing biological wastewater treatment processes, for which long-term robust and efficient sludge performance remains one of the main bottlenecks. Here, we overview the new knowledge on the molecular structure of sludge EPS gained over the past few years and discuss the future challenges and opportunities for further advancing EPS study and engineering. The structural and functional features of several macromolecules in sludge EPS and their important structural roles in granular sludge are analyzed in detail. The EPS-pollutant interactions and environment-dependent regulation machinery on EPS production are deciphered. Lastly, the remaining knowledge gaps are identified, and the future research needs that may lead to molecular-level understanding and precise engineering of sludge EPS are highlighted.
Correlation and mechanism of extracellular polymeric substances (EPS) on the effect of sewage sludge electro-dewatering

Abstract:Electro-dewatering (ED) is an attractive technology for enhancing dewaterability of waste activated sludge (WAS), and the distribution and composition of EPS and secondary structure of extracellular protein in sludge has a great influence on the sludge dewaterability. Therefore, in order to optimize and regulate sludge ED process, it is necessary to study the influence of EPS components and composition on the efficiency of sludge ED. In this study, the effects of distribution and composition of EPS and the structure of extracellular proteins on the ED rates and performance were analyzed using eight sludges from different sewage treatment process. The results showed that ED rates at both electrodes were significantly negatively correlated with the concentration of soluble EPS (S-EPS) and loosely bound EPS (LB-EPS), and this correlation was gradually weakened with the in-depth structure of layered EPS structure. High concentration of S-EPS and LB-EPS decreased the initial current and the pH gradient, deteriorating the anodic oxidation and acidification, which affect the release and degradation of EPS. Additionally, the proteins secondary structure can be destroyed and transformed by anodic oxidation and acidification, which can impact the protein water-holding capacity and ED performance.

Abstract:Extracellular polymeric substances (EPS) play an important role in biological wastewater treatment. The influence of extracted EPS in aerobic activated sludge was investigated, with EPS extraction yields represented by total organic carbon. Polysaccharides and proteins in EPS were analyzed to the major component of EPS in activated sludge. Results showed that the difference of treatment technology and culture medium among activated sludge can induce the change of component and content of extracted EPS, especially the ratio between proteins and polysaccharides.

Abstract:Laboratory experiments were carried out on activated sludge (AS) to investigate the correlations between the content of extracellular polymeric substances (EPS) and the performance of biosolids–water separation, including sludge flocculation, sedimentation, compression, and dewatering, under non-steady-state conditions. On three stabilized AS reactors changes were made in sludge retention time (SRT), substrate composition, and loading rate, respectively, to bring about unstable operation to the reactors. A two-step heating method was used to extract from the sludge the easily extractable EPS, or loosely bound EPS (LB-EPS), and tightly bound EPS (TB-EPS), respectively. The experimental results demonstrate dynamic changes in sludge characteristic and EPS production under the non-steady-state conditions. During the early phase of transition after a change was imposed, the sludge became generally worse in flocculation, compressibility, and dewaterability. With the acclimatization of the biomass to the new process conditions, biosolids–water separation showed a general trend of improvement. Changes in AS process condition also resulted in considerable variations in EPS production. The change of the LB-EPS content appeared to be more significant than that of the TB-EPS. Throughout the non-steady-state operation, the sludge flocculating behavior, settleability, compressibility, and dewaterability had a positive correlation with the LB-EPS content; however, no correlation could be found between these properties and the TB-EPS content. The results suggest that although EPS is essential to biofloc formation, excessive EPS in the form of LB-EPS would weaken cell attachment and deteriorate the AS floc structure, resulting in poor biosolids–water separation.

Abstract:Extracellular polymeric substances (EPS) are major high-molecular-weight secretions from microorganisms, binding with cells to form a vast net-like structure for protecting cells against external stress. In this study, the level and function of EPS during the granulation of aerobic sludge used to treat p-chloroaniline (p-ClA) wastewater were investigated. The content of extracellular proteins (PN) increased relative to the exopolysaccharides (PS) content during the sludge granulation (Stage I), implying that PN contributed significantly to the formation of aerobic granular sludge (AGS). Under the condition of high p-ClA shock loading (Stage II), the PS content increased from 122.7±22.0mgg−1 VSS to 482.6±5.4mgg−1 VSS, which might be a regulating mechanism of AGS to address high p-ClA loading. The results of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) demonstrated that PN bands increased with the development of sludge granulation, particularly the proteins with molecular weight located at 44.3–97.2kDa. Fourier transform infrared (FTIR) spectra of inoculated activated sludge and matured granular sludge indicated that microorganisms appeared to aggregate in the presence of tyrosine protein, aspartic acid protein and proteins with secondary structures including 3-turn helix and β-sheets. Five types of proteins were identified by two-dimensional electrophoresis (2-DE), and these proteins were related to the granulation of aerobic sludge and EPS secretion. Furthermore, the analysis of sludge microbial community revealed that the enrichment of Zoogloea spp. and Thauera spp. as p-ClA degrading and EPS secreting microbes was conducive to the formation of granular sludge and degradation of p-ClA.

Abstract:Extracellular polymeric substances (EPS) are the key components determining the dewatering behavior of wastewater sludge. However, current technical optimization of sludge conditioning for dewatering is limited by the poor understanding of the conditioner-EPS interactions at molecular levels. Herein, a combination of molecular dynamic (MD) simulations, dewaterability assessment and EPS characterization was used to reveal the sludge dewatering mechanisms using dual conditioning processes (prevalent inorganic (poly aluminum chloride (PAC)) and organic (poly dimethyl diallyl ammonium chloride (PDDA)). Results suggested that PAC and PDDA bridged the biopolymers mainly through electrostatic interactions, promoting the agglomeration of biopolymers and reducing their contact probability with water molecules. Water molecules were tightly bound to EPS mainly through hydrogen bonding with polar oxygen-containing functional groups. The adsorption of PAC and PDDA on hydrophilic components reduced the molecular polarity of biopolymers and altered the conformation of water molecules in the hydration shell, resulting in a decreased hydration capacity of EPS and the release of bound water, and sludge dewaterability was improved. PAC was found to be more effective than PDDA in disrupting the hydrogen bonding between water molecules and EPS, especially the protein β-sheet structure inside the molecular clusters with its high charge strength and diffusivity. Sludge bound water decreased by 73.16 % after PAC conditioning. In addition, PDDA exhibited superior agglomeration ability to biopolymers and promoted the electrostatic interaction between PAC and polar groups during dual conditioning. The strength and hydrophobicity of EPS molecular clusters were thus enhanced, and the conditioning efficiency was improved. This study offers molecular-level insights into the coagulation treatment process of sludge and provides theoretical references for process optimization and new conditioner development.

Abstract:Activated sludge is a complex wastewater purification process involving microorganisms, extracellular polymeric substances (EPS), and organic matter. Although this process has been utilized for over a century, our understanding of the relationships between the components of activated sludge and wastewater treatment plant (WWTP) metrics is still evolving. Polysaccharides play significant roles in water retention within loose-bound EPS and soluble EPS, negatively impacting dewaterability and flocculation. Conversely, proteins, primarily found in tightly bound EPS, are sensitive to pH and contribute positively to settleability and flocculation. Studies on eDNA are limited but suggest it has key functions in stabilizing the matrix and interacting with polymers. There is considerable information on how proteins and sugars affect sludge volume index, capillary suction time, and flocculation. Additionally, some studies investigated the biochemistry and molecular biology of activated sludge across temporal, geographical, and laboratory settings. However, there are few studies identifying trends between specific sugars, proteins, bacterial taxa, and WWTP metrics. This review explores the intermolecular forces binding these components together and the roles of proteins, polysaccharides, eDNA, and bacterial taxa in the sludge. It also examines the relationships between these sludge components and settleability, flocculation, dewaterability, type of WWTP, and environmental trends. The review provides a holistic and integrative perspective on EPS in activated sludge systems, emphasizing the significance of different EPS fractions (TB EPS and LB EPS) and their contributions to key WWTP metrics such as dewaterability, settleability, and flocculation.
Multiple drivers and mechanisms of solid-water interfacial interactions in sludge dewatering: Roles of polarity and molecular structure of extracellular polymeric substances

Abstract:Water occurrence states in sewage sludge, influenced by sludge physicochemical properties, are crucial for sludge dewaterability and have recently been regarded as a research hotspot. Here, the multifold characteristics of sludge flocs during hydrothermal treatment, including rheological properties, solid-water interfacial interactions, and the polarity distribution and molecular structure of extracellular polymeric substances (EPS), were systematically investigated, and the impact of these characteristics on sludge dewaterability was explored in depth. Hydrothermal treatment at 80 °C and 100 °C induced the conversion of free water into bound water, while an increase in temperature to 180 °C resulted in a significant decrease in bound water content, approximately 4-fold lower than at 100 °C. In addition to the conventional view of decreased sludge surface hydrophilicity at high temperatures, the decline in bound water was associated with the reduction in sludge apparent viscosity. XAD resin fractionation identified the hydrophobic/hydrophilic EPS (HPO-/HPI) ratio as an important factor determining water occurrence states. Especially, hydrolysis of HPI-related hydrophilic proteins and subsequent increase in HPO-related tryptophan-like substances played a dominant role in reducing sludge viscosity and facilitating the release of bound water. Protein conformational analysis revealed that the disruption of α-helix structures and disulfide bonds significantly reduced EPS water-holding capacity, providing strong evidence for the potential of targeting these dense structure units to enhance sludge dewaterability. These findings provide a holistic understanding of multidimensional drivers of water occurrence states in sludge, and guide directions for optimizing sludge treatment efficiency through EPS modification.

Abstract:Results in over 200 publications related to constituents, quantities, and characteristics of extracellular polymeric substances (EPS) in activated sludge and their influences on sludge flocculation, settling, and dewatering are compiled and analyzed. Constituents and quantities of EPS are strongly dependent on the extraction process. Most results show that EPS in activated sludge comprise substantially more proteins than carbohydrates. EPS and sludge flocs carry negative zeta potentials and negative charge. Sludge hydrophobicity increases with protent content of the EPS, and an increased hydrophobicity generally leads to a better flocculation. Most data show that sludge settles less well, as measured by sludge volume index, with increased amount of EPS. Sludge settling and dewatering characteristics are affected by the zeta potential, surface charge, and hydrophobicity, as well as the presence of multivalent mineral cations. Process conditions of activated sludge such as sludge retention time, dissolved oxygen, and nutrient levels affect the EPS properties and, thus, the sludge flocculation and settling characteristics. The effects of EPS quantities, ratio of protein and carbohydrate in EPS, sludge digestion, mineral cations, chelating reagent, nutrient level, and pretreatment on sludge dewaterability are discussed.

Abstract:As the importance of fecal sludge management (FSM) is increasingly being realized, the need for adequately designed and functioning fecal sludge (FS) treatment plants is also increasing. Research to fill this gap is only emerging and dewatering is a key challenge for developing sustainable treatment solutions. This study evaluated the effect of extracellular polymeric substances (EPS) on dewaterability of FS, and how EPS and dewaterability change during anaerobic storage (as a proxy for time in onsite containment). EPS was extracted from FS and activated sludge using Na 2 CO 3 and sonication and added to sludge samples to determine the effect on dewaterability. The results confirmed that an increase in EPS had a direct impact of decreasing FS dewaterability (as capillary suction time). In this context, we evaluated FS degradation during anaerobic storage, the effect of anaerobic storage time on EPS, EPS fractions and particle size distribution, and the effect of variations in these factors on FS dewaterability. Variations in EPS, EPS fraction and particle size distribution during anaerobic storage were less than expected and average VS reduction of 20% was recorded over 7 weeks. Although anaerobic digestion was verified (biogas production), the results indicate that kinetics of degradation of FS is different from wastewater sludges. Comparatively, EPS fractions in FS were 70 – 75% lower and with higher fractions of humic-like substances than wastewater sludges. Although EPS significantly affects FS dewaterability, anaerobic storage time is not a predictor of dewaterability.

Abstract:The activated sludge process is widely used for wastewater treatment, and the settleability and dewaterability of activated sludge are crucial to the operation of wastewater treatment plants. In this study the surfactant-mediated settleability and dewaterability of activated sludge were investigated. It is found that dose of anionic surfactant sodium dodecyl sulfate (SDS) caused substantial release of extracellular polymer substances (EPS), especially proteins, from sludge. Laser light scattering (LLS) results reveal that the released EPS became stretched and more expanded when the SDS dosage was increased. The sludge dewaterability was significantly deteriorated at a dosage of 2.084g/L SDS, and meanwhile the capillary suction time (CST) and specific resistance of filtration (SRF) increased by 6.3 and 5.1 times, respectively, than the raw sludge. The correlations between the EPS contents and the sludge properties, i.e., CST, SRF, moisture content of filter-cake and sludge volume index, were evaluated and the significant roles of proteins in governing the EPS properties as well as the settleability and dewaterability of sludge were further confirmed. This study elucidates how the settleability and dewaterability of activated sludge were affected by surfactants via changing the properties and structure of the released EPS from sludge.

Abstract:Ultrasound can be used to disintegrate waste activated sludge (WAS), resulting in changes in sludge dewaterability. To observe and explain the change in sludge dewatering properties before and after ultrasonic disintegration, the specific resistance to filtration (SRF), capillary suction time (CST), content of biopolymers (proteins + polysaccharides), and free water in the sludge before and after disintegration were examined. Despite the degradation of extracellular polymeric substances (EPS), the sludge dewatering properties worsened after disintegration. This result is attributed to two changes in the sludge, one being the effect of the water retained by the biopolymers and the other being the adsorption effect of the fine sludge particles for free water.

Abstract:<p>Extracellular polymeric substances (EPS) released by bacteria play a crucial role in microbial aggregation during wastewater treatment. Various environmental factors including substrate, anaerobic and aerobic conditions might contribute to the formation and properties of EPS, and thereby affect the properties of microbial aggregation and sludge. To reveal the relationship between these environmental factors and EPS properties, the characteristics of EPS generated in four types of sludge were investigated, including anammox granular sludge (ANMX), aerobic granular sludge (AGS) fed with glucose as carbon source (AGS-GLUC), AGS fed with liquor wastewater as carbon source (AGS-LIQ), and flocculent sludge from a real sewage plant (FLOC). Results indicated a positive correlation of EPS contents with granulation of sludge. As the dominant content of EPS, an increased proteins (PN) concentration facilitated the formation of microbial aggregates and the granulation of sludge. Bacteria in anaerobic environment (i.e., ANMX) or in aerobic environment with easily biodegradable carbon sources (i.e., AGS-GLUC) would generate more PN and EPS; yet, these conditions restrained the generation of humic acid (HA). The sludge from the sewage treatment plant (i.e., FLOC) had the lowest EPS and PN content, but the highest HA, showing flocculent structure sludge. In addition, a higher protein/polysaccharide (PN/PS) ratio and a lower zeta potential of EPS were conductive to sludge granulation. The hydrophobicity of EPS confirmed via analysis of chemical structures by FTIR would improve the microbial aggregation.</p>

Comprehensive Investigation of the Relationship Between Organic Content and Waste Activated Sludge Dewaterability.

Abstract:The relationship between sludge organic fraction and its dewaterability is well known in practice. However, the formal study to reveal the underlying reason is limited. To improve understanding of the nature of organic content on sludge dewatering process, this study systematically evaluated the effects of sludge organic content on its dewaterability and revealed the underlying mechanism. Analysis of 10 waste activated sludge (WAS) samples with varying organic contents showed that capillary suction time (CST) increased linearly from 34.90 ± 0.10 s to 104.90 ± 0.30 s (R2 = 0.92, p < 0.01), whereas the solid content of centrifuge cake decreased from 21.23 %±0.45 % to 12.52 %±0.14 % (R2 = 0.89, p < 0.01) when organic fractionincreased from 35.72 % to 61.11 %. These results first confirmed that WAS dewatering performance was negatively correlated to its organic content. Then, the underlying mechanism was revealed by studying the basic physicochemical properties of WAS with various organic content. The results showed that sludge with a higher organic content generally had greater extracellular polymeric substances (EPS) content, lower density and higher negative zeta potential, which hinder the aggregation and flocculation of floc particles. These properties endow the WAS with a higher organic content generally possessed more bound water content, small pores, poorer fluidity, and stronger network strength. These characteristics can hamper the separation of water from sludge cake during dewatering. Based on which, this study discussed the potential of organic fraction as a surrogate of EPS for evaluating WAS dewaterability and indicated the organic fraction can be a useful and strong indicator of WAS dewaterability.
