Abstract:Free radicals and oxidants play a dual role as both toxic and beneficial compounds, since they can be either harmful or helpful to the body. They are produced either from normal cell metabolisms in situ or from external sources (pollution, cigarette smoke, radiation, medication). When an overload of free radicals cannot gradually be destroyed, their accumulation in the body generates a phenomenon called oxidative stress. This process plays a major part in the development of chronic and degenerative illness such as cancer, autoimmune disorders, aging, cataract, rheumatoid arthritis, cardiovascular and neurodegenerative diseases. The human body has several mechanisms to counteract oxidative stress by producing antioxidants, which are either naturally produced in situ, or externally supplied through foods and/or supplements. This mini-review deals with the taxonomy, the mechanisms of formation and catabolism of the free radicals, it examines their beneficial and deleterious effects on cellular activities, it highlights the potential role of the antioxidants in preventing and repairing damages caused by oxidative stress, and it discusses the antioxidant supplementation in health maintenance.

Abstract:Free radicals have acquired growing importance in the fields of biology and medicine. They are produced during many different endogenous and exogenous processes. Mitochondria are the main source of endogenous reactive oxygen species (ROS) produced at cell level. The overproduction of free radicals can damage macromolecules such as nucleic acids, proteins and lipids. This leads to tissue damage in various chronic and degenerative diseases. Antioxidants play a crucial role in the body’s defense against free radicals. This review concerns the main properties of free radicals, their sources and deleterious effects. It highlights the potential role of the dietary supplementation of antioxidants and discusses unsolved problems regarding antioxidant supplements in the prevention and therapy of diseases.

Abstract:In recent years, there has been a great deal of attention toward the field of free radical chemistry. Free radicals reactive oxygen species and reactive nitrogen species are generated by our body by various endogenous systems, exposure to different physiochemical conditions or pathological states. A balance between free radicals and antioxidants is necessary for proper physiological function. If free radicals overwhelm the body's ability to regulate them, a condition known as oxidative stress ensues. Free radicals thus adversely alter lipids, proteins, and DNA and trigger a number of human diseases. Hence application of external source of antioxidants can assist in coping this oxidative stress. Synthetic antioxidants such as butylated hydroxytoluene and butylated hydroxyanisole have recently been reported to be dangerous for human health. Thus, the search for effective, nontoxic natural compounds with antioxidative activity has been intensified in recent years. The present review provides a brief overview on oxidative stress mediated cellular damages and role of dietary antioxidants as functional foods in the management of human diseases.

Abstract:Free radicals (FRs) are unstable molecules that cause reactive stress (RS), an imbalance between reactive oxygen and nitrogen species in the body and its ability to neutralize them. These species are generated by both internal and external factors and can damage cellular lipids, proteins, and DNA. Antioxidants prevent or slow down the oxidation process by interrupting the transfer of electrons between substances and reactive agents. This is particularly important at the cellular level because oxidation reactions lead to the formation of FR and contribute to various diseases. As we age, RS accumulates and leads to organ dysfunction and age-related disorders. Polyphenols; vitamins A, C, and E; and selenoproteins possess antioxidant properties and may have a role in preventing and treating certain human diseases associated with RS. In this review, we explore the current evidence on the potential benefits of dietary supplementation and investigate the intricate connection between SIRT1, a crucial regulator of aging and longevity; the transcription factor NRF2; and polyphenols, vitamins, and selenium. Finally, we discuss the positive effects of antioxidant molecules, such as reducing RS, and their potential in slowing down several diseases.

Abstract:Free radicals may be involved in the aetiology of cancer and cardiovascular diseases. In epidemiological studies poor plasma levels of all essential antioxidants are associated with increased relative risks; in particular, low levels of carotene and vitamin E with the risk of cancer and ischemic heart disease, respectively. The studies suggest that for optimal synergistic protection the plasma antioxidant levels should simultaneously exceed the threshold values of 28-30 mumol/l lipid-standardized vitamin E, 40-50 mumol/l vitamin C, 0.4-0.5 mumol/l carotene and 2.2-2.8 mumol/l lipid-standardized vitamin A. However the preventive efficacy of an optional antioxidant status is still to be proven in randomized intervention trials. Although these antioxidant micronutrients may be the primary protective components of vegetable-rich 'preventive' diets, the potentials of other plant components await exploration, eg carotenoids other than beta-carotene, bioflavonoids and oxygen-sensitive B-vitamins.

Abstract:Free radicals were generally regarded as highly reactive, transient and harmful species. In fact, some of the free radicals can also be inactive, long-lived and beneficial for our health. These properties of free radicals provide future possibilities for their application in various fields. Owning to their open-shell electronic structure, free radicals exhibit unique advantages in biomedical applications, such as high reactivity, photoacoustic and photothermal conversion ability, molecular magnetic. In this review, recent progress on free radicals and their applications in cancer theranostics are presented. Typical materials that exhibit controlled generation of free radicals and their applications for photodynamic therapy (PDT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), gas therapy, hypoxic cancer treatment, photothermal therapy (PTT), photoacoustic imaging (PAI) and magnetic resonance imaging (MRI) are summarized and discussed.

Abstract:Rationale: Now a days, prescribing antioxidant drugs has become very much common. Their usefulness in certain long term, chronic conditions where oxidative stress plays a vital role is well explained. But in many other conditions, their role is yet not fully discovered or explained as well their duration of use, type of antioxidant to be used, their pros and cons as well as role of free radicals in our body need to be understood by the prescribing physicians. Up to date research on this and knowledge will change attitude and practice and therefore will have positive impact on physical, mental and economic wellbeing of the patients. Aim: It is to study the use of antioxidants in clinical practice and analyze the rationality in terms of their appropriate use, duration, cost, safety and efficacy. Methodology: This review article has been prepared by going through various literature like research articles, text books and drug indexes available on the same topic. This review article focuses on various research on the pharmacological activities of natural and synthetic antioxidant molecules as well as addresses their rationality in current practice. Discussion and Conclusion: Free radicals (FR) are atoms or molecules with unpaired electrons. Endogenous production of free radicals takes place in mitochondria, peroxisomes, endoplasmic reticulum, etc. Exogenous factors like cigarette smoking, chronic alcohol intake, toxic environmental products, etc. also contribute to the production of FR. When pro-oxidative processes overwhelm cellular antioxidant defense, there is a development of oxidative stress. Antioxidants protect cells from damages caused by free radicals. Antioxidants act as free radical scavengers and further classified into endogenous & exogenous antioxidants. Antioxidants prolong or delay the occurrence of neurodegenerative disorders, eye diseases, liver diseases, cancer and ageing. Also, antioxidants are found useful in some dermatological conditions, endometriosis and periodontitis. But studies conducted throughout world shows that free radicals are harmful but are also important in human body for many physiological processes and antioxidants though considered protective and useful may have their own harmful effects. So a balance has to be maintained to get a positive outcome in clinical practice

Abstract:This study emphasizes the critical role of antioxidants in protecting human health by counteracting the detrimental effects of oxidative stress induced by free radicals. Antioxidants-found in various forms such as vitamins, minerals, and the phytochemicals abundant in fruits and vegetables-neutralize free radicals by stabilizing them through electron donation. Specifically, flavonoid compounds are highlighted as robust defenders, addressing oxidative stress and inflammation to avert chronic illnesses like cancer, cardiovascular diseases, and neurodegenerative diseases. This research explores the bioactive potential of flavonoids, shedding light on their role not only in safeguarding health, but also in managing conditions such as diabetes, cancer, cardiovascular diseases, and neurodegenerative diseases. This review highlights the novel integration of South African-origin flavonoids with nanotechnology, presenting a cutting-edge strategy to improve drug delivery and therapeutic outcomes. Abstract:Introduction: Free radicals are reactive oxygen species that constantly circulate through the body and occur as a side effect of many reactions that take place in the human body. Under normal conditions, they are removed from the body by antioxidant processes. If these natural mechanisms are disrupted, radicals accumulate in excess and contribute to the development of many diseases. Methodology: Relevant recent information on oxidative stress, free radicals, reactive oxidative species, and natural and synthetic antioxidants was collected by researching electronic databases such as PubMed / Medline, Web of Science, and Science Direct. Results: According to the analysed studies, this comprehensive review provided a recent update on oxidative stress, free radicals and antioxidants and their impact on the pathophysiology of human diseases. Discussion: To counteract the condition of oxidative stress, synthetic antioxidants must be provided from external sources to supplement the antioxidant defense mechanism internally. Because of their therapeutic potential and natural origin, medicinal plants have been reported as the main source of natural antioxidants phytocompounds. Some non-enzymatic phytocompounds such as flavonoids, polyphenols, and glutathione, along with some vitamins have been reported to possess strong antioxidant activities in vivo and in vitro studies. Thus, the present review describes, in brief, the overview of oxidative stress-directed cellular damage and the unction of dietary antioxidants in the management of different diseases. The therapeutic limitations in correlating the antioxidant activity of foods to human health were also discussed.

Abstract:การประชมุวชิาการ ครั ง้ที ่30 ประจําป ี2557 All aerobic organisms are evolved with cellular respiration using molecular oxygen as final electron acceptor, where mitochondria are the powerhouse generating high energy phosphate compounds, ATP. The inevitable electron leakage from the electron transport chain in mitochondria is one of major sources of reactive oxygen species (ROS) formed in the cell. ROS is also generated by other systems including NADPH oxidases (NOXs), xanthine oxidases (dehydrogenase) and etc. The ROS formation is not simply the waste products from cellular metabolism, but in many conditions it is purposefully formed to serve many special functions, for instances, for defending invading microorganisms in phagocytic cells, and importantly serving as intracellular signaling molecules. In contrast, uncontrolled formation of ROS produces deleterious effects on biomolecules such as lipid peroxidation, protein oxidation and DNA damage which lead to cell dysfunction and cell death. A number of diseases are associated with oxidative stress, such as, cardiovascular disease, diabetes, metabolic syndrome, ageing and various neurodegenerative disorder and cancer. Abstract:Free radicals (FRs), also known as reactive oxygen species (ROS), are usually established in the body when adequate oxygen depletion occurs. Oxidative stress and the establishment of FRs in the body are mainly caused by high metabolic activity, the need for rapid growth, inadequate flock management, exposure to viral and bacterial microorganisms, and adverse environmental conditions. Furthermore, FRs can also be produced during the activity of phagocytes when they depend on the action of ROS to kill the engulfed pathogen. FRs have very adverse effects on all cells, particularly the cells of the immune system. They are extremely erratic and reactive molecules that directly harm DNA, cellular proteins, lipids, and carbohydrates within cells. Antioxidants are substances that can eliminate and neutralize FRs within the body and free the body from the oxidative stress that occurs due to the accumulation of FRs. Many vitamins and minerals support the activity and effect of the immune system in fighting against microbes and cancer, which mostly depend on their antioxidant elements to diminish the negative impact of FRs in the body. Examples are vitamin C, vitamin E, superoxide dismutase, selenium, glycine, cofactors of glutathione peroxidase, manganese, essential oils, and phenolic compounds.

Abstract:The increase in the formation of reactive oxygen and reactive nitrogen species of endogenous or exogenous origin causes oxidative stress due to pro-oxidant and antioxidant imbalance that causes cellular damage in metabolism. This can increase inflammation of cells, apoptosis and necrosis, damage to DNA base damage, DNA and protein cross-links, lipid membrane peroxidation, and mitochondrial dysfunction. Antioxidants can be described as a system that protects biomolecules and the organism against the harmful effects of free radicals, reduces or repairs the damage done by reactive oxygen species (ROS) to the target molecule, and this is called antioxidant defense. It is known that the mechanisms caused by the increase in ROS resulting from oxidative stress are positively related to the pathology of many diseases such as cancer, metabolic syndrome, atherosclerosis, malaria, Alzheimer's disease, rheumatoid arthritis, neurodegenerative diseases and preeclampsia.

Abstract:Free radicals are constructed by natural physiological activities in the human cells as well as in the environment. They may be produced as a result of diet, smoking, exercise, inflammation, exposure to sunlight, air pollutants, stress, alcohol and drugs. Imbalanced redox status may lead to cellular oxidative stress, which can damage the cells of the body, resulting in an incidence of various diseases. If the endogenous antioxidants do not stop the production of reactive metabolites, they will be needed to bring about a balance in redox status. Natural antioxidants, for example plants, play an important part in this context. This paper seeks to report the available evidence about oxidative stress and the application of plants as antioxidant agents to fight free radicals in the human body. For this purpose, to better understand oxidative stress, the principles of free radical production, the role of free radicals in diseases, antioxidant defense mechanisms, and the role of herbs and diet in oxidative stress are discussed.

Abstract:Oxidative injury caused by free radicals is an important cause of tissue injury now recognized to occur in inflammation, ischemia and by the action of xenobiotics. It is also recognized to induce gene mutation and promote carcinogenesis. In this review the general concept of nett free radical injury counterbalanced by antioxidants is discussed as oxidative stress. The role of oxidative stress in intestinal ischemia, radiation enteritis, inflammatory bowel disease and the promotion of gastric and colorectal cancer is discussed. The data for the role of oxidative stress in the pathogenesis of ischemic, inflammatory and radiation induced disease are strong, but interventional studies with antioxidants have shown only weak beneficial effects in the above diseases. Therefore the role of antioxidants in the therapy of gastrointestinal diseases remains controversial and should be the subject of controlled trials.

Abstract:Reactive oxygen species are involved in the pathogenesis of cancers and metabolic diseases, including diabetes, obesity, and fatty liver disease. Thus, inhibiting the generation of free radicals is a promising strategy to control the onset of metabolic diseases and cancer progression. Various synthetic drugs and natural product-derived compounds that exhibit antioxidant activity have been reported to have a protective effect against a range of metabolic diseases and cancer. This review highlights the development and aggravation of cancer and metabolic diseases due to the imbalance between pro-oxidants and endogenous antioxidant molecules. In addition, we discuss the function of proteins that regulate the production of reactive oxygen species as a strategy to treat metabolic diseases. In particular, we summarize the role of proteins such as nuclear factor-like 2, Sestrin, and heme oxygenase-1, which regulate the expression of various antioxidant genes in metabolic diseases and cancer. We have included recent literature to discuss the latest research on identifying novel signals of antioxidant genes that can control metabolic diseases and cancer.
