Abstract:Pollutants and parasites represent stressors for fish at the individual, population, or community levels. The current study outlines the seasonal infestation pattern of Oreochromis niloticus by protozoan and helminths parasites linked to some physicochemical parameters (pH, dissolved oxygen, total dissolved solids, and electrical conductivity), and metals (Cd, Pb, Fe, Zn, and Ca), at nine selected sites in River Nile (Greater Cairo area) from summer 2019 till spring 2020. Most of the studied criteria in Nile water samples showed normal levels during all seasons compared to the recommended limits of EPA. The parasitological examinations recorded three protozoan categories: Myxobolus (8 species), hemoflagellates ( Trypanosoma mukasi ) and Ciliates ( Trichodina compacta ), and three helminths; Clinostomum sp., Acanthocephala sp. and Euclinostomum ardeola . The highest incidence of infection was determined for Clinostomum sp., followed in descending order by Myxobolus sp., Acanthocephala sp., T. mukasi, E. ardeolathen, and T. compacta . Notably, this study introduces the novel identification of new species of Myxobolus in the blood of Oreochromis niloticus . The histopathological examination of gills, muscles, and kidneys reveals serious changes and the presence of encysted trematodes, metacercariae, and cysts of protozoan parasites. Additionally, the study employs cluster analysis based on site similarity in water variables and canonical correspondence analysis, explaining 98.7 % of the variables and indicating correlations between parasite infestation and environmental factors. These analytical approaches reveal the impact of land use activities on water variables and the influence of adjacent activities on fish parasite infestation patterns. In conclusion, this study provides a comprehensive perspective by considering various factors to enhance our understanding of pollutants and parasites affecting fish in the River Nile.

From Africa to Brazil: detection of African Oreochromis niloticus parasites in Brazilian fish farms

Abstract:Abstract Aim To evaluate the introduction of Oreochromis niloticus gill parasites in the Paranapanema River basin, northern Paraná, southern Brazil, as well as to inventory its occurrences in Brazilian fish farms and discuss the risks of transmission to native fauna. Methods The gills of 632 fish specimens from four fish farms in the Paranapanema Basin were analyzed. The parasites were collected, processed and identified according to specific procedure. Literature review was carried out to compile records of occurrence of gill parasites species in other Brazilian river basins. Results A total of seven (7) species of parasites were recorded, five (5) of the genus Cichlidogyrus, one (1) of Scutogyrus (Ancyrocephalidae, Monogenoidea) and one (1) of Lamproglena, Lamproglena monodi (Copepoda, Lernaeidae). All native from Africa. Some of these species have been reported in fish farms located in five other Brazilian watersheds. However, in this study a greater number of African gill parasite species was recorded in fish farms in northern Paraná (seven species), in the Paranapanema Basin, with Cichlidogyrus rognoni reported only for this basin. Conclusions The results confirm the introduction of these African parasites along with their host, O. niloticus, and the establishment of these species in Brazilian waters, since many specimens belonging to the species identified herein presented breeding activity for at least one period of the year. Although, only L. monodi has been recorded parasitizing Brazilian native species, data reported for other countries demonstrates the imminent risk of transmission of these O. niloticus parasites to native cichlids.
First report of monogenean flatworms from Lake Tana, Ethiopia: gill parasites of the commercially important Clarias gariepinus (Teleostei: Clariidae) and Oreochromis niloticus tana (Teleostei: Cichlidae)

Abstract:BackgroundLake Tana is the largest lake in Ethiopia and the source of the Blue Nile. The lake harbours unique endemic cyprinid fish species, as well as the commercially important endemic Nile tilapia subspecies Oreochromis niloticus tana and the North African catfish Clarias gariepinus. Its endemicity, especially within the Labeobarbus radiation, its conservation importance and its economic indispensability attract scientific interest to the lake’s ichthyofauna. Fish parasites of Lake Tana, however, are hitherto poorly known, and no formal report exists on its monogenean flatworms. For sustainable aquaculture and fisheries development, it is essential to study monogenean fish parasites in these economically most important fish species. Moreover, it remains to be verified whether this unique ecosystem and its endemicity gave rise to a distinct parasite fauna as well.ResultsNile tilapia and North African catfish hosts were collected from Lake Tana in 2013. Nine species of monogenean parasites of two orders, Gyrodactylidea Bychowsky, 1937 and Dactylogyridea Bychowsky, 1937, were recovered. Gyrodactylus gelnari Přikrylová, Blažek & Vanhove, 2012, Macrogyrodactylus clarii Gussev, 1961, Quadriacanthus aegypticus El-Naggar & Serag, 1986 and two undescribed Quadriacanthus species were recovered from C. gariepinus. Oreochromis niloticus tana hosted Cichlidogyrus cirratus Paperna, 1964, C. halli (Price & Kirk, 1967), C. thurstonae Ergens, 1981 and Scutogyrus longicornis (Paperna & Thurston, 1969).ConclusionsExcept for M. clarii, all species represent new records for Ethiopia. This first study on the monogenean fauna of Lake Tana revealed that the lake’s North African catfish, as well as its endemic Nile tilapia subspecies, harbour parasites that are known from these host species elsewhere in Africa.

Abstract:A parasite study was conducted from August 2004 to July 2005 to highlight the distribution of gill Monogenean parasites from Oreochromis niloticus. A total of 370 fish collected from Ayamé man-made Lake, Côte d’Ivoire were examined for Monogenean species using routine parasitological techniques. The study revealed the presence of six species of Monogeneans (Cichlidogyrus thurstonae, Cichlidogyrus halli, Cichlidogyrus tilapiae, Cichlidogyrus rognoni, Cichlidogyrus cirratus and Scutogyrus longicornis). These species showed an aggregate pattern within the host population. Four species of parasites showed positive and significant correlation with parasitic abundance and the relative condition factor. The prevalence and intensity of the infection of these parasites were highest in the long rainy season and lowest in the dry season. The large sized fishes were more subjected to parasitic infection than smaller ones. In contrast, no sex effect was observed on the infestation of O. niloticus by the Monogeneans. No preference for the right or left side of the fish host was observed and the parasite species mostly colonized the middle arches (II and III). This could lead to more informed aquacultural practises, and maximization of the production of O. niloticus. Key words: Disease, season, sex, infestation, gill.
Prevailing parasitic diseases affecting Oreochromis aureus in Lake Burullus

Abstract:Lake Burullus is an important source of fish production in Egypt; it produces 20.5% of the Egyptian fisheries' production. There is intense controversy about the heavy metal pollution in Burullus water and its effects on fish health and safety for human consumption. Heavy metals represent a major concern for aquatic life and could negatively affect fish health. Agricultural and industrial water drainage represents a considerable part of the lake water supply. The present work was conducted to determine heavy metal concentrations in lake water and blue tilapia Oreochromis aureus musculature. Water samples were collected from six locations to determine cadmium, copper, lead, zinc and iron. Sixty O. aureus fish samples were also collected from the same sampling points to assess the prevalent parasites infesting fish and to determine the heavy metal (Cd, Cu, Pb, Zn and Fe) concentrations in fish musculature then study the relationship between heavy metals concentration and prevailing fish parasites. Results indicated that 53.34% of the examined fish were infested with encysted metacercaria. Centrocestus formosanus, Prohemistomum vivax, and Euclinostomum heterostomum were retrieved from gills, musculature, hepatopancreas and the posterior kidney. The parasitic intensity in fish tissues was between 1 and 9 cyst g-1. Centrocestus formosanus was identified using the polymerase chain reaction in the gill tissues of 16 fish. The gill parasitic copepod Lamproglena monodi was identified in one fish. Degenerative changes such as thickening, corrugation, and destruction of gill filament are the most dominant pathological changes in infested fish gills. Heavy metal concentrations in water samples were at normal levels, except for copper and iron in the southern part of the lake. All heavy metals in fish musculature were below the permissible limits. The parasitic infestation was more dominant in the northern part of the lake than in the southern region; this could be due to elevated copper concentration in the southern part of the lake that could negatively affect the survival of the first intermediate host and parasite cercaria. In conclusion, captured fish from Lake Burullus were safe for human consumption, and heavy metal pollution in lake water does not represent a severe risk.

Abstract:Parasites are a significant component of biodiversity. They negatively affect fish appearance, growth, and reproduction. In this study, the prevalence of infection, diversity, and mean intensity of parasites were examined in 9 freshwater fish species (45 samples per fish species). Ecto-parasites were examined on the skin, gills, and fins with a hand lens. Wet mounts were prepared using mucosal scrapings from all the external and internal organs of the sampled fish. Microscopy, muscle compression, and the pepsin-HCL artificial digestion technique were also performed. In this study, 26 species of parasites were identified including three taxa belonging to 9 species of protozoan parasites, 11 treamtodes, and 6 monogenean parasites. The identified protozoan parasites were Entamoeba histolitica, Chilodonella sp., Coccidia sp., Costia sp., Cryptobia sp., Ichthyopthiris-multifilis, Microsporidia, Piscinoodinium sp., and Ichthyobodo necator. The identified trematode parasites were Fasciola gigantica, Echinostoma revolutum, Fasciola hepatica, Haplorchis pumilio, Brachylaima cribbi, Echinostoma cinetorchis, Neascus sp., Deropegus sp., Trematode Soldier, Centrocestus formosanus, and Clinostomum marginatum. The identified monogenean parasites were Dactylogyrus limipopoensis, Dactylogyrus anchoratus, Dactylogyrus myersi, Dactylogyrus vastator, Gyrodactylus salaris, and Ancyrocephalus. The diversity of parasites was maximum at the Okara site. The host's organs that were targeted for parasitic infection included the intestine, liver, gills, fins, skin, and kidneys. The majority of the parasites were identified in Labeo rohita followed by Hypophthalmichthys molitrix, Ctenopharyngodon idella, Oreochromis niloticus, Cyprinus carpio, and Wallagu attu. Two species appeared to be resistant species because none of the parasites were observed in Notopterus notopterus or Sperata seenghala. This study also concluded that the prevalence of parasites increased with increasing length, size, and age of fish.
Endohelminth diversity in the invasive Clarias gariepinus (Burchell, 1822) from two freshwater lakes: Naivasha and Ol'Bolossat, Kenya

Abstract:Fish parasitology contributes to our understanding of the potential risks posed by diverse groups of parasitic organisms on fish stocks in either wild and culture systems. This study was conducted in May 2023 and aimed at assessing the diversity of endohelminths in the invasive North African catfish Clarias gariepinus (Burchell, 1822) obtained from two freshwater lakes, Naivasha and Ol'Bolossat, in Kenya. Parasitological examination of 66 and 35 fish samples collected from the two lakes respectively was achieved using light and scanning electron microscopy methods. Results revealed endohelminth diversity broadly classified as four digeneans, two nematodes, and one cestode. Seven taxa of endohelminths were found in C. gariepinus samples, but only four of these taxa could be identified up to the species level. Six of the taxa ( Diplostomum sp., Tylodelphys mashonense , Plagiorchioidea sp., Paracamallanus cyathopharynx , Contracaecum sp., and Tetracampos ciliotheca ) were common in samples from the two lakes. Glossidium pedatum only occurred in samples from Lake Ol'Bolossat. Parasite prevalence ranged from 8.6 ( T. mashonense ) to 100% ( Diplostomum sp., T. ciliotheca , and Contracaecum sp.) and mean intensity from 1.4 ( T. mashonense ) to 16.9 ( Diplostomum sp.). The diversity and richness indices were comparatively higher in fish samples from Lake Ol'Bolossat and attributed to the occurrence of G. pedatum in the Ol'Bolossat. However, parasitic infestation of fish samples from the two lakes depicted close similarity, both in diversity and prevalence. These findings form an important baseline data for further follow-up studies, and they suggest the need for further molecular analyses to fully describe three of the taxa only identified up to the genus level.

Abstract:This study provides the first investigation on acquisition of parasites in invasive O. niloticus by parasite species of native Cichlidae from the Igarapé Fortaleza basin, Northern Brazil. There were examined 576 specimens of 16 species of native cichlids and invasive O. niloticus collected in the main channel and the floodplain area of this tributary of Amazon River. The invasive O. niloticus was poorly parasitized having only Ichthyophthirius multifiliis, Trichodina centrostrigeata, Paratrichodina africana, Trichodina nobilis (Protozoa) and Cichlidogyrus tilapiae (Monogenoidea), and this host has not acquired any parasite species common to the native ichthyofauna region. In contrast, species of native cichlids showed rich fauna of parasites with predominance of Monogenoidea species, larvae and adults of Nematoda, Digenea, Cestoidea and Acanthocephala, besides four species of Protozoa and four Crustacea. However, only T. nobilis was acquired by native fish, the Aequidens tetramerus, which is a new host for this exotic Trichodinidae. In O. niloticus, well established in the region, the small number of helminth species may be associated with its rusticity, good adaptation in the new environment and also the presence of native parasites with relative specificity, but without ability to complete its life cycle in this invasive host of this ecosystem.
A multidisciplinary study on Clinostomum infections in Nile tilapia: micro-morphology, oxidative stress, immunology, and histopathology

Abstract:Yellow grub disease, caused by Clinostomum metacercaria, is an endemic zoonotic infection in freshwater fish, responsible for Halzoun syndrome transmitted through the consumption of raw infected fish. This study aimed to conduct a multidisciplinary investigation integrating detailed morphology, oxidative stress, immunology, and histopathology alteration to advance our understanding of Clinostomum infection. In this annual study, 400 Nile tilapia (Oreochromis niloticus) were collected from the Nile River at Al Bahr Al Aazam, Giza Governorate to assess Clinostomum infection prevalence. Of the examined fish, 160 individuals (40.0%) harboured larval Clinostomum infections. Clinostomum metacercariae were observed in various anatomical locations, with 135 fish (33.8%) in buccal cavities, 21 fish (5.25%) in gill chambers, and 4 fish (1.0%) on the skin. Infection intensity ranged from 2 to 12 cysts per fish, averaging 5 cysts, notably with skin infections characterized by a single cyst in each fish. Macroscopic encysted metacercariae were collected from buccal cavities, gills, and skin. Micro-morphology revealed distinct features in C. complanatum, including an elliptical oral sucker with collar-like rings and large sensory papilla-like structures, contrasting with the absence of these features in C. phalacrocoracis. Oxidative stress, assessed through malondialdehyde (MDA) and nitric oxide levels, revealed an elevation in MDA to 35.13 ± 6 nmol/g and nitric oxide to 25.80 ± 3.12 µmol/g in infected fish. In infected fish, MHC-I gene expression increased approximately 13-fold, MHC-II peaked at 19-fold, and IL-1β significantly upregulated by 17-fold, compared to control levels. Histopathology detailed associated lesions, such as cyst encapsulation and eosinophilic infiltration. Clinstomiasis and its impacts on fish hosts offer crucial insights to control this emerging fish-borne zoonotic disease, threatening wildlife, aquaculture, and human health.
Isolation and identification of major bacteria from three Ethiopian rift valley lakes live and processed fish, and water samples: implications in sanitary system of fish products

Abstract:Bacterial pathogens are a great threat to fish production. Gram-negative bacteria are among the major bacterial fish pathogens and are zoonotic with the potential to infect humans. This cross-sectional study was conducted to isolate and identify major gram-negative bacteria from live and processed fish, and water samples from Lakes Hawassa, Langanoo and Ziway. A total of 674 different types of samples: 630 tissue samples (210 samples for each intestine, Kkidney and liver collected from 210 live fish ( Oreochromis niloticus , Cyprinus carpio and Clarias gariepinus ), 20 processed fish samples from lake Ziway fish processing center and 24 lake water samples were included in the study from each lake. The mean values of pH, temperature, dissolved oxygen and nitrate in all water samples were within the normal range at which most freshwater fish species become non-stressed. Of a total of 674 samples included in the study, bacteria were isolated from 154(22.8%) samples with significant difference ( P < 0.05) observed in some isolates with respect to sample origin. Of these 154 isolates, 103(66.8%) isolates were gram-negative bacteria consisting of 15 species based on morphology and a range of biochemical tests. From live fish samples, Escherichia coli was the dominant species with 15 isolates followed by Edwardsiella tarda (12), Salmonella Paratyphi (10), Salmonella Typhi (9), Shigella dysenteriae (7), Shigella flexneri (7), Klebsiella pneumonia (7), Enterobacter aerogenes (6), Enterobacter cloacae (5), Pseudomonas aeruginosa (5), Vibrio parahemolyticus (5), Aeromonas sobria (4), Citrobacter freundii (4), Citrobacter koseri (4) and Plesiomonas shigelloides (3). The detection of the common fecal coliforms ( E. coli , K. pneumoniae and E. aerogenes ) and Salmonella spp . in processed fish indicates the potential danger of passage of pathogenic bacteria and/or their poisons to humans via infected and/or contaminated fish products. Human infection by pathogenic fish bacteria and food poisoning is possible through contamination of fish product in fish production chain due to inadequate handling, poor hygiene and contact with contaminated water. Therefore, producers, consumers and all other stakeholders need to be cautious during handling, processing and consumption of fish harvested from the study lakes.

Impact of water quality and morphometric indices on the spatio-temporal prevalence of fish endo-parasites and diversity in the Ase River, Niger-Delta, Nigeria

Abstract:Three stations along the Ase River, Delta State Nigeria provided water and 85 fish samples which were analyzed. The fish were measured and examined for endoparasites according to established protocol. All water quality parameters investigated were within the WHO-acceptable values for surface waters. Station variation of physicochemical parameters was not statistically significant (p > 0.05). Fish body conformation indices positively correlated with the prevalence of parasites in Clarias gariepinus, Heterobranchus longifilis, Parachana africana, Chromidotilapia guntheri guntherii, and Denticeps clupeodes. The overall parasite prevalence of 63.53% was established with the most abundant parasite being Trichodina mutabillis. The parasites had a predilection for the gastrointestinal tract with a high occurrence of 307 individuals. Stations 1, 2, and 3 had 326, 213, and 259 parasites, respectively, out of a total of 798 parasites detected. P. laevis was absent in station 1. All parasites were found in stations 2 and 3. Statistically, there was a significant difference (p < 0.05) in the prevalence in all stations. The correlation index of T. mutabillis and R. congolensis in stations 1 and 3 was positively strong (p < 0.05) with the concentrations of water quality. However, water conditions in stations 1 and 3 had a deleterious impact on P. laevis. T. mutabillis maintained a high positive correlation with physicochemical water quality in all three stations. Shannon-Weiner's index in station 3 (H = 1.337) shows that the parasites were more diverse. PCA and biodiversity indices have enabled us to comprehend how parasite-host-environment systems interact.
[Larval Clinostomum infection of cichlid fish in the lake of Kompienga in Burkina Faso: a menace to haleutic exploitation and public health]

Abstract:Clinostomum sp. is a trematode, a parasitic worm, of which the larva infest a number of species of wild fish. In intertropical Africa and particularly in artificial lakes in the east of Burkina Faso, the infestation is especially observed in the cichlids fish. Because cichlids constitute 80 to 90% of the fish catches, enzootic infestation could infringe upon the fisheries, and also adversely affect public health. For this epidemiological study conducted during the first three quarters of 1994, we examined a total of 932 cichlids, a mix of all species. Prevalence (Pr) and intensity (I, number of parasites per fish) were the two indicators of the infestation level. 387 fish were carriers of the parasite, comprising 41.5% of the total. Differences were observed between the species of cichlids. Oreochromis niloticus was the most infested (Pr, 56%; I, 17), followed by Sarotherodon galilaeus (Pr, 44%; I,6); and Hemichromis fasciatus (Pr, 20%; I, 5). In 89% of the carriers, the parasite was localized on the internal face of the operculum and in the pericardial septum, compared to 11% of the localizations in subcutaneous and muscular tissues. These last two localizations render the fish repugnant, and can cause the fresh fish to be rejected. If the parasitized fish is not rejected and then insufficiently cooked, as occurs at public grills with multitudes of consumers, the living parasite can penetrate and stay in the upper digestive tract and constitute a threat for consumer health.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:Fish are a resource with great economic, nutritional and recreational benefits to humans on a global scale. In Kenya, fish represent an important source of food and income through trade and employment to many communities. In fact, fish are an important alternative source of animal protein, especially in famine‐tolerant arid and semi‐arid regions such as Turkana, Marsabit, Pokot and Baringo Counties, where annual drought periodically hampers traditional livestock keeping destitute. Kenya freshwater fishes have remained vulnerable to a variety of parasites and related diseases. Approximately 119 fish parasites have been reported in the country, with about 83 being identified to species level and 35 to genus level. Out of the reported parasites, 77% were detected in wild fish, 8% in farm fish and 15% in both wild and farmed fish. The parasites identified by various studies include protozoa (7), myxozoa (7), nematode (20), monogenean (33), digenean (16), cestoda (15), acanthocephalan (6) and crustacean (15). The highly commercialized fishes, such as Oreochromis niloticus and Clarias gariepinus, harbour the highest number of parasites, which greatly hinder fisheries and aquaculture productivity through retarded growth, mechanical damages, reduced reproduction rates and increased mortality of the fish hosts. Thus, there is need for adequate information on the taxonomy and ecology of these parasites as a basis for developing appropriate management and policies to control them. This review article is meant to provide an overview of the distribution, occurrence of fish parasites and their impacts on inland water fisheries and aquaculture in Kenya, while also highlighting the available gaps warranting further studies, with the goal of developing appropriate and accurate control measures to improve the region's fisheries and aquaculture productivity and food security.

Abstract:This study aimed to conduct a multidisciplinary investigation integrating detailed morphology, molecular characterization, water parameters, histopathology alteration, and the trials of treatment of Clinostomum spp. In this study, 300 Nile tilapia (Oreochromis niloticus) were collected from the farmed and wild Nile River at Al Bahr Al Aazam, Giza Governorate to assess Clinostomid infection prevalence. Fish and water samples were collected from private fish farms, and water drains at Dakahlia, and Giza, Egypt. Analysis of the water revealed inadequate water quality, particularly in the fish farms. Snails and piscivorous birds were abundant at fish collection sites. The recovered Clinostomid MCs morphological characteristics and COI gene sequence analysis identified them as Clinostomum complanatum, C. phalacrocoracis, and Euclinostomum heterostomum. Clinostomid MCs disturbed the fish's hematological and biochemical blood parameters. Bath treatment of parasitized fish with praziquantel (2 mg/L for 24 h) revealed a significant reduction in the number of vital MCs vs. infected fish (non-treated). Praziquantel (PZQ) is an effective and safe therapy for controlling Clinostomid infections affecting farmed Nile tilapia. The current findings indicate a link between poor environmental conditions and Clinostomum infections in tilapia. The study highlights the impacts of Clinostomid MCs on fish health and recommends bath treatment with PZQ as an efficient control method for these dangerous parasites to protect human and fish health.
The synergy between serious parasitic pathogens and bacterial infestation in the cultured Nile tilapia (Oreochromis niloticus): a severe threat to fish immunity, causing mass mortality and significant economic losses

Abstract:Stress-induced epidemic parasites and bacterial diseases are becoming a typical occurrence in fish aquaculture across the globe, those consider deadly infectious pathogens. To prevent losses on fish farms, it is crucial to obtain information on the infection severity and the characteristics of those diseases which have been peaked for fish farms and lead to mass mortality. This study investigates the effect of dual parasitic and bacterial infection on farmed tilapia, which leads to losses in farmed fish; focuses on the harmful effect of polluted water on speared infections in farming. During the production season, water and fish specimens were taken from private fish farms in El-Sharkia and Kafr El-Sheikh, Egypt, particularly during significant mortality times. The infected and uninfected fish samples were examined to determine bacteriological, parasitological, haemato-biochemical indices, immune responses, and pathologic changes besides water analysis in fish farming in the study area. It was discovered that fish were significantly infected with Centrocestus formosanus encysted metacercariae and/or Cichlidogyrus tilapiae , Heterophyes spp. Also, three species of gram-negative: Aeromonas hydrophila , A. sobria , Pseudomonas aeruginosa, and two Gram-positive cocci: Streptococcus agalactiae and Enterococcus faecalis . Identification of bacterial isolates was confirmed by using 16S rRNA gene sequencing. Results demonstrated substantial ( P < 0.05 ) output losses in both farms. Results displayed negative effects of dual infection on the lost weight, whole-body biochemical, haemato-biochemical indices, digestive enzyme, immune responses, gene expression, and abnormal features histopathology in different organs of infected Nile tilapia. At the same time, most of the water Indices strayed considerably from standards. Thus, the obtained data indicated the detrimental impacts of dual infection and water quality deterioration on the health condition of Nile tilapia and might help us acquire a deeper understanding of yield loss as potentially severe economic repercussions for fish farms.

Abstract:Schizocypris altidorsalis is a fish of very restricted distribution, with its populations being known only from few water reservoirs on the border of Iran and Afghanistan. The aim was to investigate the occurrence and diversity of host-specific Monogenea on this fish. In total, 400 fish were purchased from the market and examined for the presence of Monogenea. Specimens were characterised morphologically and genetically. In total, 26.5% of fish were found to be infected with Monogenea. Further morphological and molecular studies based on the sequences of the 18S , 28S and ITS2 regions suggested that they all belong to a new species, herein named as Paradiplozoon jalalii . Our results suggest that this monogenean is a highly specific parasite of Schizocypris altidorsalis . Monogeneans are highly specialised host-specific parasites infecting fish gill. They can adversely affect respiration and excretory functions in fish, making them more vulnerable to illness, leading to population declines. Determining effective strategies for safeguarding endangered fish can be challenging in conflict zones, such as the area studied here. Additionally, the parasite identified in our study, with its blood-feeding habits, could contribute to the extinction of its fish host, subsequently leading to its own extinction. An unanswered question lingers regarding the potential extinction of other symbionts of Schizocypris altidorsalis if the fish faces extinction.

The Role of Nile Tilapia (Oreochromis niloticus) in the Life Cycle of Toxocara spp

Abstract:The present study aimed to experimentally assess Nile tilapia as potential paratenic host of Toxocara spp. A total of 15 Nile tilapia (Oreochromis niloticus) were fed with 300 embryonated Toxocara canis eggs by oral gavage, while five others of the control group received distilled water. The fish were individually analyzed at 16, 24, 48, 72, and 240 h after inoculation. Water contamination was assessed, and tissue migration by liver, gastrointestinal tract (GIT), eyes, and central nervous system. A murine model was used as the paratenic host for egg infectivity assessment. Eggs and larvae were found in plastic tank water and fish GIT, ranging from 23 to 86% per fish. Eggs and larvae were recovered from the tank water (76.3%) and fish GIT (23.7%). The counting of eggs and larvae observed was negatively correlated with number of eggs and larvae in the water tank (rho = -0.698, p = 0.003). Shedding of embryonated eggs was first detected at 16 and up to 240 h, with significant egg and larvae yield decrease on water-shedding (p = 0.001) and in the GIT (p = 0.007). Although no T. canis larva was recovered in fish tissues, egg infectivity after fish GIT transit was experimentally confirmed by mice assessment. In conclusion, despite shedding viable embryonated eggs through the gastrointestinal tract, tilapias may not play a role as a suitable paratenic hosts for Toxocara spp., posing low risk of zoonotic transmission by fish meat consumption.

Abstract:The recent discovery of disease caused by Nucleospora braziliensis in Nile tilapia ( Oreochromis niloticus ) is important as it has highlighted the high prevalence of infection and associated mortality in cultured fish. Thus, this study conducted an experimental infection of this microsporidium to evaluate pathological alterations and conduct proteomic analysis. For pathological observation, samples of brain, eyes, gall bladder, gut, heart, kidney, liver, muscle, skin, spleen, and stomach tissue, were collected, and liquid chromatography–mass spectrometry (LC-MS/MS) was performed for proteomic analysis. The most prevalent lesions were brownish color of the liver, gill filament fusion, gut ischemia, hemorrhage of the lips and fins, hepatomegaly, spleen atrophy, splenomegaly, and stomach congestion. The most common microscopic lesions were degeneration, hemorrhage, and inflammation in the brain, gills, gut, kidney, liver, muscle, spleen, and stomach. The digested peptides were identified by LC-MS/MS and the intersection of each group showed that in the spleen there were 121 exclusive proteins in the infected sample and 252 in the control, while in the kidney, 129 proteins were identified in the infected specimen compared to 83 in the control. In conclusion, this study demonstrates the proteome profile of O. niloticus kidney and spleen tissue in response to infection with N. braziliensis .

Abstract:Background: Streptococcus agalactiae (Group B streptococcus, GBS) induces a serious infection that can harm not only aquatic life but also humans and other animals. In a fish farm in southern Egypt, Nile tilapia (Oreochromis niloticus) has developed an epidemic with clinical symptoms resembling piscine streptococcosis. Results: Initial microscopic inspection of the affected fish brain and kidney indicated the presence of Gram-positive cocci. S. agalactiae was effectively isolated and identified using nucleotide homology of the 16S rRNA and species-specific PCR. The partial 16S rRNA sequence was deposited in the GenBank database at the NCBI and given the accession number MW599202. Genotyping using RAPD analysis indicated that the isolates in the present study belonged to the same genotypes and had the same origin. The challenge test, via immersion (9.2 × 107, 9.2 × 106, and 9.2 × 105 CFU/ml for 1 h) or intraperitoneal injection (4.6 × 107, 4.6 × 106, and 4.6 × 105 CFU/fish), elicited clinical symptoms resembling those of naturally infected fish with a mortality rate as high as 80%. The ability to create a biofilm as one of the pathogen virulence factors was verified. Zinc oxide nanoparticles and the ethanolic leaf extracts of nine medicinal plants demonstrated considerable antibacterial activities against the tested S. agalactiae strain with low minimum bactericidal concentrations (MBC) and minimum inhibitory concentrations (MIC). The ethanolic leaf extracts from Lantana camara and Aberia caffra showed potent antibacterial activity with MBC values of 0.24 and 0.485 mg/ml, and MIC values of 0.12 & 0.24 mg/ml, respectively. Conclusion: This study isolated S. agalactiae from O. niloticus mortalities in a fish farm in Assiut, Egypt. The pathogen persists in fish environments and can escape through biofilm formation, suggesting it cannot be easily eliminated. However, promising findings were obtained with in vitro control employing zinc oxide nanoparticles and medicinal plant extracts. Nevertheless further in vivo research is needed.

Abstract:Nile-Tilapia (Oreochromis niloticus) aquaculture represents one of the most important cultivation species in Egypt. However, Tilapia fish farming is challenged by some problems. Of those, the presence of bacterial pathogens resulting in high fish mortalities and huge economic losses. Thus, the current investigation aimed to isolate, identify, and characterize the pathogenic bacteria from Nile tilapia fish farm in El-Abassa village, Egypt and to investigate their antibiotic susceptibility as a primary step for controlling diseases. 182 bacterial isolates were obtained from one hundred Tilapia fish samples. The microbiological and biochemical analysis of the examined fish indicated the presence of only 5 bacterial genera. Three of them are Gram-negative bacteria (representing 86.26% of total isolates) including Aeromonas spp. (46.70 %), Pseudomonas spp. (23.08 %), and Vibrio spp. (16.48 %). While two genera are Gram-positive bacteria (representing 13.74% of the total isolates) including Streptococcus spp. (8.79 %) and Staphylococcus spp. (4.95 %). This indicates that Gram-negative bacteria are the main cause of high fish mortalities in the studied area while Aeromonas hydrophila exhibited the highest prevalence in infected tilapia. Antibiogram test revealed high levels of resistance expressed by all isolates to ampicillin, amoxicillin, and erythromycin. On the other hand, norfloxacin was effective against all isolated bacteria followed by ciprofloxacin; therefore, norfloxacin should be recommended as a supplement in fish fed-diets to control the bacterial infection. Establishing effective control methods for pathogenic isolates would greatly enhance fish production.
