Abstract:The self-healing bioconcrete, or bioconcrete as concrete containing microorganisms with self-healing capacities, presents a transformative strategy to extend the service life of concrete structures. This technology harnesses the biological capabilities of specific microorganisms, such as bacteria and fungi, which are integral to the material's capacity to autonomously mend cracks, thereby maintaining structural integrity. This review highlights the complex biochemical pathways these organisms utilize to produce healing compounds like calcium carbonate, and how environmental parameters, such as pH, temperature, oxygen, and moisture critically affect the repair efficacy. A comprehensive analysis of recently published peer-reviewed literature, and contemporary experimental research forms the backbone of this review with a focus on microbiological aspects of the self-healing process. The review assesses the challenges facing self-healing bioconcrete, including the longevity of microbial spores and the cost implications for large-scale implementation. Further, attention is given to potential research directions, such as investigating alternative biological agents and optimizing the concrete environment to support microbial activity. The culmination of this investigation is a call to action for integrating self-healing bioconcrete in construction on a broader scale, thereby realizing its potential to fortify infrastructure resilience and sustainability.

Abstract:This review explores the burgeoning field of microbially enhanced construction materials, with a special focus on self-healing concrete, through the lens of microbial biotechnology. Central to this discourse is the innovative use of bacteria, particularly Bacillus species, to address the pervasive issue of microcracks in concrete, a fundamental material in the construction industry. Traditional remedies, such as chemical admixtures and fiber reinforcements, offer partial solutions; however, self-healing concrete represents a paradigm shift, harnessing the natural calcite-precipitating ability of bacteria to autonomously repair cracks, thereby augmenting structural durability and longevity. Delving into the mechanics, the bacteria, embedded within the concrete matrix, remain dormant until crack formation triggers their metabolic pathways, leading to calcite production that effectively seals the fissures. This bio-mediated repair mechanism not only enhances the structural integrity of concrete but also aligns with sustainable construction practices by minimizing maintenance requirements and material wastage. The review extends beyond self-healing phenomena, encompassing broader applications of microbial technology in construction, including bio-concrete, bio-cement, and soil stabilization methods. These applications underscore the versatility of microbes in enhancing material properties such as compressive strength, tensile resilience, and water impermeability. Bio-concrete: Unveiling Self-Healing Properties Beyond Crack-Sealing

Abstract:Do microbes categorically heal the cracks in concrete or only seal the cracks - this study aims to answer, based on a few experimental findings, highlighting the self-healing concepts in cementbased materials. Alkaliphilic spore-forming Bacillus cohnii microbes (105-1010 cells/ml) are used in the cement-concrete (bio-concrete) in addition to varying nutrient amounts. The healing in the form of regaining its structural efficiency is evaluated by its mechanical (compressive, flexural) strength and durability properties. Bio-mineralized calcium carbonate seals 37% of 20-50 nm dimensioned pores and 87% of 50-200 nm dimensioned pores resulting in 43-48% total voids reduction within 28 days of the healing period. Microbes only degrade the nutrients to precipitate healing agents (calcium carbonate) without affecting the C-S-H structure in concrete. Statistical analysis reveals that calcite deposition mostly influences the strength of self-healed bio-concrete whereas experimental findings show 5.8-6.3 wt% calcium carbonate deposition augments the 40-60% compressive strength of bio-concrete compared to reference concrete. The complete-to60% sealing of cracks of 0.02-2 mm width is examined using visual inspection of concrete skins. Microbes reduce porosity, sealing cracks intensifies mechanical strength, and the durability of the concrete validates self-healing rather than only sealing the cracks.

Abstract:This study presents a pioneering approach that enables remarkable crack closure up to 0.8 mm through stimulated biomineralization aiming to enhance the self-healing capacity of concrete. The innovative synergy tackles the pressing environmental challenges head-on investigates the intriguing germination process of endospores in a cementitious environment and underscores the function of calcareous fly ash in promoting Microbially Induced Calcium Carbonate Precipitation (MICP) by providing additional Ca2+ ions, that are converted into carbonate phases through bacterial metabolic processes. These phenomena are thoroughly examined using an in-situ model particularly concerning MICP in bio-concrete, and thermodynamic modelling is performed to forecast the influence of fly ash on the quantity of Ca-bearing (carbonation sensitive) phases present in the hydrated phases of fly ash-blended cement system. Incorporating 30 % class C fly ash significantly amplifies the biomineralization activity, with 6.5 % calcium carbonate deposition in bio-concrete leds to significant healing activity. Concurrently, MICP contributes to a significant reduction in pore sizes, thereby enhancing the overall durability of concrete. Beyond structural improvements, this innovative bio-concrete formulation showcases an impressive 39 % reduction in CO2 emissions, aligning with sustainability goals. Additionally, repurposing fly ash, typically consigned to landfills, seamlessly adheres to the tenets of a circular economy, despite incurring a 7 % rise in the initial production cost. That promising advanced methodology can deliver transformative environmental benefits, ushering in a paradigm shift in sustainable construction practices, and finding the synergistic interplay between microbial activity and calcium-dependent fly ash.

Abstract:The integration of green construction practices within the built environment has been significantly advanced by biotechnological innovations, among which microbially induced biomineralization (MIB), predominantly facilitated by various strains of spore-forming bacilli, emerges as a pivotal mechanism for the self-healing of concrete. However, the practical deployment of this technology faces challenges, notably the compromised viability of bacterial spores due to germination triggered by severe shear stress during concrete mixing. To address this limitation, a water-insoluble polymer (extracellular polymeric substance) produced by Cellulomonas flavigena was utilized to encapsulate and protect the spores. The encapsulation process was rigorously verified through physicochemical methodologies, including X-ray diffraction (XRD) analysis, which revealed alterations in the interlayer spacings of the extracellular polymeric substance (EPS) structure during the encapsulation process, indicating successful EPS coating of the spores. Furthermore, a proof of concept for the enhanced biomineralization capacity of EPS-coated spores was demonstrated. Standard analytical techniques confirmed the precipitation of calcite and vaterite among other minerals, underscoring the effectiveness of this novel approach. This breakthrough paves the way for the development of innovative, sustainable bioconcrete applications, aligning with broader environmental objectives and advancing the field of green construction technology.IMPORTANCEDevelopment of bioconcrete with self-healing capability through MIB constitutes an important sustainable construction biotechnology approach for restoration and repair of built environment. Like every promising technology, MIB also suffers from certain shortcomings in terms of compromised viability of the microbial cells after premature germination of the spores on exposure to shear stress caused during concrete mixing. In this study, these challenges were adequately addressed by successfully providing a protective coating of indigenously extracted EPS to the bacterial spores and elucidating the interactive mechanisms between them. The results showed stable encapsulation of the spores while providing mechanistic insights of the encapsulation phenomenon. The data also showed enhanced rate of biomineralization by encapsulated microbes when subjected to stress conditions.

Abstract:Cracks in concrete structures can significantly decrease their lifespan by exposing reinforcement to outside environment, leading to concrete degradation. To address this issue, self-healing techniques have been developed, including biomineralization-based-self healing, where bacteria are employed to initiate microbially induced calcium precipitate (MICP), promoting the healing of cracks. This article explores the extreme environmental conditions including pH and temperature which can reduce sustainability and self-healing potential of bacteria. Furthermore, it explores the implementation techniques of using bacteria in concrete aimed at mitigating the adverse effects of the concrete environment on bacteria, thereby enhancing their self-healing capabilities. Notably, studies have found that the mechanical strength of concrete can be increased at cell concentrations of 10 5 to 10 8 cells/ml. Based on this review, it is found that the self-healing of concrete depends on factors such as environmental conditions of pH and temperature, as well as the implementation methods of bacteria in concrete. Moreover, there exists a direct correlation between bacterial cell concentration and alterations in the mechanical properties of concrete. The incorporation of bacteria in concrete leads to increment in strength properties, with strength enhancement of up to 42.8 %. Understanding the interplay between environmental factors and bacterial sustainability is crucial in optimizing biomineralization-based self-healing and enhancing the durability of concrete infrastructure.

Abstract:Concrete's weak tensile strength renders it susceptible to cracking under prolonged loads, leading to reduced load-bearing capacity and reinforcing bar corrosion. This study investigates the effectiveness of microbial-based self-healing in high-strength concrete, focusing on two bacterial strains: Sporosarcina koreensis and Bacillu s. Results demonstrate significant enhancements in micro- and macro-physical properties of high-strength bacterial concrete with Bacillus flexus and S. koreensis , surpassing the control. Bacillus flexus -infused concrete exhibits a remarkable 21.8% increase in compressive strength at 7 days and 11.7% at 56 days. Similarly, S. koreensis -treated concrete shows 12.2% and 7.4% increases at 7 and 56 days, respectively. Enhanced crack healing occurs due to calcite precipitation, confirmed by X-ray diffraction and scanning electron microscopy. Both bacterial strains achieve crack closure within 42 days, with widths of 259.7 μm and 288.7 μm, respectively. Moreover, bacterial concrete from these strains excels in durability against water, acid, and salt exposure, surpassing control concrete. These findings emphasize microbial-based self-healing's potential in high-strength concrete, providing a practical strategy to enhance structural resilience and extend concrete infrastructure lifespan.

Abstract:Concrete, one of the most important building materials, has been in use for nearly two hundred years and will likely continue to be the dominant building material for the foreseeable future. Therefore, its role in the world economy is crucial. However, cracks are almost inevitable in fresh and hardened concrete due to load-independent or load-related reasons. Furthermore, it is often difficult to detect and repair these cracks and pose a threat to the safety and durability of concrete infrastructures, especially in those with stringent sealing requirements. Therefore, bacteria-based remediation using microorganisms is a promising solution for both repairing cracks without manual intervention and sustainable concrete maintenance. In this respect, in this study, the effects of parameters such as biomineralization processes and environmental conditions, bacterial species, type of inclusion in the system, nutrient medium, crack width, microbiological and durability properties of bacterial concrete on microbial self-healing performance are discussed in detail.

Abstract:The most commonly used building material in the construction industry is concrete. However, the weak features of concrete are its low ductility and limited tension capacity and hence crack development with the increase in load. These cracks get more worsened by the intrusion of water and salt present in the composition and hence causing deterioration and reducing the longevity of the material. This study focuses on an innovative approach to mitigate concrete’s fractures and flaws by utilizing microbiologically induced calcite (CaCO3) precipitation (MICP) excited by Escherichia coli (E. coli) bacteria to improve the performance of cementitious building materials. The study investigated the development of microbiological concrete in plain water using only one culture density (OD600 0.5 ± 0.1). In this study, two water-to-bacterial mix ratios (75 : 25 and 50 : 50) were used and compared to the conventional concrete (100 : 0). 100-mm cube-sized specimens cured for a period of 7, 28, 90, and 365 days were tested for compressive strength, water absorption capacity, ultrasonic pulse velocity (UPV), and SEM analysis, which were performed on the samples at regular intervals. According to the results of these experiments, microbial concrete with the 50 : 50 ratio exhibited the highest strength for all curing times. From the water absorption test of samples, it is found that the absorption of the materials got reduced due to the infusion of microorganisms in concrete. On the other hand, the UPV test showed high velocity than the control samples for specimens with an OD600 0.5 ± 0.1. Scanning electron microscope (SEM) analysis performed on distinct concrete groups at the age of 28 days showed fewer voids in the concrete lumps due to the increase in water substitution rate caused by microbial culture.

Abstract:The production of cement accounts for 5 to 7% of carbon dioxide emissions in the world, and its broad-scale use contributes to climate imbalance. As a solution, biotechnology enables the cultivation of bacteria and fungi for the synthesis of calcium carbonate as one of the main constituents of cement. Through biomineralization, which is the initial driving force for the synthesis of compounds compatible with concrete, and crystallization, these compounds can be delivered to cracks in concrete. Microencapsulation is a method that serves as a clock to determine when crystallization is needed, which is assisted by control factors such as pH and aeration. The present review addresses possibilities of working with bioconcrete, describing the composition of Portland cement, analysis methods, deterioration, as well as environmental and energetic benefits of using such an alternative material. A discussion on carbon credits is also offered. The contents of this paper could strengthen the prospects for the use of self-healing concrete as a way to meet the high demand for concrete, contributing to the building of a sustainable society.

Abstract:Developing bio-based self-healing concrete aims to minimize durability problems related to cracking. In this study, MICP was used as a smart and eco-friendly approach to produce bio-based durable materials. Bacillus pasteurii (BP) and Bacillus sphaericus (BS) were added into mortar mixtures with 0.25% and 0.5% cement weight. All treated samples exhibited a significant decline in water uptake, capillary permeability, and volume of permeable voids, as compared to control with no bacteria. All treated samples showed significant increase in compressive strength by 28-50%, after 28 days of curing. At the age of 120 days, the flexural strength of all treated samples was significantly increased by 19.29-65.94%. SEM imaging and EDAX confirmed that treated samples were denser with less voids due to MICP. DTA verified that the calcite amount and the crystallinity degree were improved in treated samples. Load deflection of bacterial Reinforced-Laminates had less deformation than control. Reloaded bacterial Reinforced-Laminates exhibited excellent restoration of physico-mechanical properties and performance, after 28, 90, and 120 days, confirming the healing process. Microbial self-healing is an innovative approach for continuous repair of micro-cracks in concrete, improving its durability, thus can reduce the maintenance costs.

Abstract:Self-healing of cracks improves durability and reduces maintenance of concrete. Microbial induced calcite precipitation is a novel approach to engage self-healing in concrete, and bacteria spores are protected from direct contact with the surrounding cement matrix to maintain their viability. This study proposes a novel single bacteria spore capsule via layer-by-layer (LbL) self-assembly of poly(dimethyldiallyl ammonium chloride) and silica nanoparticles to enhance the consistency of healing as well as to minimize the negative impact on the mechanical properties of the resulting concrete. The resulting single bacteria spore capsule has a size of 1 μm and the inclusion of the capsules dose not compromise the compressive strength of the matrix. Cement paste incorporating the capsules shows complete closure of large crack of few hundred microns and complete recovery of transport property. Healing products are observed along the entire crack from the surface to the interior.

Abstract:A large amount of anthropogenic CO 2 emissions are derived from Portland cement production, contributing to global warming, which threatens human health and exposes flora and fauna to ecological imbalance. With concerns about the high maintenance and repair costs of concrete, the development of microbially induced calcium carbonate precipitation (MICP)-based self-healing concrete has been extensively examined. Bacterial carriers for microcrack healing could enhance the concrete's self-healing capacity by maintaining bacterial activity and viability. To reduce cement consumption, the development of sustainable engineered living materials (ELMs) based on MICP has become a promising new research topic that combines synthetic biology and material science, and they can potentially serve as alternatives to traditional construction materials. This review aims to describe bacterial carriers and the ongoing development of advanced ELMs based on MICP. We also highlight the emerging issues linked to applying MICP technology at the commercial scale, including economic challenges and environmental concerns.

Abstract:Microbially induced calcium carbonate precipitation (MICP) has been widely explored and applied in the field of environmental engineering over the last decade. Calcium carbonate is naturally precipitated as a byproduct of various microbial metabolic activities. This biological process was brought into practical use to restore construction materials, strengthen and remediate soil, and sequester carbon. MICP has also been extensively examined for applications in self-healing concrete. Biogenic crack repair helps mitigate the high maintenance costs of concrete in an eco-friendly manner. In this process, calcium carbonate precipitation (CCP)-capable bacteria and nutrients are embedded inside the concrete. These bacteria are expected to increase the durability of the concrete by precipitating calcium carbonate in situ to heal cracks that develop in the concrete. However, several challenges exist with respect to embedding such bacteria; harsh conditions in concrete matrices are unsuitable for bacterial life, including high alkalinity (pH up to 13), high temperatures during manufacturing processes, and limited oxygen supply. Additionally, many biological factors, including the optimum conditions for MICP, the molecular mechanisms involved in MICP, the specific microorganisms suitable for application in concrete, the survival characteristics of the microorganisms embedded in concrete, and the amount of MICP in concrete, remain unclear. In this paper, metabolic pathways that result in conditions favorable for calcium carbonate precipitation, current and potential applications in concrete, and the remaining biological challenges are reviewed.
Physiological suitability of sulfate-reducing granules for the development of bioconcrete

Abstract:Regular monitoring and timely repair of concrete cracks are required to minimize further deterioration. Self-healing of cracks has been proposed as an alternative to the crack maintenance procedures. One of the proposed techniques is to use axenic cultures to exploit microbial-induced calcite precipitation (MICP). However, such healing agents are not cost-effective for in situ use. As the market for bio-based self-healing concrete necessitates a low-cost bio-agent, nonaxenic sulfate reducing bacterial (SRB) granules were investigated in this study through cultivation in an upflow anaerobic sludge blanket reactor. The compact granules can protect the bacteria from adverse conditions without encapsulation. This study investigated the microbial activities of SRB granules at different temperatures, pH, and chemical oxygen demand concentrations which the microbes would experience during the concrete casting and curing process. The attenuation and recovery of microbial activities were measured before and after the exposure. Moreover, the MICP yield was also tested for a possible use in self-healing bioconcrete. The results consistently showed that SRB granules were able to survive starvation, high temperature (50-60°C), and high pH (12), together with scanning electron microscope/energy dispersive spectrometry/X-ray diffraction analysis evidence. Microbial staining analysis demonstrated the formation of spores in the granules during their exposure to harsh conditions. SRB granule was thus demonstrated to be a viable self-healing nonaxenic agent for low-cost bioconcrete.

Abstract:The self-healing of concrete through the use of bacteria is an ecological, sustainable, and environmentally friendly method that can replace time-consuming and costly repairs. The aim of this research was to isolate and select spore-forming bacilli with potential for concrete self-healing. Soil samples from the limestone mines of Simbal, Trujillo, La Libertad, Peru were used. These samples were processed to primarily isolate spore-forming bacteria. The self-healing potential of these bacteria was evaluated through their ability to produce the urease enzyme, grow at an alkaline pH, and produce calcium carbonate crystals in a culture medium with urea and CaCl 2 at pH 8.0. The produced crystals were visualized with scanning electron microscopy and their calcium content was confirmed through EDS analysis. The isolated bacteria were identified by 16 S rRNA gene amplification by PCR as Bacillus sp and Streptomyces sp. It was observed that Bacillus sp. produces CaCO 3 crystals with vaterite morphology, while Streptomyces sp. produces rhombohedral calcite crystals. It is concluded that the isolated bacteria have the potential to be used in concrete self-healing processes.

Abstract:A total of 12 billion tons of concrete materials are produced annually, about 2 tons per person. More sustainable buildings are thus needed to decrease the carbon footprint of concrete infrastructures in the context of climate change. Crack formation is a major flaw of concrete structures. Although cracks are usually small and do not necessarily induce building collapse, cracks reduce the life span and sustainability of buildings. Therefore, research has developed self-healing materials that are capable of repairing narrow cracks automatically. Here we review self-healing technologies such as adding mineral mixtures, bacteria, and adhesive liquids. Mineral healing is economically positive, yet relies on suitable conditions such as the presence of water, and is less efficient to heal larger cracks. By contrast, the bacterial encapsulation is promising due to the uniform characteristics of bacteria in the alkaline environment of concrete.

Abstract:Cementitious materials deteriorate progressively with the formation of cracks that occur due to diverse physical, chemical, thermal, and biological processes. Numerous strategies have been adopted to obtain cement-based self-healing materials and determine the novel self-healing mechanisms. The uses of microbes have been established to improve the thickness of the healed crack and mechanical properties of the concrete, a phenomenon seldom addressed in the literature. Based on these factors, this article comprehensively appraises the smart bio-agents-based autonomous healing performance of concrete to demonstrate the recent progress, expected benefits, and ongoing challenges. The fundamentals, design strategies, and efficacy of the smart bio-agents-activated self-healing cementitious materials are the recurring themes of this overview. Furthermore, the effects of various processing parameters on the performance of cementitious self-healing smart bio-agents are discussed in-depth. The achievements, knowledge gaps, and needs for future research in this ever-evolving area for the sustainability and resilience of the built environment are highlighted.

Abstract:The bioinspired synthesis of construction material, known as biocement, represents a significant advancement in addressing the environmental sustainability issues associated with traditional cement use in the built environment. Biocement is produced through the process of microbially induced bio-mineralization (MIBM), which offers a promising alternative or supplement to conventional cement, potentially reducing its consumption. Despite extensive literature on the application of biocement in construction biotechnology, the fundamental mechanisms underlying its ability to enhance concrete quality remain poorly understood. This study focuses on the kinetics of biomineral synthesis by two Bacillus species; Bacillus megaterium RB05 and Bacillus foraminis DRG5, to identify the most effective strain for biomineralization. Bioconcrete specimens were created by adding inoculum containing Bacillus megaterium RB05 cells with a nutrient solution to the concrete mixture in a layer-by-layer approach. After 28 days of water curing, nanoparticles of CaCO3, ranging in size from 27 to 82 nm, were produced in the bioconcrete specimens. The resulting concrete, containing nanocrystalline biogenic calcite, demonstrated significant improvements in mechanical properties. Specifically, compressive and tensile strengths of the bioconcrete, tested using a universal testing machine (UTM), increased by 7.69 ± 0.08% and 22 ± 0.1%, respectively, after 72 h of curing. Additionally, the biocement was found to exhibit an organic-inorganic hybrid nature, as identified by TEM, EDAX, FESEM, FTIR, and XRD analyses. The enhanced mechanical properties were attributed to the high surface-to-volume ratio and hybrid nature of the calcite nanoparticles. The findings of this investigation are encouraging, suggesting the potential development of future green and self-sustainable construction materials or bioconcrete.

Abstract:The Cracking of concrete is a common phenomenon without immediate and proper treatment, cracks in concrete structures tends to expand further and eventually require costly repairs. Even though it is possible to reduce the extent of cracking by available modern technology, remediation of cracks in concrete has been the subject of research for many years. Cracks and fissures are a common problem in building structures, pavements, and historic monuments. Cement mortar durability is the function of its internal pore structure and distribution, porosity, and its permeation properties. Research has shown that some specific bacterial species isolated from soil can tolerate harsh and challenging alkaline environment and can be used in remediating cracks in cement mortar structures. This state-of-the-art microbial based crack healing mechanism is one such phenomenon on which studies were carried out to investigate the role of calcite mineral precipitation in improvement of durability in bacteria integrated cement mortar. The primary goal of this study is to explore the potential of biomineralization microbial calcium carbonate deposition in cement mortar, to develop sustainable construction materials. The idea has led to the conception of energy efficient and sustainable construction material called ‘Bacterial Cement mortar’. This paper primarily focuses on the studies related to the characterization of bacteria produced calcium carbonate crystals using various nano characterization techniques such as Scanning Electron Microscope (SEM), X-ray diffraction (XRD), and Thermo gravimetric analysis (TGA) to validate that cracks/pore were sealed up by calcite crystals grown due to complex metabolic mechanism of nitrogen cycle by Bacillus subtilis JC3. In concrete, cracking is a common phenomenon developed due to relatively low tensile strength. High tensile strength may be developed in concrete due to external loads, imposed deformations, plastic shrinkage, plastic settlement and expensive reaction. Proper and immediate treatment should be done to prevent expansion of cracks which may eventually be of higher cost. For crack repair, a variety of traditional repair systems are available which possess several disadvantages aspect such as different thermal expansion coefficient, environmental and hazards of health. Bacterially induced calcium carbonate precipitation has been proposed as an alternative and environmentally friendly crack repair technique. It is found that microbial mineral precipitation as a result from metabolic activities of favorable bacteria in concrete improved the overall behavior of concrete. It is expected that further development of this technique will result in a more durable, sustainable, and crack free concrete that can be used efficiently for construction in wet atmosphere where corrosion of reinforcement affects the durability, permeability, and strength of concrete.
