Abstract:Rosin, an abundant raw material from pine trees, was used as a starting material directly for the synthesis of rosin acid starch. The esterification reaction was catalyzed by lipase (Novozym 435) under mild conditions. Based on single factor experimentation, the optimal esterification conditions were obtained as follows: rosin acid/anhydrous glucose unit in the molar ratio 2 : 1, reaction time 4 h at 45°C, and 15% of lipase dosage. The degree of substitution (DS) reaches 0.098. Product from esterification of cassava starch with rosin acid was confirmed by FTIR spectroscopy and iodine coloration analysis. Scanning electron microscopy and X-ray diffraction analysis showed that the morphology and crystallinity of the cassava starch were largely destroyed. Thermogravimetric analysis indicated that thermal stability of rosin acid starch decreased compared with native starch.

Effect of ultrasonic pre‐treatments on the octenyl succinic anhydride substitution of potato starch and its physicochemical and emulsifying properties

Abstract:To investigate whether the ultrasonic pre-treatments of starch will affect the octenyl succinic anhydride (OSA) esterification, different power ultrasonic pre-treatments (150, 300, 450 and 600 W) were performed on potato starch, then the OSA esterification was performed and the physicochemical as well as the emulsifying properties of the OSA starch were analysed. Results indicated that ultrasonic pre-treatments decreased the degree of substitution and reaction efficiency by 30%, regardless of the pre-treatments condition, while little differences were observed in the crystalline type and the long-range ordered structure. DSC results demonstrated that low-power ultrasound decreased the values of To and Tp, but increased Delta H, while high-power ultrasound had the opposite effect. In addition, high-power sonication improved the emulsifying capacity of OSA starch, which ranged from 189.08 +/- 0.23 m(2)/g to 334.70 +/- 0.35 m(2)/g, while emulsion stability decreased from 277.02 +/- 10.83 min to 42.75 +/- 0.08 min. Overall, the result of the present work implied that OSA starch exhibited varied thermal and emulsifying properties under different ultrasonic pre-treatments, and our work might provide basic theory for the preparation of OSA starch.

Abstract:Phosphorylated amphoteric starch was prepared from enzyme-modified tapioca starch in the aqueous alcoholic-alkaline medium. Optimization of the reaction parameters for maximum degree of substitution (DS) of phosphorylated amphoteric starch were carried out using response surface methodology (RSM). Optimum modification conditions were shown as following: NaOH concentration 3.71 g/100 g starch (dry basis), CHPTAC concentration 6.34 mL/100 g starch (dry basis), STP concentration 6.13 g/100 g starch (dry basis), water/ethanol ratio 1.16, reaction temperature 50°C, reaction time 3 h, and the starch concentration 30%. Under the optimal condition, the maximum anion degree of substitution (DS A) and cation degree of substitution (DS C) were 0.0274 and 0.0408, respectively. Compared with the DS of the amphoteric starch prepared from native tapioca starch (NPTAS), the DS of the amphoteric starch from enzyme-pretreated tapioca starch (EPTAS) was higher at the same reaction conditions, suggesting enzymatic pretreatment improved the reaction processes significantly. Scanning electron microscopy and X-ray diffraction data showed that the granular shape and crystalline structure of EPTAS did not change. Compared with NPTAS, the pasting stability of EPTAS with the same DS was also significantly improved. © 2012 Elsevier Ltd. All rights reserved.

Abstract:Cornstarch granules were allowed to react with α-amylase and then esterified with octenyl succinic anhydride (OSA). The paste properties and the distribution of octenyl succinate (OS) groups of OS-starch granule were studied. Results show that the α-amylase pretreatments significantly decreased the degree of substitution (DS) of OS-starch compare with a non-pretreated control group, while the retrogradation properties, freeze-thaw stability and shearing resistant stability of α-amylase pretreated OS-starch didn't change markedly. Enzymatic pretreatment appears to influence the distribution of the OS groups from being mostly on the surface and amorphous part of the starch granules to being in the inner amorphous and crystalline regions of the granules, as shown by X-ray photoelectron spectroscopy (XPS) and lintnerisation studies.
Effects of Combined Pretreatment of Dilute Acid Pre-Extraction and Chemical-Assisted Mechanical Refining on Enzymatic Hydrolysis of Lignocellulosic Biomass

Abstract:An efficient enzymatic hydrolysis of lignocellulosic biomass into fermentable sugars depends greatly on the pretreatment of raw materials. In this study, a combination of dilute acid pre-extraction and chemical-assisted mechanical refining was used to pretreat wood lignocellulosic biomass for subsequent enzymatic hydrolysis. This work analyzed the surface lignin concentration, specific surface area, crystallinity, fines content, fiber length, and kink index of the resultant pulp substrates and their effects on the enzymatic hydrolysis. The results showed that the combined pretreatment significantly enhanced the enzymatic hydrolysis efficiency, and the maximum glucose conversion yield and glucose concentration were 93.32% and 21.41 g L-1, respectively. It is found that the surface lignin concentration, specific surface area, and fines content significantly affected the enzymatic hydrolysis.

Abstract:Extrusion process using 1-ethyl-3-methylimidazolium acetate ([EMIM]Ac) in co-solvent, dimethyl sulfoxide (DMSO), was conducted to improve the pretreatment effect on the enzymatic saccharification of pussy willow (<i>Salix gracilistyla</i> Miq.). The use of a twin screw extruder capable of continuous processing increased the solid loading to 50% in [EMIM]Ac and its co-solvent DMSO. The water-soluble fraction was increased by increasing the [EMIM]Ac amount in co-solvent and the extrusion temperature but decreasing the screw rotating speed and solid loading. The surface morphologies of the extruded product showed remarkable disruption of the cell wall structures and fibrillation with increasing amounts of [EMIM]Ac in co-solvent. With increasing [EMIM]Ac amount in co-solvent and extrusion temperature and with decreasing screw rotating speed and solid loading, the crystalline structure of cellulose was more disrupted, resulting in the increase in saccharification yield. The highest yield of glucose and xylose was 99.0% and 99.5%, respectively, from the pretreated products using only [EMIM]Ac under solid loading of 15% at 160 °C extrusion temperature and 5 rpm screw rotating speed. The initial saccharification rate was faster in the pretreated product precipitated in alcohol and acetone than in water.

Abstract:Enzymatic modification of starch was conducted by lipase-coupling esterification with octenyl succinic anhydride (OSA). Parameters affecting the esterification were systematically studied. Products were characterized by Fourier transform infrared (FT-IR) spectroscopy, scanning electron microscopy (SEM), X-ray diffraction, differential scanning calorimetry (DSC) and viscosity analysis (VA). Optimum condition for lipase-coupling OSA starch preparation was as follows: starch pretreatment at 65 °C for 15 min, starch concentration 35%, amount of lipase and OSA, 0.6% and 3%, reaction pH, temperature and time, 8.0, 40 °C and 30 min respectively, which resulted in 0.0195 of the degree of substitution and 84.05 ± 2.07% of the reaction efficiency. FT-IR spectroscopy confirmed the formation of OSA starch. SEM and X-ray diffraction showed apparent surface change, but no crystalline change. DSC and VA results indicated the synthesized OSA starch gelatinized rapidly with high viscosity. Attractively, reaction time drastically reduced to 30 min, showing vast potential for scale production of OSA starch. © 2011 Elsevier Ltd. All rights reserved.

Abstract:Corn starch was dually modified using thermostable α-amylase and pullulanase to prepare resistant starch (RS). The concentration of starch liquid, the amount of added thermostable α-amylase, the duration of enzymatic hydrolysis and the amount of added pullulanase were optimized using RSM to increase RS content of the treated sample. The optimum pretreatment conditions were 15% starch liquid, 3 U/g thermostable α-amylase, 35 min of enzymatic hydrolysis and 8 U/g pullulanase. The maximum RS content of 10.75% was obtained, and this value was significantly higher than that of native corn starch. The degree of polymerization (DP) of the enzyme-modified starch decreased compared with that of native starch. The scanning electron microscopy (SEM) and differential scanning calorimetry (DSC) were performed to assess structural changes in native and pretreated starch. The effect of dual enzyme pretreatment on the structure and properties of corn starch was significant. Unlike the untreated one, the pretreated corn starch showed clear pores and cracks. Significant differences in RS contents and structural characterization between starch pretreated and untreated with dual enzymes demonstrated that the dual enzyme modification of corn was effective in enhancing RS contents.

Abstract:Upon Dess‐Martin periodinane oxidization, starch macromolecule chain was cleaved, promoting esterification reaction level. After esterification, viscosity and gelatinization temperature decreased but transparency increased. A combined middle‐level DMP oxidization and esterification can generate low‐viscosity starch. Esterification is a normal method for starch modification while reaction condition is not clear. Here, pure corn starch and vinyl acetate were chosen for starch esterification. Reaction conditions involving esterifying agent content, pH, reaction time and temperature were explored. Dess‐Martin periodinane (DMP) pretreatment was adopted to accelerate esterification. Main results reveal that a content of 30 wt% vinyl acetate, pH = 9, time as 1 h and temperature as 45°C is an optimized condition for vinyl acetate esterification and DMP oxidization can enhance it. After modification, variations in microstructure, surface morphology and macroscopic properties such as viscosity, transparency, adhesion property were explored by FT‐IR, XRD, SEM, TGA, UV–Vis, viscosity, and slushing experiments. Main results reveal that after esterification carbonyl groups were introduced, lattice plane occurred expansion, surface morphology was destroyed, water absorption capacity, viscosity, and gelatinization temperature decreased while transparency, thermal‐stability and adhesion property increased. DMP treatment enhanced the previous variations. It seems via a middle‐level DMP pretreatment and vinyl acetate esterification starch solution viscosity and gelatinization temperature decreased greatly while transparency and adhesion force increased greatly, possibly having a potential use in textile industry. For instance, using as slurry its manufacture cost can be decreased with decreased energy consumption, economical.

Abstract:Fenton's reagent, ultrasound, and the combination of Fenton's reagent and ultrasound were used to pretreat rice straw (RS) to increase its enzymatic digestibility for saccharification. The characterization shows that compared with ultrasound, Fenton's reagent pretreatment was more efficient in increasing the specific surface area and decreasing the degree of polymerization (DP) of RS. The enzymatic hydrolysis results showed that the RS pretreated by ultrasound-assisted Fenton's reagent (U/F-RS), which exhibited the largest specific surface area and the lowest DP value, had the highest enzymatic activity, and the amount of reducing sugar released from U/F-RS at 48h of enzymatic saccharification is about 4 times as large as that from raw RS and 1.5 times as large as that from Fenton's reagent pretreated RS. The ultrasound-assisted Fenton process provides a reliable and effective method for RS pretreatment.

Abstract:<p>To reduce the cost of lignocellulosic pretreatment, rice straw was ensiled with dilute formic acid (FA, 0, 0.2, 0.4, and 0.6%) for 3, 6, 9, 15 and 30 days, and evaluated its effects on fermentation dynamics, lignocellulosic degradation and enzymatic hydrolysis. The results showed that the application of FA, especially at 0.6% level, reduced total fermentation losses of the resulting silages, as evidenced by low dry matter loss, ammonia nitrogen and ethanol content. Meanwhile, the 0.6% FA application promoted hemicellulose removal (232.41 vs 187.52 g/kg DM) and xylose production (0.35 vs 2.80 g/kg DM). The glucose yield and cellulose convertibility of rice straw increased after 30 days of ensiling, and further enhanced by the 0.6% FA application. In conclusion, the 0.6% FA-assisted ensiling pretreatment improved both biomass preservation, hemicellulose removal and enzymatic hydrolysis of rice straw, which is beneficial to the subsequent biofuel production chain.</p>

Abstract:Dodecenylsuccinic anhydride (DDSA) has been widely used to obtain amphiphilic starches. In this study, we investigated the functionalities of synchronous intermissive multi-ultrasound-assisted esterified starch. Compared to native starch (NS), it was deduced that multi-ultrasound-modified starch (US), esterified starch (ES), and multi-ultrasound-assisted esterified starch (UES) exhibited increased viscosities but reduced gelatinization temperatures and thermal stabilities. The viscoelastic moduli, retrogradation behaviors and hydrophobicity of the ES and UES species significantly altered. Moreover, the results of structural characterization suggested that esterification reduced the molecular weight and structural order of starch, whereas the intermissive ultrasonication treatment did not aggravate the structural disruption of ES. Additionally, compared with NS and US, the emulsification abilities of the ES and UES specimens were improved, leading to the desirable effect of stabilizing astaxanthin. Overall, this study provides a method for preparing amphiphilic starch, which can be exploited as a potential emulsifier and emulsion stabilizer for bioactive compounds.

Preparation and characterization of rice starch citrates by superheated steam: A new strategy of producing resistant starch

Abstract:In this study, starch citrates were synthesized by superheated steam (SS) treatment without changing the granule structure of starch. The degree of substitution (DS) of starch citrates prepared in present study was ranged from 0.015 to 0.064. Fourier transform infrared spectroscopy confirmed that citric acid (CA) and starch were successfully esterified with an absorption peak appeared at 1735 cm−1. In vitro digestion results revealed that the content of resistant starch (RS) was significantly increased by citric acid esterification, which was attributed to the cross-linked structure of starch and the inhibition of gelatinization and swelling. According to the physicochemical properties analysis, the pasting temperature of rice starch citrates were significantly increased by citric acid treatment, while the swelling power and gelatinization enthalpy were significantly decreased and the short- and long-range molecular order of the starch was disrupted after the esterification reaction. These results indicated that it is a feasible method to synthesize starch citrate by superheated steam to increase resistant starch, and it could be used as an alternative approach for preparing starch citrate to conventional hot air treatment.
Hydrogen Peroxide-Assisted Sodium Carbonate Pretreatment for the Enhancement of Enzymatic Saccharification of Corn Stover

Abstract:In this work, the effectiveness of hydrogen peroxide-assisted sodium carbonate (HSC) pretreatment of corn stover to produce fermentable sugars was investigated. It was found that the addition of H2O2 (a green oxidative agent) in Na2CO3 pretreatment could enhance the lignin removal and increase the hydrophilicity of the residual lignin, leading to the improved enzymatic digestibility and the yield of fermentable sugars. Also, results showed that lignin removal (in the range of 12–65%) was positively and linearly correlated with final total sugar yields. Post PFI refining could further improve the enzyme accessibility by increasing porosity of substrates. After HSC pretreatment (40 wt % Na2CO3 and 15 wt % H2O2, 120 °C for 60 min) and enzymatic hydrolysis, the final total sugar yield could achieve 79%, which was about 10% higher in comparison with the conventional Na2CO3 pretreatment under the same conditions. HSC pretreatment will have less environmental impact and less barriers for large scale production.

Abstract:The starch suspension was treated by ultra‐high pressure homogenization under various pressures, the starch granules were broken. Subsequent pullulanase debranching produced large amounts of short linear amylose. The final temperature‐cycled crystallization promoted the formation of the double helix structure by hydrogen bonds between starch chains. This study examined the structural characterization and in vitro digestibility of rice starch processed with the combination of ultra‐high pressure homogenization (UHPH), debranching, and temperature‐cycled crystallization under various pressures (0–160 MPa). After UHPH treatment, the degree of branching (DB) of processed starches (0.65%–2.03%) was lower than that of the native rice starch (NS, 2.96%). Assessment of chain length distribution suggested that the considerable amylose was produced after treatments. Processed starches exhibited an irregular fragmented structure, and the crystalline structure was transformed from A‐type (native rice starch) into B + V‐type (processed starches). Moreover, the relative crystallinity (RC) and the absorbance ratio of 1047/1022 cm−1 (R1047/1022) of processed starches decreased from NS (RC, 22.99%; R1047/1022, 0.79) to 13.47%–18.21% (RC) and 0.51–0.78 (R1047/1022), respectively. Iodine binding curves revealed that processed starches possessed the higher degree of polymerization (DP). Furthermore, the contents of slowly digestible starch (SDS) and resistant starch (RS) increased in all processed starches, and the RS content of starch treated under 160 MPa (35.33%) was notably greater than that of native starch (9.37%). These results suggested that the UHPH can be used as a pretreatment strategy to reduce the digestibility of starch products. Practical applications UHPH pretreatment disrupts starch molecular chains to produce the large amounts of short linear amylose. Debranching followed by temperature‐cycled crystallization can increase the content of resistant starch (RS), yielding a raw material suitable for boosting the health of the digestive system and overall health. Therefore, UHPH pretreatment has a high application value in the field of starch modification.

Abstract:Rice starch was subjected to enzymatic treatment with pullulanase and the physicochemical properties of the modified rice starch were investigated as a function of the degree of hydrolysis which ranged from 0.9 to 10.2%. The enzymatic hydrolysis of rice starch caused a decrease in the fraction of rapidly digestible starch (RDS) (78.31→44.65%), whereas the levels of slowly digestible starch (SDS) (0.8→22.18%) and resistant starch (RS) (20.79→34.43%) increased, consequently reducing in vitro starch digestibility. In addition, the swelling factor of rice starch decreased with increasing enzyme concentration while an increase in its syneresis was observed. In rapid visco analyser (RVA) measurement, dramatic decreases in the peak viscosity (452.5→12.0 cp), breakdown (307.0→4.5 cp), and setback (207.0→3.0 cp) were observed in the enzyme-treated rice starch of which X-ray diffraction showed that there was the coexistence of A-and V-type crystallinity.
