Abstract:Owing to flexible structural design and rapid prototyping manufacturing, 3D printable hydrogels with solid-like and fluid-like characters have emerged as a promising candidate for biomedical engineering, soft robotics, and flexible electronics. However, existing 3D printing of hydrogel place substantial constraints on hydrogel high-fidelity of the printed hydrogel complex structures due to the shape deformation and collapse caused by water evaporation. Herein, we propose a mixed solvent printing strategy followed with water dialysis to achieve vat photopolymerization 3D printing of high-fidelity hydrogel structure by restraining water evaporation. Concretely, dimethyl sulfoxide (DMSO) is introduced into the hydrogel ink as co-solvent to improve the anti-dehydration performance during 3D printing. The as-prepared gels are then transformed into hydrogels with good fidelity and excellent performance through solvent exchange and water dialysis. Enabled by these characteristics, various functional devices with complex structures are demonstrated with the present high-fidelity 3D printing of diverse hydrogels. The solvent assisted high-fidelity hydrogel 3D printing protocol combining with the compatibility to various high-performance hydrogels is believed to be promising for wearable devices, wound dressings, electronic skin, and intelligent robots.

Abstract:Light-based hydrogel crosslinking is a new approach in rapid and high-resolution 3D printing; however, using this method in tissue engineering is challenging due to the toxicity of photoinitiators, their solvents, and their low efficiency. Herein, a novel, water-soluble photoinitiator with high efficiency in light-based 3D printing is introduced. Low-cost photoinitator, 2,4,6-trimethylbenzoylphenyl phosphinate, is converted into nanoparticles via a microemulsion method and dispersed in water. Cell toxicity assays were performed to prove that these nanoparticles are non-toxic and can be used in biomedical applications. Finally, the nanoparticles were utilized in the high-accuracy 3D printing of hydrogels. The results of this study indicate that these particles are potent to be used in bioprinting.

Abstract:In this article, a preparation method of a water-dispersible photoinitiator, which can be used for three-dimensional (3D) printing of the hydrogel and can be initiated under visible light is proposed. The oil-soluble photoinitiator 2,4,6-trimethylbenzoyl-diphenylphosphine oxide (TPO) is nano-wrapped with surfactant Tween 20 to obtain the oil-in-water (O/W) miniemulsion, and then it is successfully transformed into water-dispersible nanospheres without the use of organic solvents or crystallization inhibitors. The morphology and optical properties were characterized by transmission electron microscopy (TEM), particle size analysis and ultraviolet-visible (UV-Vis) spectroscopy. Subsequently, the water-dispersible nano-photoinitiator was used to realize the photopolymerization of acrylamide/N,N '-methylenebisacrylamide hydrogel under visible light (405 nm). Then, the micromorphology of the photocuring process of the hydrogel was observed by TEM and the photocuring activity of the initiator was evaluated. Finally, a viscosifier suitable for AM/MBA hydrogel system was screened out, and the direct ink writing (DIW) 3D printing of the hydrogel was successfully realized by using this nano-photoinitiator.
General One-Pot Method for Preparing Highly Water-Soluble and Biocompatible Photoinitiators for Digital Light Processing-Based 3D Printing of Hydrogels

Abstract:We report a facile but general method to prepare highly water-soluble and biocompatible photoinitiators for digital light processing (DLP)-based 3D printing of high-resolution hydrogel structures. Through a simple and straightforward one-pot procedure, we can synthesize a metal-phenyl(2,4,6-trimethylbenzoyl)phosphinates (M-TMPP)-based photoinitiator with excellent water solubility (up to ∼50 g/L), which is much higher than that of previously reported water-soluble photoinitiators. The M-TMPP aqueous solutions show excellent biocompatibility, which meets the prerequisite for biomedical applications. Moreover, we used M-TMPP to prepare visible light (405 nm)-curable hydrogel precursor solutions for 3D printing hydrogel structures with a high water content (80 wt %), high resolution (∼7 μm), high deformability (more than 80% compression), and complex geometry. The printed hydrogel structures demonstrate great potential in flexible electronic sensors due to the fast mechanical response and high stability under cyclic loadings.

Abstract:The direct ink writing (DIW) technology is probably the most feasible method for 3D printing hydrogels. The collaborative development of photosensitive hydrogel and DIW printing equipment based on specific application goals is a continuous research hotspot. To avoid UV damage to the skin, the development of an aqueous photoinitiator that can be initiated under visible light is of great significance for 3D printing hydrogel technology. A surfactant with π - π stacking ability is selected, and the water-based photoinitiator (Aqu-PI) is formed by supramolecular assembly with TPO without VOC emission by solvents. The particle size of the Aqu-PI is around 15 nm, and the absorption peak of its UV-visible spectrum reaches the visible region. The photosensitive hydrogel is prepared by Aqu-PI and aqueous monomers including mono functional monomer (AM) and bifunctional monomer (MBA). Then the tackifier is added which makes the viscosity enhance into the photosensitive hydrogel, so hydrogels with better mechanical properties are obtained. Then the photosensitive hydrogel is photocured by a 405 nm LED laser on the extrusion device of DIW printer and forms the interpenetrating network (IPN) structure, thus a 3D printout hydrogel is obtained consistent with the shape of the digital design model.

Abstract:Although 3D printing can easily control the macroscopic structure of printed subjects based on the digital modeling, the control of micro-structure of polymer network was still a challenge. Herein, we report the using of macroinitiator induced rapid photoATRP to regulate the 3D printed hydrogel network. This approach consists of synthesizing of macroinitiator through CDs catalyzed photoATRP, and photocuring in the same pot by subsequent adding of crosslinker. By varying the chain length of the macroinitiator, we demonstrated the effect of physical entanglements of on the formation of hydrogel, as well as the mechanical property and swelling ratio. All the 3D printing objects with different macroinitiators exhibited outstanding print resolution. With PNIPAM macroinitiator, the printed hydrogel shows the characteristics of temperature-sensitive response. Overall, the findings of this work enable an interesting platform for control the mechanical property and functional of 3D printing hydrogel.

Abstract:3D printing technology has been widely explored for the rapid design and fabrication of hydrogels, as required by complicated soft structures and devices. Here, a new 3D printing method is presented based on the rheology modifier of Carbomer for direct ink writing of various functional hydrogels. Carbomer is shown to be highly efficient in providing ideal rheological behaviors for multifunctional hydrogel inks, including double network hydrogels, magnetic hydrogels, temperature-sensitive hydrogels, and biogels, with a low dosage (at least 0.5% w/v) recorded. Besides the excellent printing performance, mechanical behaviors, and biocompatibility, the 3D printed multifunctional hydrogels enable various soft devices, including loadable webs, soft robots, 4D printed leaves, and hydrogel Petri dishes. Moreover, with its unprecedented capability, the Carbomer-based 3D printing method opens new avenues for bioprinting manufacturing and integrated hydrogel devices.

Abstract:3D printing hydrogels with a good resolution, diversified geometries, and a tunable mechanical performance is still a big challenge, which is mainly due to the low photo-reactivity of the high-water containing precursor and the low modulus of the photo-cured hydrogel. In the present work, an acryloylmorpholine - acrylic acid - PEG diacrylate drygel was firstly printed with a good fidelity and subsequently post- treated with an aqueous solution containing specific ions, during which hydration of the hydrophilic network and coordination between the carboxyl and ions occurred simultaneously. The obtained dual-crosslinked hydrogel combined a complex geometry and a good stretchability. This approach decoupled the printability and mechanical properties of the hydrogels. To furtherly show this advantage over conventional direct hydrogel 3D printing, ultrasoft hydrogel lattice structures which cannot be self-supported and purely ionic crosslinked hydrogels with a good self-weldability were demonstrated.

Abstract:Stimuli-responsive hydrogels and/or composite hydrogels have been of great interest for various printing applications including four-dimensional printing. Although various responsive hydrogels and/or composite hydrogels have been found to respond to given stimuli and change shapes as designed, the fabrication of three-dimensional (3D) structures from such responsive hydrogels is still a challenge due to their poor 3D printability, and most of the responsive material-based patterns are two-dimensional (2D) in nature. In this study, Laponite nanoclay is studied as an effective additive to improve the self-supporting printability of N-isopropylacrylamide (NIPAAm), a thermoresponsive hydrogel precursor while keeping the responsive functionality of NIPAAm. Graphene oxide (GO) is further added as a nanoscale heater, responding to near-infrared radiation. Due to the different shrinking ratios and mechanical properties of the poly( N-isopropylacrylamide) (pNIPAAm)-Laponite and pNIPAAm-Laponite-GO nanocomposite hydrogels, printed 2D patterns deform in a predictable way. In addition, 3D microfluidic valves are directly printed and cured in air, which can effectively control the flow directions in response to different stimuli as validated in a microfluidic system. Because Laponite nanoclay can be mixed with various responsive hydrogel precursors to improve their 3D printability, the proposed Laponite nanoclay-based nanocomposite hydrogels can be further expanded to prepare various 3D printable responsive nanocomposite hydrogels.

Abstract:The rapid development of additive manufacturing techniques in the field of tissue regeneration offers unprecedented success for artificial tissue and organ fabrication. However, some limitations still remain for current bioinks, such as the compromised cell viability after printing, the low cross-linking efficiency leading to poor printing resolution and speed due to the relatively slow gelation rate, and the requirement of external stimuli for gelation. To address these problems, herein, a biocompatible and printable instant gelation hydrogel system has been developed based on a designed hyperbranched poly(ethylene glycol) (PEG)-based multihydrazide macro-cross-linker (HB-PEG-HDZ) and an aldehyde-functionalized hyaluronic acid (HA-CHO). HB-PEG-HDZ is prepared by the postfunctionalization of hyperbranched PEG-based multivinyl macromer via thiol-ene chemistry. Owing to the high functional group density of HB-PEG-HDZ, the hydrogel can be formed instantly upon mixing the solutions of two components. The reversible cross-linking mechanism between the hydrazide and aldehyde groups endows the hydrogel with shear-thinning and self-healing properties. The minimally toxic components and cross-linking chemistry allow the resulting hydrogel to be a biocompatible niche. Moreover, the fast sol-to-gel transition of the hydrogel, combining all of the advanced characteristics of this platform, protects the cells during the printing procedure, avoids their damage during extrusion, and improves the transplanted cell survival.
Preparation of a Photocurable Hydrogel with Adjustable Mechanical Properties for 3D Printing

Abstract:Purpose This paper aims to show a series of hydrogels with adjustable mechanical properties, which can be cured quickly with visible light. The hydrogel is prepared conveniently with hydroxyethyl acrylate, cross-linker, gelatin and photoinitiator, and can be printed into certain 3D patterns with the direct ink write (DIW) 3D printer designed and developed by the research group. Design/methodology/approach In this paper, the authors designed a composite sensitization initiation system that is suitable for hydrogels. The concentration of photoinitiator, gelatin and cross-linker was studied to optimize the curing efficiency and adjust the mechanical properties. A DIW 3D printer was designed for the printing of hydrogel. Pre-gel solution was loaded into printer for printing into established models. The models were made and sliced with software. Findings The hydrogels can be cured efficiently with 405-nm visible light. While adding various content of gelatin and cross-linker, the mechanical properties of hydrogels show from soft and fragile (elastic modulus of 121.18 kPa and work of tension of 218.11 kJ·m−3) to rigid and tough (elastic modulus of 505.15 kPa and work of tension of 969.00 kJ·m−3). The hydrogels have high capacity of water absorption. With the DIW 3D printer, pre-gel hydrogel solution can be printed into objects with certain dimension. Originality/value In this work, a composite sensitization initiation system was designed, and fast curing hydrogels with adjustable mechanical properties had been prepared conveniently, which has high equilibrium water content and 3D printability with the DIW 3D printer.

Abstract:Abstract Additive manufacturing technologies based on stereolithography rely on initiating spatial photopolymerization by using photoinitiators activated by UV-visible light. Many applications requiring printing in water are limited since water-soluble photoinitiators are scarce, and their price is skyrocketing. On the contrary, thermal initiators are widely used in the chemical industry for polymerization processes due to their low cost and simplicity of initiation by heat at low temperatures. However, such initiators were never used in 3D printing technologies, such as vat photopolymerization stereolithography, since localizing the heat at specific printing voxels is impossible. Here we propose using a thermal initiator for 3D printing for localized polymerization processes by near-infrared and visible light irradiation without conventional photoinitiators. This is enabled by using gold nanorods or silver nanoparticles at very low concentrations as photothermal converters in aqueous and non-aqueous mediums. Our proof of concept demonstrates the fabrication of hydrogel and polymeric objects using stereolithography-based 3D printers, vat photopolymerization, and two-photon printing.

Using DLP to print hydrogel structures with high resolution requires the addition of water-soluble photo-absorbers to absorb excess light and confine photopolymerization to the desired area. However, the current photo-absorbers for hydrogel printing are neither efficient to absorb the excess light nor water-soluble. Herein, we report a volatile microemulsion template method that converts a wide range of commercial non-water-soluble photo-absorbers including Sudan orange G, quercetin, and many others to water-soluble nanoparticles with solubility above 1.0 g mL-1. After using these water-soluble photo-absorber nanoparticles, the highest lateral and vertical resolutions of printing high-water-content (70-80 wt%) hydrogels can be improved to 5 μm and 20 μm, respectively. Moreover, the quercetin nanoparticle can be easily washed out so that we achieve colorless and transparent printed hydrogel structures with excellent mechanical deformability and biocompatibility as well as thermally controllable variations on transparency and actuation. The proposed methods pave a new efficient way to develop water-soluble photo-absorbers, which helps to greatly improve the printing resolution of the high-water-content hydrogel structure and would be beneficial to extend the application scope of hydrogels.

Abstract:As the biocompatible materials, hydrogels have been widely used in three- dimensional (3D) bioprinting/organ printing to load cell for tissue engineering. It is important to precisely control hydrogels deposition during printing the mimic organ structures. However, the printability of hydrogels about printing parameters is seldom addressed. In this paper, we systemically investigated the printability of hydrogels from printing lines (one dimensional, 1D structures) to printing lattices/films (two dimensional, 2D structures) and printing 3D structures with a special attention to the accurate printing. After a series of experiments, we discovered the relationships between the important factors such as air pressure, feedrate, or even printing distance and the printing quality of the expected structures. Dumbbell shape was observed in the lattice structures printing due to the hydrogel diffuses at the intersection. Collapses and fusion of adjacent layer would result in the error accumulation at Z direction which was an important fact that could cause printing failure. Finally, we successfully demonstrated a 3D printing hydrogel scaffold through harmonize with all the parameters. The cell viability after printing was compared with the casting and the results showed that our bioprinting method almost had no extra damage to the cells.

Abstract:A simple ink preparation strategy and a reliable 3D‐printing protocol are developed to enable the precise creation of multifunctional conducting polymer composite hydrogels. The 3D‐printed hydrogels exhibit high electrical conductivity, excellent mechanical properties, and biocompatibility, as well as good stability and durability in water, holding great potential in electromagnetic interference shielding, pressure sensing, and tissue engineering applications. Functional conductive hydrogels are widely used in various application scenarios, such as artificial skin, cell scaffolds, and implantable bioelectronics. However, their novel designs and technological innovations are severely hampered by traditional manufacturing approaches. Direct ink writing (DIW) is considered a viable industrial‐production 3D‐printing technology for the custom production of hydrogels according to the intended applications. Unfortunately, creating functional conductive hydrogels by DIW has long been plagued by complicated ink formulation and printing processes. In this study, a highly 3D printable poly(3,4‐ethylenedioxythiophene):polystyrene sulfonate (PEDOT:PSS)‐based ink made from fully commercially accessible raw materials is demonstrated. It is shown that complex structures can be directly printed with this ink and then precisely converted into high‐performance hydrogels via a post‐printing freeze–thawing treatment. The 3D‐printed hydrogel exhibits high electrical conductivity of ≈2000 S m−1, outstanding elasticity, high stability and durability in water, electromagnetic interference shielding, and sensing capabilities. Moreover, the hydrogel is biocompatible, showing great potential for implantable and tissue engineering applications. With significant advantages, the fabrication strategy is expected to open up a new route to create multifunctional hydrogels with custom features, and can bring new opportunities to broaden the applications of hydrogel materials.

Abstract:Shear-thinning hydrogels are often employed in direct-write 3D printing, however, the viscoelastic behaviors that define a printable hydrogel have not been fully established. Herein, we demonstrate a library of hydrogel inks based on the incorporation of water-soluble reactive meth(acrylate) monomers into F127-dimethacrylate hydrogels. This strategy afforded printed hydrogels with a broad range of chemical functionalities and mechanical properties. A systematic investigation was also performed to correlate the printability and mechanical properties to the viscoelastic properties of the hydrogel ink formulations. The materials with a high dynamic yield stress afforded extruded filaments that correlated well with the inner diameter of the printing nozzle. The static yield stress of the material was correlated to the extrusion pressure and print speed required for optimal printing. Thus, this study provides a guide for the future development of hydrogel inks for direct-write 3D printing along with a new set of functional hydrogel inks.

Abstract:Double-network hydrogels have attracted more and more attention in the field of hydrogels due to their high toughness and strength. Up to now, such hydrogels were processed only via model casting and extrusion-based three-dimensional (3D) printing, greatly restricting the freedom of design and manufacture. Herein, we provide a pre-hydrogel ink material composed of commercially available compounds, such as acrylamide and stearyl methacrylate, which can not only be suitable for vat photopolymerization, but also produces a hydrogel with double-network structures. Various hydrogel objects can be printed with a good CAD fidelity, verifying the good printability of the ink material. The printed hydrogels are based on an interpenetrated polymeric network consisting of covalent bond and hydrophobic association, enabling excellent toughness, puncture resistance, and self-recovering performance of the printed objects. The proposed approach may provide an effective way to design the ink of double-network hydrogels suitable for photocurable 3D printing, which may have great application prospects in the fields of customizing wearable sensors, robotics and biomedicine.

Abstract:Composite hydrogels have gained great attention as three-dimensional (3D) printing biomaterials because of their enhanced intrinsic mechanical strength and bioactivity compared to pure hydrogels. In most conventional printing methods for composite hydrogels, particles are preloaded in ink before printing, which often reduces the printability of composite ink with little mechanical improvement due to poor particle-hydrogel interaction of physical mixing. In contrast, the in situ incorporation of nanoparticles into a hydrogel during 3D printing achieves uniform distribution of particles with remarkable mechanical reinforcement, while precursors dissolved in inks do not influence the printing process. Herein, we introduced a "printing in liquid" technique coupled with a hybridization process, which allows 3D freeform printing of nanoparticle-reinforced composite hydrogels. A viscoplastic matrix for this printing system provides not only support for printed hydrogel filaments but also chemical reactants to induce various reactions in printed objects for in situ modification. Nanocomposite hydrogel scaffolds were successfully fabricated through this 3D freeform printing of hyaluronic acid (HAc)-alginate (Alg) hydrogel inks through a two-step crosslinking strategy. The first ionic crosslinking of Alg provided structural stability during printing, while the secondary crosslinking of photo-curable HAc improved the mechanical and physiological stability of the nanocomposite hydrogels. For in situ precipitation during 3D printing, phosphate ions were dissolved in the hydrogel ink and calcium ions were added to the viscoplastic matrix. The composite hydrogels demonstrated a significant improvement in mechanical strength, biostability, as well as biological performance compared to pure HAc. Moreover, the multi-material printing of composites with different calcium phosphate contents was achieved by adjusting the ionic concentration of inks. Our method greatly accelerates the 3D printing of various functional or hybridized materials with complex geometries through the design and modification of printing materials coupled with in situ post-printing functionalization and hybridization in reactive viscoplastic matrices.

Abstract:3D printing of hydrogels finds widespread applications in biomedicine and engineering. Artificial cartilages and heart valves, tissue regeneration and soft robots, require high mechanical performance of complex structures. Although many tough hydrogels have been developed, complicated synthesis processes hinder their fabrication in 3D printing. Here, a strategy is proposed to formulate hydrogel inks, which can be printed into various strong and tough particle-based double-network (P-DN) hydrogels of arbitrary shapes without any rheological modifiers. These hydrogel inks consist of microgels and a hydrogel precursor. The microgels are individual highly cross-linked networks. They are prepared by swelling dried microparticles in the hydrogel precursor that consists of monomers, initiators, and cross-linkers. Microgels regulate the rheological properties of the hydrogel ink and enable the direct printing. After printing and curing, the precursor forms a sparsely cross-linked network that integrates the microgels, leading to a P-DN hydrogel. The proposed hydrogel inks allow 3D printing of multifunctional hydrogel structures with high mechanical performance and strong adhesion to diverse materials.
