Abstract:Five green solvents were assessed as alternatives to N,N‐dimethylformamide in microwave‐assisted solid‐phase peptide synthesis (SPPS): Cyrene, ethyl acetate, 1,3‐dioxolane, tetrahydro‐2‐methylfuran, and N‐Butylpyrrolidinone (NBP). Combinations of NBP with either ethyl acetate or tetrahydro‐2‐methylfuran demonstrated excellent results. Solid‐phase peptide synthesis (SPPS) is the prevailing method for synthesizing research peptides today. However, SPPS is associated with a significant environmental concern due to the utilization of hazardous solvents such as N,N‐dimethylformamide (DMF) or N‐methylpyrrolidone, which generate substantial waste. In light of this, our research endeavors to identify more environmentally friendly solvents for SPPS. In this study, we have assessed the suitability of five green solvents as alternatives to DMF in microwave assisted SPPS. The solvents evaluated include Cyrene, ethyl acetate, 1,3‐dioxolane, tetrahydro‐2‐methylfuran, and N‐Butylpyrrolidinone (NBP). Our investigation encompassed all stages of the synthesis process, from resin swelling, dissolution of reagents, culminating in the successful synthesis of five diverse peptides, including the challenging ACP 65–74, Peptide 18A, Thymosin α1, and Jung‐Redemann peptide. Our findings indicate that NBP emerged as a strong contender, performing on par with DMF in all tested syntheses. Furthermore, we observed that combinations of NBP with either ethyl acetate or tetrahydro‐2‐methylfuran demonstrated excellent results. This research contributes to the pursuit of more sustainable and environmentally conscious practices in peptide synthesis.

Abstract:N-benzyl-3-pyrrolidinone was synthesized starting from ethyl acrylate via addition reaction, substitution reaction, Dieckmann cyclization and hydrolytic decarboxylzation. Starting materials (raw and processed materials) could be obtained easily from commercials, operations were simple and the yields were stable in the technology. The Dieckmann cyclization was improved in the presence of the sodium in the form of granule, and the yield was increased from 44. 7% to 64.0% greatly. Other steps of the synthetic reaction were also optimized, and the gross yield of target product was 57. 2% based on ethyl acrylate. This route was better for industrial production of N-benzyl-3-pyrrolidinone.

Abstract:We herein report that the highly hydrophobic ether 4‐MeTHP constitutes a promising resource for the development of sustainable synthetic methodologies grounded on the use of organometallic reagents. The beneficial effects of 4‐MeTHP as reaction media are illustrated in the organolithium‐promoted anionic ortho‐Fries rearrangement of 1,1'‐bi‐2‐naphthol (BINOL)‐based carbamates under bench‐type conditions. The use of 4‐MeTHP induces a remarkable and unexpected stability of the organolithiums species, enabling the efficient preparation of chiral (bis)carboxamide catalysts. Furthermore, superior performances of 4‐MeTHP than other environmentally responsible solvents are observed using the synthesized BINOL catalysts in the asymmetric addition of organozinc reagents to aldehydes. Spectroscopic studies in solution suggest that 4‐MeTHP plays a key role in these reactions by inducing the preferential formation of a reactive monomeric dinuclear complex. This methodology allows for the asymmetric assembly of enantioenriched secondary alcohols in good yields and high stereoselectivity, working at 0 °C and under air.

Abstract:The alkylation of 1,4-diketo-3,6-arylpyrrolo[3,4-c]pyrroles (ArDPP) is one of the most important steps in the synthesis of soluble materials based on these molecules and the polymers derived from them (that are employed widely in putative organic solar cells). Here we report an improvement in their method of synthesis replacing habitual solvent and base. Compared with more usual conditions, we employed acetonitrile as solvent to give higher or similar yields, with less toxic and hazardous waste, lower reaction time and temperature, and allows recycling of unreacted starting materials. Unlike dimethylformamide and N-methylpyrrolidone, which are the most commonly employed solvents. Our reaction conditions have been tested on three different ArDPPs (Ar = thiophene, phenyl and 4-methoxyphenyl) with a variety of linear and branched alkyl reagents. The results show similar and improved results in comparison with the published reports while reducing the waste and hazard of the reaction, as well as simplifying the purification of the products in many cases. Overall this method has lower environmental impact, is more cost effective and requires neither the use of dry solvent nor inert atmosphere

Abstract:Dipolar aprotic and ethereal solvents comprise just over 40% of all organic solvents utilized in synthetic organic, medicinal, and process chemistry. Unfortunately, many of the common "go-to" solvents are considered to be "less-preferable" for a number of environmental, health, and safety (EHS) reasons such as toxicity, mutagenicity, carcinogenicity, or for practical handling reasons such as flammability and volatility. Recent legislative changes have initiated the implementation of restrictions on the use of many of the commonly employed dipolar aprotic solvents such as dimethylformamide (DMF) and <i>N</i>-methyl-2-pyrrolidinone (NMP), and for ethers such as 1,4-dioxane. Thus, with growing legislative, EHS, and societal pressures, the need to identify and implement the use of alternative solvents that are greener, safer, and more sustainable has never been greater. Within this review, the ubiquitous nature of dipolar aprotic and ethereal solvents is discussed with respect to the physicochemical properties that have made them so appealing to synthetic chemists. An overview of the current legislative restrictions being imposed on the use of dipolar aprotic and ethereal solvents is discussed. A variety of alternative, safer, and more sustainable solvents that have garnered attention over the past decade are then examined, and case studies and examples where less-preferable solvents have been successfully replaced with a safer and more sustainable alternative are highlighted. Finally, a general overview and guidance for solvent selection and replacement are included in the Supporting Information of this review.

Abstract:A sustainable, metal-free, and highly efficient protocol for the synthesis of dihydro indeno [1, 2-b] pyrrole derivatives via a tandem one-pot three-component reaction in aqueous medium at room temperature using an effective and reusable mesoporous heterogeneous organocatalyst, MPC@But-SO 3 H has been developed. MPC@But-SO 3 H was easily synthesized using a melamine-based covalent organic polymer and 1,4-butane sultone. The organocatalyst was well characterized by numerous spectroscopic techniques such as Fourier Transform Infrared (FTIR), Brunauer-Emmett-Teller (BET), X-ray photoelectron spectroscopy (XPS), Powder X-ray diffraction (PXRD), Scanning Electron Microscope (SEM), Energy Dispersive X-ray (EDX), elemental mapping, and Thermal Gravimetric (TG) analyses. The catalyst displayed excellent catalytic potential, high thermochemical stability, and reusability for up to eight catalytic cycles. It offered the title compounds in excellent yields (˃90%) in a short reaction time (9-14 min). The synthesized compounds were characterized through FTIR, 1 H, and 13 C Nuclear Magnetic Resonance (NMR), and the molecular structure of 2-(benzylamino)-3a,8b-dihydroxy-1-methyl-3-nitro-3a,8b-dihydroindeno [1,2-b] pyrrol-4(1H)-one (4a) was confirmed by the Single Crystal XRD (SCXRD) analysis. The key features of the present protocol include the use of water as a green solvent, zero involvement of any metal, sustainable reaction conditions, easy catalyst recovery, and a simple work-up procedure. The calculations for green metrics parameters further supported the greenness and sustainability of the protocol.

Abstract:Background: In recent times, there is an on-going interest in developing convenient and environmentally friendly synthetic methods in organic chemistry. The use of ionic liquid catalysts in organic synthesis is a developing area that allows reactions to be run at low temperature and without solvents. Literature overview revealed that room temperature supported ionic liquid catalysis is a developing field in catalytic science with huge application in organic synthesis. Hence in this current article our focus is on the one-pot synthesis of arylidene derivatives with the use of ([bmim]OH) ionic liquid. Objectives: We describe here the use of an ionic liquid catalyst, 1-n-butyl-3-methylimidazolium hydroxide, [bmim]OH), in the convenient one pot synthesis of arylidene derivatives by the reaction of the active methylene compound, malononitrile, with pyrazole aromatic aldehydes under microwave irradiation. Methods: The functionalized ionic liquid, 1-n-butyl-3-methylimidazolium hydroxide ([bmim]OH) catalyzed Knoevenagel condensation reactions of pyrazole aromatic aldehydes with active methylene compound malononitrile carried out under microwave irradiation. The reaction progress was monitored by thin layer chromatography and the synthesized compounds were further characterized by NMR spectroscopy. Results: This proposed work demonstrate the utility of the use of the ionic liquid catalyst [bmim]OH, in the suitable, high yield (80-95%) microwave assisted reactions of pyrazole aromatic aldehydes with the active methylene compound malononitrile. Conclusion: An eco-friendly synthesis of pyrazole derivatives has been demonstrated using ([bmim]OH) ionic liquid as a catalyst for the Knoevenagel condensation reactions of pyrazole aromatic aldehydes and malononitrile with microwave irradiation. The advantages of this green method are its convenience, mild reaction conditions, and high product yields (80-95%).

Abstract:A new catalytic application of tetraethylammonium L-prolinate for the facile and cleaner synthesis of 4,4′-(arylmethylene)bis(3-methyl-1-phenyl-1H-pyrazol-5-ol)s is developed. This amino acid-based ionic liquid could be recycled up to five runs, and a negligible reduction of catalytic activity was detected within the experimental error. A variety of substituted 4,4′-(arylmethylene)bis(3-methyl-1-phenyl-1H-pyrazol-5-ol)s were obtained in good to high yields under eco-friendly conditions. Tetraethylammonium L-prolinate is easy to prepare, as well as it shows moderate thermal stability and good solubility in water. This work revealed that this amino acid-based ionic liquid, containing L-prolinate anion, can catalyze the tandem Knoevenagel-Michael condensation reaction due to hydrogen bond donor and Lewis base moieties. The current methodology has other advantages, including (a) wide substrate-scope, (b) relatively short reaction times, (c) the use of a nontoxic, biocompatible, biodegradable and metal-free ionic liquid, and (d) simple workup procedure.

Abstract:Dihydrolevoglucosenone (Cyrene) is a bio-based molecule, derived in two simple steps from cellulose, which demonstrates significant promise as a dipolar aprotic solvent. The dipolarity of dihydrolevoglucosenone is similar to NMP, DMF and sulpholane. Dihydrolevoglucosenone demonstrates similar performance to NMP in a fluorination reaction and the Menschutkin reaction.

Abstract:The syntheses of pyrimidinone derivatives by the Biginelli reaction in N -methylpyridinium methyl sulfate [mPyrMeSO 4 ] as an ionic liquid medium have been reported. This protocol provides a highly efficient and selective route for the synthesis of structurally diverse pyrimidinones under milder reaction conditions. The proposed conditions tolerate differently substituted aldehydes and ketones and afford products in synthetically useful yields. Furthermore, camphor, benzil, benzoin, and ethyl acetoacetate were utilized for the Biginelli reaction for the first time, and the resulting pyrimidinones were obtained in excellent yields.

Abstract:Acidic ionic liquid N-methyl-2-pyrrolidonium dihydrogen phosphate [NMP]H2PO4 was prepared and used as efficient catalyst and reaction medium to synthesize β-alkoxyketones by the oxa-Michael addition reactions for the first time. The effect of anions and cations, amount of ionic liquid on the reaction was investigated. Various alcohols and phenols proceeded smoothly and led to corresponding β-alkoxyketones with high yields under mild reaction conditions, O-selectivity addition of aminoethanols was also achieved in this paper. Compared with traditional imidazolium ionic liquids, [NMP]H2PO4 gave the better results. The ionic liquid was stable and could be reused at least five times with a slight loss of activity.

Abstract:Two novel pyrrolizinone-fused BOPYINs (DAB-M and DAB-U) were prepared in one-step through the oxidative and/or oxidative nucleophilic substitution of hydrogen (ONSH) reaction. Selective methoxyl substitution of alpha-unsubstituted BOPYIN was developed, in which C-O and C=O bonds were formed and demonstrated by the single crystal structure of DAB-M. The absorption and emission spectra of both compounds showed red shift and large Stokes Shift compared with starting material DAB. Based on the TD-DFT calculation, the electron transfer direction is totally different from DAB due to the carbonyl group on pyrrolizinone unit. Besides, a reasonable mechanism has been proposed and evidenced by quantum calculation in which the relative free energy has been exhibited.
Sulfolane: A Versatile Dipolar Aprotic Solvent

Abstract:Sulfolane is a highly stable and attractive alternative to other more common dipolar aprotic solvents such as DMSO, DMF, DMAC, and NMP. Sulfolane is more toxic than the other solvents but has a very low skin penetration in comparison to that of the others. A high skin penetration is not beneficial for organic solvents as dissolved toxic compounds might be transferred through the skin. Sulfolane has a high dipole moment (μ = 4.7 debye), elevated relative permittivity (ευ = 43.4), and a high Hildebrand solubility parameter (δ = 27.2 [MPa]1/2). This means a high solvency power for reactions containing polarizable intermediates. Sulfolane is capable of strong solvation of cations by the oxygens in the sulfone group which increases the nucleophilicity of the corresponding less solvated anions. With its high stability against strong acids and bases as well as its thermal stability sulfolane is a solvent that can be operated within a wide range of reaction conditions. This makes it the solvent of choice for different acid-catalyzed reactions at elevated temperatures. Sulfolane has also found use in halo exchange reactions for the formation of various fluoroaromatic compounds. It has also been used as solvent for oxidations, nitrations, rearrangements, phosphonylations, and condensation reactions.

Abstract:Herein we describe the synthesis of dipyrrolopyrazines via a tandem-Sonogashira coupling with subsequent direct cyclisation of the resulting bisalkynes. The key precursor, di-tert-butyl (3,6-dichloropyrazine-2,5-diyl)dicarbamate, can be easily obtained on a large scale. Bidirectional cross-couplings yield either the diyne or dipyrrolopyrazine scaffolds selectively. When the intermediate bisalkynes are cyclised with IPrAuNTf2, an in situ deprotection of the Boc-group is observed, giving access to the N-unsubstituted dipyrrolopyrazines. Functionalisation of the pyrrolo-CH or NH-moiety allows further adjustment of solubility, processability and optoelectronic properties. Photophysical studies demonstrate remarkable stability and high quantum yields.

Abstract:(S,S)-PDP (5d) and its backbone (2S,2′S)-bipyrrolidine (1) have been extensively applied as the scaffold of various chiral ligands in catalytic asymmetric syntheses. In this study, new short asymmetric syntheses of these two important C2-symmetrical nitrogen heterocycles have been accomplished employing economically available l-prolinol derivative 10 as the starting material. Excellent diastereoselectivity was achieved of the key Grignard addition to imine intermediate utilizing (S)-N-tert-butanesulfinamide as the chiral auxiliary.

Abstract:Significant efforts have been made in recent years to identify more environmentally benign and safe alternatives to side-chain protection and deprotection in solid-phase peptide synthesis (SPPS). Several protecting groups have been endorsed as suitable candidates, but finding a greener protecting group in SPPS has been challenging. Here, based on the 2-(o-nitrophenyl) propan-1-ol (Npp-OH) photolabile protecting group, a structural modification was carried out to synthesize a series of derivatives. Through experimental verification, we found that 3-(o-Nitrophenyl) butan-2-ol (Npb-OH) had a high photo-release rate, high tolerance to the key conditions of Fmoc-SPPS (20% piperidine DMF alkaline solution, and pure TFA acidic solution), and applicability as a carboxyl-protective group in aliphatic and aromatic carboxyl groups. Finally, Npb-OH was successfully applied to the synthesis of head–tail cyclic peptides and side-chain–tail cyclic peptides. Moreover, we found that Npb-OH could effectively resist diketopiperazines (DKP). The α-H of Npb-OH was found to be necessary for its photosensitivity in comparison to 3-(o-Nitrophenyl)but-3-en-2-ol (Npbe-OH) during photolysis-rate verification.

Abstract:The tert-butyloxycarbonyl (Boc) group is one of the most widely used amine-protecting groups in multistep reactions in synthetic organic chemistry as well as in peptide synthesis. Traditional methods to remove the Boc group have disadvantages in terms of high acidity, the use of expensive reagents, excessive amounts of catalysts and harmful solvents as well as high temperatures, making them environmentally unsustainable. Therefore, more efforts must be stepwise tightened to make Boc removal practical, clean, and minimize any potential impact. We describe an efficient and sustainable method for N-Boc deprotection by means of a choline chloride/p-toluenesulfonic acid deep eutectic solvent (DES), which is used as a reaction medium plus catalyst. The adopted conditions allow the deprotection of a wide variety of N-Boc derivatives in excellent yields. The strategy has found advantages in greening, simplicity, and short reaction times, resulting in a useful alternative to standard methods.

Abstract:An efficient protocol for the synthesis of differently substituted 1, 4-dihydropyridines in deep eutectic solvents under solvent-free conditions is reported herewith. Excellent yields of the resultant products have been obtained. Recyclability studies have also been performed for deep eutectic solvents with very little loss in activity up to five recycles.

Abstract:Three contiguous stereocenters with syn-anti structure on the 3-(1-hydroxyalkyl)pyrrolidinone were constructed in one step with high yields and high diastereoselectivity by an Et2AlCl-catalyzed group transfer radical cyclization reaction from N-alkenyl-beta-hydroxyalkanamides with 125-W UV lamp irradiation. The relative stereochemistry was confirmed by X-ray analysis.
3-Methoxybutan-2-one as a sustainable bio-based alternative to chlorinated solvents

Abstract:Methylation of acetoin with dimethyl carbonate was performed in a sustainable one-step process, with improved process mass intensity (PMI) and atom economy compared to previously published methods. The resulting product, 3-methoxybutan-2-one (MO) was successfully evaluated as a bio-based solvent, while both Kamlet-Taft solvatochromic parameters and Hansen solubility parameters demonstrate its potential viability in the substitution of chlorinated solvents. MO exhibited a low peroxide forming potential and a negative Ames mutagenicity test and was successfully used as a solvent in a Friedel-Crafts acylation (79% yield compared to 77% in dichloromethane) and for N-alkylations. MO is a renewable oxygenated solvent, with the potential ability to substitute carcinogenic halogenated solvents in some applications.
