Constitutive expression of thaumatin-like protein (TLP-3) in transgenic tobacco plants leads to enhance resistance to Alternaria alternata

Abstract:In nature, each plant is attacked by thousands of micro-organisms, but only small number of these micro-organisms cause disease in plants. One of the important groups of defensive genes is pathogenesis-related (PR) proteins. The importance of these proteins is especially because of their ability to fight with different kinds of micro-organisms. Thaumatin-like proteins (TLPs) belong to the PR proteins and play an important role in plant defense against pathogen invasions. In this study, thaumatin-like TLP-3 gene from 1-year-old alfalfa was transferred into tobacco plants through Agrobacterium-mediated technology. The polymerase chain reaction (PCR) and reverse-transcriptase-PCR assays confirmed the presence of TLP-3 gene and TLP-3 mRNA, respectively, in transgenic tobacco plants. The bioassay results showed the activity of TLP-3 against Alternaria alternata in transgenic tobacco plants. This is the first report of enhanced resistance to A. alternata in transgenic tobacco plants with constitutive expression of TLP-3.
