Abstract:The functions of the nervous system are mediated by neural circuits that are formed during development and modulated by experiences. Central to the assembly of neural circuits is the regulation of synaptic connectivity by synaptic molecules and neuronal activity. Extensive studies have focused on identifying molecules involved in synapse formation. Besides factors promoting synaptogenesis, several antisynaptogenic factors have been discovered. These factors act in concert to ensure the timing and specificity of circuit formation. Moreover, progress has been made in revealing how neuronal activity governs the balance of excitatory and inhibitory synapses. Intriguingly, several transcription factors, as well as activity-dependent transcription of BDNF through promoter IV, have been shown to selectively regulate cortical inhibitory circuits by promoting GABAergic synapse formation.

Abstract:<p>The brain is the most complicated organ in the human body with more than ten thousand genes expressed in each region. The molecular activity of the brain is divergent in various brain regions, both spatially and temporally. The function of each brain region lies in the fact that each region has different gene expression profiles, the possibility of differential RNA splicing, as well as various post-transcriptional and translational modification processes. Understanding the overall activity of the brain at the molecular level is essential for a comprehensive understanding of how the brain works. Fortunately, the development of next generation sequencing technology has made it possible to measure the molecular activity of a specific tissue as a daily routine approach of research. Therefore, at the molecular level, the application of sequencing technology to investigate the molecular organization of the brain has become a novel field, and significant progress has been made recently in this field. In this paper, we reviewed the major computational methods used in the analysis of brain transcriptome, including the application of these methods to the research of human and non-human mammal brains. Finally, we discussed the utilization of transcriptome methods in neurological diseases.</p>

Abstract:Precise connectivity in neuronal circuits is a prerequisite for proper brain function. The dauntingly complex environment encountered by axons and dendrites, even after navigation to their target area, prompts the question of how specificity of synaptic connections arises during development. We review developmental strategies and molecular mechanisms that are used by neurons to ensure their precise matching of pre- and postsynaptic elements. The emerging theme is that each circuit uses a combination of simple mechanisms to achieve its refined, often complex connectivity pattern. At increasing levels of resolution, from lamina choice to subcellular targeting, similar signaling concepts are reemployed to narrow the choice of potential matches. Temporal control over synapse development and synapse elimination further ensures the specificity of connections in the nervous system.

Abstract:The brain is composed of electrically excitable neuronal networks regulated by the activity of voltage-gated ion channels. Further portraying the molecular composition of the brain, however, will not reveal anything remotely reminiscent of a feeling, a sensation or a conscious experience. In classical physics, addressing the mind-brain problem is a formidable task because no physical mechanism is able to explain how the brain generates the unobservable, inner psychological world of conscious experiences and how in turn those conscious experiences steer the underlying brain processes toward desired behavior. Yet, this setback does not establish that consciousness is non-physical. Modern quantum physics affirms the interplay between two types of physical entities in Hilbert space: unobservable quantum states, which are vectors describing what exists in the physical world, and quantum observables, which are operators describing what can be observed in quantum measurements. Quantum no-go theorems further provide a framework for studying quantum brain dynamics, which has to be governed by a physically admissible Hamiltonian. Comprising consciousness of unobservable quantum information integrated in quantum brain states explains the origin of the inner privacy of conscious experiences and revisits the dynamic timescale of conscious processes to picosecond conformational transitions of neural biomolecules. The observable brain is then an objective construction created from classical bits of information, which are bound by Holevo's theorem, and obtained through the measurement of quantum brain observables. Thus, quantum information theory clarifies the distinction between the unobservable mind and the observable brain, and supports a solid physical foundation for consciousness research.

Abstract:There are at least two approaches to the definition of consciousness. In the first case, certain aspects of consciousness, called qualia, are considered inaccessible for research from a third person and can only be described through subjective experience. This approach is inextricably linked with the so-called "hard problem of consciousness", that is, the question of why consciousness has qualia or how any physical changes in the environment can generate subjective experience. With this approach, some aspects of consciousness, by definition, cannot be explained on the basis of external observations and, therefore, are outside the scope of scientific research. In the second case, a priori constraints do not constrain the field of scientific investigation, and the best explanation of the experience in the first person is included as a possible subject of empirical research. Historically, in the study of cause-and-effect relationships in biology, it was customary to distinguish between proximate causation and ultimate causation existing in biological systems. Immediate causes are based on the immediate influencing factors [1]. Proximate causation has evolutionary explanations. When studying biological systems themselves, such an approach is undoubtedly justified, but it often seems insufficient when studying the interaction of consciousness and the brain [2,3]. Current scientific communities proceed from the assumption that the physical substrate for the generation of consciousness is a neural network that unites various types of neurons located in various brain structures. Many neuroscientists attach a key role in this process to the cortical and thalamocortical neural networks. This question is directly related to experimental and clinical research in the field of disorder of consciousness. Progress in this area of medicine depends on advances in neuroscience in this area and is also a powerful source of empirical information. In this area of consciousness research, a large amount of experimental data has been accumulated, and in this review an attempt was made to generalize and systematize.

Abstract:The neuronal basis of consciousness is the greatest challenge to the scientific worldview. Much relevant empirical work is carried out on the minimal neuronal mechanisms underlying any one specific conscious percept. Two broad approaches are popular among brain scientists: electrophysiological recordings from individual neurons in the cortex of behaving monkeys or behavior combined with functional brain imaging in humans. However, many aspects of consciousness are problematic or remain off-limits to the former approach, while the latter one lacks sufficient spatial and temporal resolution to monitor individual neurons that are key to perception, thought, memory, and action. It is here that neurosurgeons, probing the living human brain on a daily basis, can play a decisive role. This article explores the contributions of neurosurgeons to this quest and outlines some of the results that have already been achieved.

Abstract:Abstract Consciousness is one of final questions for humans to tackle in neuroscience. Due to a lack of understanding of basic brain networks and mechanisms of functions, our knowledge of consciousness mainly stays at a theoretical level. Recent studies using brain imaging in humans and modern neuroscience techniques in animal studies reveal the basic brain network for consciousness. The projection from the thalamus to different cortical regions forms a network of activities to maintain consciousness in humans and animals. These feedback and feedforward circuits maintain consciousness even in certain brain injury conditions. Pterions and ion channels that contribute to these circuit neural activities are targets for drugs and manipulations that affect consciousness such as anesthetic agents. Synaptic plasticity that trains synapses during learning and information recall modified the circuits and contributes to a high level of consciousness in a certain population.

Abstract:Viewing the brain as a complex computer of simple neurons cannot account for consciousness nor essential features of cognition. Single cell organisms with no synapses perform purposeful intelligent functions using their cytoskeletal microtubules. A new paradigm is needed to view the brain as a scale-invariant hierarchy extending both upward from the level of neurons to larger and larger neuronal networks, but also downward, inward, to deeper, faster quantum and classical processes in cytoskeletal microtubules inside neurons. Evidence shows self-similar patterns of conductive resonances repeating in terahertz, gigahertz, megahertz, kilohertz and hertz frequency ranges in microtubules. These conductive resonances apparently originate in terahertz quantum dipole oscillations and optical interactions among pi electron resonance clouds of aromatic amino acid rings of tryptophan, phenylalanine and tyrosine within each tubulin, the component subunit of microtubules, and the brain's most abundant protein. Evidence from cultured neuronal networks also now shows that gigahertz and megahertz oscillations in dendritic-somatic microtubules regulate specific firings of distal axonal branches, causally modulating membrane and synaptic activities. The brain should be viewed as a scale-invariant hierarchy, with quantum and classical processes critical to consciousness and cognition originating in microtubules inside neurons.

Abstract:The mechanisms underlying major aspects of the human brain remain a mystery. It is unknown how verbal episodic memory is formed and integrated with sensory episodic memory. There is no consensus on the function and nature of dreaming. Here we present a theory for governing neural activity in the human brain. The theory describes the mechanisms for building memory traces for entities and explains how verbal memory is integrated with sensory memory. We infer that a core function of dreaming is to move charged particles such as calcium ions from the hippocampus to association areas to primary areas. We link a high level of calcium ions concentrations to Alzheimer's disease. We present a more precise definition of consciousness. Our results are a step forward in understanding the function and health of the human brain and provide the public with ways to keep a healthy brain.

Abstract:During early telencephalic development, intricate processes of regional patterning and neural stem cell (NSC) fate specification take place. However, our understanding of these processes in primates, including both conserved and species-specific features, remains limited. Here, we profiled 761,529 single-cell transcriptomes from multiple regions of the prenatal macaque telencephalon. We deciphered the molecular programs of the early organizing centers and their cross-talk with NSCs, revealing primate-biased galanin-like peptide ( GALP ) signaling in the anteroventral telencephalon. Regional transcriptomic variations were observed along the frontotemporal axis during early stages of neocortical NSC progression and in neurons and astrocytes. Additionally, we found that genes associated with neuropsychiatric disorders and brain cancer risk might play critical roles in the early telencephalic organizers and during NSC progression.

Abstract:There are many blank areas in understanding the brain dynamics and especially how it gives rise to consciousness. Quantum mechanics is believed to be capable of explaining the enigma of conscious experience, however till now there is not good enough model considering both the data from clinical neurology and having some explanatory power. In this paper is presented a novel model in defence of macroscopic quantum events within and between neural cells. The synaptic beta-neurexin/neuroligin-1 adhesive protein complex is claimed to be not just the core of the excitatory glutamatergic CNS synapse, instead it is a device mediating entanglement between the cytoskeletons of the cortical neurons. Thus the macroscopic coherent quantum state can extend throughout large brain cortical areas and the subsequent collapse of the wavefunction could affect simultaneously the subneuronal events in millions of neurons. The neuroligin-1/beta-neurexin/synaptotagmin-1 complex also controls the process of exocytosis and provides an interesting and simple mechanism for retrograde signalling during learning-dependent changes in synaptic connectivity. A brief outlook of the molecular machinery driving neuromediator release through exocytosis is provided with particular emphasis on the possibility for vibrationally-assisted tunneling.

These maps are crucial for identifying disease biomarkers, predicting treatment responses, and developing targeted therapies. Molecular biology and genetics are also driving progress. Researchers are uncovering the genetic basis of neurological disorders, providing clues about disease susceptibility and progression. Molecular imaging techniques visualise neurotransmitter systems and cellular processes, shedding light on disease mechanisms. The integration of neuroscience with computer modelling and AI is revolutionising research. Abstract:The evolution of the modern human brain was accompanied by distinct molecular and cellular specializations, which underpin our diverse cognitive abilities but also increase our susceptibility to neurological diseases. These features, some specific to humans and others shared with related species, manifest during different stages of brain development. In this multi-stage process, neural stem cells proliferate to produce a large and diverse progenitor pool, giving rise to excitatory or inhibitory neurons that integrate into circuits during further maturation. This process unfolds over varying time scales across species and has progressively become slower in the human lineage, with differences in tempo correlating with differences in brain size, cell number and diversity, and connectivity. Here we introduce the terms 'bradychrony' and 'tachycrony' to describe slowed and accelerated developmental tempos, respectively. We review how recent technical advances across disciplines, including advanced engineering of in vitro models, functional comparative genetics and high-throughput single-cell profiling, are leading to a deeper understanding of how specializations of the human brain arise during bradychronic neurodevelopment. Emerging insights point to a central role for genetics, gene-regulatory networks, cellular innovations and developmental tempo, which together contribute to the establishment of human specializations during various stages of neurodevelopment and at different points in evolution.

Abstract:How our brains work is one of the major unsolved problems of biology. This paper describes some of the techniques of molecular biology that are already being used to study the brains of animals. Mainly as a result of the human genome project many new techniques will soon become available which could decisively influence the progress of neuroscience. I suggest that neuroscientists should tell molecular biologists what their difficulties are, in the hope that this will stimulate the production of useful new biological tools.

Abstract:We examine the hypothesis that consciousness can be understood as a state of matter, “perceptronium”, with distinctive information processing abilities. We explore four basic principles that may distinguish conscious matter from other physical systems such as solids, liquids and gases: the information, integration, independence and dynamics principles. If such principles can identify conscious entities, then they can help solve the quantum factorization problem: why do conscious observers like us perceive the particular Hilbert space factorization corresponding to classical space (rather than Fourier space, say), and more generally, why do we perceive the world around us as a dynamic hierarchy of objects that are strongly integrated and relatively independent? Tensor factorization of matrices is found to play a central role, and our technical results include a theorem about Hamiltonian separability (defined using Hilbert–Schmidt superoperators) being maximized in the energy eigenbasis. Our approach generalizes Giulio Tononi’s integrated information framework for neural-network-based consciousness to arbitrary quantum systems, and we find interesting links to error-correcting codes, condensed matter criticality, and the Quantum Darwinism program, as well as an interesting connection between the emergence of consciousness and the emergence of time.

Abstract:The Neurobiology Of Thinking, Identity, And Geniality Abstract: Mathematically the axioms of representation are subtle, and critical. The CNS expresses its function via its internal neuronal networks in multidimensional, intrinsic frames. CNS operation can be represented by tensor transformations within and among general frames of reference. So-called metaorganization principle is transforming the dual representations of intention from one to another. The property of intentionality refers to the fact that consciousness is about objects or events, and is modulated by attention. To construct a new neural framework we must outgoing basicly from the competitive interactions among widely distributed groups of neurons. A major system in this regard is the thalamocortical system. The integrative dynamics of conscious experience suggests that the thalamocortical system behaves as a functional cluster that interacts mainly with itself.

Abstract:While many core biological processes are conserved across species, the human brain has evolved with unique capacities. Current understanding of the neurobiological mechanisms that endow human traits as well as associated vulnerabilities remains limited. However, emerging data have illuminated species divergence in DNA elements and genome organization, in molecular, morphological, and functional features of conserved neural cell types, as well as temporal differences in brain development. Here, we summarize recent data on unique features of the human brain and their complex implications for the study and treatment of brain diseases. We also consider key outstanding questions in the field and discuss the technologies and foundational knowledge that will be required to accelerate understanding of human neurobiology.

Abstract:The underlying physiological mechanisms of generating conscious states are still unknown. To make progress on the problem of consciousness, we will need to experimentally design a system that evolves in a similar way our brains do. Recent experimental data show that the multiscale nature of the evolving human brain can be implemented by reprogramming human cells. A hybrid system can be designed to include an evolving brain equipped with digital computers that maintain homeostasis and provide the right amount of nutrients and oxygen for the brain growth. Shaping the structure of the evolving brain will be progressively achieved by controlling spatial organization of various types of cells. Following a specific program, the evolving brain can be trained using substitutional reality to learn and experience live scenes. We already know from neuroelectrodynamics that meaningful information in the brain is electrically (wirelessly) read out and written fast in neurons and synapses at the molecular (protein) level during the generation of action potentials and synaptic activities. Since with training, meaningful information accumulates and is electrically integrated in the brain, one can predict, that this gradual process of training will trigger a tipping point for conscious experience to emerge in the hybrid system.
