Abstract:Surface-enhanced Raman scattering (SERS) is a promising analytical tool in nanoscale detection because of its high sensitivity and selectivity. This review focuses on recent advances in SERS-based detection of DNA and RNA. First, nanostructure-based SERS-active substrates are introduced. Using label-free and labeled SERS, target biomolecules such DNA, RNA and microRNA have been successfully detected. Finally, applications in pathogen diagnosis are discussed. The prospects and challenges of SERS-based bioanalysis are highlighted.

Abstract:Surface-enhanced Raman scattering (SERS) has emerged as a valuable technique for molecular identification. Due to the characteristics of high sensitivity, excellent signal specificity, and photobleaching resistance, SERS has been widely used in the fields of environmental monitoring, food safety, and disease diagnosis. By attaching the organic molecules to the surface of plasmonic nanoparticles, the obtained SERS tags show high-performance multiplexing capability for biosensing. The past decade has witnessed the progress of SERS tags for liquid biopsy, bioimaging, and theranostics applications. This review focuses on the advances of SERS tags in biomedical fields. We first introduce the building blocks of SERS tags, followed by the summarization of recent progress in SERS tags employed for detecting biomarkers, such as DNA, miRNA, and protein in biological fluids, as well as imaging from in vitro cell, bacteria, tissue to in vivo tumors. Further, we illustrate the appealing applications of SERS tags for delineating tumor margins and cancer diagnosis. In the end, perspectives of SERS tags projecting into the possible obstacles are deliberately proposed in future clinical translation.

Abstract:Surface-enhanced Raman scattering (SERS) technique has shown extraordinary features for biomedical applications. The implementation of SERS nanotags has opened a new era for bioimaging and detections. As a powerful tool, SERS nanotags provide favorable properties such as fingerprint spectrum, narrow peak linewidth, good photostability, and high spatial resolution accompanied by various rational designs of nanoparticles. They have proven as useful imaging agents for in vivo, ex vivo, and in vitro detection of cancerous cells and tissues. This tutorial provides the basic principles of SERS and SERS nanotags, including recent progress of SERS-based bioimaging applications, as well as the outlooks into the future developments toward practical clinical SERS.

Abstract:Surface-enhanced Raman spectroscopy (SERS) inherits the rich chemical fingerprint information on Raman spectroscopy and gains sensitivity by plasmon-enhanced excitation and scattering. In particular, most Raman peaks have a narrow width suitable for multiplex analysis, and the measurements can be conveniently made under ambient and aqueous conditions. These merits make SERS a very promising technique for studying complex biological systems, and SERS has attracted increasing interest in biorelated analysis. However, there are still great challenges that need to be addressed until it can be widely accepted by the biorelated communities, answer interesting biological questions, and solve fatal clinical problems. SERS applications in bioanalysis involve the complex interactions of plasmonic nanomaterials with biological systems and their environments. The reliability becomes the key issue of bioanalytical SERS in order to extract meaningful information from SERS data. This review provides a comprehensive overview of bioanalytical SERS with the main focus on the reliability issue. We first introduce the mechanism of SERS to guide the design of reliable SERS experiments with high detection sensitivity. We then introduce the current understanding of the interaction of nanomaterials with biological systems, mainly living cells, to guide the design of functionalized SERS nanoparticles for target detection. We further introduce the current status of label-free (direct) and labeled (indirect) SERS detections, for systems from biomolecules, to pathogens, to living cells, and we discuss the potential interferences from experimental design, measurement conditions, and data analysis. In the end, we give an outlook of the key challenges in bioanalytical SERS, including reproducibility, sensitivity, and spatial and time resolution.

Abstract:Detection of nucleic acid and protein targets related to human health and safety has attracted widespread attention. Surface-enhanced Raman scattering (SERS) is a powerful tool for biomarker detection because of its ultrahigh detection sensitivity and unique fingerprinting spectra. In this review, we first introduce the development of nanostructure-based SERS-active substrates and SERS nanotags, which greatly influence the performance of SERS biosensors. We then focus on recent advances in SERS biosensors for DNA, microRNA and protein determination, including label-free, labeled and multiplex analyses as well as in vivo imaging. Finally, the prospects and challenges of such nanostructure-based SERS biosensors are discussed.

Abstract:Surface-enhanced Raman scattering (SERS) is a phenomenon that occurs on nanoscale-roughed metallic surface. The magnitude of the Raman scattering signal can be greatly enhanced when the scatterer is placed in the very close vicinity of the surface, which enables this phenomenon to be a highly sensitive analytical technique. SERS inherits the general strongpoint of conventional Raman spectroscopy and overcomes the inherently small cross section problem of a Raman scattering. It is a sensitive and nondestructive spectroscopic method for biological samples, and can be exploited either for the delivery of molecular structural information or for the detection of trace levels of analytes. Therefore, SERS has long been regarded as a powerful tool in biomedical research. Metallic nanostructure plays a key role in all the biomedical applications of SERS because the enhanced Raman signal can only be obtained on the surface of a finely divided substrate. This review focuses on progress made in the use of SERS as an analytical technique in bio-imaging, analysis and detection. Recent progress in the fabrication of SERS active nanostructures is also highlighted.

Abstract:Plasmonic sensing has been an important multidisciplinary research field and has been extensively used in detection of trace molecules in chemistry and biology. The sensing techniques are typically based on surface-enhanced spectroscopies and surface plasmon resonances (SPRs). This review article deals with some recent advances in surface-enhanced Raman scattering (SERS) sensors and SPR sensors using either localized surface plasmon resonances (LSPRs) or propagating surface plasmon polaritons (SPPs). The advances discussed herein present some improvements in SERS and SPR sensing, as well as a new type of nanowire-based SPP sensor.

Abstract:We present a rapid and straightforward procedure for the synthesis of core-shell surface-enhanced Raman scattering (SERS) tags. SERS tags are combinations of Raman active dyes with metallic nanoparticles inducing the enhancement. Because of their significantly higher stability to environmental conditions, the majority of SERS tags are designed in core-shell geometry. The main disadvantage of this approach up to now was the time-consuming (normally days) and complex preparation of these labeled core-shell particles. The presented approach can be performed within hours on a daily base. The main characteristics and the stability of the fabricated SERS tags in extreme pH values, high ion strength, or in organic solvent is demonstrated in this communication. Furthermore, the SERS tags are functionalized with anti-Salmonella antibody as a model to present a potential application of the core-shell particles in bioimaging.

Abstract:Surface‐enhanced Raman scattering (SERS) is among the most widely applied analytical techniques due to its easy execution and extreme sensitivity. Target molecules can be detected and distinguished based upon the fingerprint spectra that arise when absorbed on the SERS substrates surface, particularly on the SERS‐active hotspots. Thus, rational fabricating the enhancing substrates plays a key role in broadening SERS application. Programmable DNA functionalized plasmonic nanoassemblies, where DNA acts as both structure basis and functional unit, combine the specificity of DNA recognition, and modulate the assembly of plasmonic nanoparticles (NPs). Specifically designed DNA not only improves the selectivity to target molecules but also promotes the sensitivity of the optical signals through precisely regulating the distance between the molecule and the substrate. A variety of DNA‐functionalized SERS sensors have been reported and obtained well performance in the analysis of heavy metal ions in water, toxins, pesticide residues, antibiotics, hormones, illicit drugs, or other small molecules. This review places an emphasis on the design and sensing strategies of the DNA‐functionalized plasmonic nanoassemblies, as well as basic principles of Raman enhancement, and recent advances for environmental analysis. The current challenges and potential trends in the development of DNA‐functionalized SERS sensors for environmental pollutant monitoring in complicated scenarios are subsequently discussed.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has great potential for the analysis of molecules adsorbed on metals with rough surfaces or substrates with micro-/nanostructures. Plasmonic coupling between metal nanoparticles and the morphology of the rough metal surface can produce "hot spots" that enhance Raman scattering by adsorbed molecules, typically at micro- to nanomolar concentrations, although high enhancement factors can also facilitate single-molecule detection. This phenomenon is widely applicable for chemical analysis and sensing in various fields. In this review, the latest research progress on SERS micro-/nanosensors is evaluated, and the sensors are classified according to their individual functions. Furthermore, the design principles and working mechanisms of reported SERS-active micro-/nanostructured substrates are analyzed, and the design features adopted to overcome the difficulties associated with precision detection are explored. Finally, challenges and directions for future development in this field are discussed. This review serves as a design guide for novel SERS-active substrates.

Abstract:Plasmon-free surface-enhanced Raman scattering (SERS) substrates have attracted tremendous attention for their abundant sources, excellent chemical stability, superior biocompatibility, good signal uniformity, and unique selectivity to target molecules. Recently, researchers have made great progress in fabricating novel plasmon-free SERS substrates and exploring new enhancement strategies to improve their sensitivity. This review summarizes the recent developments of plasmon-free SERS substrates and specially focuses on the enhancement mechanisms and strategies. Furthermore, the promising applications of plasmon-free SERS substrates in biomedical diagnosis, metal ions and organic pollutants sensing, chemical and biochemical reactions monitoring, and photoelectric characterization are introduced. Finally, current challenges and future research opportunities in plasmon-free SERS substrates are briefly discussed.

Abstract:Surface-enhanced Raman scattering (SERS) has attracted great interest due to its remarkable enhancement, excellent sensitivity, and the "fingerprinting" ability to produce distinct spectra for detecting various molecules. Noble metal nanomaterials have usually been employed as SERS-active substrates because of their strong SERS enhancement originated from their unique surface plasmon resonance (SPR) properties. Because the SPR property depends on metal material's size, shape, morphology, arrangement, and dielectric environment around metal nanostructures, the key to wider applications of SERS technique is to develop plasmon-resonant structures with novel geometries to enhance Raman signals and to control the periodic ordering of these structures over a large area to obtain reproducible Raman enhancement. This review presents a general view on the theory background of SERS effect and several basic concepts and focuses on recent progress in engineering metallic nanostructures with various morphologies using versatile methods for improving SERS properties. Their potential applications in the field of chemical detection and biological sensing are overviewed.

Abstract:Surface-enhanced Raman scattering (SERS) continues to attract great excitement because it is one of the very few techniques that provide both ultrahigh sensitivity and molecular specificity. For biomedical applications, there are two emerging trends. One is to exploit the intrinsically label-free capabilities of SERS to detect and differentiate complete biomolecular signatures, often aided by artificial intelligence or deep learning algorithms. The second trend is to develop SERS nanoparticles as ultrabright detection tags (called nanotags). In this article, we focus on the development of SERS nanoparticles as encoded spectroscopic tags and will discuss their applications in diagnostics, biomolecular imaging, and spectroscopy-guided cancer surgery. In contrast to fluorescence, SERS signals are much more stable against photobleaching, and their peak widths are often less than a few nanometers. Also, SERS probes with different spectral signatures can all be excited at a single wavelength and detected with simple instrumentation for multiplexing. To put these concepts in perspective, we will start this article by briefly discussing the principles of SERS, follow by recent progress in developing in-vitro and in-vivo SERS tags. The article will then provide highlights of current and emerging applications in biology and medicine. The article will conclude with a discussion of the challenges and opportunities in translating SERS nanotags for clinical use.

Abstract:Surface-enhanced Raman scattering (SERS) is a highly sensitive and selective tool for the identification of biological and chemical analytes based on Raman scattering. Due to its unique characteristics of high sensitivity, no interference from water, high resolution and stability, SERS has been widely used in biological analysis and biomedical research. In recent years, great development has been achieved in the application of SERS in the field of gene assay. This review is presented on the latest development of SERS for gene analysis, including label-free detection and label-dye detection. It emphasizes on the metal-nanoparticle- and the tip-enhanced-Raman-scattering-based SERS, and the SERS signal amplification on the basis of Raman-active dyes, PCR technology, molecular beacon, substrates and SERS tags. Finally, the multiplexed SERS detection and the future trends of SERS are outlined.

Abstract:Surface-enhanced Raman scattering (SERS) is a kind of popular non-destructive and water-free interference analytical technology with fast response, excellent sensitivity and specificity to trace biotargets in biological samples. Recently, many researches have focused on the preparation of various magnetic nanoparticle-based SERS substrates for developing efficient bioanalytical methods, which greatly improved the selectivity and accuracy of the proposed SERS bioassays. There has been a rapid increase in the number of reports about magnetic SERS substrates in the past decade, and the number of related papers and citations have exceeded 500 and 2000, respectively. Moreover, most of the papers published since 2009 have been dedicated to analytical applications. In the paper, the recent advances in magnetic nanoparticle-based SERS substrates for bioanalysis were reviewed in detail based on their various morphologies, such as magnetic core–shell nanoparticles, magnetic core–satellite nanoparticles and non-spherical magnetic nanoparticles and their different functions, such as separation and enrichment, recognition and SERS tags. Moreover, the typical application progress on magnetic nanoparticle-based SERS substrates for bioanalysis of amino acids and protein, DNA and RNA sequences, cancer cells and related tumor biomarkers, etc., was summarized and introduced. Finally, the future trends and prospective for SERS bioanalysis by magnetic nanoparticle-based substrates were proposed based on the systematical study of typical and latest references. It is expected that this review would provide useful information and clues for the researchers with interest in SERS bioanalysis.

By tuning the number of shells or by changing the Raman reporters in different gap layers, their Raman intensities and spectral bands were tunable. Abstract:Surface‐enhanced Raman scattering (SERS) with greatly amplified fingerprint spectra holds great promise in biochemical and biomedical research. In particular, the possibility of exciting a library of SERS probes and differentially detecting them simultaneously stimulates widespread interest in multiplexed biodetection. In this review, we summarize the recent progress in developing SERS‐active plasmonic nanostructures for cellular and intracellular detection. We highlight the development of the nanosensors with tailored plasmonic and multifunctional properties for profiling molecular and pathological processes. Future challenges towards the routine use of SERS technology in quantitative bioanalysis and clinical diagnostics are further discussed.

Abstract:Since 2000, there has been an explosion of activity in the field of plasmon-enhanced Raman spectroscopy (PERS), including surface-enhanced Raman spectroscopy (SERS), tip-enhanced Raman spectroscopy (TERS) and shell-isolated nanoparticle-enhanced Raman spectroscopy (SHINERS). In this Review, we explore the mechanism of PERS and discuss PERS hotspots — nanoscale regions with a strongly enhanced local electromagnetic field — that allow trace-molecule detection, biomolecule analysis and surface characterization of various materials. In particular, we discuss a new generation of hotspots that are generated from hybrid structures combining PERS-active nanostructures and probe materials, which feature a strong local electromagnetic field on the surface of the probe material. Enhancement of surface Raman signals up to five orders of magnitude can be obtained from materials that are weakly SERS active or SERS inactive. We provide a detailed overview of future research directions in the field of PERS, focusing on new PERS-active nanomaterials and nanostructures and the broad application prospect for materials science and technology.
