Abstract:Profenofos (PFF) is a commonly used organophosphorus insecticide that requires strict monitoring due to its potential environmental, ecological, and human health risks originating from residues in soil and water systems, as well as accumulation in crops. In this study, a novel monoclonal antibody (mAb) targeting PFF was prepared for the first time and investigated the recognition mechanism through molecular simulation. Subsequently, a mAb-based colloidal gold immunochromatographic assay (GICA) was developed for the rapid screening of PFF in fruit and vegetable samples. The mAb exhibited an IC50 value of 12.9 ng/mL, and limit of detection (LOD) of 4.6 ng/mL, respectively in indirect competitive immunosorbent enzyme-linked immunosorbent assay (ic-ELISA). After optimization, the developed GICA exhibited a visual limit of detection (vLOD) of 20 ng/mL and a quantitative of detection (qLOD) of 5.2 ng/mL, with a linear range from 10.0 to 83.8 ng/mL. Good correlation was observed between the results of GICA and standard Gas Chromatography-Tandem Mass Spectrometry (GC-MS/MS) in matrix and recovery test. The developed GICA can be used for rapid sample detection within 15 minutes, which is an excellent tool for screening PFF in foods and environmental samples.

Abstract:Profenofos, a highly poisonous organophosphorus pesticide, has been widely used in agricultural production. These pesticide residues have seriously influenced food security and threatened human health, and new methods with high sensitivity are greatly needed to detect profenofos. Here, we developed an aptamer-based microcantilever-array sensor operated in stress mode to detect profenofos, with advantages of being a label-free, highly sensitive, one-step immobilization method capable of quantitative and real-time detection. The microcantilevers were functionalized with a profenofos-specific aptamer (SS2-55), and then the specific binding of profenofos to aptamer induced a deflection of the microcantilever, which was monitored using an optical method in a real-time manner. The microcantilever deflection showed a positive relationship with profenofos concentration, and the detection limit was low to 1.3 ng mL(-1) (3.5 nM) for profenofos, which was much lower than other aptamer-based detection methods. The selectivity of the sensor was verified with another organophosphorus pesticide. Additionally, we successfully detected profenofos dissolved in vegetable-soak solution. Our results showed that this aptamer-based microcantilever-array sensor is a convenient and label-free method for detecting profenofos in small amounts and has great potential for food-security applications. Rapid and sensitive detection of quizalofop-p-ethyl by gold nanoparticle-based lateral flow immunoassay in agriproducts and environmental samples

Abstract:Quizalofop-p-ethyl is a widely used herbicide that poses a threat to human health and environmental safety. In this study, anti-quizalofop-p-ethyl monoclonal antibodies (mAbs) were prepared and used to develop a gold nanoparticle-based lateral flow immunoassay (AuNP-LFIA) for the detection of quizalofop-p-ethyl in agriproducts and environmental samples. Four hybridoma cell lines were obtained, among which 5B6D10E11 secreted mAb with the highest sensitivity, with a 50 % inhibition concentration of 4.57 ng/mL in the indirect competitive enzyme-linked immunosorbent assay. After optimization, the AuNP-LFIA strip based on the mAb (5B6D10E11) showed a visual detection limit of 10 ng/mL, and the results could be directly determined by the naked eye within 8 min. The cross-reactivity of the AuNP-LFIA for analogs of quizalofop-p-ethyl was negligible except for quizalofop-p-acid. The established AuNP-LFIA was proven to be accurate and precise based on the recovery test. Furthermore, the detection results of AuNP-LFIA were consistent with those of ultra-high-performance liquid chromatography tandem mass spectrometry.

Abstract:An amperometric method for the detection of organophosphorus pesticide based on acetylcholinesterase inhibition is reported. A silver electrode is used and shows an inherent advantage in determination of pesticides, which can monitor the activity of the enzyme in real time by electrochemical oxidization of thiocholine at low potential. Cyclic voltammetry and amperometry were used to evaluate the performance of silver electrode for thiocholine detection. Spectrophotometry was conducted to optimize the parameters for the enzymatic reaction. Cyclic voltammetry shows the advantages of silver over others (platinum, glassy carbon and gold) electrodes, when detecting thiocholine at low potentials. The silver electrode could linearly respond to thiocholine in the range from 5.2 x 10(-7) to 2.6 x 10(-5) M at 0.08 V. The measurement displayed a detection limit of 6.2 ppb. The analytical feasibility was investigated further via determination of paraoxon in Chinese cabbages and Fuji apples, which shows recovery rates of 92.05% and 106.11%, respectively. The results indicate the capability of the proposed method for pesticides monitoring in real samples.

Abstract:An uncomplicated method for the detection of organophosphorous pesticide characterized by a thione moiety (P = S) was developed, and chlorpyrifos was used as an example. It was based on the detection of thiocholine generated by acetylcholinesterase. Organophosphorothionates slightly inhibited acetylcholinesterase, and P = S in chlorpyrifos was oxidized into P = O to improve the toxicity. Optimized amount of n -bromosuccinimide (4 ppm) and ascorbic acid (20 ppm) were used for the conversion. The influence of the excessive ascorbic acid and the high overpotential of the thiocholine was addressed by using a screen-printed silver electrode. According to cyclic voltammetry and amperometry, oxidization of thiocholine occurred on the electrode at a low potential while the ascorbic acid was not oxidized. The working potential was minimized to 0 V, and a detection limit of 2.5 ppb for chlorpyrifos was obtained. This method was further used to detect chlorpyrifos in Fuji apple and cucumber.
Spectrophotometric Determination of Profenofos Using an Aptamer-Gold Nanorod (GNR)-Based Biosensor

Abstract:A biosensor based on the optical signal of gold nanorods (GNRs) and an aptamer as the recognition element is reported to perform the rapid and selective spectrophotometric determination of profenofos. The GNR synthesized by the seed growth method have unique structures which binds with the aptamer through electrostatic adsorption and is well dispersed in a salt-containing solution. Upon the addition of profenofos, specific recognition caused aggregation of the GNRs. The optical signal therefore decreased and the response linearly related to the concentration of profenofos. The results demonstrate that ascorbic acid as the reducing agent plays an important role in the morphology of the GNRs. GNRs with the length - diameter ratio (L/D) of 12 stably combined with the aptamer. For the optimum conditions, spectrophotometric determination of profenofos was achieved using the biosensor with a linear relationship between the absorbance of GNR at 508 nm and the concentration of profenofos from 40 to 460 μg/L. The limit of detection of the method was 31.93 μg/L. In addition, this approach shows good selectivity for the analysis of fruits and vegetable. A valuable new method is reported for the determination of profenofos.

Abstract:Quantitative detection of pesticide residues in food and environmental samples using an improved lateral flow immunoassay (LFIA) is of considerable importance for real-time analysis. This paper proposes a highly sensitive LFIA platform based on a hierarchical magneto-colorimetric compact. This compact serves as both the target magnetic enrichment substrate and a photosensitive label. Initially, a large porous dendritic silica template is prepared and doped with superparamagnetic ferric oxide nanoparticles (Fe3O4 NPs) and colloidal gold nanoparticles (AuNPs) at high densities within its vertical channels. The sequential assembly of central-radial channels allow for the three-dimensional integration of these two components, enabling independent control of their discrete functions without mutual interference. Following alkyl organosilicon encapsulation and silica sealing, the composite spheres are then applied in LFIA to detect chlorothalonil residues. Fe3O4 NPs enhance the binding efficiency to target analytes, while AuNPs amplify the signal, leveraging their high loading densities and robust optical properties. The developed LFIA platform exhibited a detection limit of 0.34 ng/mL for chlorothalonil and a linear range of 0.0085-824 ng/mL. The recoveries varied between 85.1 % and 103.1 %, and the relative standard deviations were 1.25%-8.84 %. This LFIA approach demonstrates high sensitivity, specificity, reproducibility and flexible detection modes, making it highly suitable for the on-site monitoring of pesticide residues.

Abstract:The co-contamination of pesticide residues and mycotoxins in agricultural products is a global concern, with the potential for cumulative and synergistic damaging effects, imposing substantial health and economic burdens to the public. The dosage-sensitive and simultaneous detection of multiple pollutants, with a heightened sensitivity in real samples, poses a significant demand and challenge. Herein, we propose a portable detection method integrating surface-enhanced Raman scattering (SERS)-with lateral flow immunoassay (LFIA), offering high sensitivity and multiplex analysis capabilities. This approach enables the simultaneous detection of imidacloprid (IMI), pyraclostrobin (PYR) and aflatoxin B1 (AFB1) through a single test strip. Utilizing the immune-specific binding between antigen and antibodies, we immobilised antibody- conjugated SERS nanotags on three test lines of the strips to generate Raman signal amplification in the proposed biosensor. Accurate quantitative analysis was performed by measuring the SERS signal intensity on the test lines. The limits of detection were 8.6 pg/mL for IMI, 97.4 pg/mL for PYR and 8.9 pg/mL for AFB1, exhibiting sensitivities 12-fold, 102-fold and11-fold higher than the colorimetric signals, respectively. Importantly, the SERS-LFIA immunosensor demonstrated robust performance when applied to real samples, yielding recoveries ranging from 86.16 % to 115.0 %, with relative standard deviation values below 8.67 %. These results underscore the excellent stability, high selectivity and reliability the proposed SERS-LFIA immunosensor. Consequently, it holds promise for the detection of multiple pesticides and mycotoxins in both environmental and agricultural samples.

Abstract:We present a novel lateral flow immunoassay (LFIA) for the simultaneous detection of the pesticides imidacloprid, chlorpyrifos-methyl and isocarbophos based on three competitive immunoreactions. In contrast to previously reported LFIAs, the method is based on the use of four strips. Each has three red channels (three test lines dispensed with different capture reagent) to detect imidacloprid, chlorpyrifos-methyl and isocarbophos respectively. Different channels on each strip are the key to multi-detection, and four strips of LFIA are needed for visual and semi-quantitative read-outs. Under optimized conditions, the LFIA was applied to the determination of three pesticides. The detection time is within 7 min and the detection limits are 50, 100, and 100 μg L−1, respectively. Abstract:As a commonly used crop fungicide with proven adverse effects against various organisms, the excessive use of flutriafol (FTF) has resulted in high residues found in crops. In this study, a novel hapten was designed by deriving a long interval arm on its benzene ring allowing sufficient exposure of the FTF active site, for the production of a highly sensitive and specific monoclonal antibody with a half-inhibitory concentration of 1.77 ng/mL. Furthermore, for the first time, a GNP-based lateral flow immunoassay (LFIA) was developed for the quantitative detection of FTF in foods with visual limit of detection in rice, tomatoes, and grapes of 0.5 mg/kg, 0.2 mg/kg and 0.5 mg/kg, respectively and a cutoff value was 4 mg/kg, and calculated limit of detection of 0.045 mg/kg, 0.035 mg/kg and 0.060 mg/kg respectively. The recoveries in spiked samples ranged from 90.17% to 110.05% and these results indicated that the GNP-based LFIA developed in this study for the rapid detection of FTF in food was a reliable on-site detection method.

Abstract:In this work, we propose an electrochemical DNA aptasensor for the detection of profenofos, an organophosphorus pesticide, based on a competitive format and disposable graphite screen-printed electrodes (GSPEs). A thiol-tethered DNA capture probe, which results to be complementary to the chosen aptamer sequence, was immobilised on gold nanoparticles/polyaniline composite film-modified electrodes (AuNPs/PANI/GSPE). Different profenofos solutions containing a fixed amount of the biotinylated DNA aptamer were dropped onto the realized aptasensors. The hybridisation reaction was measured using a streptavidin-alkaline phosphatase enzyme conjugate, which catalyses the hydrolysis of 1-naphthyl -phosphate. The 1-naphtol enzymatic product was detected by means of differential pulse voltammetry (DPV). The aptasensor showed itself to work as a signal off sensor, according to the competitive format used. A dose response curve was obtained between 0.10 μM and 10 μM with a detection limit of 0.27 μM.

Abstract:The residues of organophosphorus pesticides in agricultural production and soil constitute a significant threat to both human health and ecosystems, underscoring the critical importance of developing effective detection methods for these pesticides. Here, we introduce an innovative method for detecting trichlorfon that integrates hydrogen peroxide (H 2 O 2 )-induced etching of silver-coated gold nanostars (AuNS@Ag) with an enzymatic reaction cascade. By precisely controlling the thickness of the silver layer and the morphology of the gold nanoparticle core, we enhance the sensitivity of the H 2 O 2 etching process. In this detection system, acetylcholinesterase (AChE) catalyzes the hydrolysis of acetylcholine chloride (ACh) into choline, which is then oxidized by choline oxidase (ChOx) to produce H 2 O 2 . The generated H 2 O 2 subsequently etches the silver layer of AuNS@Ag, leading to a pronounced red-shift in the localized surface plasmon resonance (LSPR) wavelength, from 590 nm to 735 nm. However, the presence of trichlorfon inhibits the activity of AChE, resulting in a reduced production of H 2 O 2 and a subsequent attenuation of the LSPR wavelength shift. Under optimized experimental conditions, this method enables the linear detection of trichlorfon within a concentration range of 0.1–5.0 μg mL −1 and has been successfully applied to detect trichlorfon in cabbage and tap water samples, showcasing its promising potential in food safety and environmental monitoring.

Abstract:Organophosphorus (OP) pesticides are widely used to control pests because of their high activity. This study described a rapid and sensitive lateral flow immunochromatographic (LFIC) assay based on up-converting nanoparticles (UCNPs) for multi-residue detection of three OP pesticides. The developed assay integrated novel fluorescent material UCNPs labeled with a broad-specific monoclonal antibody. Based on the competitive platform by immobilized antigen in the test zone, the optimized UCNPs-LFIC assay enabled sensitive detection for parathion, parathion-methyl, and fenitrothion with IC50 of 3.44, 3.98, and 12.49 ng/mL (R 2 ≥ 0.9776) within 40 min. The detectable ability ranged from 0.98 to 250 ng/mL. There was no cross-reactivity with fenthion, phoxim, isocarbophos, chlorpyrifos, or triazophos, even at a high concentration of 500 ng/mL. Matrix interference from various agricultural products was also studied in food sample detection. In the spiked test, recoveries of the three OP pesticides ranged from 67 to 120% and relative standard deviations were below 19.54%. These results indicated that the proposed strip assay can be an alternative screening tool for rapid detection of the three OP pesticides in food samples.

Abstract:Extensive use of pesticides in agricultural production has been causing serious health threats to humans and animals. Among them, phorate is a highly toxic organophosphorus insecticide that has been widely used in planting. Due to its harmful effects on human and animal health, it has been restricted for use in many countries. Analytical methods for the rapid and sensitive detection of phorate residues in agricultural products are urgently needed. In this study, a new method was developed by combining surface-enhanced Raman spectroscopy (SERS) and immunochromatography assay (ICA). Hybrid magnetic Fe3O4@Au@DTNB-Ab nanoprobes were prepared by modifying and growing Au nanoseeds on an Fe3O4 core. SERS activity of the nanoprobe was optimized by adjusting the concentration of the Au precursor. A rapid and sensitive assay was established by replacing the traditional colloidal gold-based ICA with hybrid SERS nanoprobes for SERS-ICA. After optimizing parameters including coating antibody concentrations and the composition and pH of the buffer solution, the limit of detection (LOD) for phorate could reach 1 ng/mL, with a linear range of 5~100 ng/mL. This LOD is remarkably lower than the maximum residue limit in vegetables and fruits set by the Chinese government. The feasibility of this method was further examined by conducting a spiking test with celery as the real sample. The result demonstrated that this method could serve as a promising platform for rapid and sensitive detection of phorate in agricultural products.

Abstract:Fipronil (FP) is a very effective phenylpyrazole insecticide and is now widely used in agriculture. At the same time, the water and soil in the environment are polluted by FP. For the rapid detection of FP toxicants in food and the environment, we have designed an entirely novel electrochemical immunosensor that employs the combined functionalities of a cMWCNTs-AgNPs-CS-FAb-BSA layer to modify an SPCE by the freeze-drying technique. The high porosity of chitosan (CS) coupled with an excellent electron transfer enabled by the cMWCNTs and AgNPs increased the surface area for anti-fipronil (FAb) antibody immobilization and enhanced the current signal of the immunosensor. Cyclic voltammetry (CV) was applied for the quantitative determination of FP under optimized conditions (0.1 M PBS, pH 7.5, 35 °C incubation temperature, and 40 min incubation duration). The modified electrochemical immunosensor displayed excellent analytical performance, including a wide linear concentration range from 0.1 to 1000 ng mL −1 with a very low limit of detection of 0.021 ng mL −1 and good reproducibility (RSD = 2.58%, n = 6), stability (80.4% sensitivity after 5 days), and selectivity. Not only could the modified electrochemical immunosensor be applied in the FP residue analysis of agricultural products, but the present immobilization strategy can also potentially be applied to different biomolecules.

[Establishment of a rapid detection method of colloidal gold immunochromatographic competition assay for organophosphorus pesticides]

Abstract:Objective To establish a colloidal gold immunochromatography and develop the corresponding test strip for detecting organophosphorus compounds including omethoate, phoxim, dipterex, and parathion in fruits, vegetables and drinking water. Methods Artificial antigen molecules of organophosphorus compounds were synthesized using N-hydroxysuccinimide esters. Acetylcholinesterase antigen was prepared and purified, and the serum containing the corresponding antibody was prepared, purified, and labeled. The working parameters of the test strip were optimized, and the performance evaluation of it was conducted. Results The titer of the antisera ranged from 1:32 to 1:64, with a protein content of approximately 2 mg/mL. The purified polyclonal antibodies displayed target bands at relative molecular masses (Mr) of 25 000 and 55 000, indicating satisfactory purity. The reaction time of the test strips was between 5 to 10 minutes, with a detection limit for samples at 200 ng/mL. Both specificity and accuracy were satisfactory, and the test strip remained valid for 6 months. Conclusion A simple and rapid colloidal gold immunochromatography is established successfully for detecting several organophosphorus compounds and may be useful for on-site preliminary screening of samples in large quantities.
Novel and portable test strip platform for rapid and sensitive on-site detection of procymidone pesticide

Abstract:A novel and portable detection platform for procymidone (PRM) was developed by combining simple sample pretreatment, lateral flow test strips based on multi-branched gold nanoparticle (LFTS-MBGNP), and a smartphone. Based on the large surface area of MBGNPs, rapid detection of PRM was realized by simple naked eye observation. By utilizing a smartphone as a portable signal analyzer, ultrasensitive quantitative detection of PRM in red wine was realized with the limits of detection (LOD) of 1.60 ng/mL, which was 3000 times lower than the US limit (5 ppm). Moreover, rapid detection of four kinds of fruits and vegetables was achieved within 10 min, with LODs of 4.34 ng/g, 6.93 ng/g, 8.99 ng/g, and 5.03 ng/g, respectively, which could meet the PRM limit of the European Union (10 ng/g). Integrating the optimized QuEChERS pretreatment method, the developed platform realized a simple and sensitive on-site detection of PRM pesticide in foods and red wine within 45 min. This platform provides a useful tool and new idea for rapid screening and detection of pesticide residues in food.

Abstract:A novel dual-signal fluorescent sensor was developed for detecting organophosphorus pesticides (OPs). It relies on the catalytic activities of acetylcholinesterase (AChE) and choline oxidase (ChOx) to generate hydrogen peroxide (H 2 O 2 ) through the conversion of acetylcholine (ACh) to choline·H 2 O 2 then oxidizes ferrocene-modified tetraphenylethylene (TPE-Fc) to its oxidized state (TPE-Fc + ), resulting in enhanced cyan fluorescence due to aggregation. Simultaneously, ferrocene oxidation generates hydroxyl radicals (•OH), causing a decrease in orange fluorescence of glutathione-synthesized gold nanoclusters (GSH-AuNCs). The presence of OPs restricts AChE activity, reducing H 2 O 2 production. Increasing OPs concentration leads to decreased cyan fluorescence and increased orange fluorescence, enabling visual OPs detection. The sensor has a linear dynamic range of 10–2000 ng/mL with a detection limit of 2.05 ng/mL. Smartphone-based color identification and a WeChat mini program were utilized for rapid OPs analysis with successful outcomes.

Abstract:Fenvalerate residues in fruits and vegetables may result in biological immune system disorders. Current sensor detection methods are harsh due to the shortcomings of antibody preparation and preservation conditions. Therefore, developing a recognition material with strong specificity, good stability, and low cost is of practical significance in designing a sensitive, simple, and rapid method. This study used precipitation polymerization to synthesize molecularly imprinted polymers (MIPs). The MIP was prepared into a fiber membrane using the electrostatic spinning method. After that, the fenvalerate hapten-mouse IgG-Eu fluorescent probe was synthesized, and the side flow chromatography strip was constructed to determine fenvalerate in vegetables using the immunocompetition method. The results showed that the adsorption capacity of MIP to fenvalerate was 3.65, and the adsorption capacity on MIPFM (an electrospinning membrane containing the fenvalerate MIPs) was five times that of free MIP. The test strip showed good linearity with R 2 = 0.9761 within the range of 50 μg/L-1,000 μg/L. In conclusion, substituting fenvalerate monoclonal antibodies with a molecularly imprinted electrospinning membrane is ideal for rapid onsite detection of pyrethroids.
