Abstract:Oxygen sensing attracts high research interest because it is applicable to early diagnosis and health monitoring. Herein, based on photonic crystals (PCs), a novel fluorescence enhancement effect (FEE) was explored to achieve highly sensitive and accurate oxygen detection. A flexible oxygen sensor was fabricated from heterogeneous polydimethylsiloxane (PDMS) and polystyrene (PS) PCs embedded with platinum(II) octaethylporphyrin (PtOEP) oxygen-sensitive dye. Two different fluorophore incorporation (PtOEP-in and PtOEP-out) PCs were compared in terms of their FEE and oxygen-sensing performance. Simultaneously, photoluminescence enhancement was experimentally and theoretically examined and was determined to be the main factor in improving the performance of the oxygen sensor. In particular, the optimized PC oxygen sensor showed a 12-fold higher PL intensity than PS microspheres (control group). Moreover, the optimized sensor demonstrated high sensitivity to oxygen, with excellent accuracy, photostability and flexibility. This work is expected to provide a universal route to design flexible PC sensors for health monitoring through fluorescent ultratrace detection.

Abstract:We review the current state of optical methods for sensing oxygen. These have become powerful alternatives to electrochemical detection and in the process of replacing the Clark electrode in many fields. The article (with 694 references) is divided into main sections on direct spectroscopic sensing of oxygen, on absorptiometric and luminescent probes, on polymeric matrices and supports, on additives and related materials, on spectroscopic schemes for read-out and imaging, and on sensing formats (such as waveguide sensing, sensor arrays, multiple sensors and nanosensors). We finally discuss future trends and applications and summarize the properties of the most often used indicator probes and polymers. The ESI (with 385 references) gives a selection of specific applications of such sensors in medicine, biology, marine and geosciences, intracellular sensing, aerodynamics, industry and biotechnology, among others.

Abstract:Dynamic analysis of oxygen (O₂) has been limited by the lack of a real-time, quantitative, and biocompatible sensor. To address these demands, we designed a ratiometric optode matrix consisting of the phosphorescence quenching dye platinum (II) octaethylporphine ketone (PtOEPK) and nanocystal quantum dots (NQDs), which when embedded within an inert polymer matrix allows long-term pre-designed excitation through fluorescence resonance energy transfer (FRET). Depositing this matrix on various glass substrates allowed the development of a series of optical sensors able to measure interstitial oxygen concentration [O₂] with several hundred millisecond temporal resolution in varying biological microdomains of active brain tissue.

Abstract:Knowledge on the availability of dissolved oxygen inside microfluidic cell culture systems is vital for recreating physiological-relevant microenvironments and for providing reliable and reproducible measurement conditions. It is important to highlight that in vivo cells experience a diverse range of oxygen tensions depending on the resident tissue type, which can also be recreated in vitro using specialized cell culture instruments that regulate external oxygen concentrations. While cell-culture conditions can be readily adjusted using state-of-the-art incubators, the control of physiological-relevant microenvironments within the microfluidic chip, however, requires the integration of oxygen sensors. Although several sensing approaches have been reported to monitor oxygen levels in the presence of cell monolayers, oxygen demands of microfluidic three-dimensional (3D)-cell cultures and spatio-temporal variations of oxygen concentrations inside two-dimensional (2D) and 3D cell culture systems are still largely unknown. To gain a better understanding on available oxygen levels inside organ-on-a-chip systems, we have therefore developed two different microfluidic devices containing embedded sensor arrays to monitor local oxygen levels to investigate (i) oxygen consumption rates of 2D and 3D hydrogel-based cell cultures, (ii) the establishment of oxygen gradients within cell culture chambers, and (iii) influence of microfluidic material (e.g., gas tight vs. gas permeable), surface coatings, cell densities, and medium flow rate on the respiratory activities of four different cell types. We demonstrate how dynamic control of cyclic normoxic-hypoxic cell microenvironments can be readily accomplished using programmable flow profiles employing both gas-impermeable and gas-permeable microfluidic biochips.

Abstract:Chemical gradients are essential in biological systems, affecting processes like microbial activity in soils and nutrient cycling. Traditional tools, such as microsensors, offer high-resolution data but are limited to one-dimensional measurements. Planar optodes allow for two-dimensional (2D) and three-dimensional (3D) chemical imaging but are often sensitive to temperature changes. This study presents an advanced dual-emission optical sensor that simultaneously measures temperature and oxygen using a modified platinum(II) meso-tetrakis(3,5-ditert-butylphenyl)-tetra(2-tert-butyl-1,4-naphthoquinono)porphyrin. The ratio between thermally activated delayed fluorescence and phosphorescence was optimized by modifying platinum(II) naphthoquinonoporphyrin with tert-butyl groups which simultaneously improved solubility in apolar solvents and polymer matrix (polystyrene). This dual-function sensor enables two-parameter chemical imaging with a consumer-grade RGB camera or a hyperspectral camera. We demonstrated 2D visualization of temperature and oxygen distribution in a model soil system. The RGB camera provided rapid and cost-effective imaging, while the hyperspectral camera offered more detailed spectral information despite some limitations. Our findings revealed the formation of a stable temperature gradient and oxygen depletion, driven by water content and temperature-sensitive microbial activity. This dual O2/T sensor, with further potential improvements, shows considerable promise for advanced multiparameter sensing in complex biological and environmental studies, providing deeper insights into dynamic microenvironments.
Characterization of planar phosphorescence based oxygen sensors on a TCSPC-PLIM macro-imager

Abstract:A set of phosphorescent materials were evaluated using a new macro-imager based on fast time-stamping Tpx3Cam camera with phosphorescent lifetime imaging (PLIM) capabilities. The near-infrared emitting O2-sensitive dye PtBP was embedded in several polymers having different permeability for O2 and applied as solid-state coatings on polyester film support. These sensor materials were placed on a temperature-controlled stage of the imager and imaged under pulsed excitation with a 627 nm LED. The resulting PLIM images showed good uniformity, accuracy and contrast of measured lifetime values for the different phosphorescent materials, and the anticipated lifetime changes with altered temperature or oxygenation conditions, proving the dynamic mechanism of quenching by O2. By imaging the coatings with decreasing dye concentration, stability and limits of sensitivity of the PLIM mode were determined and compared with intensity-based imaging. Several commercial O2 sensors were also imaged and compared with the hand-made sensors, while lifetime measurements on the new imager were compared with those performed on the confocal TCSPC-PLIM microscope.

Abstract:In recent years, significant progress has been achieved in the sensing and imaging of molecular oxygen (O2) in biological samples containing live cells and tissue. We review recent developments in the measurement of O2 in such samples by optical means, particularly using the phosphorescence quenching technique. The main types of soluble O2 sensors are assessed, including small molecule, supramolecular and particle-based structures used as extracellular or intracellular probes in conjunction with different detection modalities and measurement formats. For the different O2 sensing systems, particular attention is paid to their merits and limitations, analytical performance, general convenience and applicability in specific biological applications. The latter include measurement of O2 consumption rate, sample oxygenation, sensing of intracellular O2, metabolic assessment of cells, and O2 imaging of tissue, vasculature and individual cells. Altogether, this gives the potential user a comprehensive guide for the proper selection of the appropriate optical probe(s) and detection platform to suit their particular biological applications and measurement requirements.

Abstract:Oxygen is ubiquitous in nature and it plays a key role in several biological processes, such as cellular respiration and food deterioration, to name a few. Currently, reversible and non-destructive oxygen sensing is usually performed with sensors produced by photosensitization of phosphorescent organometallic complexes. In contrast, we propose a novel route of optical oxygen sensing by fluorescence-based quenching of oxygen. We hereby developed for the first time a set of multi-emissive purely organic emitters. These were produced through a one-pot hydrothermal synthesis using p-phenylenediamine (PPD) and urea as starting materials. The origin of the multi-emission has been ascribed to the diversity of chemical structures produced as a result of oxidative oligomerization of PPD. A Bandrowski’s base (BB, i.e., trimer of PPD) is reported as the main component at reaction times higher than 8 h. This indication was confirmed by electrospray-ionization quadrupole time-of-flight (ESI-QTOF) and liquid chromatography-mass spectrometry (LC-MS) analysis. Once the emitters are embedded within a high molecular weight poly (vinyl alcohol) matrix, the intensities of all three emission centers exhibit a non-linear quenching provoked by oxygen within the range of 0–8 kPa. The detection limit of the emission centers are 0.89 kPa, 0.67 kPa and 0.75 kPa, respectively. This oxygen-dependent change in fluorescence emission is reversible (up to three tested 0–21% O2 cycles) and reproducible with negligible cross-interference to humidity. The cost-effectiveness, metal-free formulation, cross-referencing between each single emission center and the relevant oxygen range are all appealing features, making these sensors promising for the detection of oxygen, e.g., in food packaged products.

Abstract:A significantly large scope is available for the scientific and engineering developments of high-throughput ultra-high sensitive oxygen sensors. We give a perspective of oxygen sensing for two physical states of matters - solid-state nanomaterials and plasma. From single-molecule experiments to material selection, we reviewed various aspects of sensing, such as capacitance, photophysics, electron mobility, response time, and a yearly progress. Towards miniaturisation, we have highlighted the benefit of lab-on-chip-based devices and showed exemplary measurements of fast real-time oxygen sensing. From the physical-chemistry perspective, plasma holds a strong potential in the application of oxygen sensing. We investigated the current state-of-the-art of electron density, temperature, and design issues of plasma systems. We also show a numerical aspects of low-cost approach towards developing plasma-based oxygen sensor from household candle flame. In this perspective, we give an opinion about a diverse range of scientific insight together, identifies the short comings, and opens the path for new physical-chemistry device developments of oxygen sensor along with providing a guideline for innovators in oxygen sensing.

Abstract:Cells require sufficient oxygen for survival and differentiation, for example, during bone tissue regeneration. With the aid of probe molecules, two-photon phosphorescence lifetime microscopy (2PLM) can image oxygen distributions, although not in tissues lacking vasculature. Schilling et al. use electrospin polymer meshes by employing a coaxial approach to embed 2PLM probes into the cores of fibers. The fibers support bone marrow stromal cell growth and differentiation, while enabling real-time high-resolution probing of partial pressures of oxygen during the early stages of tissue repair both in vitro and in vivo.

Developing a Wearable Sensor for Continuous Tissue Oxygenation Monitoring: A Proof of Concept Study

Abstract:Objective Technologies facilitating continuous free tissue flap monitoring such as near infrared spectroscopy (NIRS) have been shown to improve flap salvage rates. However, the size and associated costs of such technology create a barrier to wider implementation. The aim of this study was to develop and validate a wearable sensor for continuous tissue oxygenation monitoring. Materials and Methods A forearm ischemia model was designed by using a brachial pressure cuff inflation protocol. Twenty healthy subjects were recruited. The forearm tissue oxygenation of each subject was monitored throughout the pressure cuff protocol by using a new optical sensor (Imperial College London), and a gold standard tissue spectrometry system (O2C, Medizintecknik, LEA, Germany). Data were processed to allow quantitative deoxygenation episode comparisons between inflations and sensor modalities. Results The correlation between O2C and optical sensor oxygenation measurements was moderate (average R = 0.672, p < 0.001). Incremental increases in cuff inflation duration resulted in a linear increase in deoxygenation values with both O2C and optical sensors, with significant differences recorded on consecutive inflations (wall shear rate, p < 0.005). The presence or absence of pulsatile blood flow was correctly determined throughout by both sensor modalities. Conclusion This study demonstrates the ability of a small optical sensor to detect and quantify tissue oxygenation changes and assess the presence of pulsatile blood flow. Low power, miniaturized electronics make the device capable of deployment in a wearable form which may break down the barriers for implementation in postoperative flap monitoring.

Abstract:Oxygen is essential for many forms of life and its depletion in the body and the environment can lead to deleterious effects. Low oxygen conditions, even anoxia, are associated with eutrophication of lakes and rivers, wherein an over abundance of nutrients often caused by pollution result in excessive plant growth and decay, threatening water quality, ecosystem balance, and aquatic life. In the body, low oxygen conditions or hypoxia may be generalized, as can occur at high altitude or during strenuous exercise, or localized in particular tissues, when there is a mismatch between oxygen supply and demand. Hypoxia is present in many important diseases as well. Low oxygen levels in tumors are often associated with biochemical changes, increased invasiveness, cancer progression, and resistance to radiation and chemotherapies. Vascular blockage in strokes, heart attacks, and peripheral artery disease, which is common in diabetes, are other situations where oxygen levels can drop precipitously and cause great damage to affected tissues. Clearly, innovative sensing technologies that provide new insight into these many oxygen dependent processes can impact global society in significant ways.

Abstract:The accurate measurement of arterial blood oxygen partial pressure often plays an important role in the clinical assessment of patients with respiratory conditions such as an acute exacerbation of chronic obstructive pulmonary disease and acute lung injury/adult respiratory distress syndrome. An oxygen-sensitive fluorescence indicator with high biocompatibility was synthesized and then fabricated to the end of an optical fiber. The properties and accuracy of this oxygen sensor were investigated in vitro using physiologic solutions under varying conditions or human blood, and in vivo by obtaining measurements after inserting this optical sensor into the collateral circulation system of rabbits. The sensitivity of the oxygen sensor was relatively stable during altered conditions of pH, PCO2, osmolality, and protein concentration in solutions and during alterations of oxygen and nitrogen content in human blood. There was a linear correlation between the reciprocal value of the fluorescence intensity and PaO2 in both in vivo and in vitro experiments (animal arterial circulation and human blood). Our results suggest that this oxygen-sensitive fluorescence indicator, which has a high biocompatibility, may have the potential for use in real-time monitoring of blood oxygen partial pressure in various clinical settings.

Abstract:The technique of near infrared spectroscopy has been found to be a non invasive method for monitoring tissue oxygenation. The change of absorption spectroscopy caused by change of oxygenated hemoglobin, deoxygenated hemoglobin made it possible. In this article, mean path length of photon migration in 0.5% intralipid solution is simulated. The concentration change of haemoglobin and deoxygenated haemoglobin can be determined by modified Beer Lambert's law. On the basis of this theory, a portable oxygen monitor is designed for quantitation of haemoglobin, deoxygenated haemoglobin concentration changes in human tissues.

Abstract:A free flap is a tissue reconstruction procedure where healthy tissue is harvested to cover up vital structures after wound debridement. Microvascular anastomoses are carried out to join the arteries and veins of the flap with recipient vessels near the target site. Continuous monitoring is required to identify the flap failure and enable early intervention to salvage the flap. Although there are medical instruments that can assist surgeons in monitoring flap viability, high upfront costs and time-consuming data interpretation have hindered the use of such technologies in practice. Surgeons still rely largely on the clinical examination to monitor flaps after operations. This paper presents a low-cost, low-power (6.6 mW), and miniaturized Hamlyn StO 2 (tissue oxygen saturation) sensor that can be embodied as a plaster and attached to a flap for real-time monitoring. Similar to the design of oxygen saturation (SpO 2 /SaO 2 ) sensors, the Hamlyn StO 2 sensor was designed based on photoplethysmography (PPG), but with a different target of quantifying tissue perfusion rather than capturing pulsatile flow. To understand the spectral response to oxygenation/deoxygenation and vascular flow, an active in vitro silicone phantom was developed. The new sensor was validated using the silicone phantom and compared with a commercially available photospectroscopy and laser Doppler machine (O2C, LEA, Germany). In addition, in vivo experiments were conducted using a brachial pressure cuff forearm ischemia model. The experiment results have shown a high correlation between the proposed sensor and the O2C machine (r = 0.672 and p <; 0.001), demonstrating the potential value of the of the proposed low-cost sensor in post-operative free flap monitoring.

Abstract:Oxygen (O2) quantification is essential for assessing cell metabolism, and its consumption in cell culture is an important indicator of cell viability. Recent advances in microfluidics have made O2 sensing a crucial feature for organ-on-chip (OOC) devices for various biomedical applications. OOC O2 sensors can be categorized, based on their transducer type, into two main groups, optical and electrochemical. In this review, we provide an overview of on-chip O2 sensors integrated with the OOC devices and evaluate their advantages and disadvantages. Recent innovations in optical O2 sensors integrated with OOCs are discussed in four main categories: (i) basic luminescence-based sensors; (ii) microparticle-based sensors; (iii) nano-enabled sensors; and (iv) commercial probes and portable devices. Furthermore, we discuss recent advancements in electrochemical sensors in five main categories: (i) novel configurations in Clark-type sensors; (ii) novel materials (e.g., polymers, O2 scavenging and passivation materials); (iii) nano-enabled electrochemical sensors; (iv) novel designs and fabrication techniques; and (v) commercial and portable electrochemical readouts. Together, this review provides a comprehensive overview of the current advances in the design, fabrication and application of optical and electrochemical O2 sensors.
Oxygen imaging of hypoxic pockets in the mouse cerebral cortex

Abstract:Consciousness is lost within seconds upon cessation of cerebral blood flow. The brain cannot store oxygen, and interruption of oxidative phosphorylation is fatal within minutes. Yet only rudimentary knowledge exists regarding cortical partial oxygen tension ( P o 2 ) dynamics under physiological conditions. Here we introduce Green enhanced Nano-lantern (GeNL), a genetically encoded bioluminescent oxygen indicator for P o 2 imaging. In awake behaving mice, we uncover the existence of spontaneous, spatially defined "hypoxic pockets" and demonstrate their linkage to the abrogation of local capillary flow. Exercise reduced the burden of hypoxic pockets by 52% compared with rest. The study provides insight into cortical oxygen dynamics in awake behaving animals and concurrently establishes a tool to delineate the importance of oxygen tension in physiological processes and neurological diseases.

Abstract:Imaging methods by the means of optical sensors are applied in diverse scientific areas such as medical research and diagnostics, aerodynamics, environmental analysis, or marine research. After a general introduction to the field, this review is focused on works published between 2012 and 2022. The covered topics include planar sensors (optrodes), nanoprobes, and sensitive coatings. Advanced sensor materials combined with imaging technologies enable the visualization of parameters which exhibit no intrinsic color or fluorescence, such as oxygen, pH, CO2, H2O2, Ca2+, or temperature. The progress on the development of multiple sensors and methods for referenced signal read out is also highlighted, as is the recent progress in device design and application formats using model systems in the lab or methods for measurements’ in the field.
