Abstract:Changes in the oxidant/antioxidant environment of aging Leydig cells have been shown to be correlated with the reduced ability of these cells to produce testosterone. We hypothesized that an altered redox environment might result in increased susceptibility to pro-oxidants, and that this might have an impact on steroidogenic function. To begin to test this hypothesis, we experimentally manipulated intracellular glutathione (GSH) concentration in MA-10 Leydig cells, and examined whether the susceptibility of the GSH-depleted cells to oxidative stress was affected. Incubation of MA-10 cells with buthionine sulfoximine (BSO, 100 µM, 18 h) or diethyl maleate (DEM, 200 µM, 30 min) reduced GSH content by more than 70%, while treatment of the cells with 3H-1,2-dithiole-3-thione (D3T, 200 µM, 24 hr) increased the GSH concentration to twice that of the control cells. Depletion of GSH significantly increased the susceptibility of the cells to oxidative stress induced by tert-butyl hydroperoxide (t-BuOOH). In contrast, increasing GSH greatly protected the cells from the same stressor. Thus, the ED50 of progesterone production was reduced from 400 µM t-BuOOH in the control cells to less than 50 µM in GSH-depleted cells. Increasing GSH with D3T greatly increased the ED50 from 400 µM to more than 800 µM. Cell viability, assayed by MTT reduction and ATP production, was significantly affected by t-BuOOH only at concentrations of 100 µM or higher whereas progesterone production was affected by t-BuOOH at much lower (12.5 µM) concentration. Intracellular ROS concentration, as measured by 2, 7-dichlorodihydrofluorescein diacetate (DCF) fluorescence, was significantly increased in GSH-depleted cells in response to t-BuOOH at 50-100 µM. The changes in progesterone and ROS production induced by GSH were prevented by restoration of the cellular GSH pool with glutathione ethyl ester (GSHEE), or by treating the cells with the antioxidant N-acetyl-cysteine (NAC). Two steps of the steroidogenic pathway were particularly sensitive to oxidative stress following GSH depletion: StAR protein synthesis and the conversion of cholesterol to pregnenolone by P450scc within the mitochondria. The results of these studies, taken together, are consistent with the hypothesis that alteration in the oxidant/antioxidant environment may play a significant role in the increased sensitivity of MA-10 cells to pro-oxidant stress, and thus perhaps to the age-related reduced ability of testicular Leydig cells to produce testosterone. Influences of flavones on cell viability and cAMP-dependent steroidogenic gene regulation in MA-10 Leydig cells

Abstract:Testicular Leydig cells are major contributors of androgen synthesis and secretion, which play an important role in testis development, normal masculinization, maintenance of spermatogenesis, and general male fertility. The rate-limiting step in testosterone biosynthesis involves the transfer of cholesterol to the mitochondrial inner membrane by the steroidogenic acute regulatory (Star) protein, a critical factor in steroid hormone biosynthesis. Once inside the mitochondria, cholesterol is metabolized by the steroidogenic enzyme Cyp11a1 to pregnenolone, which is further converted to testosterone by the action of other steroidogenic enzymes. Interestingly, the Star protein level declines during Leydig cell aging, resulting in defective mitochondrial cholesterol transfer and lower testosterone production. It is possible to delay the age-related decline in testosterone production by increasing Star and/or Cyp11a1 gene expression using supplementation with flavonoids, a group of polyphenolic compounds widely distributed in fruits and vegetables. In this study, we examined whether the distribution of hydroxyl groups among flavones could influence their potency to stimulate steroidogenesis within Leydig cells. Low levels of apigenin, luteolin, chrysin, and baicalein (10 μM) stimulated cAMP-dependent Star, Cyp11a1, and Fdx1 promoters’ activation and may increase steroidogenesis within Leydig cells. Indeed, luteolin effectively increased cAMP-dependent accumulation of progesterone from MA-10 Leydig cells, possibly through activation of Star and Fdx1 transcription. Thus, dietary luteolin could be potentially effective to maintain steroid production within aging males.

Abstract:The present study aims to investigate the roles of steroidogenic acute regulatory protein (StAR) in Yangjing Capsule (YC) induced anti-apoptotic effects on Leydig cells and the related mechanism. Leydig tumor cells (MLTC-1) were cultured and treated with YC, and immunofluorescence assay was performed to examine the expression of StAR; furthermore, luciferase reporter assay was conducted to evaluate the impact of YC on StAR promoter; next, MLTC-1 cells were treated with StAR small interfering RNA (siRNA), and flow cytometry was carried out to examine the effect of StAR siRNA on the apoptosis of the cells; furthermore, quantitative (q)RT-PCR and Western blot methods was used to determine the expression of StAR and apoptosis related molecules Bcl-2, Bax and Caspase-3 on both mRNA and protein levels in different groups; finally the secretion of testosterone in different groups was examined by radioimmunoassay. We observed that the YC can increase the expression of StAR in a dose-dependent manner, and YC can activate the promoter of StAR; moreover, transfection of StAR siRNA can block YC induced anti-apoptotic effects and increased production of testosterone. In conclusion, our results suggested that YC might suppress the apoptosis of MLTC-1 cells and enhance the production of testosterone through regulating the expression of StAR.
Anthocyanins Prevent AAPH-Induced Steroidogenesis Disorder in Leydig Cells by Counteracting Oxidative Stress and StAR Abnormal Expression in a Structure-Dependent Manner

Abstract:Testosterone deficiency may increase the risk of sexual dysfunction and the failure of spermatogenesis. Oxidative stress that is derived from the destruction of homeostasis, disease, and exposure to contaminants can damage the steroidogenicity process in Leydig cells, resulting in a reduction in testosterone synthesis. Anthocyanins are a group of innoxious antioxidants widely recognized in food sources, and are an ideal candidate to relieve oxidative stress-related steroidogenesis disorder. However, there is still a major gap in our knowledge of the structure-function relationship of anthocyanin on the activity mentioned above. In the present study, four anthocyanins including cyanidin-3-glucoside (Cy-3-glu), delphinidin-3-glucoside (Dp-3-glu), pelargonidin-3-glucoside (Pg-3-glu), and cyanidin-3,5-diglucoside (Cy-3,5-diglu) were applied to reverse testosterone generation after employing 2,2'-Azobis(2-amidinopropane)-dihydrochloride (AAPH) as the inducer of oxidative stress in R2C cells. The results demonstrated that all four kinds of anthocyanins can inhibit ROS generation, alleviate mitochondrial membrane potential damage, and contribute to increased testosterone. Among them, Cy-3,5-diglu with diglycoside performed best on antioxidative ability and improved cell dysfunction and upregulated the expression of the steroidogenic acute regulatory protein (StAR). The molecular docking further revealed the direct combination between anthocyanins and StAR, suggesting that anthocyanins with monosaccharide were more likely to interact with StAR than with diglycoside. Taken together, these data indicate that recipient R2C cells under oxidative stress submitted to anthocyanins exhibited improved steroidogenesis in a structure-dependent manner. Anthocyanins could be considered the ideal ingredients against oxidative stress-induced testosterone deficiency.
Study on Yangjing Capsule promoting testosterone synthesis through regulating StAR promoter

Abstract:Objective To explore the mechanism of Yangjing Capsule increasing the synthesis of testosterone in Leydig cells.Methods Leydig cells (MLTC-1 cell line) were incubated for 24 h with different dose of Yangjing Capsule extract.T in culture supernatant was determined using chemiluminescence.The expression of StAR was detected by immunofluorescence microscopy.RT-PCR and western blot were used to detect the expression of StAR mRNA and protein.The activity of StAR promoter was detected by double luciferase reporter assay.Results After 24 h,the low,medium and high dose of Yangjing capsule extract all promoted the synthesis of testosterone in MLTC-1 cells.The Yangjing Capsule extract forced the immunofluorescence expression of StAR,and also upregulated the expression of StAR mRNA and protein in MLTC-1 cells.Simultaneously,Yangjing Capsule extract enhanced the activity of StAR promoter in MLTC-1 cells.Conclusion Yangjing Capsule can strengthen the activity of StAR promoter and increase the expression of StAR in Leydig cells,and ultimately improve the function of testosterone synthesis.
[The Preparation of StAR Gene Probe and Its Expression in Mice Leydig Cells under Stress].

Abstract:OBJECTIVE:To prepare the cDNA probe of the steroidogenic acute regulatory (StAR) gene, and to investigate the StAR mRNA expression in stressed C57BL/6 mice leydig cells.METHODS:cDNA fragment encoding mouse StAR was amplified by RT-PCR from the total RNA prepared from the testis, and then the RT-PCR product was cloned into pCR2. 1-TOPO vector. After StAR gene cDNA was sequenced, the Dig-labeled cRNA probes for mouse StAR gene were prepared by in vitro transcription from cDNA fragment. With the specific cRNA probes, in situ hybridization analysis was conducted in stressed mice testis and controls.RESULTS:The results demonstrated that StAR mRNA levels were significantly lower in stressed leydig cells than that in controls (P < 0.05).CONCLUSIONS:The decreased StAR mRNA levels induced by stress may result in a reduced production of StAR protein, which compromised the transportation of cholesterol, the substrate for androgen biosynthesis, into mitochondria, resulting in a poor T production in leydig cells and finally a declined T levels.

Abstract:Androgen production primarily occurs in Leydig cells located in the interstitial compartment of the testis. In aging males, testosterone is crucial for maintaining muscle mass and strength, bone density, sexual function, metabolic health, energy levels, cognitive function, as well as overall well-being. As men age, testosterone production by Leydig cells of the testes begins to decline at a rate of approximately 1% per year starting from their 30s. This review highlights recent findings concerning the use of natural polyphenolics compounds, such as flavonoids, resveratrol, and phenolic acids, to enhance testosterone production, thereby preventing age-related degenerative conditions associated with testosterone insufficiency. Interestingly, most of the natural polyphenolic antioxidants having beneficial effects on testosterone production tend to enhance the expression of the steroidogenic acute regulatory protein (Star) gene in Leydig cells. The STAR protein facilitates the entry of the steroid precursor cholesterol inside mitochondria, a rate-limiting step for androgen biosynthesis. Natural polyphenolic compounds can also improve the activities of steroidogenic enzymes, hypothalamus-pituitary gland axis signaling, and testosterone bioavailability. Thus, many polyphenolic compounds such as luteolin, quercetin, resveratrol, ferulic acid phenethyl ester or gigantol may be promising in delaying the initiation of late-onset hypogonadism accompanying aging in males.

Abstract:OBJECTIVE:To investigate the changes of morphology and steroidogenic function in aged human Leydig cells and to understand the mechanism of late onset hypogonadism (LOH).METHODS:Ten young and ten aged male subjects were enrolled in this study. AMS (Aging Male's Symptoms) scale was used for symptom evaluation. Testes species with LOH were utilized as the research model. Then the histological changes in testis and ultrastructure of Leydig cells were observed by HE staining and electron microscopy (EM), respectively. The serum total testosterone concentrations were measured by an ELISA kit. The expressions of steroidogenic acute regulatory (StAR) protein and cholesterol-side-chain cleavage enzyme (P450scc) were detected by western blot.RESULTS:The scores of AMS in the aged group were higher than those in the young group with decreased serum testosterone levels (61.25 ± 7.08 vs. 20.75 ± 3.73,P<0.001). And the serum testosterone level of the aged human was lower than that of the young human [(3.12 ± 0.58) μg/L vs. (6.29 ± 1.17) μg/L,P<0.05]. HE staining showed that degenerative changes occurred in the aged human testes. And many swollen mitochondria with mitochondrial cristae that disappeared were found in Leydig cells of the aged human by EM. The serum total testosterone level of the aged human was significantly lower than that of the young group. And the expressions of StAR and P450scc protein in the aged group were significantly lower than those of the young group.CONCLUSION:Mitochondrial swelling and decreased expressions of StAR and P450scc were closely related to the reduced ability of testosterone synthesis in aged males. And the exact mechanism needs further investigation.
Study on morphology and function of aged rat Leydig cells

Abstract:Objective: To study the morphological and functional of aged Leydig cells. Methods Ten young (3 months) and ten aged (24 months old) male SD rats of clean grade were used in the study as a research model. After anesthesia, blood samples were collected by vena cava puncture, and serum total testosterone concentrations were measured by ELISA. Histological changes of testis and ultrastructure changes of Leydig cells were observed by HE staining and electron microscopy (EM) respectively. Leydig cells were isolated from rat testes by density gradient centrifugation. After luteinizing hormone (LH) stimulation, the level of testosterone in cell culture supernatant was measured by ELISA and the expression of steroidogenic acute regulatory (StAR) in Leydig cells was detected by western blot. Results: Degenerative changes were showed in aged rat testes by HE staining, and many swollen mitochondria with mitochondrial cristae disappeared were found in Leydig cells of aged rats by EM. The serum testosterone level of aged rats was significantly lower than that of young group (P<0.05). In primary Leydig cells, with or without LH stimulation, the supernatant testosterone level and StAR protein expression in aged group was significantly lower than that of young group (P<0.01 and P<0.05 respectively). Conclusion: Mitochondrial swelling and low expression of LH-induced StAR protein might impaire the ability of testosterone synthesis in aged Leydig cells.
Transduction of Star Decreases Lipid Accumulation and Atherosclerotic Lesions in Apolipoprotein E-Deficient Mice

Abstract:We have previously demonstrated that steroidogenic acute regulatory protein (StAR) improves lipid clearance in endothelial cells. The present study was designed to evaluate the effect of altering StAR activities on the development of atherosclerosis in a well-established mouse model of atherogenesis. StAR mRNA and protein levels in different tissues as well as blood lipids were determined in wild-type and apoE-deficient (apoE−/−) mice at different ages. As previously seen, apoE−/− mice had significantly higher levels of triglyceride, total cholesterol and LDL cholesterol with concordant lower HDL-cholesterol. Older apoE−/− mice had lower level of high-density lipoprotein-bound cholesterol and higher levels of triglyceride, total cholesterol and low density lipoprotein-bound cholesterol. Hepatic StAR expression increased with aging when apoE−/− mice were younger than 3 months but decreased afterward. However, lipids components and StAR expression in the wild-type mice did not change significantly with aging. Injection of Ad-CMV-StAR via tail vein significantly reduced aortic atherosclerotic lesion and lipid accumulation in the blood and in the liver of apoE−/− mice compared to the mice receiving control virus encoding enhanced green fluorescent protein or normal saline. The enhanced StAR expression is associated with decreased hepatic and blood lipids and aortic plaque formation in a mouse model of atherosclerotic disease. These findings are the first direct evidence linking this transporter to the risk and severity of cardiovascular disease.
The roles of p38 MAPK → COX2 and NF-κB → COX2 signal pathways in age-related testosterone reduction

Abstract:In our study, we explored changes in the redox status and inflammatory response in the testes of the SAMP8 model of varying ages (2, 4, 8, 10 months old) compared with control mice SAMR1 by the methods of immunohistochemical staining, Western blotting, RT-PCR and Luminex multi-analyte cytokine profiling. We found that as ROS and inflammation levels increased during aging, steroidogenic enzymes (StAR and P450scc) reduced and led to the decline of testosterone production eventually. The pathways of P38 MAPK → COX2 and NF-κB → COX2 were detected by using specific inhibitors of SB203580 and Bay 11-7082 in isolated Leydig cells. These results indicated that activation of both p38 MAPK → COX2 and NF-κB → COX2 signaling pathways are functionally linked to the oxidative stress response and chronic inflammation during aging, and mediate their inhibitory effects on testosterone production.
Testosterone May Inhibit Its Synthesis by Decreasing the Expressing of StAR Protein

Abstract:In order to study the autocrine mechanism of testosterone synthesis, Leydig cells from 2-3 week old piglets were used as material. Difference doses of testosterone (0, 10 mg/mL, 20 mg/mL, 50 mg/mL,100 mg/mL) was added into these cultivating bottles and cocultivated with Leydig cells for 48 h. Then, changing cultivating Liquid and certain concentrations of hCG were added to each bottle and go on cultivating up to 4 h. On the other hand, some precursors including cholesterol(5 mg/mL), pregnenolone (500 ng/mL), dehydroepiandrosterone (500 ng/mL), androstenedione (500ng/ml) and 22R-hydroxychol-esterol (3μg/mL) and testosterone(50 mg/mL) were added into these cultivating bottles and cocultivated with leydig cells for 48 h. Then,changing cultivating Liquid and certain concentrations of hCG were added to each bottle and go on cultivating up to 4 h. The concentrations of testosterone, hCG and cAMP were measured by radioimmunoassay, but the level of steroidogenic acute regulatory(StAR) protein was measured by immunoblotting. The results were as follows: (1) with the increase of concentration of testosterone, the inhibiting effect of testosterone was gradually increasing. when the concentration of testosterone was 50 mg/mL, the inhibiting effect is the maximum. The decreasing percentages of concentration of testosterone, hCG binding rate and concentration of cAMP were 69. 93% ,61. 24% and 52. 4%;(2) although the conversion from cholesterol to testosterone,androstenedione to testosterone could significantly be inhibited, the conversion from other precursors to testosterone were not affected significantly; (3) the levels of protein was decreased by testosterone in 4 h. The results showed that testosterone may inhibit its synthesis by decreasing the expressing of StAR protein.
The Role of StAR2 Gene in Testicular Differentiation and Spermatogenesis in Nile Tilapia (oreochromis Niloticus)

Abstract:Sex steroids play critical roles in sex differentiation and gonadal development in teleosts. Steroidogenic acute regulatory protein (StAR), transporting cholesterol (the substrate for steroidogenesis) from the outer mitochondrial membrane to the inner membrane, is the first rate-limiting factor of steroidogenesis. Interestingly, two StAR genes (named as StAR1 and StAR2) have been isolated from non-mammalian vertebrates. To characterize the functions of the novel StAR2 gene in the gonadal differentiation and fertility, we generated a StAR2 homozygous mutant line in Nile tilapia (Oreochromis niloticus). StAR2 gene knockout in male tilapia impeded meiotic initiation, associate with the down-regulation of meiosis related gene expressions of vasa, sycp3 and dazl at 90 days after hatching (dah). Meanwhile, cyp11b2 expression and serum 11-KT production significantly declined in StAR2- /- XY fish at 90 dah. From 120-300 dah, spermatogenesis gradually recovered, and so did the expressions of vasa, sycp3 and dazl in StAR2-/- XY fish testes. However, seminiferous lobules arranged disorderly in StAR2-/- XY fish testes at 300 dah. The number of Leydig cells and expressions of downstream steroidogenesis enzymes including cyp11a1, 3 beta-HSD-I, 3 beta-HSD-II, cyp17a1 and cyp17a2 decreased in StAR2-/- XY fish testes at 300 dah. Serum testosterone and 11-KT levels were significantly lower in StAR2-/- XY fish than that of their control counterparts. Furthermore, significantly elevated ar, fsh and lh expressions in StAR2-deficient XY fish testes and pituitaries were found when compared with the control XY fish. Testes degeneration and spermatogenic cell apoptosis were observed, while no sperm were squeezed out in StAR2-/- XY fish testes at 540 dah. Taken together, our results suggest that StAR2 has a role in testicular development, spermatogenesis and spermiation by regulating androgen production in tilapia, but may not be essential and could be compensated.
Integrated stress response mediates HSP70 to inhibit testosterone synthesis in aging testicular Leydig cells

Abstract:The integrated stress response (ISR) is implicated in age-related diseases, while the molecular chaperone heat shock protein 70 (HSP70) can facilitate proper protein folding. However, the regulatory mechanism of ISR in insufficient testosterone synthesis of aging Leydig cells (LCs) remains unclear. This study aims to elucidate the regulatory role of ISR in inadequate testosterone synthesis of aging LCs. We observed a positive correlation between testosterone and HSP70 levels, which were found to be decreased in elderly men. ISR was detected in testicular tissue from old mice. The expression of testosterone synthesis related protein and the content of testosterone decreased in testicular tissue of old mice. Conversely, inhibition of the integrated stress response in testicular tissue led to an increase in steroid synthase expression among old mice. Furthermore, inhibiting ISR specifically within aging LCs resulted in enhanced protein translation efficiency and increased expression levels of new HSP70 and steroidogenic acute regulatory protein (StAR). These findings suggest that ISR occurrence within aging LCs affects StAR protein expression through regulation of HSP70-mediated translation, consequently impairing testosterone synthesis.
Luteolin modulates gene expression related to steroidogenesis, apoptosis, and stress response in rat LC540 tumor Leydig cells

Abstract:In males, androgens are mainly produced by Leydig cells from the testis. A critical and highly regulated step of steroidogenesis involves the importation of cholesterol within the mitochondria by the steroidogenic acute regulatory (STAR) protein. During aging, STAR protein levels in Leydig cells gradually decrease, leading to a reduced entry of cholesterol into mitochondria and lower testosterone production. In addition to preserving its steroidogenic capacity, tumor Leydig cells can also be excellent models for evaluating the mechanisms of action of anticancer agents. In this study, we examined whether polyphenolics having structural similarities to luteolin could promote steroidogenic and cancer-related gene expressions within rat L540 tumor Leydig cells. In this cell model, luteolin activated Star expression and increased progesterone as well as testosterone productions. Interestingly, luteolin decreased gene expression related to cholesterol biosynthesis, possibly inhibiting membrane synthesis and cell proliferation. In addition, increased expression of genes such as Fas, Cdkn1a, Atp7b, and Tp53, as well as increased accumulation of cleaved caspase 3 and PARP, in response to luteolin treatment indicates that apoptosis is being activated. Luteolin also modulated the expression of genes involved in stress response, such as glutathione-S transferases Gsta1 and Gstt2, and the unfolded protein response. Thus, dietary luteolin may be effective in Leydig cell tumor chemoprevention and in maintaining steroidogenesis in aging males.

Effect of Annexin 5 on StAR and Testosterone Synthesis Related Enzyme in Rat Leydig Cells

Abstract:OBJECTIVE To investigate the effect of annexin 5 on the expressions at mRNA levels and protein levels of StAR, P450scc, 3β-HSD, 17α-hydroxylase and 17β-HSD in rat Leydig cells. METHODS The primary rat Leydig cells were cultured for 24 h and then stimulated with 10(-9) mol/L annexin 5 for 12 h and 24 h respectively. Cellular total RNA and total protein were extracted respectively. The expressions of StAR, P450scc, 3β-HSD, and 17α-hydroxylase and 17β-HSD(10) mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR)and the protein levels were detected by Western blotting. RESULTS Compared with the control group, at the mRNA level, after being treated with annexin 5 for 12 h, only 17β-HSD(10) expression had a 26% increase (P<0.05) while the others had no significant difference. The expressions of StAR, P450scc and 3β-HSD elevated 55%, 69% and 59%(P<0.05) respectively, and 17β-HSD(10) increased 104%(P<0.01) while 17α-hydroxylase had no significant difference after being treated with annexin 5 for 24 h. At the protein level, after being treated with annexin 5 for 12 h, 17β-HSD expression had a 39% increase (P<0.05). After 24 h, P450scc, 3β-HSD and 17β-HSD elevated 35%, 88% (P<0.05) and 47% (P<0.01) respectively while StAR had no significant difference. CONCLUSION Annexin 5 regulates testosterone synthesis by affecting the expressions of P450scc, 3β-HSD and 17β-HSD at gene and protein levels.

Abstract:With aging, men develop testosterone-deficiency syndrome (TDS). The development is closely associated with age-related mitochondrial dysfunction of Leydig cell and oxidative stress-induced reactive oxygen species (ROS). Testosterone-replacement therapy (TRT) is used to improve the symptoms of TDS. However, due to its various side effects, research on functional ingredients derived from natural products that do not have side effects is urgently needed. In this study, using the mitochondrial dysfunction TM3 (mouse Leydig) cells, in which testosterone biosynthesis is reduced by H 2 O 2 , we evaluated the effects of elderberry extract and monosaccharide-amino acid (fructose–leucine; FL) on mRNA and protein levels related to steroidogenesis-related enzymes steroidogenic acute regulatory protein (StAR), cytochrome P450 11A1(CYP11A1, cytochrome P450 17A1(CYP17A1), cytochrome P450 19A1(CYP19A1, aromatase), 3β-hydroxysteroid dehydrogenase (3β-HSD), and 17β-hydroxysteroid dehydrogenase(17β-HSD). We analyzed elderberry extract and extract-derived FL for changes in ROS scavenging activity and testosterone secretion. Elderberry extract and FL significantly reduced H 2 O 2 -induced intracellular ROS levels, improved testosterone secretion, and increased the mRNA and protein expression levels of steroidogenesis-related enzymes (StAR, 3b-HSD, 17b-HSD, CYP11A1, CYp17A1). However, the conversion of testosterone to estradiol was inhibited by elderberry extract and extract-derived FL, which reduced the mRNA and protein expression of CYP19A1. In conclusion, elderberry extract and FL are predicted to have value as novel functional ingredients that may contribute to the prevention of TDS by ameliorating reduced steroidogenesis.

Abstract:Leydig cells contain significant amounts of constitutively produced steroidogenic acute regulatory protein (STAR; STARD1). Hormone-induced STAR plays an essential role in inducing the transfer of cholesterol into the mitochondria for hormone-dependent steroidogenesis. STAR acts at the outer mitochondrial membrane, where it interacts with a protein complex, which includes the translocator protein (TSPO). Mutations in STAR cause lipoid congenital adrenal hyperplasia (lipoid CAH), a disorder characterized by severe defects in adrenal and gonadal steroid production; in Leydig cells, the defects are seen mainly after the onset of hormone-dependent androgen formation. The function of constitutive STAR in Leydig cells is unknown. We generated STAR knockout (KO) MA-10 mouse tumor Leydig cells and showed that STAR KO cells failed to form progesterone in response to dibutyryl-cAMP and to TSPO drug ligands, but not to 22(R)-hydroxycholesterol, which is a membrane-permeable intermediate of the CYP11A1 reaction. Electron microscopy of STAR KO cells revealed that the number and size of lipid droplets were similar to those in wild-type (WT) MA-10 cells. However, the density of lipid droplets in STAR KO cells was drastically different than that seen in WT cells. We isolated the lipid droplets and analyzed their content by liquid chromatography–mass spectrometry. There was a significant increase in cholesteryl ester and phosphatidylcholine content in STAR KO cell lipid droplets, but the most abundant increase was in the amount of diacylglycerol (DAG); DAG 38:1 was the predominantly affected species. Lastly, we identified genes involved in DAG signaling and lipid metabolism which were differentially expressed between WT MA-10 and STAR KO cells. These results suggest that constitutive STAR in Leydig cells is involved in DAG accumulation in lipid droplets, in addition to cholesterol transport. The former event may affect cell functions mediated by DAG signaling.

Protective effect of TNFAIP3 on testosterone production in Leydig cells under an aging inflammatory microenvironment

Abstract:Background: The aging inflammatory microenvironment surrounding Leydig cells is linked to reduced testosterone levels in males. Tumor necrosis factor alpha-induced protein 3 (TNFAIP3) acts as a critical anti-inflammatory factor in various aging-related diseases. This study aims to investigate the protective effect of TNFAIP3 on testosterone production in Leydig cells under an aging inflammatory microenvironment. Methods: Bioinformatics analysis examined TNFAIP3 expression differences in aging rat testes and validated the findings in aging mouse testes. In vitro models of inflammation were established using two Leydig cell lines, with tumor necrosis factor alpha (TNF-α) as the inflammatory factor. Lentiviral transduction was utilized to manipulate TNFAIP3 expression in these cell lines. Transcriptomic sequencing identified differentially expressed genes in TNFAIP3-overexpressing cells. Results: Bioinformatics analysis and validation experiments revealed increased inflammatory signaling and elevated TNFAIP3 expression in aging rat and mouse testes. TNFAIP3 knockdown worsened testosterone synthesis inhibition and apoptosis in cells, while TNFAIP3 overexpression reversed these effects. Transcriptome analysis identified alterations in the P38MAPK pathway following TNFAIP3 overexpression. TNFAIP3 knockdown enhanced TNF-induced P38MAPK signaling, whereas its overexpression attenuated this effect. TNFAIP3 was found to regulate testosterone synthesis by upregulating CEBPB expression. Conclusions: TNFAIP3 exhibits inhibitory effects on apoptosis and promotes testosterone production in Leydig cells. The protective influence of TNFAIP3 on Leydig cells within an inflammatory microenvironment is likely mediated through by inhibiting the P38MAPK pathway and upregulating CEBPB expression.

Abstract:The structure,gene character and distribution of the steroidogenic acute regulatory protein(StAR) were summarized,and the transport functions of StAR and it′s homologous protein were also expounded.In steroid hormone cells,StarD4 transports cholesterol to outer mitochondrial membrane,however,StAR transports cholesterol from outer mitochondria membrane to inner mitochondrial membrane.The influencing factors on StAR expression,includeing the positive and negative factors,indirectly affect the steroidogenesis.The suggestion on conformity of the transport function of StAR homologous protein and regulative mechanism of StAR in the steroidogenesis was proposed.It should be noticed that some factors affecting StAR expression result in functional abnormity of reproductive endocrine.
