Abstract:Abstract Background Plants are the prime source of vital secondary metabolites (SMs) which are medicinally important for drug development, and these secondary metabolites are often used by plants in the various important tasks like defense against herbivory, interspecies defenses and against different types of stresses. For humans, these secondary metabolites are important as medicines, pigments, flavorings and drugs. Because most of the pharmaceutical industries are highly dependent on medicinal plants and their extraction, these medicinal plants are getting endangered. Main body Plant cell culture technologies are introduced as a viable mechanism for producing and studying SMs of plants. Various types of in vitro strategies (elicitation, hairy root culture system, suspension culture system, etc.) have been considerably used for the improvement of the production of SMs of plants. For the enhancement of SM production, suspension culture and elicitation are mainly used, but hairy root culture and other organ cultures are proved to satisfy the demand of secondary metabolites. Now, it is easy to control and manipulate the pathways that produce the plant secondary metabolites. Conclusions Techniques like plant cell, tissue and organ cultures provide a valuable method for the production of medicinally significant SMs. In recent years, most of the in vitro strategies are used due to knowledge and regulation of SM pathway in commercially valuable plants. In future, these things will provide a valuable method to sustain the feasibility of medicinal plants as the renewable sources of medicinally important compounds, and these methods will provide successful production of desired, important, valuable and also unknown compounds.

Abstract:Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites. Adventitious roots culture of Panax ginseng and Echinacea purpure has reached the scale of 1-10 kL. Some molecular biological techniques, such as transgenic technology and genetic stability are increasingly used in the studies on plant tissue cultures. The studies on elicitors have deepened into the induction mechanism, including signal molecules, functional genes, and so on. More and more biological elicitors, such as A. niger and yeast are used to increase the active compounds in plant tissue cultures. We also discussed the application of synthetic biology in the studies on biosynthesis of artemisinin, paclitaxel, and tanshinon. The studies on active ingredients biosynthesis of medicinal plants provide unprecedented possibilities to achieve mass production of active ingredients. Plant tissue cultures can not only produce active ingredients but also as experimental materials for biosynthesis. In order to improve the contents of active compounds in medicinal plants, following aspects could be carried out gene interference or gene silencing, gene overexpression, combination with chemical synthesis, application of elicitors, and site-directed mutagenesis of the key enzymes.

Abstract:Plants are the sources of many bioactive secondary metabolites which are present in plant organs including leaves, stems, roots, and flowers. Although they provide advantages to the plants in many cases, they are not necessary for metabolisms related to growth, development, and reproduction. They are specific to plant species and are precursor substances, which can be modified for generations of various compounds in different plant species. Secondary metabolites are used in many industries, including dye, food processing and cosmetic industries, and in agricultural control as well as being used as pharmaceutical raw materials by humans. For this reason, the demand is high; therefore, they are needed to be obtained in large volumes and the large productions can be achieved using biotechnological methods in addition to production, being done with classical methods. For this, plant biotechnology can be put in action through using different methods. The most important of these methods include tissue culture and gene transfer. The genetically modified plants are agriculturally more productive and are commercially more effective and are valuable tools for industrial and medical purposes as well as being the sources of many secondary metabolites of therapeutic importance. With plant tissue culture applications, which are also the first step in obtaining transgenic plants with having desirable characteristics, it is possible to produce specific secondary metabolites in large-scale through using whole plants or using specific tissues of these plants in laboratory conditions. Currently, many studies are going on this subject, and some of them receiving attention are found to be taken place in plant biotechnology and having promising applications. In this work, particularly benefits of secondary metabolites, and their productions through tissue culture-based biotechnological applications are discussed using literature with presence of current studies.

Abstract:Plant secondary metabolites exhibit a variety of biological activities and therefore serve as valuable therapeutics or flavoring compounds. However, the small amounts isolated from plants often cannot meet market demands. This led to the exploration of other, more profitable methods for their production, including plant cell culture systems, chemical synthesis and biotechnological production in microbial hosts. The biotechnological production can be pursued by reconstructing metabolic pathways in selected microbial systems. But due to their complexity, most of these pathways are not completely understood and require the expression of a multitude of genes in a foreign organism. Recently, next generation sequencing data and advances in gene silencing in plants allowed the elucidation of some biosynthetic pathways in more detail. Thus, the de novo production of some natural products, including morphine, strictosidine, artemisinin, taxol® and resveratrol, in extensively engineered microbial hosts has become feasible. This review highlights the reconstruction of these pathways, missing pieces and novel techniques employed.

Abstract:The development of plant tissue (including organ and cell) cultures for the production of secondary metabolites has been underway for more than three decades. Plant cell cultures with the production of high-value secondary metabolites are promising potential alternative sources for the production of pharmaceutical agents of industrial importance. Medicinal plant cell suspension cultures (MPCSC), which are characterized with the feature of fermentation with plant cell totipotency, could be a promising alternative "chemical factory". However, low productivity becomes an inevitable obstacle limiting further commercialization of MPCSC and the application to large-scale production is still limited to a few processes. This review generalizes and analyzes the recent progress of this bioproduction platform for the provision of medicinal chemicals and outlines a range of trials taken or underway to increase product yields from MPCSC. The scale-up of MPCSC, which could lead to an unlimited supply of pharmaceuticals, including strategies to overcome and solution of the associated challenges, is discussed.

Abstract:Plants have been used throughout the world for its medicinal powers since ancient time. The pharmacological properties of plants are based on their phytochemical components especially the secondary metabolites which are outstanding sources of value added bioactive compounds. Secondary metabolites have complex chemical composition and are produced in response to various forms of stress to perform different physiological tasks in plants. They are used in pharmaceutical industries, cosmetics, dietary supplements, fragrances, flavors, dyes etc. Extended use of these metabolites in various industrial sectors has initiated a need to focus research on increasing the production by employing plant tissue culture (PTC) techniques and optimizing their large scale production using bioreactors. PTC techniques being independent of climatic and geographical conditions will provide an incessant, sustainable, economical and viable production of secondary metabolites. This review article intends to assess the advantages of using plant tissue culture, distribution of important secondary metabolites in plant families, strategies involved for optimal metabolite production and the industrial importance of selected secondary metabolites.

Abstract:Plants display an amazing ability to synthesize a vast array of secondary metabolites that are an inexhaustible source of phytochemicals, bioactive molecules some of which impact the human health. Phytochemicals present in medicinal herbs and spices have long been used as natural remedies against illness. Plant tissue culture represents an alternative to whole plants as a source of phytochemicals. This approach spares agricultural land that can be used for producing food and other raw materials, thus favoring standardized phytochemical production regardless of climatic adversities and political events. Over the past 20 years, different strategies have been developed to increase the synthesis and the extraction of phytochemicals from tissue culture often obtaining remarkable results. Moreover, the availability of genomics and metabolomics tools, along with improved recombinant methods related to the ability to overexpress, silence or disrupt one or more genes of the pathway of interest promise to open new exciting possibilities of metabolic engineering. This review provides a general framework of the cellular and molecular tools developed so far to enhance the yield of phytochemicals. Additionally, some emerging topics such as the culture of cambial meristemoid cells, the selection of plant cell following the expression of genes encoding human target proteins, and the bioextraction of phytochemicals from plant material have been addressed. Altogether, the herein described techniques and results are expected to improve metabolic engineering tools aiming at improving the production of phytochemicals of pharmaceutical and nutraceutical interest.

Abstract:The paper covers the questions of secondary metabolite modulation in medicinal plants by means of gene engineering. It is demonstrated that cutting-edge tools of contemporary biotechnology tools made it possible to manage the biosynthesis of important bioactive substances, modify the secondary metabolism, enabling plants to synthesize and produce new compounds, as well as eliminate metabolic pathways of synthesizing harmful substances. Currently, large-scale production of bioactive substances (BAS) requires highly productive plants to produce them. Applying methods of gene engineering to medicinal plants is a promising way to reduce the resource consumption and increase their productivity, quality and the product's marketability [1]. Traditional growing and collecting techniques are challenged by resource shortage, environmental damage, etc. [2]. Gene engineering helps to increase pest, disease and herbicide resistance, gain greater yields and higher BAS content [3]. Using transgenic medicinal plants (TMP) as BAS producers in the pharmaceutical industry is crucial for metabolic engineering. Current research of the secondary metabolism modulation in TMP enables to modify the key BAS biosynthesis and the secondary metabolism, so that plants can produce new substances, or, on the contrary, silence the metabolic pathways for harmful ones. This way, greater TMP biomass with higher BAS content can be obtained in bioreactors. This would require rather modest investments — an advantage for biopharmacy. Nowadays, TMP are grown in vitro as calluses or suspension cell cultures. Biotechnology can modify the secondary metabolism in TMP to produce surplus amounts of necessary BAS, reduce the content of toxic compounds or even synthesize new substances. The versatility of transcription and translation mechanisms in medicinal plants enables them to accumulate homologous substances and synthesize heterologous ones. It is known that in TMP, MYB transcription factors are involved in gene regulation in secondary metabolic pathways, regulation of genes engaged in developmental processes, etc. [4]. In conclusion, we should emphasize the relative biosafety of BAS obtained from TMP, for human use, as they are chemically pure and are not connected with biological hazards.
Metabolic engineering in hairy roots: An outlook on production of plant secondary metabolites

Abstract:Plants are one of the vital sources of secondary metabolites. These secondary metabolites have diverse roles in human welfare, including therapeutic implication. Nevertheless, secondary metabolite yields obtained through the exploitation of natural plant populations is insufficient to meet the commercial demand due to their accumulation in low volumes. Besides, in-planta synthesis of these important metabolites is directly linked with the age and growing conditions of the plant. Such limitations have paved the way for the exploration of alternative production methodologies. Hairy root cultures, induced after the interaction of plants with Rhizobium rhizogenes (Agrobacterium rhizogenes), are a practical solution for producing valuable secondary metabolite at low cost and without the influence of seasonal, geographic or climatic variations. Hairy root cultures also offer the opportunity to get combined with other yield enhancements strategies (precursor feeding, elicitation and metabolic engineering) to further stimulate and/or enhance their production potential. Applications of metabolic engineering in exploiting hairy root cultures attracted the interest of several research groups as a means of yield enhancement. Currently, several engineering approaches like overexpression and silencing of pathway genes, and transcription factor overexpression are used to boost metabolite production, along with the contextual success of genome editing. This review attempts to cover metabolic engineering in hairy roots for the production of secondary metabolites, with a primary emphasis on alkaloids, and discusses prospects for taking this research forward to meet desired production demands.

Abstract:Plants are the main source of secondary metabolites, which can be used in different sectors such as pharmaceutics, food, cosmetics, agriculture, etc. turning them into an attractive source of income. Primary metabolites (carbohydrates, lipids, and proteins) have been linked to vital processes such as growth, development, and fruiting, while the secondary metabolites (phenols, flavonoids, and carotenoids, to name a few) are the result of adaptation and evolution of the plants with respect to changes in the environment. Therefore, it can be said that the secondary metabolites are secreted when the plant is under biotic and abiotic stresses. These secondary metabolites possess several bio-active functions and, hence, are well recognized for industrial applications. Due to difficulties for extracting these natural plant bio-active compounds, they are usually produced alternatively through plant cell/tissue culture methods. Further, several approaches, such as the use of plant and tissue cell cultures, the application of metabolism-inducing factors or elicitation, control of biological factors, such as intensity of light, induction by sound waves, and application of nanoparticles are used to alter/enhance in vitro production of these bio-active metabolites. Overall, these approaches interact with the biochemical routes of the plant either in positive or negative ways to produce secondary metabolites in high quantities. Although there is information on this aspect, the effect of these strategies cannot be generalized, since it has been observed that the metabolism of the plant response depends on the study model, the concentration and time of use of the stimulus, as well as its nature. Considering the above facts, this chapter discusses on the most current strategies for the production of secondary metabolites in plants in a continuous and reliable manner.

Abstract:For centuries plants have been intensively utilized as reliable sources of food, flavoring, agrochemical and pharmaceutical ingredients. However, plant natural habitats are being rapidly lost due to climate change and agriculture. Plant biotechnology offers a sustainable method for the bioproduction of plant secondary metabolites using plant in vitro systems. The unique structural features of plant-derived secondary metabolites, such as their safety profile, multi-target spectrum and "metabolite likeness," have led to the establishment of many plant-derived drugs, comprising approximately a quarter of all drugs approved by the Food and Drug Administration and/or European Medicinal Agency. However, there are still many challenges to overcome to enhance the production of these metabolites from plant in vitro systems and establish a sustainable large-scale biotechnological process. These challenges are due to the peculiarities of plant cell metabolism, the complexity of plant secondary metabolite pathways, and the correct selection of bioreactor systems and bioprocess optimization. In this review, we present an integrated overview of the possible avenues for enhancing the biosynthesis of high-value marketable molecules produced by plant in vitro systems. These include metabolic engineering and CRISPR/Cas9 technology for the regulation of plant metabolism through overexpression/repression of single or multiple structural genes or transcriptional factors. The use of NMR-based metabolomics for monitoring metabolite concentrations and additionally as a tool to study the dynamics of plant cell metabolism and nutritional management is discussed here. Different types of bioreactor systems, their modification and optimal process parameters for the lab- or industrial-scale production of plant secondary metabolites are specified.

Quantification and comparison of major secondary metabolites and antioxidant efficacy in Gardenia jasminoides mother plant, callus and suspension culture: An exploratory study to enhance bioactive compounds

Abstract:Gardenia jasminoides suspension culture has gained recognition as a functional approach for bioactive component development in the pharmaceutical industries but exhibits limited biomass accumulation and secondary metabolite production. This study presents the first record of maximum biomass production and demonstrates the cumulative levels of phenols, flavonoids and terpenoids observed through the growth trajectory of G. jasminoides suspension culture. Successful callus induction was obtained from leaf explants cultured on Murashige and Skoog (MS) medium augmented with a standardized conjunction of 1 mg/L of 2,4-Dichlorophenoxyacetic acid (2,4-D) and 0.5 mg/L kinetin (KT). The experimental outcomes revealed that on the 35th day, the in vitro suspension culture exhibited the highest biomass accumulation which was 5.43 times greater than the initial inoculation level. The study quantified total phenols, flavonoids, and terpenoids present in leaf explants, callus cultures, and suspension cultures and determined antioxidant efficacy. Findings suggest that an optimized growth regulator in G. jasminoides suspension culture significantly increases biomass accumulation. Quantification of secondary metabolites offers a promising path for future enhancement of their yield through elicitation and holds the potential to achieve extensive yield of cost-effective bioactive components.

Abstract:Nature provides an array of medicinal plants that serve the society by treating a wide range of diseases. Many plants are on the verge of extinction due to their extensive use, ignorance, and restricted habitat. Thus, generation of in vitro cultures via “plant tissue culture techniques” is necessary for maintenance and large-scale propagation of flora. Additionally, these in vitro cultures also provide constant production of a wide variety of complex and structurally diverse secondary metabolites, possessing a range of therapeutic properties and biological activities. This chapter emphasizes on the establishment of in vitro plant cell cultures for secondary metabolite production and their applications in various biological assays. Furthermore, a brief description of in vitro assays is given to elucidate a particular type of bioactivity and its mechanism of action.

Abstract:Phenolic compounds represent big group of plant secondary metabolites that influence flavor, color, and texture and can be used as food additives, nutraceuticals, and pharmaceuticals.However, there are some limitations in obtaining sufficient amount of these bioactive compounds from plants, because they are rather seldom or occur naturally in plant tissues only at very low concentrations. Alternatively, it is possible to synthesize them chemically, but this way if oft technologically not possible or very sophisticated and economically infeasible.Plant in vitro cultures provide an attractive route to produce high-value plant-derived products and therefore can be an alternative source of valuable phenolics.Moreover, compounds synthesized by plant in vitro cultures are natural products and therefore can be more easily accepted by consumers as artificially synthetized substances.The synthesis of phytochemicals by plant in vitro cultures in contrast to these in plants is not depending on environmental conditions and can be regulated through standard physical and chemical conditions in bioreactor, which helps to avoid qualitative and quantitative fluctuations in product yield.The process of obtaining valuable phytochemicals can be represented as a multistage technology, each link of which can vary individually in dependence of specific requirements of in vitro cultures (e.g., phytohormones, nutrients, light) or properties of end product (e.g., antioxidative potential, stability).For the establishment of high-producing and fast-growing cell lines, the parent plants should be selected (Murthy et al. Abstract:Secondary metabolites are gaining an increasing importance in various industries, such as pharmaceuticals, dyes, and food, as is the need for reliable and efficient methods of procuring these compounds. To develop sustainable and cost-effective approaches, a comprehensive understanding of the biosynthetic pathways and the factors influencing secondary metabolite production is essential. These compounds are a unique type of natural product which recognizes the oxidative damage caused by stresses, thereby activating the defence mechanism in plants. Various methods have been developed to enhance the production of secondary metabolites in plants. The elicitor-induced in vitro culture technique is considered an efficient tool for studying and improving the production of secondary metabolites in plants. In the present review, we have documented various biosynthetic pathways and the role of secondary metabolites under diverse environmental stresses. Furthermore, a practical strategy for obtaining consistent and abundant secondary metabolite production via various elicitation agents used in culturing techniques is also mentioned. By elucidating the intricate interplay of regulatory factors, this review paves the way for future advancements in sustainable and efficient production methods for high-value secondary metabolites.

Functional Genomic Approaches to Explore Secondary Metabolites in Medicinal Plants

Abstract:Extensive genomic data concerning medicinal plants are rather scarce and insights of the secondary metabolic pathways and their regulatory mechanism are insufficient, hampering the broad application of cell or tissue cultivation and metabolic engineering to producing high-value secondary metabolites. The integration of cDNA-AFLP based transcript profiling and metabolomics, a new development of functional genomic approaches could establish correlations between the changes of secondary metabolites and expressions of related genes. It has manifested widely applicative prospects in seeking genes involved in biosynthesis of secondary metabolites and exploring secondary metabolic pathways. Functional genomic approaches are promising trends in the field of medicinal plants secondary metabolites research and will lead to better utilization of natural medicinal resources.

Abstract:Plant-derived natural products are a class of secondary metabolites with complex structures and diverse properties, which are widely used in food, medicine, cosmetics and other fields. At present, the main source of plant-derived natural products is direct extraction from plants, which is time consuming and farming land occupying. Microbial synthesis has been studied to produce plant-derived natural products for the advantages of short growth cycle, mature manipulating methods and controllable large-scale fermentation. Nowadays, it has attracted many attentions to achieve biosynthesis and transformation of plant-derived natural products employing microbial cells. Many kinds of natural products such as terpenoids, flavonoids, alkaloids and saponins have been successfully synthesized in microbes. In this review, the applications of microbial cell factories in the synthesis of plant-derived natural products have been summarized as well as different strategies, which will be helpful for the systematic and in-depth study in the synthesis and transformation of plant-derived natural products.

Abstract:Main conclusionPlant tissue culture as an important tool for the continuous production of active compounds including secondary metabolites and engineered molecules. Novel methods (gene editing, abiotic stress) can improve the technique.Humans have a long history of reliance on plants for a supply of food, shelter and, most importantly, medicine. Current-day pharmaceuticals are typically based on plant-derived metabolites, with new products being discovered constantly. Nevertheless, the consistent and uniform supply of plant pharmaceuticals has often been compromised. One alternative for the production of important plant active compounds is in vitro plant tissue culture, as it assures independence from geographical conditions by eliminating the need to rely on wild plants. Plant transformation also allows the further use of plants for the production of engineered compounds, such as vaccines and multiple pharmaceuticals. This review summarizes the important bioactive compounds currently produced by plant tissue culture and the fundamental methods and plants employed for their production.
