Metalloproteinases and Regulation of Endometrial Function

Abstract:Matrix metalloproteinases (MMP) form a family of homologous enzymes (Table 1) which have a Zn2+ atom in their active site (see Matrisian 1990 for review). They are secreted as inactive pro enzymes (zymogens) which become activated upon partial hydrolysis whereby they loose their propeptide. They are classified in three subfamilies according to their substrate specificity: Gelatinases are represented by two enzymes, gelatinase A and B (72 and 92 kDa gelatinases, or MMP-2 and MMP-9, respectively). These proteases digest collagen type IV (the major constituent of basement membranes) and denatured collagen (gelatin). Collagenases include three proteases: the interstitial collagenase (MMP-1 or collagenase), the neutrophil collagenase (MMP-8), and collagenase 3 (MMP-13). These enzymes digest collagen types I, II, III, VII, and X. They are thus directed at digesting the collagen of the extracellular matrix of the interstitium.
