Abstract:Aryl carboxylic acids are among the most abundant substrates in chemical synthesis and represent a perfect example of a traceless directing group that is central to many processes in the preparation of pharmaceuticals, natural products and polymers. Herein, we describe a highly selective method for the direct step-down reduction of carboxylic acids to arenes, proceeding via well-defined Pd(0)/(ii) catalytic cycle. The method shows a remarkably broad substrate scope, enabling to direct the classical acyl reduction towards selective decarbonylation by a redox-neutral mechanism. The utility of this reaction is highlighted in the direct defunctionalization of pharmaceuticals and natural products, and further emphasized in a range of traceless processes using removable carboxylic acids under mild, redox-neutral conditions orthogonal to protodecarboxylation. Extensive DFT computations were conducted to demonstrate preferred selectivity for the reversible oxidative addition and indicated that a versatile hydrogen atom transfer (HAT) pathway is operable.

Abstract:We describe the direct synthesis of organophosphorus compounds from ubiquitous aryl and vinyl carboxylic acids via decarbonylative palladium catalysis. The catalytic system shows excellent scope and tolerates a wide range of functional groups (>50 examples). The utility of this powerful methodology is highlighted in the late-stage derivatization directly exploiting the presence of the prevalent carboxylic acid functional group. DFT studies provided insight into the origin of high bond activation selectivity and P(O)-H isomerization pathway.

Abstract:In this anniversary issue, we present a DFT study of the mechanism of decarbonylative Hirao cross-coupling of carboxylic-phosphoric anhydrides to afford aryl phosphonates. Traditionally, the direct activation of carboxylic acids to participate in decarbonylative couplings is performed in the presence of carboxylic acid anhydride activators. We discovered that direct dehydrogenative decarbonylative phosphorylation of benzoic acid can be performed in high yield via dehydrogenative and decarbonylative coupling in the presence of phosphite as dual activating and nucleophilic reagent, enabling direct decarbonylative phosphorylation. Control studies demonstrated that carboxylic-phosphoric anhydride (acyl phosphate) is an intermediate in this process. DFT studies were conducted to gain insight into this decarbonylative process and compare the selectivity of C-O and P-O bond activations. Considering the utility of ubiquitous carboxylic acids, this alternative activation pathway may find applications in decarbonylative coupling of carboxylic acids for the synthesis of valuable molecules in organic synthesis.

Abstract:Decarbonylative borylation of carboxylic acids is reported. Carbon electrophiles are generated directly after reagent-enabled decarbonylation of the in situ accessible sterically-hindered acyl derivative of a carboxylic acid under catalyst controlled conditions. The scope and the potential impact of this method are demonstrated in the selective borylation of a variety of aromatics (>50 examples). This strategy was used in the late-stage derivatization of pharmaceuticals and natural products. Computations reveal the mechanistic details of the unprecedented C-O bond activation of carboxylic acids. By circumventing the challenging decarboxylation, this strategy provides a general synthetic platform to access arylpalladium species for a wide array of bond formations from abundant carboxylic acids. The study shows a powerful combination of experiment and computation to predict decarbonylation selectivity.

Abstract:Cooperative bimetallic catalysis is a fundamental approach in modern synthetic chemistry. We report bimetallic cooperative catalysis for the direct decarbonylative heteroarylation of ubiquitous carboxylic acids via acyl C-O/C-H coupling. This novel catalytic system exploits the cooperative action of a copper catalyst and a palladium catalyst in decarbonylation, which enables highly chemoselective synthesis of important heterobiaryl motifs through the coupling of carboxylic acids with heteroarenes in the absence of prefunctionalization or directing groups. This cooperative decarbonylative method uses common carboxylic acids and shows a remarkably broad substrate scope (>70 examples), including late-stage modification of pharmaceuticals and streamlined synthesis of bioactive agents. Extensive mechanistic and computational studies were conducted to gain insight into the mechanism of the reaction. The key step involves intersection of the two catalytic cycles via transmetallation of the copper-aryl species with the palladium(II) intermediate generated by oxidative addition/decarbonylation.

Abstract:Formic acid is efficiently used as a C1 source to directly carboxylate allylic alcohols in the presence of a low loading of palladium catalyst and acetic anhydride as additive to afford β,γ‐unsaturated carboxylic acids with excellent chemo‐, regio‐, and stereoselectivity. The reaction proceeds through a carbonylation process with in situ‐generated carbon monoxide under mild conditions, avoiding the use of high‐pressure gaseous CO. A bisphosphine ligand with a large bite angle (4,5‐bis{diphenylphosphino}‐9,9‐dimethylxanthene, Xantphos) was found to be uniquely effective for this transformation. The regio‐ and stereoconvergence of this reaction is ascribed to the thermodynamically favored isomerization of the allylic electrophile in the presence of the palladium catalyst.

Abstract:In the absence of directing auxiliaries, the catalytic addition of carbogenic groups to unactivated alkenes with control of regioselectivity remains an ongoing challenge in organic chemistry. Here we describe a directing-group-free, nickel-catalysed strategy that couples a broad array of unactivated and activated olefins with aryl-substituted triflates and organometallic nucleophiles to afford diarylation adducts in either regioisomeric form, in up to 93% yield and >98% site selectivity. By switching the reagents involved, the present strategy may be extended to other classes of dicarbofunctionalization reactions. Mechanistic and computational investigations offer insights into the origin of the observed regiochemical outcome and the utility of the method is highlighted through the concise syntheses of biologically active molecules. The catalyst control principles reported are expected to advance efforts towards the development of general site-selective alkene functionalizations, removing the requirement for neighbouring activating groups.

Abstract:Abstract Transition-metal-catalyzed homogeneous dehydrogenation and isomerization are common organic molecular activation reactions. Palladium hydrides are good olefin isomerization catalysts but are usually short-lived species under redox-active dehydrogenation conditions. Here, we show that Pd-H in the presence of an N-heterocyclic carbene ligand and an alkene regulator enables transfer-dehydroaromatization, avoiding the homo-disproportionation pathway. The desired product is obtained with up to 99:1 selectivity, and the exo-to-endo olefin isomerization can be carried out in one pot. In contrast to previously reported methods that rely on the efficient removal of Pd-H, the approach reported herein benefits from the steric effects of the N-heterocyclic carbene and the choice of alkene to regulate the competing reactivity of allylic C‒H activation and hydropalladation. This method circumvents the challenges associated with tedious olefin separation and a low exo-to-endo olefin isomerization ratio and expands the scope to include challenging endo- and exo-cyclic olefins under mild, neutral, and oxidant-free conditions. Overall, herein, we provide a strategy to synthesize (hetero)aromatic compounds via chemoselective dehydrogenation of cyclic alkenes over ketones and the dehydrogenative Diels-Alder reaction of a cyclic enamine.

Abstract:The synthesis of (deuterated) 1,1-disubstituted alkenes via Pd-catalyzed decarboxylative hydroarylation of alkynyl carboxylic acids with arylboronic acids has been developed. The reaction features excellent regioselectivity, a broad substrate scope and gram-scale synthetic ability and offers a general synthetic method to synthesize 1,1-dideuterio olefins. Preliminary mechanism investigations indicate that 1,1-disubstituted alkenes are formed by hydroarylation of terminal alkynes generated by in situ decarboxylation of alkynyl carboxylic acids.

Abstract:Currently used directing groups for selective aliphatic β-functionalization of carbonyl compounds show excellent reactivity and selectivity with an amide as a linker. Described herein is 2-piconimide, used for the first time with commercially available 2-picolinamide/2-picolic acid as precursors, to direct C-H arylation/alkenylation by palladium catalysis. The directing group is essential for promoting the sequnetial primary and secondary C(sp(3))-H arylation with different aryl iodides in one substrate. The directing group was easily removed under simple reaction conditions at room temperature.

Abstract:A regioselective Pd-catalyzed hydrocarboxylation of vinyl arenes with oxalic acid is described. A wide variety of either linear or branched carboxylic acids can be readily obtained with high regioselectivities under mild reaction conditions. The reaction process is operationally simple and requires no handling of toxic CO. Besides the ligand, the counteranion of the Pd catalyst system plays an important role in the regioselectivity.

Abstract:Decarbonylation of aldehydes is a basic organic transformation, which has been developed for more than six-decade. However, as comparing to well-studied aromatic aldehydes, fewer examples for catalytic decarbonylation of aliphatic aldehydes were reported, mainly on simple or special substrates. For α -bulky or highly functionalized ones, stoichiometric Rh(I) were usually required for decent yields. Herein, we present a rare example of Ir(I)-catalyzed direct decarbonylation of α -quaternary aldehydes with broad substrate scope and good functional group compatibility via judicious selection of ligand. The α -chirality is memorized in this decarbonylation process. In addition, we report a broad-spectrum decarbonylation of α -secondary and α -tertiary aldehydes containing multifunctional groups with an improved Rh(I)/DPPP recipe. Finally, we realized selective decarbonylation of α -tertiary aldehydes in the presence of α -quaternary one via the reactivity differences.

Abstract:Herein, we present a catalytic system comprising Pd(OAc)2/Xantphos/Al(OTf)3, which exhibits excellent reactivity and linear selectivity in catalyzing the hydroesterification of unsaturated acids/esters, which were electron‐deficient, aliphatic alkenes, functionalized styrenes, and phenylacetylenes. Mechanistic studies indicate that the reaction proceeds via the palladium hydride pathway. Herein, we introduce a catalytic system consisting of Pd (OAc)2/Xantphos/Al (OTf)3, which exhibits excellent reactivity in catalyzing the hydroesterification of a wide range of substrates, including electron‐deficient unsaturated acids/esters, aliphatic alkenes, functionalized styrenes, and phenylacetylenes. The catalytic system featured high reactivity and selectivity, low catalyst loading, wide substrate scope, and low CO pressure. Importantly, it eliminated the need for corrosive strong Brønsted acid additives, yielding alkyl‐ or aryl‐substituted dicarboxylic acid esters and monocarboxylic acid ester derivatives that hold significant industrial and pharmaceutical value. Mechanistic studies indicate that the reaction proceeds via the palladium hydride pathway.

Abstract:Direct conversion of carboxylic acids to alkanes by decarboxylation on PdH x is an effective way to defunctionalize biomass-derived feedstock. This catalytic decarboxylation generally suffers from low activity and selectivity loss by competing hydrogenation routes. Anchoring the carboxylate group with K + on PdH x /C increases the rate of decarboxylation by more than one order of magnitude. Temperature-programmed desorption of CO 2 shows the modification to increase the surface basicity. In turn, this induces dissociative adsorption of the acid carboxylic group. The resulting adsorption as carboxylate facilitates the C-C bond cleavage by weakening the bond adjacent to the carboxyl group.

Abstract:Employing density functional theory (DFT) calculations, the study meticulously analyzes the mechanistic details of the Pd-catalyzed dual-γ-1,1-C(sp3)–H activation of aliphatic carboxylic acids. To explore the intrinsic nature of double C(sp3)–H in aliphatic carboxylic acids, we compared the reaction mechanisms of two coupling agents: allyl alcohol and methyl acrylate. The calculations indicated that allyl alcohol favors the second C(sp3)–H activation due to the absence of conjugation effects, which typically hinder C(sp3)–H activation. In contrast, methyl acrylate favors the Michael addition mechanism due to reduced steric hindrance and the formation of a stabilizing Na−O coordination interaction. We also computed the reaction mechanism of the coupling agent but-3-en-2-yl acetate and compared it with that of allyl alcohol to determine the reasons for the occurrence of β-H elimination. For allyl alcohol, β-H elimination is favored due to the lack of steric hindrance in the transition state, whereas for but-3-en-2-yl acetate, β-OAc elimination is preferred because of a stabilizing N-H⋯O hydrogen bond in the transition state. The study also highlights the significant role of Na2HPO4 base in enhancing the reaction's efficiency. Specifically, we found that the presence of Na2HPO4 base significantly facilitates the hydroxyl deprotonation process. To further explore the reaction's diastereoselectivity, we calculated the diastereoselectivity-determining steps for two substrates with significantly different d.r. ratios and analyzed the driving forces behind the high diastereoselectivity. Our analysis revealed that the diastereoselectivity of the reaction improves as the disparity in steric hindrance between the two substituents at the β-position of the carboxylic acid increases. Overall, the mechanistic insights gained in this study may prove valuable in the ongoing advancement and application of metal-catalyzed dual-1,1-C(sp3)−H activation.

Abstract:Oxidant-controlled selective mono-/di-alkenylation of aryl C-H bonds via Pd-catalysis is reported. The substrate scopes for both the mono- and di-alkenylation were good. Thioether was used as the directing group and the product can be transformed to a useful sulfoxide-olefin ligand by simple oxidation in quantitative yield.
