Abstract:A new method of conjugating horseradish peroxidase with proteins was developed. The carbohydrate moiety of fluorodinitrobenzene-blocked peroxidase was oxidized with sodium periodate to form aldehyde groups. The peroxidase-aldehyde was then bound to free amino groups of proteins unidirectionally at high efficiencies. Peroxidase-labeled immunoglobulin retained its immunologic as well as enzymatic activities.

Improving the sensitivity of traditional Western blotting via Streptavidin containing Poly‐horseradish peroxidase (PolyHRP)

Abstract:Immunoassays such as enzyme‐linked‐immunosorbent assays (ELISAs) and Western blotting have been the common choice for protein validation studies for the past several decades. Technical advancements and modifications are continuously being developed to enhance the detection sensitivity of these procedures. Among them, streptavidin containing Poly‐horseradish peroxidase (PolyHRP) based detection strategies have been shown to improve signals in ELISA. The use of commercially available streptavidin and antibodies conjugated with many HRPs (PolyHRPs) to potentially enhance the detection sensitivity in Western blotting has not been previously investigated in a comprehensive manner. The use of PolyHRP‐secondary antibody instead of HRP‐secondary antibody increased the Western blotting sensitivity by up to 85% depending on the primary antibody used. The use of a biotinylated secondary antibody and commercially available streptavidin conjugated with HRP or PolyHRP all resulted in increased sensitivity with respect to antigen detection. Utilizing a biotinylated secondary antibody and streptavidin conjugated PolyHRP resulted in as much as a 110‐fold increase in Western blotting sensitivity over traditional Western blotting methods. Quantification of troponin I in rat heart lysates showed that the traditional Western blotting method only detected troponin I in ≥ 2 μg of lysate while streptavidin conjugated PolyHRP20 detected troponin I in ≥ 50 ng of lysate. A modified blocking procedure is also described that eliminated the interference caused by the endogenous biotinylated proteins. These results suggest that streptavidin conjugated PolyHRP and PolyHRP secondary antibodies are likely to be commonly utilized for Western blots in the future.This article is protected by copyright. A Specific Sensitive and Rapic ELISA for Determination of Trace Amounts of IgG

Abstract:A specific, sensitive and rapic ELISA(double antibody sandwich technique)with anti-human IgG and corresponding Horse radish peroxidase conjugate was described for the quantitative determination of trace amounts of IgG.The chequerboard titrations were applied for selection of optimal con centration of coating antibody and conjugate, they are 10rr/ml and 1:2000 respectively. The sensitivity can reach as low as 3.0 ng/ml.It is suitable for research work as well as clinical study.Several factors affected the method have been investigated. Reproduci- bility and specificity of the method were identified.

Abstract:Antigen was identified histochemically without the use of labeled antibodies by the sequential application of (a) specific rabbit antiserum, (b) sheep antiserum to rabbit immunoglobulin G, (c) specifically purified, soluble horseradish peroxidase-anti-horseradish peroxidase complex (PAP), (d) 3,3'-diaminobenzidine and hydrogen peroxide and (e) osmium tetroxide. A simple method for preparation of high yields of PAP consisted of precipitation of antibody from specific rabbit antiserum with horseradish peroxidase (PO) at equivalence, solubilization of the washed precipitate with excess PO at pH 2.3, 1°C, followed by immediate neutralization and separation of PAP from PO by half-saturation with ammonium sulfate. The ratio of PO to anti-PO in PAP was 3:2 irrespective of the source of antiserum. PAP was heterogeneous on electrophoresis, homogeneous on sedimentation, diffusion and electron microscopy and consisted of pentagons with diameters of 205 Å. s 20, w , 11.98 x 10 –13 ; d 20, w , 2.48 x 10 –7 ; molecular weight by sedimentation velocity, 429,000, and equilibrium, 413,000. Sensitivity and specificity of immunohistochemical staining of spirochetes was about 100- to 1000-fold that of immunofluorescence. The unexpected ratio of PO to anti-PO is presumed to be due to stabilization by the pentagonal shape in which three corners are suspected to be PO and two antibody fragment Fc.

Abstract:Salmonella anatis and the antibody (Ab) against human cytomegalovirus (HCMV) in serum were detected by chemiluminescent photographic detection (CPD) on nitrocellulose (NC) membranes using horseradish peroxidase (HRP) as the label and X-ray film, HRP was first estimated on NC membranes with a limit of detection of 0.5-2.5 amol. When used to detect the Ab against HCMV, the antigen was mobilized on the NC membrane before the incubation of human serum, then the HRP labelled sheep Ab against human IgM was incubated again, Finally, the washed membrane was put into direct contact with a piece of filter-paper that had previously absorbed the CL substrate mixture and was exposed to the X-ray film, Fifty-seven human serum samples were thus detected for, the Ab against HCMV. The results were in good agreement with the routine enzyme-linked immunosorbent assay method, but the CPD procedures used an HRP-labelled sheep Ab solution 1000 times more dilute. Salmonella anatis was detected with a limit of detection of 100 bacteria.
Preparation of goat and rabbit anti-camel immunoglobulin G whole molecule labeled with horseradish peroxidase

Abstract:AIM: As the labeled anti-camel immunoglobulins (Igs) with enzymes for enzyme-linked immunosorbent assay (ELISA) are unavailable in the Egyptian market, the present investigation was directed for developing local labeled anti-camel IgG with horseradish peroxidase (HRP) to save hard curacy.MATERIALS AND METHODS: For purification of camel IgG whole molecule, camel sera was preliminary precipitated with 50% saturated ammonium sulfate and dialyzed against 15 mM phosphate-buffered saline pH 7.2 then concentrated. This preparation was further purified by protein A sepharose affinity column chromatography. The purity of the eluted camel IgG was tested by sodium dodecyl sulfate polyacrylamide gel electrophoresi. Anti-camel IgG was prepared by immunization of goats and rabbits separately, with purified camel IgG. The anti-camel IgG was purified by protein A sepharose affinity column chromatography. Whole molecule anti-camel IgG was conjugated with HRP using glutraldehyde based assay. Sensitivity and specificity of prepared conjugated secondary antibodies were detected using positive and negative camel serum samples reacted with different antigens in ELISA, respectively. The potency of prepared conjugated antibodies was evaluated compared with protein A HRP. The stability of the conjugate at -20°C during 1 year was assessed by ELISA.RESULTS: The electrophoretic profile of camel IgG showed four bands of molecular weight 63, 52, 40 and 33 kDa. The recorded sensitivity and specificity of the product are 100%. Its potency is also 100% compared to 58-75% of commercial protein A HRP. The conjugates are stable for 1 year at -20°C as proved by ELISA.CONCLUSION: Collectively, this study introduces goat and rabbit anti-camel IgG whole molecules with simple, inexpensive method, with 100% sensitivity, 100% specificity and stability up to 1 year at -20°C. The important facet of the current study is saving hard curacy. Future investigations are necessary for preparation of IgG subclasses.
An Enzyme-Catalyzed Reaction System Using Puerarin As Substrates for Horseradish Peroxidase and Its Application in Immunoassays

Abstract:An enzyme-catalyzed reaction system puerarin-horseradish peroxidase (HRP)-hydrogen peroxide, which used puerarin as substrates for HRP, was developed. In enzymatic reaction procedure, HRP catalyze the polymerization of puerarin by H2O2, and the puerarin is partly converted to dimers, a significantly fluorescent species. The increase of the fluorescence of the HRP-enzymatic reaction solution at emission of 478 nm (excitation: 315 nm) is proportional to the concentration of HRP. The enzyme-linked fluoroimmunoassay based on the principle mentioned above and competitive immunoassay model for the determination of Brucella melitensis antibodies (BrAb) was established. The results of the property investigation of puerarin demonstrate the advantages in stability and reactivity with HRP over conventional substrates such as p-hydroxyphenylacetic acid (p-HPA), Amplex red and homovanillic acid. The experimental parameters such as BrAb-HRP, temperature were optimized. The proposed method has been successfully used for analysis of BrAb in rabbit plasma sample. The linear range of determination for BrAb is 1.3 similar to 120 ng/mL with the relative standard deviation of 13.8%. The detection limit is 1.3 ng/rnL (3 sigma).

Abstract:This paper describes the combination of electrochemical immunosensor using gold nanoparticles (GNPs)/carbon nanotubes (CNTs) hybrids platform with horseradish peroxidase (HRP)-functionalized gold nanoparticle label for the sensitive detection of human IgG (HIgG) as a model protein. The GNPs/CNTs nanohybrids covered on the glass carbon electrode (GCE) constructed an effective antibody immobilization matrix and made the immobilized biomolecules hold high stability and bioactivity. Enhanced sensitivity was obtained by using bioconjugates featuring HRP labels and secondary antibodies (Ab2) linked to GNPs at high HRP/Ab2 molar ratio. The approach provided a linear response range between 0.125 and 80ng/mL with a detection limit of 40pg/mL. The immunosensor showed good precision, acceptable stability and reproducibility and could be used for the detection of HIgG in real samples, which provided a potential alternative tool for the detection of protein in clinical laboratory.

Abstract:A novel method of oriented immobilization was presented: affinity Langmuir-Blodgett (LB) technique. Firstly, a long carbon chain was bond to a ligand of Horseradish Peroxidase (HRP). The ligand derivative appears surface activity with the hydrophobic carbon chain oriented to air and the hydrophilic ligand faced to water. Then, this derivative was put onto the water/air surface to assemble a LB film and formed the affinity interaction with the active site of HRP. After that, the affinity LB film with the enzyme was transferred onto the support to obtain the oriented immobilized HRP. The specific activity of HRP immobilized by affinity LB (182.1 +/- 14 U/mg) was higher than that by adsorption (40.5 +/- 5 U/mg). HRP immobilized by affinity LB could maintain a more native conformation, compared to that by adsorption. This method could be effectively used to immobilize protein with orientation and show widely promising applications in many fields including biosensor and bioreactor. (C) 2016 Elsevier B.V. Abstract:The multi-detection size exclusion chromatography (SEC) has been recognized as an advanced analytical technique for the characterization of macromolecules and process control, as well as the manufacturing and formulation of biotechnology products. It reveals reproducible molecular characterization data, such as molecular weight and its distribution, and the size, shape, and composition of the sample peaks. The aim of this work was to investigate the potential and suitability of the multi-detection SEC as a tool for surveillance over the molecular processes during the conjugation reaction between the antibody (IgG) and horseradish peroxidase (HRP), and demonstrate the plausibility of its application in the quality control of the final product, the IgG-HRP conjugate. Guinea pig anti-Vero IgG-HRP conjugate was prepared using a modified periodate oxidation method, based on periodate oxidation of the carbohydrate side chains of HRP, followed by the formation of Schiff bases between the activated HRP and amino groups of the IgG. The quantitative molecular characterization data of the starting samples, intermediates, and final product were obtained by multi-detection SEC. Titration of the prepared conjugate was performed by the ELISA and its optimal working dilution was determined. This methodology proved to be a promising and powerful technology for the IgG-HRP conjugate process control and development, as well as for the quality control of the final product, as verified by the analysis of several commercially available reagents.

Abstract:A disposale amperometric immunosensor which is fabricated by screen printing technique for the detection of horseradish peroxidase (HRP). Three-electrode structure was designed with carbon working electrode and silver pseudo-reference electrode. The reproducibility of those transducers was assessed by electron transfer coefficient based on cyclic voltammetry in ferrocene solution. And-HR-P antibody was immobilized on the carbon electrode using glutaraldehyde. After binding with HR-P, the response of immunosensor was determined by amperometric and colorimetric methods in substrate solution containing H2O2 and o-Phenylenediamine. The results show that the limit of detection for HRP is about 10 mu g/L in amperometric method and 2 mg/L in colorimetric method.

Abstract:To generate and characterize a monoclonal antibody (mAb) against humanin (HN), BALB/c mice were immunized with the purified pet-44a-HN in adjuvant and their splenic lymphocytes were fused with myeloma SP2/0 cells. The hybridoma cell lines were screened for HN-specific antibodies by indirect enzyme-linked immunosorbent assay (ELISA), and anti-HN mAb-producing hybridoma clones were obtained using a limiting dilution assay. The specificity and affinity of the antibodies were characterized by western blot assays and indirect ELISA. Following fusion, screening and cloning, four hybridoma clones were obtained, and the clone 5A8H3 was demonstrated to stably produce anti-HN IgG2a. Further characterization of 5A8H3 revealed that this mAb specifically recognized HN, the fusion proteins of pet-44a-HN protein and pGEMEX-1-HN, but not control (Escherichia coli proteins). This mAb interacted with HN at an affinity constant (Ka) of 2.0 x 10(8)M(-1). The HN-specific IgG2a mAb was successfully generated. It interacted with HN specifically and sensitively, providing a valuable tool for further study of the biological functions of HN.

Abstract:Three hapten derivatives of Sudan I were designed. Hapten-M and hapten-T shared a similar structure, differing only in spacer length. Hapten-X was obtained by reacting with the hydroxyl of Sudan I. Then polyclonal antibodies were produced and different combinations of antibodies and coating antigens were designed. The results showed that it was not the spacer length of hapten but the hydroxyl played an indispensable role in inducing antibody production for Sudan I. More importantly, we verified the experimental results theoretically by calculating the charge distribution of atoms. Next, monoclonal antibody was prepared and the optimal combination of monoclonal antibody and coating antigen was selected. The IC50 was 0.65 ng/mL for Sudan I, and good cross-reactivity of antibody to other azo compounds was obtained. Finally, we acquired the recoveries of 82.2–116.0% and coefficients of variation of 4.3– 10.9% through the analysis of chilli sauce, tomato paste and chilli powder.

Abstract:In this study, two methods were examined to optimize the immobilization of antibodies on paper when conducting a paper-based enzyme-linked immunosorbent assay (P-ELISA). Human IgG, as a test-capture protein, was immobilized on paper via the formation of Schiff bases. Aldehyde groups were introduced onto the surface of the paper via two methods: NaIO 4 and 3-aminopropyltriethoxysilane (APTS) with glutaraldehyde (APTS-glutaraldehyde). In the assay, horseradish peroxidase-conjugated anti-human IgG (HRP-anti-IgG) binds to the immobilized human IgG, and the colorimetric reaction of 3,3′,5,5′-tetramethylbenzyzine (TMB) produces a blue color in the presence of H 2 O 2 and HRP-anti-IgG as a model analyte. The immobilization of human IgG, the enzymatic reaction conditions, and the reduction of the chemical bond between the paper surface and immobilized human IgG all were optimized in order to improve both the analytical performance and the stability. In addition, the thickness of the paper was examined to stabilize the analytical signal. Consequently, the APTS-glutaraldehyde method was superior to the NaIO 4 method in terms of sensitivity and reproducibility. Conversely, the reduction of imine to amine with NaBH 4 proved to exert only minimal influence on sensitivity and stability, although it tended to degrade reproducibility. We also found that thick paper was preferential when using P-ELISA because a rigid paper substrate prevents distortion of the paper surface that is often caused by repeated washing processes.

Abstract:We have developed a simple and sensitive competitive enzyme-linked immunosorbent assay (ELISA) to determine aflatoxin B1 (as a model small analyte) and using streptavidin-polymeric horseradish peroxidase complex (SApolyHRP) as a label for signal amplification. The performance of the assay was evaluated by comparing it with the classical indirect competitive ELISA using HRP labeled anti-mouse IgG as the tracer antibody. The results indicate that the SApolyHRP-based competitive ELISA exhibits a typically 2.4-fold steeper slope of the linear working range of the calibration curve compared to the monomeric HRP based classical ELISA, i.e., the sensitivity was increased. The SApolyHRP conjugate causes a typically 19-fold stronger signal generation in comparison to the traditional HRP labeled anti-mouse IgG at the same concentration (25 ng mL−1). Moreover, the SApolyHRP-based assay has a much wider linear range and a 3.8-fold better signal-to-noise ratio. Considering its simplicity, sensitivity and ease of operation, this competitive ELISA is considered to be a promising tool for small molecule immunodetection.

Abstract:The aim of this study was to utilize the antibody produced using Indonesia local horses (Equuscaballus) to make the conjugate of ELISA kit for detection of hepatitis B virus (HBV) surface antigen(HBsAg). The polyclonal antibodies were isolated and purified from local horses immunized repeatedlyusing isolated and purified HBsAg from Indonesia. The antibodies were conjugated with horseradishperoxidase by a modified method of Nakane and Kawaoi. The conjugate activities were performed usingthe principle of ELISA test conducted by the researchers as well as by independent laboratory.Commercial conjugate for HBsAg ELISA was used as a comparison study. The results of this studyindicated that the antibody produced from local horses can be used to make conjugates that werecomparable to commercial HBsAg ELISA kit.

Abstract:Objective To prepare universal secondary antibodies those can bind to the IgG from mice and rabbits, and use the antibodies in a variety of immunoassays. Methods The fusion genes of staphylococcal protein A (SPA), streptococcal protein G (SPG), and horseradish peroxidase (HRP) were synthesized, and cloned into the vector pcDNATM3.1 to generate the eukaryotic expression plasmids. The plasmids were transiently transfected into HEK293F cells for expression. The fusion protein expressed in the plasmid was detected by SDS-PAGE and Western blotting, and its immunoactivity was measured by Western blotting, ELISA, and immunohistochemical staining. Results Restriction enzyme digestion and gene sequencing showed the pPA-HRP, pPG-HRP, and pPA/G-HRP plasmids were successfully created. Coomassie brilliant blue staining and Western blotting indicated that the fusion proteins PA-HRP, PG-HRP, and PA/G-HRP successfully expressed in HEK293F cells. Western blotting, ELISA, and immunohistochemical staining showed that IgGs derived from mice and rabbits could be recognized and bound by the three kinds of fusion protein, of which the fusion protein PA/G-HRP exhibited the highest affinity. Conclusion The fusion protein PA/G-HRP with high and universal IgG affinity is successfully prepared. The PA/G-HRP can replace traditional secondary antibodies against mouse and rabbit IgG in a variety of immunological assays.

Abstract:A new approach based on potential control was firstly used for the immobilization of horseradish peroxidase (HRP) as the model protein. The self-assembly monolayer (SAM) was prepared with 2-aminoethanethiol (AET) on the gold electrode. The charge on HRP was adjusted by means of the acidity of the phosphate buffer solution (PBS) for dissolving the HRP. The influence of electric potential on HRP immobilization was investigated by means of colorimetric immunoassay of enzyme-substrate interaction (CIESI) using an automatic plate reader. The HRP modified electrodes were characterized with X-ray photoelectron spectroscopy (XPS) as well as atomic force microscope (AFM) on template-stripped gold surface. The potential for maximum immobilization of HRP was near the zero charge potential. The result indicates that controlled potential can affect the course of HRP immobilization without the loss of enzymic activity. It is of great significance for the control of biomolecular self-assembly and the intrinsic electric device.
Enzyme-Linked Immunosorbent Assay: Types and Applications

Abstract:Enzyme-linked immunosorbent assay (ELISA) is an immunological assay widely used in basic science research, clinical application studies, and diagnostics. The ELISA technique relies on the interaction between the antigen (i.e., the target protein) versus the primary antibody against the antigen of interest. The presence of the antigen is confirmed through the enzyme-linked antibody catalysis of the added substrate, the products of which are either qualitatively detected by visual inspection or quantitatively using readouts from either a luminometer or a spectrophotometer. ELISA techniques are broadly classified into direct, indirect, sandwich, and competitive ELISA-all of which vary based on the antigens, antibodies, substrates, and experimental conditions. Direct ELISA relies on the binding of the enzyme-conjugated primary antibodies to the antigen-coated plates. Indirect ELISA introduces enzyme-linked secondary antibodies specific to the primary antibodies bound to the antigen-coated plates. Competitive ELISA involves a competition between the sample antigen and the plate-coated antigen for the primary antibody, followed by the binding of enzyme-linked secondary antibodies. Sandwich ELISA technique includes a sample antigen introduced to the antibody-precoated plate, followed by sequential binding of detection and enzyme-linked secondary antibodies to the recognition sites on the antigen. This review describes ELISA methodology, the types of ELISA, their advantages and disadvantages, and a listing of some multifaceted applications both in clinical and research settings, including screening for drug use, pregnancy testing, diagnosing disease, detecting biomarkers, blood typing, and detecting SARS-CoV-2 that causes coronavirus disease 2019 (COVID-19).
