Abstract:With the development of commercial use of transgenic plants, ecological risk assessment on transgenic plants had been discussed, such as the likelihood of genes dispersal from transgenic plants to related wild species, the occurrence of super-weeds and the tolerance to insect-resistant transgenic plants, the influence on bio-diversity etc. This paper reviewed several major approaches of genes escape from transgenic plants. Global Patterns of Insect Resistance to Transgenic Bt Crops: The First 25 Years

Abstract:Crops genetically engineered to produce insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) have improved pest management and reduced reliance on insecticide sprays. However, evolution of practical resistance by some pests has reduced the efficacy of Bt crops. We analyzed global resistance monitoring data for 24 pest species based on the first 25 yr of cultivation of Bt crops including corn, cotton, soybean, and sugarcane. Each of the 73 cases examined represents the response of one pest species in one country to one Bt toxin produced by one or more Bt crops. The cases of practical resistance rose from 3 in 2005 to 26 in 2020. Practical resistance has been documented in some populations of 11 pest species (nine lepidopterans and two coleopterans), collectively affecting nine widely used crystalline (Cry) Bt toxins in seven countries. Conversely, 30 cases reflect no decrease in susceptibility to Bt crops in populations of 16 pest species in 10 countries. The remaining 17 cases provide early warnings of resistance, which entail genetically based decreases in susceptibility without evidence of reduced field efficacy. The early warnings involve four Cry toxins and the Bt vegetative insecticidal protein Vip3Aa. Factors expected to favor sustained susceptibility include abundant refuges of non-Bt host plants, recessive inheritance of resistance, low resistance allele frequency, fitness costs, incomplete resistance, and redundant killing by multi-toxin Bt crops. Also, sufficiently abundant refuges can overcome some unfavorable conditions for other factors. These insights may help to increase the sustainability of current and future transgenic insecticidal crops.

Abstract:Transgenic crops have revolutionized insect pest control, but evolution of resistance by pests threatens their continued success. The primary strategy for combating pest resistance to crops producing insecticidal proteins from Bacillus thuringiensis (Bt) uses refuges of non-Bt host plants to allow survival of susceptible insects. The prevailing paradigm is that refuges delay resistance that is rare and recessively inherited. However, we discovered refuges countered resistance to Bt cotton that was neither rare nor recessive. In a 15-year field study of the cotton bollworm, the frequency of a mutation conferring dominant resistance to Bt cotton surged 100-fold from 2006 to 2016 yet did not rise from 2016 to 2020. Computer simulations indicate the increased refuge percentage from 2016 to 2020 is sufficient to explain the observed halt in the evolution of resistance. The results also demonstrate the efficacy of a Bt crop can be sustained by non-Bt refuges of other crops.

Abstract:Abstract Background While the rapidly increasing global population has led to a dramatically increased demand for the agricultural production, there have been heavy economic losses owing to various pest attacks on different food crops. The advancement of various biotechnological techniques have come as a boon in addressing the global concern and leads to the development of novel varieties that have proven to be highly economical, pesticide resistant and environmentally safe. Main body The present review was aimed to update the recent developments that have taken place in the field of crop production. Major focus was laid predominantly on such genes that have demonstrated positive effects and proved to be of commercial success at the market primarily due to the development of pest-resistant transgenic food crops with expression of Bacillus thuringiensis toxins. This technology has been effective against a wide range of pests including coleopterans, lepidopterans, hemipterans, dipterans, strongylida (nematodes) and rhabditida. In similar lines various plant derived toxic proteins were also discussed along with different genes that code for insect resistant proteins such as δ -endotoxins and secreted toxins. This article also helps in understanding the structural features of the genes that are endowed with insect resistance followed by their mechanism of action on pests. Further the role of secondary metabolites in controlling the pests was addressed. The Pros and Cons of existing tools of insect pest management were demonstrated. Conclusions Novel technologies are necessary in crop improvement to progress the pace of the breeding programs, to confer insect resistance in crop plants. Therefore, the future aim of crop biotechnology is to engineer a sustainable, multi-mechanistic resistance to insect pests considering the diversity of plant responses to insect attack.

Abstract:Insecticidal transgenes, when incorporated and expressed in plants, confer resistance against insects by producing several products having insecticidal properties. Protease inhibitors, lectins, amylase inhibitors, and chitinase genes are associated with the natural defenses developed by plants to counter insect attacks. Several toxin genes are also derived from spiders and scorpions for protection against insects. Bacillus thuringiensis Berliner is a microbial source of insecticidal toxins. Several methods have facilitated the large-scale production of transgenic plants. Bt-derived cry, cyt, vip, and sip genes, plant-derived genes such as lectins, protease inhibitors, and alpha-amylase inhibitors, insect cell wall-degrading enzymes like chitinase and some proteins like arcelins, plant defensins, and ribosome-inactivating proteins have been successfully utilized to impart resistance to insects. Besides, transgenic plants expressing double-stranded RNA have been developed with enhanced resistance. However, the long-term effects of transgenes on insect resistance, the environment, and human health must be thoroughly investigated before they are made available for commercial planting. In this chapter, the present status, prospects, and future scope of transgenes for insect pest management have been summarized and discussed.

Abstract:Plants are challenged incessantly by several biotic and abiotic stresses during their entire growth period. As with other biotic stress factors, insect pests have also posed serious concerns related to yield losses due to which agricultural productivity is at stake. In plants, trait modification for crop improvement was initiated with breeding approaches followed by genetic engineering. However, stringent regulatory policies for risk assessment and lack of social acceptance for genetically modified crops worldwide have incited researchers toward alternate strategies. Genome engineering or genome editing has emerged as a new breeding technique with the ability to edit the genomes of plants, animals, microbes, and human beings. Several gene editing strategies are being executed with continuous emergence of variants. The scientific community has unraveled the utility of various editing tools from endonucleases to CRISPR/Cas in several aspects related to plant growth, development, and mitigation of stresses. The categorical focus on the development of tools and techniques including designing of binary vectors to facilitate ease in genome engineering are being pursued. Through this Review, we embark upon the conglomeration of various genome editing strategies that can be and are being used to design insect pest resistance in plants. Case studies and novel crop-based approaches that reiterate the successful use of these tools in insects as well as in plants are highlighted. Further, the Review also provides implications for the requirement of a specific regulatory framework and risk assessment of the edited crops. Genome editing toward insect pest management is here to stay, provided uncompromising efforts are made toward the identification of amiable target genes.This article has not yet been cited by other publications.

Genome evolution in an agricultural pest following adoption of transgenic crops

Abstract:Abstract Replacement of synthetic insecticides with transgenic crops for pest management has been both economically and environmentally beneficial. These benefits have often eroded as pests evolved resistance to transgenic crops, but a broad understanding of the timing and complexity of adaptive changes which lead to field-evolved resistance in pest species is lacking. Wild populations of Helicoverpa zea , a major lepidopteran crop pest and the target of transgenic Cry toxin-expressing cotton and corn, have recently evolved widespread, damaging levels of resistance. Here, we quantified patterns of genomic change in wild H. zea collected between 2002 and 2017 when adoption rates of Cry-expressing crops expanded in North America. Using a combination of genomic approaches, we identified significant temporal changes in allele frequency throughout the genomes of field-collected H. zea . Many of these changes occurred concurrently with increasingly damaging levels of resistance to Cry toxins between 2012 and 2016, in a pattern consistent with polygenic selection. Surprisingly, none of the eleven previously described Cry resistance genes showed signatures of selection in wild H. zea . Furthermore, we observed evidence of a very strong selective sweep in one region of the H. zea genome, yet this strongest change was not additively associated with Cry resistance. This first, whole genome analysis of field-collected specimens to study evolution of Cry resistance demonstrates the potential and need for a more holistic approach to examining pest adaptation to changing agricultural practices. Significance Statement Evolution of pest resistance to management approaches in agricultural landscapes is common and results in economic losses. Early detection of pest resistance evolution prior to significant crop damage would benefit the agricultural community. It has been hypothesized that new genomic approaches could track molecular signals of emerging resistance problems and trigger efforts to pre-empt widespread damage. We tested this hypothesis by quantifying genomic changes in the pest Helicoverpa zea over a 15 year period concurrent with commercialization and subsequent loss of efficacy of transgenic Bt-expressing crops. Our results demonstrate the complex nature of evolution in agricultural ecosystems and provide insight into the potential for and pitfalls associated with use of genomic approaches for resistance monitoring. We discuss approaches for improvement.

Abstract:Annually, the cost of insect pest control in agriculture crosses billions of dollars around the world. Until recently, broad-spectrum synthetic pesticides were considered as the most effective means of pest control in agriculture. However, over the years, the overreliance on pesticides has caused adverse effects on beneficial insects, human health and the environment, and has led to the development of pesticide resistant insects. There is a critical need for the development of alternative pest management strategies aiming for minimum use of pesticides and conservation of natural enemies for maintaining the ecological balance of the environment. Host plant resistance plays a vital role in integrated pest management but the development of insect-resistant varieties through conventional ways of host plant resistance takes time, and is challenging as it involves many quantitative traits positioned at various loci. Biotechnological approaches such as gene editing, gene transformation, marker-assisted selection etc. in this direction have recently opened up a new era of insect control options. These could contribute towards about exploring a much wider array of novel insecticidal genes that would otherwise be beyond the scope of conventional breeding. Biotechnological interventions can alter the gene expression level and pattern as well as the development of transgenic varieties with insecticidal genes and can improve pest management by providing access to novel molecules. This review will discuss the emerging biotechnological tools available to develop insect-resistant engineered crop genotypes with a better ability to resist the attack of insect pests.

Abstract:Insect resistant transgenic plants have been planted commercially in many countries and play an important role in integrated pest management (IPM) and creating economic benefits. However, great concerns have been raised about the potential ecological risks of transgenic plants, and increasing attentions have also been paid for the effects of transgenic plants on non-target insects, especially on natural enemies. This paper reviewed the impact of transgenic plants resistant to insects on natural enemies from both aspects of individual and population levels. So far, most reports indicated that the transgenic plants resistant to insects do not have side effect on natural enemies, but some side effects also reported. The populations of the predators and parasitoids in transgenic plants fields were equal to or higher than those in regular plant fields, except for some reports that the number of the predators changed not clearly in individual transgenic cotton field, but the population dynamic of parasitic insects decreased significantly.
Spatio-Temporal Variation in Landscape Composition May Speed Resistance Evolution of Pests to Bt Crops

Abstract:Transgenic crops that express insecticide genes from Bacillus thuringiensis (Bt) are used worldwide against moth and beetle pests. Because these engineered plants can kill over 95% of susceptible larvae, they can rapidly select for resistance. Here, we use a model for a pyramid two-toxin Bt crop to explore the consequences of spatio-temporal variation in the area of Bt crop and non-Bt refuge habitat. We show that variability over time in the proportion of suitable non-Bt breeding habitat, Q, or in the total area of Bt and suitable non-Bt habitat, K, can increase the overall rate of resistance evolution by causing short-term surges of intense selection. These surges can be exacerbated when temporal variation in Q and/or K cause high larval densities in refuges that increase density-dependent mortality; this will give resistant larvae in Bt fields a relative advantage over susceptible larvae that largely depend on refuges. We address the effects of spatio-temporal variation in a management setting for two bollworm pests of cotton, Helicoverpa armigera and H. punctigera, and field data on landscape crop distributions from Australia. Even a small proportion of Bt fields available to egg-laying females when refuges are sparse may result in high exposure to Bt for just a single generation per year and cause a surge in selection. Therefore, rapid resistance evolution can occur when Bt crops are rare rather than common in the landscape. These results highlight the need to understand spatio-temporal fluctuations in the landscape composition of Bt crops and non-Bt habitats in order to design effective resistance management strategies.

Abstract:Transgenic cotton plants producing Bacillus thuringiensis (Bt) (Berliner) insecticidal proteins have contributed to the management of key lepidopteran pests. Larval high dispersal rates between non-Bt and Bt plants in landscapes with seed contamination can speed the evolution of insect resistance to Bt plants. We evaluated the effect of Bt and non-Bt cotton plants on the larval dispersal pattern and survival of susceptible, heterozygous and Cry1F-resistant Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) genotypes in pure and contaminated artificial micro-landscape. A computer model was used to analyse the consequences of S. frugiperda larval dispersal behaviour on resistance evolution in refuge areas with different contamination levels and migration of different adult genotype combinations. The biological data from artificial micro-landscape experiments were used in simulations of macro-landscape scales. The Cry1F-resistant genotype avoided non-Bt cotton. The heterozygote had a similar larval dispersal behaviour as the susceptible genotype when non-Bt cotton was the central plant. Our simulations provide evidence that in refuge areas contaminated with Bt cotton plants, the evolution of resistance may be > 75-fold faster in relation to a contamination-free refuge. In conclusion, S. frugiperda resistance management practices on regional scales with contamination-free refuges are important to prevent loss in different crops.

Abstract:Insecticide resistance in insects severely threatens both human health and agriculture, making insecticides less compelling and valuable, leading to frequent pest management failures, rising input costs, lowering crop yields, and disastrous public health. Insecticide resistance results from multiple factors, mainly indiscriminate insecticide usage and mounted selection pressure on insect populations. Insects respond to insecticide stress at the cellular level by modest yet significant genetic propagations. Transcriptional, co-transcriptional, and post-transcriptional regulatory signals of cells in organisms regulate the intricate processes in gene expressions churning the genetic information in transcriptional units into proteins and non-coding transcripts. Upregulation of detoxification enzymes, notably cytochrome P450s (CYPs), glutathione S-transferases (GSTs), esterases [carboxyl choline esterase (CCE), carboxyl esterase (CarE)] and ATP Binding Cassettes (ABC) at the transcriptional level, modification of target sites, decreased penetration, or higher excretion of insecticides are the noted insect physiological responses. The transcriptional regulatory pathways such as AhR/ARNT, Nuclear receptors, CncC/Keap1, MAPK/CREB, and GPCR/cAMP/PKA were found to regulate the detoxification genes at the transcriptional level. Post-transcriptional changes of non-coding RNAs (ncRNAs) such as microRNAs (miRNA), long non-coding RNAs (lncRNA), and epitranscriptomics, including RNA methylation, are reported in resistant insects. Additionally, genetic modifications such as mutations in the target sites and copy number variations (CNV) are also influencing insecticide resistance. Therefore, these cellular intricacies may decrease insecticide sensitivity, altering the concentrations or activities of proteins involved in insecticide interactions or detoxification. The cellular episodes at the transcriptional and post-transcriptional levels pertinent to insecticide resistance responses in insects are extensively covered in this review. An overview of molecular mechanisms underlying these biological rhythms allows for developing alternative pest control methods to focus on insect vulnerabilities, employing reverse genetics approaches like RNA interference (RNAi) technology to silence particular resistance-related genes for sustained insect management.

Abstract:Genetically engineered plants expressing insecticidal proteins from the bacterium Bacillus thuringiensis can provide safe and effective pest control, but they will also select intensely for insect adaptation to the toxins. Mallet & Porter (Proc. R. Soc. Lond. B 250, 165 (1992)) used a model to compare two planting strategies for delaying crop failure: ‘seed mixtures’ of toxic and toxin-free plants within fields, and - 'refugia’ of toxinfree fields. They concluded that ‘if insects can move from plant to plant, seed mixtures may actually hasten insect resistance compared with pure stands of toxic plants’. In the present study, the model of Mallet & Porter was used to evaluate pure stands of toxic plants, seed mixtures, refugia, and mixtures + refugia. Across a broad range of conditions, seed mixtures delayed development of insect resistance compared with pure stands of toxic plants. The relative merits of seed mixtures and refugia depend upon untested assumptions about pest movement, mating, and inheritance of resistance. Minimizing exposure of pests to toxins in space and time is the surest way to preserve the efficacy of transgenic plants.

Abstract:SUMMARY Pests and diseases impact on crop yield and quality, and also reduce resource-use efficiency. Improved crop protection strategies to prevent such damage and loss can increase production and make a substantial contribution to food security. DNA-based technologies are likely to greatly increase the speed, sensitivity and accuracy of pest and pathogen detection and diagnosis. Rapid sequencing of nucleic acids from infected plants will aid identification of novel disease agents. Biomarkers of disease or crop damage such as volatile chemicals or blends may also be used to detect pest outbreaks. Biosensors coupled to information networks will provide real-time monitoring and surveillance of crops or stored produce and hence early warning of emerging problems and new invasive species. Challenges remain in the dissemination of new technologies and information to resource poor farmers in developing countries, although the rapid extension of the internet, mobile phones and other communication networks will provide new opportunities. Defining the genetic and molecular basis of innate plant immunity has been a major advance in plant biology with the potential to identify new targets for intervention via novel chemistry or genetic modification (GM). Identification of regulatory genes, signal molecules, pathways and networks controlling induced plant defence should lead to the development of a new generation of defence modulators, delivered either as crop protection products, or via biological agents on seeds or in the root zone. There should also be opportunities to select more responsive crop genotypes, or to develop transgenic crops tailored to respond to specific chemical cues or molecular patterns diagnostic for particular biotic threats. Sequencing of the genomes of the major crop species and their wild relatives will expand enormously the known gene pool and diversity of genetic resources available for plant breeders to access. It should be possible to identify genomic regions and genes conferring more durable, quantitative resistance to pathogens. The breeding cycle will be accelerated by high-throughput phenotyping and more efficient selection of resistance traits using within-gene markers. GM approaches will facilitate pyramiding (combining) resistance genes with different specificities and modes of action, thereby reducing the risk of directional selection for virulence. Analysis of the genomes of plant pathogens and invertebrate pests is already providing new information on genes, gene families and processes involved in host colonization and pathogenicity. Comparative genomics of species with diverse host ranges, contrasting feeding habits and different pathogenic lifestyles will identify new targets for inhibiting pest attack and aid the development of novel antimicrobial drugs and pesticides. Understanding the natural ecology of pests and pathogens, such as the factors determining host location, resource exploitation and interactions with other organisms, will improve our ability to manipulate behaviour, or exploit natural enemies or other antagonists of pest species. Volatile signals, either from natural plant sources, or engineered in transgenic crops, will be more widely used to modify pest behaviour. It may also be possible to manipulate microbial communities regulating pathogen populations and activity, and thereby recruit and retain more effective biocontrol agents. Insights into the natural diversity and activity of soil and microbial populations in the zones surrounding roots and seeds will provide new information on mechanisms of suppression regulating pest species. Fully effective interventions are unlikely, due to the complexity and diversity of the soil system, but there should be progress towards integrated control regimes combining more resistant crop genotypes (either selected or GM) with targeted management of natural suppressive processes. Harnessing new technologies and knowledge to create more durable resistant crops and sustainable disease and pest management systems will require improved understanding of the factors driving pest and pathogen adaptation and evolution. There must also be an increased emphasis on translational research and delivery, and developing strategies appropriate for lower-input production systems, if the second ‘green revolution’ is to become a reality.

Abstract:The benefits of crops that produce insecticidal toxins from Bacillus thuringiensis ( Bt ) are threatened by the emergence of insect resistance. Working with five major crop pests, Tabashnik et al . show that previously described variant Bt toxins kill pests rendered resistant to native Bt toxins by multiple mechanisms.

Abstract:Herbivorous insects are widespread and often serious constraints to crop production. The use of insect-resistant crops is a very effective way to control insect pests in agriculture, and the development of such crops can be greatly enhanced by knowledge on plant resistance mechanisms and the genes involved. Plants have evolved diverse ways to cope with insect attack that has resulted in natural variation for resistance towards herbivorous insects. Studying the molecular genetics and transcriptional background of this variation has facilitated the identification of resistance genes and processes that lead to resistance against insects. With the development of new technologies, molecular studies are not restricted to model plants anymore. This review addresses the need to exploit natural variation in resistance towards insects to increase our knowledge on resistance mechanisms and the genes involved. We will discuss how this knowledge can be exploited in breeding programmes to provide sustainable crop protection against insect pests. Additionally, we discuss the current status of genetic research on insect-resistance genes. We conclude that insect-resistance mechanisms are still unclear at the molecular level and that exploiting natural variation with novel technologies will contribute greatly to the development of insect-resistant crop varieties.
