Abstract:The basic leucine zipper (bZIP) transcription factors have crucial roles in plant stress responses. In this study, the bZIP family gene <i>SlbZIP38</i> (GenBank accession No: XM004239373) was isolated from a tomato (<i>Solanum lycopersicum</i> cv. Ailsa Craig) mature leaf cDNA library. The DNA sequence of <i>SlbZIP38</i> encodes a protein of 484 amino acids, including a highly conserved bZIP DNA-binding domain in the C-terminal region. We found that <i>SlbZIP38</i> was differentially expressed in various organs of the tomato plant and was downregulated by drought, salt stress, and abscisic acid (ABA). However, overexpression of <i>SlbZIP38</i> significantly decreased drought and salt stress tolerance in tomatoes (Ailsa Craig). The findings that <i>SlbZIP38</i> overexpression reduced the chlorophyll and free proline content in leaves but increased the malondialdehyde content may explain the reduced drought and salt tolerance observed in these lines. These results suggest that <i>SlbZIP38</i> is a negative regulator of drought and salt resistance that acts by modulating ABA signaling.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the function of the SlbZIP38 gene in tomato (Solanum lycopersicum), especially its role in drought and salt stress tolerance. Specifically: 1. **Expression pattern of SlbZIP38**: The researchers analyzed the expression pattern of SlbZIP38 under different abiotic stress conditions, including drought, salt stress, low temperature and high temperature, as well as the expression changes under different phytohormone treatments. They found that SlbZIP38 showed different expression trends under these conditions, especially down - regulated expression under drought and salt stress. 2. **Function verification of SlbZIP38**: Through over - expression experiments, the researchers found that over - expression of SlbZIP38 significantly reduced the tolerance of tomato to drought and salt stress. Further physiological index detection showed that over - expression of SlbZIP38 led to a decrease in the content of chlorophyll and free proline in leaves, while the content of malondialdehyde (MDA) increased, which explained the reason for the decrease in its stress tolerance. 3. **Role of the ABA signaling pathway**: The study also revealed the role of SlbZIP38 in the ABA signaling pathway. ABA is an important phytohormone and plays a key role in response to abiotic stress. The study found that the expression of SlbZIP38 was negatively regulated by ABA, indicating that it may affect the stress tolerance of plants by regulating the ABA signaling pathway. In summary, the main purpose of this paper is to reveal the mechanism of action of SlbZIP38 as a negative regulatory factor for drought and salt stress tolerance and to explore its function in the ABA signaling pathway. This provides a new perspective for understanding how plants respond and adapt to environmental stress and provides a potential target for further improving the stress resistance of crops. ### Key formulas and data - **Chlorophyll content calculation formula**: \[ \text{Total Chlorophyll (µg mL}^{-1}) = 20.29 A_{646} + 8.02 A_{663} \] - **Malondialdehyde (MDA) content calculation formula**: \[ \text{MDA (nmol/mL fresh weight)} = \left( \frac{A_{532} - A_{600}}{155,000} \right) \times 10^6 \] - **Relative expression level calculation method**: Calculate using the 2−∆∆Ct method, where: \[ \Delta C_t = C_t(\text{target gene}) - C_t(\text{reference gene}) \] \[ \Delta \Delta C_t = \Delta C_t(\text{treatment group}) - \Delta C_t(\text{control group}) \] \[ 2^{-\Delta \Delta C_t} \] These formulas and methods ensure the accuracy and reproducibility of experimental data.
