Abstract:With advances in nanotechnology and the development of emerging therapeutic modalities, a surge in research on the use of nanomedicines for biomedical applications has occurred over the past three decades. Carbon dots (CDs) are new members of carbon-based nanomaterials that can be used in cancer treatment to reduce side effects of drugs and improve treatment efficiency, offering new medical opportunities for further research. Urged as such, we are encouraged to classify CDs-based cancer treatments, including photodynamic therapy (PDT), photothermal therapy (PTT), sonodynamic therapy (SDT), chemodynamic therapy (CDT), chemotherapy (CT), and synergistic therapy. The advantages of these approaches are summarized, as well as ways to improve certain shortcomings. Meanwhile, this review highlights the feasible practice of CDs in cancer treatment, which is further developed and widely used in the biomedical field.

Abstract:<p>Cancer threatens the life and health of human beings, while the effective treatment of cancer is a major challenge. The design of fluorescent probe provides the strategy for developing effective therapeutic nanomaterials that can detect and identify cancer cells as well as achieve targeted and image-guided cancer therapy. Among the fluorescent nanomaterials studied, carbon dots (CDs) have attracted increasing attention owing to their most significant advantages in terms of diverse imaging features, high drug loading as well as low toxicity, and thus they have emerged as one of the most appealing candidates in the field of imaging-guided theranostics. In this review, we firstly overview the imaging analysis of cancer cells, including single-modal imaging and multi-modal imaging. After the introduction of diverse imaging features, we focus on the design of imaging-assisted drug delivery system. Finally, the current development of CDs in imaging-guided photothermal therapy (PTT) and photodynamic therapy (PDT) are further described.</p>

Abstract:Despite the rapid development of science and technology, the effective treatment of cancer still threatens human life and health. However, the success of cancer treatment is closely related to early diagnosis, identification, and effective treatment. In recent years, with the strengthening of the development and research of nanomaterials for cancer diagnosis and treatment, researchers have found that carbon dots (CDs) have the advantages of wide absorption, excellent biocompatibility, diverse imaging characteristics, and photostability and are widely used in various fields, such as sensing, imaging, and drug/gene transportation. Recently, researchers also discovered that CDs could be used as an effective photosensitizer to generate active oxygen or convert light energy into heat under the stimulation of the external lasers, making them have the effects of photothermal and photodynamic therapy for cancer. In this review, we first outline the single-modal and multimodal imaging analysis of CDs in cancer cells. After introducing diversified imaging functions, we focused on the design and the latest research progress of CDs in phototherapy and introduced in detail the strategies of CDs in phototherapy treatment and the challenges faced by clinical applications. We hope that this overview can provide important insights for researchers and accelerate the pace of research on CDs in imaging-guided phototherapy treatment.

Abstract:Carbon dots ( CDs) with unique optical properties are a stable and highly biocompatible fluorescent material offering enormous application potential in optical imaging and photothermal cancer therapy (PTT). However, the low photothermal conversion efficiency and the high power density still impede the development of CDs in PTT. Herein, the nitrogen and oxygen co- doped CDs (N-O-CDs) with strong absorbance in the near infrared (NIR) region were prepared from 1,3,6-trinitropyrene (TNP) and N-containing polymer of branched polyethylenimine (BPEI) via a one-step molecular fusion route. The asprepared black N-O-CDs with high photostability and superb biocompatibility can be utilized not only as a new fluorescence imaging agent but also as an excellent PTT agent in vivo. The structure, optical properties, bioimaging, photothermal effect in vitro, and therapeutic efficiency in vivo of the N-O-CDs are investigated. The low power density (0.8 Wcm(-2)) and high photothermal conversion efficiency (eta = 38.3%) facilitate N-O-CDs to act as an ideal theranostic agent for fluorescence imaging and photothermal therapy in vitro and in vivo. Abstract:As a new type of fluorescent agents, carbon dots (CDs) have revealed great potential in biomedical imaging applications attributed to their unique optical properties and good biocompatibility. However, engineering a versatile CDs nanoplatform integrating multifunctional imaging and therapeutic functionalities has always been an intractable challenge in targeted cancer treatment based on carbon nanomaterials. Herein, a gadolinium-doped CD conjugated with AS1411 aptamers (AS1411-Gd-CD) was prepared via facile solvothermal approach for targeted fluorescence (FL)/MR imaging guided photothermal therapy (PTT). The synthesized AS1411-Gd-CDs exhibited uniform nanostructure, good biocompatibility, strong red-fluorescence emission, significant MR contrast and efficient photothermal conversion. Benefiting from these outstanding properties, the AS1411-Gd-CDs were successfully employed in tumor-specific FL/MR dual-modal imaging in vitro and in vivo. Furthermore, under the guidance of FL/MR imaging, the AS1411-Gd-CDs demonstrated significant tumor inhibition effect owing to the outstanding photothermal effect. Thus, this study provided a versatile CDs nanoplatform as a multifunctional theranostic agent for synergistic FL/MR imaging and PTT in cancer treatment.

Abstract:Carbon nanoparticles become photoluminescent upon surface passivation with oligomeric polymer chains. In this work, the dependence of the carbon dots photoluminescent properties on the passivation polymer selection has been demonstrated by conjugating polyethylene glycol (PEG) chains, polyethylenimide-co-polyethylene glycol-co-polyethylenimide copolymer, and 4-armed PEG molecules, respectively. The cytotoxicity and cellular internalization of the resulting three types of photoluminescent nanoformulations of carbon dots, named CD2, CD3, and CD4, were evaluated. These nanoforinulations exhibited no apparent cytotoxicity on their own and were shown to successfully target cancer cells by conjugation with transferrin. The implication to the use of carbon dots as biocompatible optical nanoprobes for in vitro cancer diagnostics is discussed.

Abstract:Carbon dots (CDs), as one new class of carbon nanomaterials with various structure and extraordinary physicochemical properties, have attracted tremendous interest for their potential applications in tumor theranostics, especially in targeted bioimaging and therapy. In these areas, CDs and its derivatives have been employed as highly efficient imaging agent for photoluminescence bioimaging of tumors cells. With unique structure, optical and/or dose attention properties, CDs have been harnessed in various nanotheranostic strategies for diverse tumors through integrating with other functional nanoparticles or utilizing their inherent physical properties. Up to now, CDs have been approved as novel biomaterials by their excellent performances in precise targeted bioimaging and therapy for tumors. Herein, the latest progress in the development of CDs in targeted bioimaging and tumor therapy are reviewed. Meanwhile, the challenges and future prospects of the application of CDs in promising nanotheranostic strategies are discussed and proposed.

Abstract:Carbon dots (CDs) are nanomaterials with excellent properties, including good biocompatibility, small size, ideal photoluminescence and surface modification, and are becoming one of the most attractive nanomaterials for the imaging, detection and treatment of tumors. Based on these advantages, CDs can be combined other materials to obtain composite particles with improved, even new, performance, mainly in photothermal and photodynamic therapies. This paper reviews the research progress of CDs and their composites in targeted tumor imaging, detection, diagnosis, drug delivery and tumor killing. It also discusses and proposes the challenges and perspectives of their future applications in these fields. This review provides ideas for future applications of novel CD-based materials in the diagnosis and treatment of cancer.

Abstract:Carbon dots (CDs) are novel carbon-based nanomaterials that have been used as photosensitizer-mediated photodynamic therapy (PDT) in recent years due to their good photosensitizing activity. Photosensitizers (PSs) are main components of PDT that can produce large amounts of reactive oxygen species (ROS) when stimulated by light source, which have the advantages of low drug resistance and high therapeutic efficiency. CDs can generate ROS efficiently under irradiation and therefore have been extensively studied in disease local phototherapy. In tumor therapy, CDs can be used as PSs or PS carriers to participate in PDT and play an extremely important role. In bacterial infectious diseases, CDs exhibit high bactericidal activity as CDs are effective in disrupting bacterial cell membranes leading to bacterial death upon photoactivation. We focus on recent advances in the therapy of cancer and bacteria with CDs, and also briefly summarize the mechanisms and requirements for PSs in PDT of cancer, bacteria and other diseases. We also discuss the role CDs play in combination therapy and the potential for future applications against other pathogens.

Abstract:Carbon nanodots have drawn a great deal of attention due to their green and expedient opportunities in biological and chemical sciences. Their high fluorescence capabilities and low toxicity for living cells and tissues make them excellent imaging agents. In addition, they have a fluorimetric response against inorganic and organic species. Boron-doped carbon nanodots (B-CDs) with high fluorescence yield were produced from phenylboronic acid and glutamine as boron and carbon sources, respectively, by a hydrothermal method. First, the effects of the temperature on their fluorescence yield and the structural characteristics of B-CDs were investigated. Second, their cytotoxicity and cell death and proliferation behaviors were examined. The cytotoxicity was evaluated by the MTT assay. The cellular properties were evaluated with the distribution of caspase 3, Ki67, lamin B1, P16, and cytochrome c after the indirect immunoperoxidase technique. After the MTT assay, 1:1 dilution of all applicants for 24 h was used in the study. After immunohistochemical analyses, the application of B-CDs synthesized at 230 °C did not change control cell (Vero) proliferation, and also apoptosis was not triggered. Colo 320 CD133+ and CD133- cell-triggered apoptosis and cellular senescence were found to be synthesis temperature dependent. In addition, Colo 320 CD133- cells were affected relatively more than CD133+ cells from B-CDs. While B-CDs did not affect the control cells, the colon cancer stem cells (Colo 320 CD133+) were affected in a time-dependent manner. Therefore, the use of the synthesized B-CD product may be an alternative method for controlling or eliminating cancer stem cells in the tumor tissue.
Facile construction of carbon dots via acid catalytic hydrothermal method and their application for target imaging of cancer cells

Abstract:To solve the problem of high temperature or long reaction time in hydrothermal synthesis of carbon dots (CDs), a novel method based on the promoting carbonization by hydrochloric acid as catalysis was developed in present work. The acid catalyzed carbon dots (ACDs) were prepared facilely from tryptophan and phenylalanine at 200 °C for 2 h. In our findings, the acids could promote significantly the formation of the ACDs’ carbon core, as a result of the accelerating of the carbonization due to the easy deoxidation. The ACDs showed an average size of 4.8 nm, and consisted of high carbon crystalline core and various surface groups. The ACDs exhibited good optical properties and pH-dependent photoluminescence (PL) intensities. Furthermore, the ACDs were safe and biocompatible. The experimental results demonstrated that such new ACDs were connected with DNA-aptamer by EDC/NHS reaction maintaining both the bright fluorescence and recognizing ability on the cancer cells, which so could be served as an effective PL sensing platform. The resultant DNA-aptamer with ACDs (DNA-ACDs) could stick to human breast cancer cells (MCF-7) specifically, and exhibited high sensitivity and selectivity, indicating the potential applications in the cancer cells targeted imaging fields.

Abstract:Photosensitizers are light-sensitive molecules that are highly hydrophobic, which poses a challenge to their use for photodynamic therapy. Hence, considerable efforts have been made to develop carriers for the delivery of PSs. Herein, we synthesized a new theranostic nanoagent (CQDs@PtPor) through the electrostatic interaction between the tetraplatinated porphyrin complex (PtPor) and the negatively charged CQDs. The size and morphology of as-prepared CQDs and CQDs@PtPor were characterized by a series of methods, such as XRD, TEM, XPS, and FTIR spectroscopy. The CQDs@PtPor composite integrates the optical properties of CQDs and the anticancer function of porphyrin into a single unit. The spectral results suggested the effective resonance energy transfer from CQDs to PtPor in the CQDs@PtPor composite. Impressively, the CQDs@PtPor composite showed the stronger PDT effect than that of organic molecular PtPor, suggesting that CQDs@PtPor is advantageous over the conventional formulation, attributable to the enhanced efficiency of 1O2 production of PtPor by CQDs. Thus, this CQDs-based drug nanocarrier exhibited enhanced tumor-inhibition efficacy as well as low side effects in vitro, showing significant application potential in the cancer therapy.

Abstract:Natural products have been an important database for anti-cancer drug development. However, low water solubility and poor biocompatibility limit the efficacy of natural products. Carbon dots (CDs), as an emerging 0D material, have unique properties in bioimaging, water solubility and biocompatibility. Here, we prepared three pentacyclic triterpenoids (PTs) included glycyrrhetinic acid (GA), ursolic acid (UA) and oleanolic acid (OA), which have anticancer activity but poor water solubility, as raw materials into CDs to improve disadvantages. Our data indicated that the active surface groups of all three CDs were largely preserved and were able to excite green fluorescence. Their carboxyl edges not only exhibited excellent water solubility, but also specifically targeted tumor cell mitochondria due to high sensitivity to ROS-induced damage and high internal oxidative stress. In cancer cells, the PT-CDs induced cell death through three pathways (apoptosis, ferroptosis, and autophagy), which is essentially the same way their raw materials induce death, but the effect was much stronger than raw materials. Notably, functionalized PT-CDs also exhibited extremely low toxicity. In summary, PT-CDs not only have improved water solubility and biocompatibility, but also retain the structure of their raw materials well and exert better efficacy, which provides new ideas for the development of anti-cancer natural product drugs.

Abstract:Carbon dots (CDs) from glucose were synthesized using two of the most common bottom-up methods, namely, microwave assisted (MW) and hydrothermal carbonization (HT). Synthetic parameters such as reaction time, temperature, and precursor concentration were changed to study the effects of each parameter on CD size, structure, surface functionalities, charge, photoluminescence behavior, quantum yield, cytotoxicity, blood–brain barrier (BBB) crossing ability and bioimaging. A detailed analysis is performed to compare the structure and properties of the CDs synthesized in ten different conditions. We show that the synthesis route drastically changes the structure, properties, and related functions of glucose-derived CDs yielding two different subtypes of CDs. Surprisingly, CDs that was synthesized via HT method showed specific anticancer activity against a neuroblastoma cell line while being non-toxic towards healthy cell lines, indicating significant potential for therapeutic applications. CDs synthesized via MW crosses the BBB in zebrafish and rat models, and accumulates in neurons. CDs synthesized via MW method showed high biocompatibility and a great potential to be used for bioimaging applications in vitro and in vivo targeting neurons. Finally, a formation mechanism of CDs is proposed for both HT and MW synthesis routes.

Abstract:Carbon Dots (CDs) are the latest members of carbon-based nanomaterials, which since their discovery have attracted notable attention due to their chemical and mechanical properties, brilliant fluorescence, high photostability, and good biocompatibility. Together with the ease and affordable preparation costs, these intrinsic features make CDs the most promising nanomaterials for multiple applications in the biological field, such as bioimaging, biotherapy, and gene/drug delivery. This review will illustrate the most recent applications of CDs in the biomedical field, focusing on their biocompatibility, fluorescence, low cytotoxicity, cellular uptake, and theranostic properties to highlight above all their usefulness as a promising tool for cancer diagnosis and therapy.

Abstract:Fluorescent carbon dots (CDs) have aroused great interest in the nanomedicine due to their excellent chemical stability and biological compatibility. However, carbon dots-based nanoplatform integrated with targeted cancer imaging and responsive drug delivery are still challenging. Herein, we synthesized strongly green-fluorescent, cancer-cell-targeted and cationic carbon dots (HP-CDs) via hydrothermal method, by using hyaluronic acid (HA) and polyetherimide together as the starting materials. The nanoprobe could be specifically internalized with cancer cells, due to the recognition of surface HA with highly-expressed CD44 receptor. Ferrocenylseleno-dopamine (FcDA) was particularly designed as new generation drug of Fenton-like reagents, assembled on the surface of HP-CDs to obtain CDs@FcDA nanoprobe. In CDs@FcDA nanoprobe, FcDA significantly quenched CDs fluorescence due to photo-induced electron transfer. Upon the cellular specific uptake, ferrocene moiety of the nanoprobe would be oxidized into ferrocenium cation by intracellular H2O2 in a typical Fenton-like reaction, which resulted in charge reversal and drug release from HP-CDs surface. The quenched nanoprobe fluorescence was thus recovered and achieved the cancer-cell-specific imaging. Meantime, the Fenton chemistry produced toxic •OH, leading to the cancer cell apoptosis. Importantly, the composites of CDs@FcDA exhibited lower IC50 value than that of free FcDA for HeLa cell lines, indicating greater therapeutic efficiency of the drug nanoplatform. Moreover, oxidized ferrocenium could react with endogenous glutathione (GSH) and be reversibly restored to the reduced ferrocene, which formed a Fenton-chemistry cycle regulated by H2O2 and GSH in cancer microenvironment. Therefore, the HA-functionalized CDs@FcDA nanoprobe proposed in this work represents an attractive tool for redox-gated imaging and cancer-cell-responsive drug delivery.

Advances in the application of biosynthesized carbon dots as fluorescent probes for bioimaging

Abstract:Abstract Carbon dots (CDs) are emerging as versatile fluorescent nanoprobes for bioimaging applications due to advantages like tunable emissions, excellent biocompatibility, facile surface functionalization, and ease of synthesis. This review summarizes recent advances in applying biosynthesized CDs for sensitive bioimaging. CDs derived from sustainable biomass sources through green techniques like hydrothermal and microwave synthesis demonstrate bright, excitation-tunable photoluminescence spanning visible to near-infrared spectra. Careful control of synthesis parameters and surface passivation strategies enhance quantum yields above 50% comparable to toxic semiconductor dots. Conjugation with polymers, peptides, and recognition elements like antibodies impart solubility and selectivity towards cancer cells and biomarkers. In vitro validation in standard lines shows targeted organelle imaging abilities. In vivo administration reveals renal clearance pharmacokinetics with preferential tumor accumulation via enhanced permeability effects. Average tumor growth inhibition around 50-80% was achieved in mouse xenografts using CDs-drug formulations through combined therapeutic effects of chemotherapy and photothermal ablation under imaging guidance. However, concerns regarding toxicity from chronic exposures, large-scale reproducible manufacturing, and multimodal imaging capabilities need redressal prior to further clinical translation.

Abstract:As an emerging fluorescent nanomaterial, carbon dots (CDs) exhibit many attractive physicochemical features, including excellent photoluminescence properties, good biocompatibility, low toxicity and the ability to maintain the unique properties of the raw material. Therefore, CDs have been intensively pursued for a wide range of applications, such as bioimaging, drug delivery, biosensors and antibacterial agents. In this review, we systematically summarize the synthesis methods of these CDs, their photoluminescence mechanisms, and the approaches for enhancing their fluorescence properties. Particularly, we summarize the recent research on the synthesis of CDs from drug molecules as raw materials and introduce the representative application aspects of these fascinating CDs. Finally, we look into the future direction of CDs in the biomedical field and discuss the challenges encountered in the current development.
Carbon Dots Derived from Tea Polyphenols as Photosensitizers for Photodynamic Therapy

Abstract:Photodynamic therapy (PDT) has become an emerging cancer treatment method. Choosing the photosensitizer (PS) compounds is one of the essential factors that can influence the PDT effect and action. Carbon dots (CDs) have shown great potential as photosensitizers in PDT of cancers due to their excellent biocompatibility and high generation of reactive oxygen species (ROS). Here, we used tea polyphenol as raw material for synthesized tea polyphenol carbon dots (T−CDs) that show dual emission bands of red and blue fluorescence and can efficiently generate hydroxyl radicals (OH) under mildly visible irradiation with a LED light (400–500 nm, 15 mW cm−2). The extremely low cytotoxicity and excellent biocompatibility of T−CDs without light irradiation were tested using MTT and hemolytic assay. Further, T−CDs have been shown by in vivo experiments, using a mouse breast cancer cell line (4T1) subcutaneously injected in the back of the mouse buttock as a model, to effectively inhibit the tumor cell proliferation in solid tumors and show an excellent PDT effect. In addition, pathological sections of the mice tissues after further treatment showed that the T−CDs had no apparent impact on the major organs of the mice and did not produce any side effect lesions. This work demonstrates that the as−synthesized T−CDs has the potential to be used as a PS in cancer treatment.
