Drug and Chemical Toxicology
Abstract:The toxic effects of ACR monomer include carcinogenesis, cellular genotoxic, and neurotoxicity. In this study, we examined the effect of acrylamide on biochemical and hematologic parameters in Wistar rats and explored the renal and hepatic function of these animals through a complementary anatomopathologic study. For it, thirty female Wistar rats aged 4 weeks and weighing 100 ± 10 g were housed six animals per cage and divided as follows: two groups were exposed for 2 months to drinking water containing 5 mg (Group 2) or 10 mg acrylamide (Group 3); one group of 12 rats received the median lethal dose of acrylamide by gavage (Group 4); and the control group (Group 1) received pure water. The results clearly showed that acrylamide affects various biochemical parameters, such as creatinine, urea, and serum globulin levels and the lipid balance, which are directly related to renal and hepatic dysfunction and disruption of the hematologic system. In addition, the data revealed changes in the complete blood count (CBC); significant increases in the number of leukocytes (9.95 ± 1.44 and 10.44 ± 1.21) and lymphocytes (6.11 ± 0.48 and 6.33 ± 0.76) in Groups 3 and 4, respectively; and decreases in total protein (88.95 ± 6.36), albumin (37.65 ± 1.65) and α-1 globulin levels (24.84 ± 2.10) in Group 3. The anatomopathologic study confirmed liver damage in the animals administered an acrylamide containing diet compared with those in the control group. The present study confirmed the effects of acrylamide on different hematologic, biochemical and immunologic parameters, with a specific focus on the liver and kidney, and on the induction of neurotoxic disorders. The results showed that oral exposure to acrylamide via drinking water or gavage induces kidney damage, hepatocellular insufficiency and chronic liver disease, resulting in primary immunodeficiency and activation of the immune system following the possible expression of certain immunoreaction genes.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the effects of oral acrylamide (ACR) on the biochemical and hematological parameters of Wistar rats, and to explore the renal and liver functions of these animals through supplementary anatomopathological studies. Specifically, the authors hope to understand whether acrylamide will have an impact on the following aspects: 1. **Biochemical parameters**: such as creatinine, urea, serum globulin levels and lipid balance, etc. These parameters are directly related to renal and liver functions. 2. **Hematological parameters**: including complete blood cell count (CBC), especially the changes of white blood cells (leukocytes), lymphocytes, red blood cells (RBCs) and other blood cells. 3. **Immune system**: evaluate the potential impact of acrylamide on the immune system, such as the expression of immune response genes. In addition, this study also pays special attention to the effects of acrylamide on the liver and kidneys, as well as the possibility of inducing neurotoxic disorders. Through experiments, researchers hope to reveal the toxicological effects of acrylamide at different doses, so as to provide a scientific basis for preventing its toxicity. ### Research background Acrylamide is a transparent, odorless, water - soluble ethylene compound, which is widely used in chemical engineering, water treatment, construction and other fields. In recent years, due to the discovery that acrylamide may be formed in high - temperature - cooked foods, this problem has attracted wide attention. Acrylamide has been recognized as a carcinogen and poses a potential risk to human health. Therefore, it is of great significance to study its specific effects on organisms. ### Experimental design The researchers used 30 female Wistar rats aged 4 weeks, with a body weight of about 100 ± 10 grams. These rats were divided into four groups: - **Control group (Group 1)**: drank pure water. - **Low - dose group (Group 2)**: drank water containing 5 mg/L acrylamide. - **High - dose group (Group 3)**: drank water containing 10 mg/L acrylamide. - **Acute exposure group (Group 4)**: acrylamide was given through a gastric tube at a half - lethal dose (LD50, 150 mg/kg body weight). The experiment lasted for two months. During this period, the body weight changes of the rats were monitored, and blood sampling and organ dissection were carried out at the end of the experiment to evaluate the changes of biochemical and hematological parameters. ### Main results The experimental results showed that acrylamide significantly affected a variety of biochemical parameters, such as creatinine, urea and serum globulin levels, as well as lipid balance. Hematological examinations showed that the numbers of white blood cells and lymphocytes increased significantly, while the levels of total protein, albumin and α - 1 globulin decreased. Anatomopathological studies confirmed that liver damage occurred in rats treated with acrylamide. ### Conclusion This study shows that oral acrylamide through drinking water or gastric tube administration can cause kidney damage, hepatocyte dysfunction and chronic liver disease, leading to primary immunodeficiency and activation of the immune system. This provides important information for further understanding the toxicity mechanism of acrylamide and provides a scientific basis for preventing its adverse health effects. ### Formula summary The formulas involved in this paper are mainly concentrated in the statistical analysis part, such as analysis of variance (ANOVA) and principal component analysis (PCA). The following is the Markdown - format representation of these methods: ```markdown ### Analysis of variance (ANOVA) \[ \text{ANOVA}=\frac{\text{Group sum of squares}}{\text{Within - group sum of squares}} \] ### Principal component analysis (PCA) \[ \text{PCA}=\text{Principal component score matrix}\times\text{Feature vector matrix} \] ``` These formulas help to explain the statistical processing methods of experimental data.
