Abstract:Polysaccharide is the main active compound of Lilium, and showed many activities, such as hypoglycemic, antioxidant, immune-modulatory. There are three types' Lilium in China market, i.e. Lilium lancifolium Thunb (JD), Lilium davidiivar. Unicolor Salisb (L. davidii var)(LZBH), and Lilium brownii F.E. Brown var. viridulum Baker (BH). Near infrared spectroscopy (NIR) technique has become popular in the fields of quality control, due to its advantages, such as fast, non-destructive, and can detect several ingredients, simultaneously. In this study, a classification model was established based on NIR technique and random forest method to accurately distinguish three types' Lilium species, and the classification accuracy reached 94.37%. Furthermore, taking the effects of neighbor wavelength into account, a new weighted partial least square algorithm was proposed to establish an accurate and quantitative model for predicting the polysaccharide contents of these samples. In the model establishing process, some signal pre-treatment methods were optimized, and the validation results with highest determination coefficient (R2) and low root mean square errors of prediction (RMSEP) were, 0.9455 and 0.9098, respectively. The obtained results showed that combined NIR technique with chemometrics was an effective and green method for quality control.
Purification, partial characterization and anti-inflammatory characteristics of lotus (Nelumbo nucifera Gaertn) plumule polysaccharides

Abstract:A novel lotus plumule polysaccharide (LPPS) was purified, characterised and cultured with RAW264.7 macrophages to evaluate its anti-inflammatory characteristics. LPPS was purified using Sepharose 6B gel filtration and dissolved into two major components, fraction-1 (F1) and fraction-2 (F2). The molecular weights of native F1 and F2 were approximately distributed at >2,000 and 25.7kDa, respectively. The total protein and carbohydrate constituent ratios in LPPS, F1, and F2 were 30.0±0.9% vs. 70.0±0.9%, 30.1±2.6% vs. 69.9±2.6%, and 96.5±6.1% vs. 3.5±6.1% (w/w), respectively, suggesting that F1 may be a major proteo-polysaccharide component and F2 a glycoprotein constituent in LPPS. Pro-/anti-inflammatory (IL-6/IL-10) cytokine secretion ratios by lipopolysaccharide-stimulated RAW264.7 macrophages were significantly decreased by F1 and F2 treatments, particularly by F2, in a dose-dependent manner under a preventive experimental model. This study suggests that purified components, F1 and F2 from LPPS, have strong anti-inflammatory effects on LPS-induced inflamed macrophages in a preventive manner.

Abstract:The in vitro activities of 39 polysaccharides from different parts of 13 lotus root varieties were evaluated and introduced into multiple linear regression analysis to explore the structure-activity relationships using their chromatographic fingerprint features as independent variables. Their 2,2-diphenyl-1-picrylhydrazyl/hydroxyl radical scavenging abilities, ferric reducing antioxidant powers and growth-inhibitory effects against HepG2 and SGC7901 cancer cells were all diverse, with the variable-coefficients ranging from 24.49% to 87.76%, while their macrophage immunostimulatory activities evaluated by nitric oxide production and tumor necrosis factor-alpha secretion showed relatively low variations. Lotus root polysaccharides (LRPs) from the peels and nodes possessed stronger activities than those from the fleshes. Their fingerprint-activity relationship models indicated that monosaccharide composition was closely related to the activities, but not molecular weight. LRPs have health-improving potentials, and their activities can be partly predicted by the quantitative fingerprint-activity relationship model.

Abstract:Lotus seed kernel (LSK) has high starch content and possesses many bioactivities, which are mainly attributed to its phenolics. In the present study, LSK phenolics were extracted using aqueous glycerol, and the obtained phenolics and its starch properties were observed for their correlations. Response surface methodology indicated significant quadratic effect of temperature and significant linear effect of glycerol on LSK phenolics (p < 0.05). Temperature also had a significant effect on LSK starch solubility and swelling power and % leaching amylose (p < 0.05). Leaching amylose in the presence of glycerol and swelling power showed positive correlations with LSK phenolics in a linear relationship (r = 0.825, p < 0.01) below 50 degrees C and a monotonic relationship (r(s) = 0.933, p < 0.01) above 60 degrees C, respectively. A building block backbone model was adopted to illustrate the interaction between amylose/amylopectin-glycerol, and most importantly, their relationship to LSK phenolics.
Extractation and Characteristics of Polysaccharides from Lotus Root Pomace

Abstract:For comprehensive use of the pomace of lotus roots, water-soluble polysaccharides and insoluble dietary fibre were extracted and their characteristics were studied as well. Polysaccharides were obtained by hot-water extraction, ethanol precipitation, sevag method removing protein and then purificated by DEAE Sepharose Fast-flow, and NPh1 and NPh2 were fractioned. HPLC showed NPh2 was a pure acid polysaccharide with relative molecule weight more than 2000 kD that mainly composed of galactose, arabinose, rhamnose, glucose, fucose and xylose with molar ratio of 26.74: 16.17: 5.69: 5.49: 2.31:1. Dietary fibre being gained by alkaline hydrolyzation and decolored by H2O2, the yield was 26.97% and its water-holding capability and swelling power were 7.58 g/g and 9.0 ml/g, respectively. Results showed that lotus roots pomace can be a good source of water-soluble polysaccharides and insoluble dietary fibre.

Abstract:Chemical fingerprinting has been widely used for the quality control of complex natural products including traditional Chinese medicines and botanical drugs. However, there is still lack of appropriate method to monitor the batch-to-batch consistency of hydrophilic constituents, such as saccharides and oligosaccharides. In the present study, we developed a novel approach based on high performance liquid chromatography with evaporative light scattering detector(HPLC-ELSD) for establishing saccharide fingerprinting of Danhong Injection(DHI), which is a widely used botanical drug for treating cardiovascular diseases. Major saccharides in DHI were isolated and four compounds including fructose, glucose and two oligosaccharides were identified. The structures of two novel saccharides named glycerol-1-O-galactfpyranosyl-(1→4)-O-arabinofuranoside, and glycerol-1-O-galactpyranosyl-(1→4) [O-arabinfuranosyl-(1→3)]-O-arabinofuranoside were confirmed by NMR, HR-ESI-MS and GC-MS for the first time. The establishment approach was successfully applied to distinguishing 12 batches of DHI from other botanical drugs with the aid of principle component analysis as well as to evaluating batch-to-batch consistency with the help of calculating similarity of fingerprints. Our findings indicate that the chemical fingerprint of saccharides can be a useful tool for the quality control of complex natural products.

Abstract:The lotus (Nelumbo nucifera Gaertn.) is the largest aquatic vegetable in Asia. The lotus seedpod (LS) is an inedible part of the mature flower receptacle of the lotus plant. However, the polysaccharide isolated from the receptacle has been less studied. The purification of LS resulted in two polysaccharides (LSP-1 and LSP-2). Both polysaccharides were found to be medium-sized HG pectin, with a Mw of 74 kDa. Their structures were elucidated via GC-MS and NMR spectrum and proposed as the repeating sugar units of GalA connected via α-1,4-glycosidic linkage, with LSP-1 having a higher degree of esterification. They have certain content of antioxidant and immunomodulatory activities. The esterification of HG pectin would have an adverse effect on these activities. Furthermore, the degradation pattern and kinetics of LSPs by pectinase conformed to the Michaelis-Menten model. There is a large amount of LS, resulting from the by-product of locus seed production, and thus a promising source for the isolation of the polysaccharide. The findings of the structure, bioactivities, and degradation property provide the chemical basis for their applications in the food and pharmaceutical industries.

Abstract:Lotus flower polysaccharide (LFP) and lotus seedpod polysaccharide (LSP) were separated by water extract–alcohol precipitation, and their structures and biological activities were investigated. The results of monosaccharide composition showed that LFP and LSP were composed of nine monosaccharides, fucose, rhamnose, arabinose, glucose, galactose, mannose, fructose, galacturonic acid, and glucuronic acid, with the molar percentages of 0.18: 0.43: 2.26: 45.22: 32.14: 4.28: 8.20: 6.28: 1.01 and 2.70: 1.02: 8.15: 45.63: 20.63: 1.44: 2.59: 16.45. LSP and LFP exhibited molecular weights of 9.37 × 104 Da and 1.24 × 106 Da, respectively. SEM showed that LFP and LSP have similar structures; XRD analysis showed that both polysaccharides had crystalline structure and amorphous structure. The results of ABTS+, DPPH, hydroxyl radical scavenging experiment, and a reducing power experiment showed that LFP and LSP had good antioxidant capacity. Cell viability findings showed that polysaccharide concentrations of lotus flower and lotus seedpod could enhance cellular proliferation ranging from 25 to 400 μg/mL without cytotoxicity. By inducing the production of crucial proteins in the TLR4/NF-κB pathway, LFP and LSP were able to induce autophagy in RAW264.7, according to the results of the RT-PCR and Western blotting assays.

Abstract:Starch extracted from ten lotus root varieties (Yeyong, MRHZ, Zhonghua, Suiningbaiou, Zaohua, Meirenhong, E9:11, Huchengyeou, Peixianyeou, Xin No. 5) from Jiangsu Province, China, were studied. The structural characteristics of lotus root starch were analyzed based on its physical and chemical properties, particle structure, crystal structure, layered structure, thermal properties and gel properties. The lotus root starches were generally milky white, with a starch content ranging from 81.72 to 99.54%, amylose content ranging from 15.44 to 23.54%, light transmittance between 15.30 and 26.90%, and swelling power and solubility at 22.15–29.72% and 2.00–6.00%, respectively. Among the varieties, “Suiningbaiou” had the whitest starch, while “Meirenhong” and “Suiningbaiou” had higher starch contents. “Huchengyeou” had the lowest amylose content, and “Zhonghua” showed the highest light transmittance. “Yeyong” and “Zaohua” had higher solubility, whereas “Zhonghua” had the highest swelling power. An optical, polarizing and scanning electron microscope revealed that most of the lotus starch was oval. However, starch from “Zhonghua”, “E9:11”, “Peixianyeou” and “Zaohua” was longer, whereas starch from “Meirenhong”, “MRHZ”, “Huchengyeou”, “Yeyong”, “Suiningbaiou” and “Xin No.5” was shorter. The X-ray powder diffraction patterns and 13C NMR spectroscopy indicated that “Zaohua” starch exhibited type A crystallization, while starch from the other varieties displayed type CA crystallization, with a relative crystallinity between 18.27 and 22.10%; “Huchengyeou” and “Yeyong” had higher crystallinity. Infrared spectroscopy and small-angle X diffraction analysis showed similar results for short-range order and lamella spacing. The thermal and gel properties of starch from the ten lotus root varieties also varied. The enthalpy change values of “Xin No.5” and “Yeyong” were higher, and starch from “Yeyong” and “Huchengyeou” showed a greater gel strength. Finally, factor score analysis ranked the starch quality of the top four lotus root varieties in Jiangsu as “Suiningbaiou”, “Yeyong”, “Huchengyeou”, and “Zaohua”.

Abstract:Polysaccharide was one of the considered major active ingredient in Polygonatum odoratum which was crucial for its quality evaluation. In this study, High performance liquid chromatography (HPLC) combined with chemometrics methods were performed to assess the quality of P. odoratum polysaccharide (POP) harvested from different locations. The methodology validation and similarity evaluation results showed that the analysis method was able to meet the requirement of fingerprint analysis, and 10 batches of POPs had a high degree of similarity based on the similarity values were greater than 0.960. The results of hierarchical cluster analysis (HCA) showed that different regions POPs could be classified by clustering analysis based on their nuances. The results of principal component analysis (PCA) showed that the mannose (58.13%∼78.18%) and glucuronic acid (2.36%∼11.72%) could be selected as herb markers for the quality control of P. odoratum . Abstract:In this paper, a sequential gas-liquid chromatography and mass spectrometry route was proposed for characterization of polysaccharides in Panax ginseng (PG), P. notoginseng (PN), and P. quinquefolius (PQ). Due to the reflection of stepped structure parameters, the resulting integrative profiles were tentatively defined as structural-fingerprinting of polysaccharides (SFP) with monosaccharide compositional fingerprinting (MCF), Smith degradation and non-degradation fingerprinting (SDF and SNF), and oligosaccharide compositional fingerprinting (OCF). The MCF, OCF and SDF did not allow for visual discrimination of the three species due to the high interspecific similarity of PG and PQ, whereas SNF could intuitively distinguish PG, PN, and PQ by the presence or absence of Rha and the peak area ratio of Glc/Gal. Similarity analysis, heatmap analysis and principal component analysis were further performed to discern three Panax species based on SNF data sets. The linear →4)-Hexp-(1 → structures were clearly identified as the common structural backbones in side chains or smooth regions of the main chain in PPG, PPN, and PPQ using HILIC-UHPLC-ESI--MS/MS for characterization of partial acid hydrolyzates. Multiple-fingerprint analysis of Poria cocos polysaccharide by HPLC combined with chemometrics methods

Abstract:In this study, the multiple fingerprints, which were integrated with HPGFC-ELSD (high performance gel filtration chromatography - evaporative light scattering detector) fingerprint, PMP-HPLC-DAD (1-phenyl-3-methyl-5-pyrazolone-high performance liquid chromatography - diode array detector) fingerprint of complete acid hydrolysates and HILIC-HPLC-ELSD (hydrophilic interaction - high performance liquid chromatography - evaporative light scattering detector) fingerprint of enzyme hydrolysates, were established to evaluate the quality of polysaccharides from Poria cocos (PCPs). The similarity evaluation showed that 16 batches of PCPs from different origins had high similarity in structural characteristics based on the multiple fingerprints. The chromatographic data of multiple fingerprints of PCPs were fused, processed and analyzed by chemometric methods including HCA (hierarchical cluster analysis), PCA (principal component analysis) and PLS-DA (partial least squares discriminant analysis). The 16 batches of PCPs were divided into 3 categories in PCA, indicating a certain relationship between the structural characteristics and the origins. PLS-DA analysis indicated that Man, Glc, Gal, Fuc, the components with m/z of 2.22 × 104∼1.53 × 104 Da and 3.46 × 103∼2.69 × 103 Da, oligosaccharides with DPs of 6 and 7, respectively, could be regarded as potential chemical markers for the classification of PCPs from different origins. According to the multiple fingerprints and chemometric analysis, the two commercial samples were proved to be adulterants.

Abstract:In this study, we extracted lotus root polysaccharide (LRP) and synthesized carboxymethylated lotus root polysaccharide (CM-LRP) using response surface methodology (RSM). The monosaccharide component of LRP was analyzed by pre-column derivatization high performance liquid chromatography (PCD-HPLC). The polysaccharide was characterized by ultraviolet (UV) spectroscopy scan, Fourier transform infrared spectroscopy (FTIR), circular dichroism spectrometer (CD), and scanning electron microscopy (SEM). It was found that the main monosaccharide component of LRP was glucose, accounting for more than 98% of the total polysaccharide component. RSM analyses revealed that the optimal conditions for CM-LRP synthesis were a reaction duration of 2.22 h, the chloroacetic acid dosage is 2.02 g, and a temperature of 46.87 °C. Under these conditions, the predicted degree of substitution (DS) was determined to be 0.5101. The superoxide anion, ferrous ion and hydroxyl radical antioxidant system were constructed to study the antioxidant activity of LRP and CMLRP. The activity of CMLRP to remove ferrous ions and hydroxyl radicals were significantly stronger than that of LRP, and it also showed a certain concentration-dependent. LRP has a better scavenging ability than CMLRP in scavenging superoxide anion free radicals. Our data revealed that CM-LRP is a promising natural antioxidant with potential value as a food supplement.

Abstract:Few studies reported the quality evaluation and gut microbiota regulation effect of polysaccharides from Frit-illaria species. In this study, polysaccharides extracted from ten Fritillaria species were compared and distin-guished through multi-levels evaluation strategy and data fusion. Furthermore, the gut microbiota regulation effect of polysaccharides among different species was analyzed and evaluated. The fingerprint profiling of IR, molecular weight distribution of polysaccharides, chromatogram of partially hydrolyzed polysaccharides (oli-gosaccharides) and completely hydrolyzed polysaccharides (monosaccharides) were similar, and no exclusive signals were observed. However, the signal strength of functional group, oligosaccharides abundance and monosaccharides proportion showed obvious differences in inter-and intra-species. Glucan may be the main component of polysaccharides in Fritillaria species, CIRR derived from CIR, PRZ, DEL, TAI, UNI possessed higher total polysaccharides content, polymerization degree, oligosaccharides abundance (DP 2-4), and glucose content than the others. Meanwhile, data fusion model was established for identification of affinis and multi-original species, the accuracy of which proved to be 100 %. In addition, Fritillaria polysaccharides could increase the bacterial community richness and diversity, regulate the gut microbiota composition and possessed potential therapeutic effects on gastrointestinal diseases and nervous system diseases.
Evaluation of Pre-Harvest Nutrient Composition and Functional Active Substances in Various Lotus Roots

Abstract:This study investigated the impact of variety and harvest time on the visual appearance, nutritional quality, and functional active substances of six lotus root cultivars: "Xinsanwu", "Wuzhi No. 2", "Baiyuzhan", "Huaqilian", "Elian No. 6", and "Elian No. 5". Samples were collected monthly from December 2023 to April 2024. A nutrient analysis revealed a decrease in the water content with a delayed harvest. The total soluble solids and soluble sugar content peaked towards the end and middle-to-late harvest periods, respectively. Starch levels initially increased before declining, while the soluble protein exhibited a triphasic trend with an initial rise, a dip, and a final increase. The vitamin C (Vc) content varied across cultivars. Functional active substances displayed dynamic changes. The total phenolics initially decreased, then increased, before ultimately declining again. The total flavonoid content varied by both cultivar and harvest time. The phenolic acid and flavonoid content mirrored the trends observed for total phenolics and total flavonoids. Gastrodin was the most abundant non-flavonoid compound across all varieties. "Wuzhi No. 2" and "Baiyuzhan" displayed higher levels of functional active substances and starch, while the Elian series and "Xinsanwu" cultivar exhibited a greater content of Vc, soluble sugar, and soluble protein. Specific harvest periods yielded optimal results: "Wuzhi No. 2" (H1 and H5), "Huaqilian" (H2), and "Baiyuzhan" (H3 and H4) demonstrated a high nutrient and functional active substance content. Overall, the lotus roots harvested in period H4 achieved the highest score. Overall, this study provides the foothold for the rapid identification of superior lotus root cultivars and the valorization of lotus root by-products via advanced processing methods. Additionally, it offers valuable insights for market participants and consumers to select optimal varieties and harvest times based on their specific needs.

Abstract:The current study sought to depict the structural feature of polysaccharides extracted from Na 2 CO 3 unextractable fraction (LUN) of lotus rhizome using galactosidase with assistance of ball milling. The extracted polysaccharides were a complex of cellulose microfibrils and the RG-I structural domain of pectin, and the top three monosaccharides were glucose, galactose and galactose uronic acid, which allowed to tune the properties of the enzyme-hydrolyzed polysaccharide from LUN after 15 and 45 min of ball milling. The data of XRD revealed that pectin has a masking effect on the diffraction peaks of cellulose components. The removing of the polysaccharides could increase the degree of crystallinity and the pectin-cellulose interaction mainly occured through the galactan side chain was speculated. Textural characterization by SEM exhibited a cross-linked rod-like structure, which is similar to the structure of cellulose microfibrils. The morphological analysis of AFM revealed that L15-P (enzyme-hydrolyzed polysaccharide from LUN after 15 min of ball milling) contained relatively ordered and uniform network structures. Overall, the present study provides an important insight into cell wall of lotus rhizome matrix polysaccharide.

Abstract:Enzymatic fingerprinting of polysaccharides from Dendrobium officinale was studied and applied to authenticate Dendrobium species. Results showed that Dendrobium officinale species from Anhui province, Fujian province, Yunnan province, Guangdong province and Guangxi province of China, could be identified by polysaccharide analysis using carbohydrate gel electrophoresis (PACE). However, the fingerprints of Dendrobium officinale from Jiangxi province, Hu’nan province and Wenzhou, Yandangshan and Fuyang in Zhejiang province were very similar. As far as the fingerprints of different Dendrobium species were concerned, the differences between Dendrobium officinale, Dendrobium huoshanense, Dendrobium moniliforme, Dendrobium devonianum, Dendrobium aphyllum, Dendrobium wilsonii and Dendrobium crystallinum were obvious. Moreover, the genetic relationships between different samples were analyzed by using principal component analysis and unweighted pair group method with arithmetic mean cluster analysis. Results suggested that polysaccharide fingerprint analysis by PACE has the potential to become a valuable new method for the identification and control of quality of herbal medicines in future.

Abstract:Lotus (Nelumbo nucifera), belonging to the family of Nelumbonaceae, is a beautiful aquatic perennial plant. It has been used as an ancient horticulture plant and famous agricultural crop for thousands of years. Modern phytochemical and pharmacological experiments have proved that polysaccharide is one of the most pivotal bioactive constituents of lotus. Hence, the systematic review covering the fundamental research advances and developing prospects of N. nucifera polysaccharides (NNPs) is an urgent demand to provide theoretical basis for their further research and application. The present review summarizes current emerging research progresses on the polysaccharides isolated from lotus, and it focuses on advanced extraction and purification methods, unique structural features, engaging biological activities, potential molecular mechanisms, as well as the relationship of structure and activity of NNPs. This review sheds light on the potential values of NNPs in affording functionally bioactive agents in food industry or therapeutically effective medicines for health care. In addition, this review will provide valuable insights for further commercial product development and promising industrial application of NNPs in both of the fundamental research communities and food or pharmaceutical industries in future.

Abstract:Nelumbo nucifera Gaertn. (lotus) is a widely distributed plant with a long history of cultivation and consumption. Almost all parts of the lotus can be used as foodstuff and nourishment, or as an herb. It is noteworthy that the polysaccharides obtained from lotus exhibit surprisingly and satisfying biological activities, which explains the various benefits of lotus to human health, including anti-diabetes, anti-osteoporosis, antioxidant, anti-inflammatory, anti-tumor, etc. Here, we systematically review the recent major studies on extraction and purification methods of polysaccharides from different parts (rhizome, seed, leaf, plumule, receptacle and stamen) of lotus, as well as the characterization of their chemical structure, biological activity and structure-activity relationship, and the applications of lotus polysaccharides in different fields. This article will give an updated and deeper understanding of lotus polysaccharides and provide theoretical basis for their further research and application in human health and manufacture development.

Abstract:Abstract It have been significantly demonstrated that Hexokinase (HXK), Granule-bound starch synthase (GBSS) and ADP-glucose pyrophosphorylase (AGPase) are three critical enzymes in the starch biosynthetic pathway and are related to starch (amylose, amylopectin and total starch) content in lotus. It is important to develop functional markers in marker-assisted selection of lotus breeding. So far there have been few reports about lotus functional markers. In this study, based on insertion-deletions (INDELs) and single-nucleotide polymorphisms (SNPs), we developed three functional markers, FMHXK-E1, FMGBSS-I8 and FMAGPL-I1. FMHXK-E1 was developed based on polymorphisms of two haplotypes of NnHXK . 26 lotus cultivars that the 320-bp fragment presented in NnHXK had a lower content of amylose and a higher content of amylopectin. FMGBSS-I8 was developed based on polymorphisms of two haplotypes of NnGBSS . The group containing 32 lotus cultivars with the 210-bp fragment had less amylose content and more amylopectin content. FMAGPL-I1 was developed based on polymorphisms of two haplotypes of NnAGPL (ADP-glucose pyrophosphorylase large subunit gene). The group containing 40 lotus cultivars with the 362-bp fragment had less amylopectin, total starch content and more amylose content. According to the study, FMHXK-E1, FMGBSS-I8 and FMAGPL-I1 are closely related to lotus starch content. It could be provided research basis for molecular assisted selection of lotus starch content improve breeding efficiency.
