Abstract:Subjective cognitive decline (SCD) is thought to have high risks for conversion to Alzheimer's disease characterized as self-experienced decline in memory without cognitive assessment deficits. Several studies have demonstrated the loss of structural morphometry in the hippocampal and frontotemporal and increased functional activation in medial temporal lobe during encoding tasks. However, the alterations of intrinsic brain activity in SCD remain largely unknown. Here, we used a combined resting-state functional MRI (R-fMRI) and structural MRI dataset to examine alterations of regional amplitude of low-frequency fluctuations (ALFF) and gray matter volume (GMV) in SCD. R-fMRI and structural MRI data of 25 SCD subjects and 61 healthy controls (HCs) were collected. After functional preprocessing (slice timing, head motion correction, spatial normalization, temporal filtering and regression of Friston 24-parameter model, global, CSF and white-matter signals) and structural preprocessing (spatial normalization, Jacobian modulation and smoothing [10mm]), we obtained voxel-wise ALFF map and GMV for each subject. Before statistical analysis, all ALFF maps were spatially smoothed (4mm). Regression analysis with age, gender and education level removed was used to examine between-group differences and the results were corrected for multiple comparisons. Finally, correlation analysis was used to determine the relationships between the imaging results and cognitive measures. SCD exhibited significantly higher ALFF in the inferior parietal lobule, right middle occipital gyrus and temporal gyrus compared with HC (Fig. 1 and Table 1). There was no significant different GMV between SCD and HC. In the SCD group, we observed significant negative correlations between the ALFF values and Auditory Verbal Learning test (AVLT)-immediate recall in the right middle occipital gyrus and AVLT-recognition in the left inferior parietal lobule (P < 0.05, corrected) (Fig. 2). SCD is associated with abnormal ALFF and normal GMV. These findings provide insights into the biological mechanism of SCD and highlight the potential of ALFF as the imaging biomarker for SCD.
