Abstract:Hydroxyapatite nanowires exhibit a great potential in biomedical applications owing to their high specific surface area, high flexibility, excellent mechanical properties, and similarity to mineralized collagen fibrils of natural bone. In this work, zinc-containing nanoparticle-decorated ultralong hydroxyapatite nanowires (Zn-UHANWs) with a hierarchical nanostructure have been synthesized by a one-step solvothermal method. The highly flexible Zn-UHANWs exhibit a hierarchical rough surface and enhanced specific surface area as compared with ultralong hydroxyapatite nanowires (UHANWs). To evaluate the potential application of Zn-UHANWs in bone regeneration, the biomimetic Zn-UHANWs/chitosan (CS) (Zn-UHANWs/CS) composite porous scaffold with 80 wt % Zn-UHANWs was prepared by incorporating Zn-UHANWs into the chitosan matrix by the freeze-drying process. The as-prepared Zn-UHANWs/CS composite porous scaffold exhibits enhanced mechanical properties, highly porous structure, and excellent water retention capacity. In addition, the Zn-UHANWs/CS porous scaffold has a good biodegradability with the sustainable release of Zn, Ca, and P elements in aqueous solution. More importantly, the Zn-UHANWs/CS porous scaffold can promote the osteogenic differentiation of rat bone marrow derived mesenchymal stem cells and facilitate in vivo bone regeneration as compared with the pure CS porous scaffold or UHANWs/CS porous scaffold. Thus, both the Zn-UHANWs and Zn-UHANWs/CS porous scaffold developed in this work are promising for application in bone defect repair.

Abstract:Bone defects are very common, and there has been a great deal of research in the field of orthopedics to find ideal materials to repair such defects. Nanohydroxyapatite is a good bone substitute material; it has a number of structural similarities to natural bone, can promote new bone formation, is noncytotoxic, and has good biodegradability and biocompatibility. The use of composite and polymeric biomaterials can overcome the problems associated with the brittleness and weak mechanical properties of nanohydroxyapatite. Nanohydroxyapatite and its composite biomaterials were confirmed to play important roles in bone defect repair. This review presents a comparison of research regarding use of nanohydroxyapatite and its composite biomaterials in repairing bone defects. The goal is to identify the artificial bone substitute materials with the best biocompatibility and clinical repairing effects for various individuals and clinical situations.

Abstract:Multifunctional biomaterials that simultaneously combine high biocompatibility, biodegradability, and bioactivity are promising for, applications in various biomedical fields such as bone defect repair and drug delivery. Herein, the synthesis of hydroxyapatite nanowire@magnesium silicate nanosheets (HANW@MS) core-shell porous hierarchical nano composites (nanobrushes) is reported. The morphology of the magnesium silicate (MS) shell can be controlled by simply varying the solvothermal temperature and the amount of Mg2+ ions. Compared with hydroxyapatite nanowires (HANWs), the HANW@MS core-shell porous hierarchical nanobrushes exhibit remarkably increased specific surface area and pore volume, endowing the HANW@MS core-shell porous hierarchical nano brushes with high-performance drug loading and sustained release. Moreover, the porous scaffold of HANW@MS/chitosan (HANW@MS/CS) is prepared by incorporating the HANW@MS core-shell porous hierarchical nano brushes into the chitosan (CS) matrix. The HANW@MS/CS porous scaffold not only promotes, the attachment and growth of rat bone marrow derived mesenchymal stem cells (rBMSCs), but also induces the expression of osteogenic differentiation related genes and the vascular endothelial growth factor (VEGF) gene of rBMSCs. Furthermore, the HANW@MS/CS porous scaffold can obviously stimulate in vivo bone regeneration, owing to its high bioactive performance on the osteogenic differentiation of rBMSCs and in vivo angiogenesis. Since Ca, Mg, Si, and P elements are essential in human bone tissue, HANW@MS core-shell porous hierarchical nanobrushes with multifunctional properties are expected to be promising for various biomedical applications such as bone defect repair and drug delivery.

Abstract:A novel carbon nanotube-collagen@hydroxyapatite (CNT-Col@HA) composite with good mechanical and biological properties was fabricated successfully by a multiin situsynthesis process, which can be used to repair or replace the damaged bone tissues. The carbon nanotube (CNT)/hydroxyapatite (HA) composite powders were firstly synthesized by thein situchemical vapor deposition method. After the acidification of CNTs, the collagen (Col) molecules were covalently grafted onto the surface of CNTsin situby the formation of amide linkages, obtaining Col-encapsulated CNTs powders. And then, a HA layer was depositedin situonto the Col-encapsulated CNTs to form HA- and Col-encapsulated CNTs, consequently the ideal CNT-Col@HA composite was fabricated by the powder metallurgy method, and its mechanical and biological properties were investigated. The results showed that, the multiin situsynthesis process ensured the homogeneous dispersion of CNTs in HA matrix, and via the intermediate layer of Col, the close chemical bonding between CNT reinforcements and HA matrix was obtained, thereby the flexural strength and fracture toughness of thein situsynthesized 3 wt.% CNT-Col@HA composite were increased by approximately 74.2% and 274.6% compared with those of pure HA bulk, and better cell adhesion, spreading and proliferation were also observed on thein situsynthesized CNT-Col@HA composites. Therefore, the obtained composites in this work have great potential to be applied as implant material in clinic.
Nano-hydroxyapatite/collagen Composite for Bone Repair

Abstract:OBJECTIVE:To develop nano-hydroxyapatite/collagen (NHAC) composite and test its ability in bone repairing.METHODS:NHAC composite was developed by biomimetic method.RESULTS:The composite showed some features of natural bone in both composition and microstructure. The minerals could contribute to 50% by weight of the composites in sheet form. The inorganic phase in the composite was carbonate-substituted hydroxyapatite (HA) with low crystallinity and nanometer size. HA precipitates were uniformly distributed on the type I collagen matrix without preferential orientation. The composite exhibited an isotropic mechanical behavior. However, the resistance of the composite to localized pressure could reach the lower limit of that of femur compacta. The tissue response to the NHAC composite implanted in marrow cavity was investigated. Knoop micro-hardness test was performed to compare the mechanical behavior of the composite and bone. At the interface of the implant and marrow tissue, solution-mediated dissolution and macrophage-mediated resorption led to the degradation of the composite, followed by interfacial bone formation by osteoblasts. The process of implant degradation and bone substitution was reminiscent of bone remodeling.CONCLUSION:The composite can be incorporated into bone metabolism instead of being a permanent implant.

Abstract:Developing composite materials containing polylactic acid (PLLA) and hydroxyapatite (HA) has been a central issue in the orthopedic implant material because of their limited mechanical property which is caused by interface separation and heterogeneous dispersion in organic-inorganic composite materials. This research constructed a kind of craze disperse stress-based electrospun fibers containing hydroxyapatite nanowire, which could increase mechanical property and promote osteogenic differentiation and intravascular differentiation. The result illustrated that these composite fibers could not only consume stress energy but also stop the generation of cracks, by silver craze produced by the deformation of Hydroxyapatite nanowire. Moreover, the mechanical property of composite scaffold could be enhanced by the stress transmission between silver craze and microfiber. In addition, HA nanowires had the ability to increase PLLA fibers' hydrophilicity and promote the tensile strength of these complex fibers without making significant influence on the thermos ability of composite scaffold. Furthermore, these HA nanowires were beneficial to adhesion and proliferation of mesenchymal stem cells, and these nanowires can act as crystal nucleus to induce the deposition and growth of HA on the surface of compounded scaffold. Therefore, the composite PLLA's capability of promoting osteogenic differentiation and intravascular differentiation has been dramatically improved.

Abstract:Bone repair accounts for a large number of surgical operations. However, artificial bone replacement materials do not present the delicate continuing adjustment ability like natural bones and cause obvious side effects. Thus, materials that induce the regeneration of bones would be an optimal choice to repair bone tissues. This study proposes a biocompatible bone repair material prepared from crosslinked porous composite of collagen and hydroxyapatite. The proposed dehydrothermal method to cross-link the composite avoids use of extra chemical reagents for crosslinking and ensures that the materials were prepared using only bio-compatible materials. By adjusting the preparative parameters such as componential ratios or heating period, materials with a large property space could be achieved. Properties including porosity, mechanical strength, and swelling ratios could be facilely adjusted, promising its applications in personalized medical treatment. Cell experiments and animal experiments demonstrate the material presented high biocompatibility and effectively induced osteanagenesis in vivo. We expect the proposed material possesses high commercialization potential and serves as an effective bone repair material in realistic applications.

Abstract:Abstract At this stage, bone defects caused by trauma, infection, tumor, or congenital diseases are generally filled with autologous bone or allogeneic bone transplantation, but this treatment method has limited sources, potential disease transmission, and other problems. Ideal bone graft materials remain continuously explored, and bone defect reconstruction remains a significant challenge. Mineralized collagen prepared by bionic mineralization combining organic polymer collagen with inorganic mineral calcium phosphate can effectively imitate the composition and hierarchical structure of natural bone and has good application value in bone repair materials. Magnesium, strontium, zinc, and other inorganic components not only can activate relevant signaling pathways to induce differentiation of osteogenic precursor cells, but also stimulate other core biological processes of bone tissue growth and play an important role in natural bone growth, and bone repair and reconstruction. This study reviewed the advances in hydroxyapatite/collagen composite scaffolds and osseointegration with natural bone inorganic components such as magnesium, strontium, and zinc.

Abstract:The repairment of critical-sized bone defects is a serious problem that stimulates the development of new biomaterials. In this study, nanohydroxyapatite (nHA)-doped porous polyetheretherketone (pPEEK) were successfully fabricated by the thermally induced phase separation method and hydrothermal treatment. Structural analysis was performed by X-ray diffraction. The water contact angles and scanning electron microscopy were measured to assess physical properties of surfaces. The mechanical strength of the composites is also determined. Microcomputed tomography is used to characterize the nHA content of the composites. The in vitro bioactivity of the composites with or without nHA was investigated by using murine pre-osteoblasts MC3T3-E1, and the results of cytotoxicity and cell proliferation assays revealed that the cytocompatibility of all specimens was good. Adherence assays were employed to examine the adhesion and morphology of cells on different materials. However, nHA-doped composites induced cell attachment and cell spreading more significantly. Osteogenic differentiation was investigated using alkaline phosphatase activity and alizarin red staining, and these in vitro results demonstrated that composites containing nHA particles enhanced osteoblast differentiation. Its effectiveness for promoting osteogenesis was also confirmed in an in vivo animal experiment using a tibial defective rat model. After 8 weeks of implantation, compared to the pure PEEK and pPEEK without nHA groups, the nHA-pPEEK group showed better osteogenic activity. The results indicate that the nHA-pPEEK composites are possibly a well-designed bone substitute for critical-sized bone defects by promoting bone regeneration and osteointegration successfully.

A Highly Interweaved HA-SS-nHAp/collagen Hybrid Fibering Hydrogel Enhances Osteoinductivity and Mineralization

Abstract:The combination of bioactive hydroxyapatite (HAp) with biomimetic bone matrix biomaterials as bone filling scaffolds is a promising strategy for bone regeneration, but the undesirable dispersion of HAp and its interfacial interaction result in inefficient mineralization, mechanical instability, incomplete osteointegration, and even repair failure. Herein, the size dispersion and stabilization of nano-hydroxyapatite (nHAp) in aqueous media were obviously improved by hydrophilic solubilisation and strong negatively charged thiolated hyaluronic acid (HA-SH). Furthermore, the highly interweaved HA-SS-nHAp/collagen hybrid fibering hydrogel exhibited significantly improved mechanical properties and structural stability due to its thickened and densified interweaved fiber network, which ensured the homogeneous dispersion of nHAp in the matrix materials and its integration with the hydrogel network structure completely by covalent self-crosslinking among the sulfhydryl groups derived from the free HA-SH polymer and the mercapto functional groups on the surface of nHAp. Compared with the physically combined micro-hydroxyapatite (μHAp) (d≤25 μm) and nHAp (∼530 nm) with injectable bionic HA-SH and collagen type I biopolymers, HA-SS-nHAp/collagen achieved the maximum efficiency in facilitating rabbit bone marrow stromal cell (rBMSC) adhesion, proliferation and osteogenic differentiation in vitro. The in vivo murine dorsal subcutaneous implantation results further confirmed that the interweaved fiber network structure in HA-SS-nHAp/collagen significantly promoted osteoinductivity and mineralization. This work provides novel insights for the development of new low invasive bone filling biomaterials.

Abstract:The clinical treatment of bone defects includes allogeneic bone transplantation and autologous bone transplantation. However, they all have their own limitations, and the scope of application is limited. In recent years, bone tissue engineering scaffolds based on a variety of materials have been well developed and achieved good bone regeneration ability. However, most scaffold materials always face problems such as high biotoxicity, leading to inflammation and poor bioactivity, which limits the bone regeneration effect and prolongs the bone regeneration time. In our work, we prepared hydroxyapatite, erythropoietin (EPO), and osteogenic growth peptide (OGP) codoped type-I collagen (Col I) polypeptide nanofiber membranes (NFMs) by electrostatic spinning. In cell experiments, the composite NFMs had low cytotoxicity and promoted osteogenic differentiation of rat bone marrow mesenchymal stem cells. Quantitative real-time polymerase chain reaction and alkaline phosphatase staining confirmed the high expression of osteogenic genes, and alizarin red S staining directly confirmed the appearance of calcium nodules. In animal experiments, the loaded hydroxyapatite formed multiple independent mineralization centers in the defect center. Under the promotion of Col I, EPO, and OGP, the bone continued to grow along the mineralization centers as well as inward the defect edge, and the bone defect completely regenerated in about two months. The hematological and histological analyses proved the safety of the experiments. This kind of design to promote bone regeneration by simulating bone composition, introducing mineralization center and signal molecules, can shorten repair time, improve repair effect, and has good practical prospects in the future.
Mechanically enhanced composite hydrogel scaffold for in situ bone repairs

Abstract:High-efficiency repair of critical bone defects is a pressing problem in clinical practice. However, most biological replacement materials do not simultaneously satisfy the dual requirements of mechanical strength and cell compatibility. In this study, chitosan methacryloyl (CSMA) and β-tricalcium phosphate (β-TCP) were subjected to photo-crosslinking to form the CSMA/β-TCP composite hydrogel, which has strong mechanical properties contributing to bone regeneration. In addition, its scaffold can alter the morphology of bone marrow mesenchymal stem cells (BMSCs), promote their proliferation, enhance the expression of alkaline phosphatase (ALP), and augment the nodular deposition of calcium. Meanwhile, the expressions of osteogenic proteins (ALP, osteocalcin, and osteopontin) were upregulated and the regulatory mechanism of the Hippo signaling pathway was verified. Moreover, animal experiments have confirmed that CSMA/β-TCP has adequate biocompatibility and bone regeneration. These results demonstrate the immense potential of the CSMA/β-TCP composite hydrogel in bone regeneration therapy.
Nanoclay mineral-reinforced macroporous nanocomposite scaffolds for in situ bone regeneration: In vitro and in vivo studies

Abstract:<p>Biomimetic organic-inorganic nanocomposite scaffolds have represented a tremendous opportunity to accelerate bone regeneration due to their excellent structural and biological cues. However, the tradeoff between biodegradability, mechanical strength, porosity and pore size, and bioactivity of the tissue engineered nanocomposites remains a challenge. To this end, we constructed a novel biodegradable nanocomposite scaffold by chemical crosslinking of hydroxyethyl cellulose (HEC)/soy protein isolate (SPI) and montmorillonite (MMT) modification. Owing to the strong interfacial interaction between nanoclay and polymer chains, the prepared nanocomposites showed enhanced mechanical and physical properties, within the ideal range for bone tissue engineering. Furthermore, the sustained sequential release of Ca and Mg ions during the degradation of nanocomposites was observed, which provided essential signaling cues for guiding bone regeneration. It was verified in vitro that the engineered nanocomposites possessed satisfactory cytocompatibility, offering a highly desirable microenvironment for cellular behaviors, including adhesion, proliferation, and spreading, as well as promoting the mineralization and osteoblastic differentiation of rat bone marrow stem cells. Notably, when employed to repair critical-sized cranial defects in rat models, the nanocomposites demonstrated not only reliable biosafety, but also significant osteointegration and bone-forming ability in vivo. Collectively, our results confirmed that the incorporation of inorganic MMT nanosheets into biocompatible HEC/SPI scaffolds is a viable strategy for fabricating a suitable biomaterial that enhances the regeneration of large bone defects.</p>

Abstract:Abstract Critical-sized bone defects represent a significant clinical challenge due to their inability to undergo spontaneous regeneration, necessitating graft interventions for effective treatment. The development of tissue-engineered scaffolds and regenerative medicine has made bone tissue engineering a highly viable treatment for bone defects. The physical and biological properties of nanocomposite biomaterials, which have optimized structures and the ability to simulate the regenerative microenvironment of bone, are promising for application in the field of tissue engineering. These biomaterials offer distinct advantages over traditional materials by facilitating cellular adhesion and proliferation, maintaining excellent osteoconductivity and biocompatibility, enabling precise control of degradation rates, and enhancing mechanical properties. Importantly, they can simulate the natural structure of bone tissue, including the specific microenvironment, which is crucial for promoting the repair and regeneration of bone defects. This manuscript provides a comprehensive review of the recent research developments and applications of structure-optimized and microenvironment-inspired nanocomposite biomaterials in bone tissue engineering. This review focuses on the properties and advantages these materials offer for bone repair and tissue regeneration, summarizing the latest progress in the application of nanocomposite biomaterials for bone tissue engineering and highlighting the challenges and future perspectives in the field. Through this analysis, the paper aims to underscore the promising potential of nanocomposite biomaterials in bone tissue engineering, contributing to the informed design and strategic planning of next-generation biomaterials for regenerative medicine.

Abstract:<p>Various materials are utilized as artificial substitutes for bone repair. In this study, a silk fibroin (SF) hydrogel reinforced by short silica nanoparticles (SiNPs)‐distributed‐silk fibroin nanofibers (SiNPs@NFs), which exhibits a superior osteoinductive property, is fabricated for treating bone defects. SF acts as the base part of the composite scaffold to mimic the extracellular matrix (ECM), which is the organic component of a native bone. The distribution of SiNPs clusters within the composite hydrogel partially mimics the distribution of mineral crystals within the ECM. Incorporation of SiNPs@NFs enhances the mechanical properties of the composite hydrogel. In addition, the composite hydrogel provides a biocompatible microenvironment for cell adhesion, proliferation, and osteogenic differentiation in vitro. In vivo studies confirm that the successful repair is achieved with the formation of a large amount of new bone in the large‐sized cranial defects that are treated with the composite hydrogel. In conclusion, the SiNPs@NFs‐reinforced‐hydrogel fabricated in this study has the potential for use in bone tissue engineering.</p>

Abstract:Objective To observe the repairing effects of bone after nano-hydroxyapatite/collagen (nHAC)being implanted in and to study its prospect in clinical application.Methods Nano-hydroxyapatite/collagen composite was developed by biomimetic method. The material was used in clinical operation,the healing effects of bone defects were assessed by methods of radiography.Results The composite showed some features of natural bone in both composition and microstructure.After implanting,the patients were not hyperpyrexial,did not have extravasate and immunoreaction,the bone defects were healing well.Conclusion The composite can serve as one of the promising materials for bone repairing.

Abstract:Hydroxyapatite (HA) whisker (HAw) represents a distinct form of HA characterized by its high aspect ratio, offering significant potential for enhancing the mechanical properties of bone tissue engineering scaffolds. However, the limited osteoinductivity of HAw hampers its widespread application. In this investigation, we observed HAw-punctured osteoblast membranes and infiltrated the cell body, resulting in mechanical damage to cells that adversely impacted osteoblast proliferation and differentiation. To address this challenge, we developed nano-zinc oxide particle-modified HAw (nano-ZnO/HAw). Acting as a reinforcing and toughening agent, nano-ZnO/HAw augmented the compressive strength and ductility of the matrix materials. At the same time, the surface modification with nano-ZnO particles improved osteoblast differentiation by reducing the mechanical damage from HAw to cells and releasing zinc ion, the two aspects collectively promoted the osteoinductivity of HAw. Encouragingly, the osteoinductive potential of 5% nano-ZnO/HAw and 10% nano-ZnO/HAw was validated in relevant rat models, demonstrating the efficacy of this approach in promoting new bone formation in vivo. Our findings underscore the role of nano-ZnO particle surface modification in enhancing the osteoinductivity of HAw from a physical standpoint, offering valuable insights into the development of bone substitutes with favorable osteoinductive properties while simultaneously bolstering matrix material strength and toughness.
