Abstract:Four deep eutectic solvents (DESs), namely, glycerol/chlorocholine (glycerol/ChCl), urea/ChCl, citric acid/ChCl, and oxalic acid/ChCl, were synthesized and their performance in the dissolution of cellulose was studied. The results showed that the melting point of the DESs varied with the proportion of the hydrogen bond donor material. The viscosity of the DESs changed considerably with the change in temperature; as the temperature increased, the viscosity decreased and the electrical conductivity increased. Oxalic acid/ChCl exhibited the best dissolution effects on cellulose. The microscopic morphology of cellulose was observed with a microscope. The solvent system effectively dissolved the cellulose, and the dissolution method of the oxalic acid/ChCl solvent on cellulose was preliminarily analyzed. The ChCl solvent formed new hydrogen bonds with the hydroxyl groups of the cellulose through its oxygen atom in the hydroxyl group and its nitrogen atom in the amino group. That is to say, after the deep eutectic melt formed an internal hydrogen bond, a large number of remaining ions formed a hydrogen bond with the hydroxyl groups of the cellulose, resulting in a great dissolution of the cellulose. Although the cellulose and regenerated cellulose had similar structures, the crystal form of cellulose changed from type I to type II.

Abstract:The relatively low solubility of cellulose in choline chloride-based deep eutectic solvents (DESs) restricts their practical application in the production industry. Here, cellulose was slightly oxidized into dialdehyde cellulose (DAC) at first, and then partial chemical bonding was introduced to intensify the dissolution process of cellulose in (choline chloride-resorcinol) DES. In this process, the solubility of cellulose in DES increased 10-fold, and the crystallinity of cellulose decreased over 24% while the oxidation level was lower than 20%. An intensified hydrogen bond network was revealed, which implied that cellulose components mainly acted as hydrogen bond donor after dissolution. A new class of DES containing cellulose was thus preliminarily established. The cellulose chains and the intensive hydrogen bond network also resulted in an increased viscosity and a conversion from Newtonian behavior to non-Newtonian behavior of dissolved samples. The phase transition behaviors of dissolved samples were gradually converted from crystallization to cool crystallization. The proposed and performed strategy is novel and scalable and will open a fresh perspective to achieve higher efficient dissolution of cellulose in DES and establish a new partially derivatized cellulose/DES system.
Deep eutectic solvent pretreatment of cellulose and development of hydrophobic foaming material

Abstract:This work aims to investigate the effects of deep eutectic solvents (DES) on the chemical and physical structure of cellulose. Choline chloride-oxalic acid and choline chloride-oxalic acid-glycerol were selected as solvents and cotton fibers was sued as raw materials to explore the difference between cotton fibers treated separately with two different DES. According to yield analysis, ternary solvents alleviated the degradation of cellulose when comparing to binary solvents, resulting in over 90 % of cellulose being obtained. Particularly, there is an esterification reaction of cellulose during treatment with the DES system, which also affects the performance of the subsequent products. Through the simple use of mechanical foaming with polyvinyl alcohol and the palm wax impregnation process, foams with a water contact angle greater than 140° and excellent mechanical properties can be obtained. The resultant foam material has 5 % linear elastic area, and prominent compressive strength providing potential use in the packaging industry in the replacement of plastic.

Abstract:As an effective substitute for ionic liquids (ILs), deep eutectic solvents (DESs) are known as a new generation of green solvents. In addition, DESs are considered to be the best solvents to replace traditional organic solvents and ILs due to their advantages of economy, simple preparation and biodegradability, which can be used to further break through the technical bottleneck in the field of biomass pretreatment. This study synthesized a series of choline chloride-based (ChCl-based) DESs and used them to pretreat microcrystalline cellulose (MCC) without dissolution, in order to achieve the purpose of improving the saccharification effect. DES-9 (ChCl/hydroquinone), DES-12 (ChCl/acetamide) and DES-16 (ChCl/acetic acid) were better pretreatment solvents. The optimum conditions for the pretreatment of MCC by DES-9 and DES-12 were both at 110 °C for 4 h, and the optimum condition of DES-16 was at 90 °C for 4 h. Compared with untreated MCC (26.66%), the saccharification yield of pretreated MCC with DES-16 increased by 20.5%. Finally, the structure and morphology changes of MCC before and after pretreatment were investigated in detail through the degree of polymerization (DP), SEM and enzyme adsorption. The results showed that DESs pretreatment made the surface of MCC become rough, which enhanced the accessibility of cellulose and enzymes, improving the saccharification effect.

Abstract:Novel and green cholinium amino acids-based deep eutectic solvents (DESs) and their aqueous mixtures were synthesized and employed in deconstructing poplar for hemicellulose fractionation. The effects of water content in DESs on hemicellulose dissociation and structural features were comprehensively investigated, along with the reusability of DESs for treatment. The integration of water into DESs could facilitate hemicellulose fractionation, and the cholinium lysine: urea with 5 wt% water (CL: U-5) demonstrated the best performance with a hemicellulose yield of 59.2%. Further structure analysis revealed that hemicelluloses with various branching degrees and molecular weights were obtained with varying water content of DESs. Furthermore, the CL: U-5 had recyclability and reusability with a 40.5% hemicellulose yield obtained after reused three times. The novel and eco-friendly cholinium amino acids-based DESs treatment provides an effective and sustainable strategy for hemicellulose fractionation from woody biomass.

Abstract:In this study, bleached chemical pulp from hardwood was treated with different carboxylic acid choline chloride-based deep eutectic solvents (CA/ChCl DESs) including formic acid, acetic acid, ethanedioic acid, propanedioic acid and choline chloride. The dissolution and degradation of cellulose and hemicellulose in chemical pulps in these DESs were analyzed, while the relationships between the polarity parameters of DESs and the degradation and dissolution of cellulosic fibers were discussed. The results show that dicarboxylic acids/ChCl were more conducive to the solubility and degradation of cellulosic fibers compared to monocarboxylic acids/ChCl at a higher temperature. In addition, the increase in alkyl of the CA could reduce the solubility to cellulosic fiber. The further study reveals that the dissolution and degradation of cellulosic fibers are generally governed both by temperature and the ability of DESs hydrogen bond acidity ( α ), while temperature shows little effect on α. However, no remarkable relationships were found between the DESs hydrogen bond basicity ( β ) and cellulose degradation and dissolution.

Abstract:Deep eutectic solvents (DESs) have emerged as a promising class of solvents for lignin dissolution, which could significantly expand the application potential of lignin. In this study, over forty ChCl-based DESs of three major types were synthesized and subjected to investigate the solubilisation of two industrial lignins in DES. The results showed that higher HBD content, shorter carbon chain length in the HBD, and fewer functional groups favored lignin solubilization. DESs containing carboxyl groups were found to be more favorable for breaking β-O-4 bonds and solubilizing lignin. Additionally, high temperature and appropriate water content were observed to promote lignin solubilization. The pretreatment of poplar and maize straw with ChC/FA(1:3), ChC/LA(1:3), ChC/Gly(1:3), ChC/EG(1:3), ChC/Urea(1:3), and ChC/TEOA(1:3) showed good solubilization of lignin, with ChCl/FA(1:3) being particularly effective in solubilizing poplar lignin and maize straw lignin, achieving lignin solubilization of 82% and 57%, respectively. Overall, these findings suggest that DESs have great potential as solvents for lignin dissolution.

Abstract:Abstract A novel lignin-derived deep eutectic solvent (DES) was synthesized from choline chloride (ChCl) and p-coumaric acid (PCA). The results showed that after pretreatment with lignin-derived DES, the dense cellulose–hemicellulose–lignin structure of herb residues of Cortex albiziae (HRCA) was effectively broken down and this facilitated subsequent enzymatic hydrolysis. The optimum pretreatment conditions were ChCl/PCA molar ratio of 1:1, 160 °C-5 h, and a solid–liquid ratio of 1:10. Compared with untreated HRCA, 72-h enzymatic digestibility increased from 48.08 to 84.62%. When a ChCl–PCA–water pretreatment was used, the 72-h digestibility level of the residue reached 99.36%. Moreover, the recycling and reusability of this novel DES were evaluated. The results showed that synthesis and utilization of green, environment-friendly and recyclable lignin-derived DES can offer a new way of utilizing lignin which has considerable potential in the refining of biomass. Choline chloride - oxalic acid dihydrate deep eutectic solvent pretreatment of Barley straw for production of cellulose nanofibers

Abstract:This study investigates the production of cellulose nanofibers (CNF) from Barley straw using ultrasound-assisted deep eutectic solvent (US-DES) treatment for biomass fractionation and subsequent high-intensity ultrasonication (HIUS) for nano-fibrillation. Two deep eutectic solvents (DES), synthesized from choline chloride (ChCl) and oxalic acid dihydrate (OAD) at 1:1 and 2:1 M ratio, achieved solubilisation of over 80 % of lignin and hemicellulose under optimal conditions. The purification of these DES-treated materials resulted in cellulose with a purity >88 %. CNFs, characterized by a size of <100 nm, a polydispersity index under 0.5, and a zeta potential lower than -30 mV, were successfully isolated through a combination of wet grinding and HIUS treatment. SEM and XRD results showed the formation of a network of interconnected fibres with a Type I cellulose structure. This research highlights Barley straw's potential as a sustainable source of high-value CNF from agricultural waste.

Abstract:This work proposed a homogeneous preparation strategy of cellulose acetate by using a deep eutectic solvent consisting of zinc chloride/phosphoric acid/water (ZnCl2/PA/H2O). The results showed that the DES (the molar ratio of ZnCl2: PA: H2O = 1:1:3) had good solubility for bamboo pulp with high degree of polymerization. The dynamic dissolution behavior of cellulose in DES was also studied. Cellulose was first moistened and swollen and then dissolved. The crystalline state of cellulose changed from type I to type II after regeneration. Under the synergistic effect of acetic anhydride (Ac2O), the dissolved cellulose in DES could be uniformly acetylated. The effects of temperature, reaction time and molar ratio of Ac2O/anhydrous glucose on degree of substitution (DS) were investigated in their respective ranges (30–50 °C, 2–10 h and 4–24). Cellulose acetate with DS of 0.42–1.81 was successfully prepared. The substituent distribution of the sample was analyzed as C-6 > C-2 > C-3. Moreover, the entire process required no additional catalysts or activation steps, reducing the difficulty of solvent recovery. The insight from this study provides a new idea for the development of greener methods for the homogeneous preparation of cellulose acetate.

Abstract:Deep eutectic solvents (DESs) are a potentially high-value lignin extraction methodology. DESs prepared from choline chloride (ChCl) and three hydrogen-bond donors (HBD)-lactic acid (Lac), glycerol, and urea-were evaluated for isolation of willow (Salix matsudana cv. Zhuliu) lignin. DESs types, mole ratio of ChCl to HBD, extraction temperature, and time on the fractionated DES-lignin yield demonstrated that the optimal DES-lignin yield (91.8 wt % based on the initial lignin in willow) with high purity of 94.5% can be reached at a ChCl-to-Lac molar ratio of 1:10, extraction temperature of 120 °C, and time of 12 h. Fourier transform infrared spectroscopy (FT-IR) , 13C-NMR, and 31P-NMR showed that willow lignin extracted by ChCl-Lac was mainly composed of syringyl and guaiacyl units. Serendipitously, a majority of the glucan in willow was preserved after ChCl-Lac treatment.

Abstract:Deep eutectic solvents (DESs) hold great potential in biorefining because they can efficiently deconstruct the recalcitrant structure of lignocellulose. In particular, inorganic salts with Lewis acids have been proven to be effective at cleaving lignin-carbohydrate complexes. Herein, a Zr-based DES system composed of metal chloride hydrate (ZrOCl2·8H2O) and ethylene glycol (EG) was designed and used for poplar powder pretreatment. Zr4+-based salts provide sufficient acidity for lignocellulose depolymerization. The acidity of the DES was analysed by the Kamlet-Taft solvatochromic parameter, and the results demonstrated that the acidity can be regulated by the DES composition. Under the optimum conditions (ZrOCl2·8H2O:EG molar ratio of 1:2), the DES pretreatment removes nearly 100 % hemicellulose and 94.7 % lignin. The recovered lignin exhibited a low polydispersity of 1.7. The cellulose residues deliver an efficiency of 94.4 % upon enzymatic digestion. Moreover, the DES can be easily recovered with high yield and purity, and the recycled DES still maintains high delignification and enzymatic hydrolysis efficiencies. The proposed DES pretreatment technology is promising for biomass valorization.

Abstract:Schematic diagram of ultrasound‐assisted deep eutectic solvent (DES) treatment of cotton straw. Deep eutectic solvent (DES) is a new type of green solvent that can be used for the separation of lignocellulosic components. In this study, ultrasound‐assisted DES pretreatment was used to treat cotton straw, which could retain most of the cellulose in cotton straw and separate hemicellulose and lignin at the same time. Here, DESs with different molar ratios were prepared using choline chloride as hydrogen bond acceptor and formic acid as hydrogen bond donor. Simultaneously changing the ultrasonic frequency and DESs synergistic extraction of lignin. Results show that the ultrasonic power is 32 kHz (room temperature), ultrasound 30 min, oil bath heat 3 h under the best conditions can significantly extract lignin, and the yield of lignin is 87.7% and the purity reached 97.3%. The characteristic peaks of FT‐IR confirmed the extraction of lignin. The structural characteristics of 2D‐HSQC also confirmed the effective separation of lignin, and x‐ray diffraction (XRD) and scanning electron microscope (SEM) confirmed the effect of ultrasound‐assisted DES pretreatment on lignocellulose structure. In short, this study provides a feasible method for the separation of lignocellulose.

Abstract:The introduction of specific choline chloride (ChCl)- and betaine (Bet)-based deep eutectic solvents (DESs) into the conventional acetic acid–sodium acetate solution led to a significant increase in cellulase activity and stability. In particular, a higher enhancement in β-glucosidase activity was obtained, which plays a key role in improving cellulase function. The DESs prolonged the half-life of cellulase. The addition of 10 % (v/v) ChCl/1,3-propanediol (1:2) and 5 % (v/v) Bet/glycerol (1:2) resulted in the most significant improvement, with the half-life of cellulase reaching three times the initial half-life value. The above both DESs allowed the cellulase to maintain an excellent level of activity over time, even after 72 h. The DESs tightened the structure of the enzyme by changing its secondary structure, which had a stabilizing effect on the overall conformation. The density functional theory calculations of DESs revealed that the DES with a more stable structure more effectively protected the active conformation. The molecular dynamics simulation on β-glucosidase indicated that the hydrogen bond donor end of the DES primarily formed hydrogen bonds with the enzyme, which induced a spatially compact conformation by increasing the α-helix content while reducing the β-sheet content. It also restricted the movement of amino acids, particularly stabilizing the active site, and increased the hydrophilic region, thereby enhancing enzyme activity and stability. In addition, the DES formed a protective layer that prevented the unproductive adsorption of lignin on the enzymes during the hydrolysis of lignocellulosic feedstocks. These multiple improvements provided by DESs significantly increased the enzymatic saccharification efficiency of corn stover and poplar.

Abstract:DESs are promising green solvents in lignocellulose pretreatment. In this work, four choline chloride based DESs were synthesized and their physicochemical properties were evaluated. Herbaceous lignocellulose (wheat straw, sorghum straw) and woody lignocellulose biomass (poplar wood, pine wood) were selected as pretreated substrates. In order to reveal the mechanism that pretreatment can improve the efficiency of enzymatic hydrolysis, the pretreated lignocellulose and lignin were analyzed. SEM, FT-IR and XRD characterization revealed the microstructural changes of pretreated lignocellulose, including the increase of surface roughness and pore erosion, changes of polysaccharide/lignin chemical groups, the increase of CrI. The efficient delignification increased the accessibility of enzyme to cellulose, the lignin content negatively affected enzymatic hydrolysis conversion. Furthermore, GPC analysis showed that the polydispersity of sulfonated lignin was reduced after pretreatment, lignin had more homogeneous molecular structures. The degree of unsaturation was calculated according to the elemental composition of sorghum straw and lignin. The unsaturation of pretreated sorghum straw decreased, that may be due to the removal of partial lignin. The elemental composition of acid insoluble lignin (separated from sorghum straw) reflects the increase of unsaturation of lignin after pretreatment.

Abstract:The depolymerization of biomass carbohydrate polymers usually happened in homogeneous medium, just a few in heterogeneous solution. Herein, holocellulose from two Chinese herb residues Cortex albiziae (HRCA) and Heteropogon contortus (HRHC) was prepared and characterized. Deep eutectic solvent (DES) of choline chloride/p-coumaric acid collaborating with water was employed for the dissociation of those two holocelluloses and selectively dissolved hemicellulose into xylose with maximal yields of 81.50 % and 72.47 %, respectively. Most cellulose remained as the polymer state with a maximum solubility of 9.38 %. The synergistic action of DES and water was investigated to unveil the depolymerization mechanism and the roles of each component in depolymerizing procedure. It was water that released hemicellulose polymers and dissolved polymers to soluble short oligosaccharides, and DES rapidly cleaved the latter to xylose. Contrarily, DES broke cellulose to only soluble cello-oligosaccharides, but water exhibited the ability to hydrolyze cellulose to glucose.

Abstract:In this work, deep eutectic solvent (DES) was prepared by mixing choline chloride (ChCl) with lactic acid (LA), and effects of cellulase non-productive binding onto DES-extracted lignin from willow and corn stover on enzymatic hydrolysis of cellulose was investigated. The correlation between hydrolysis yield of cellulose and chemical features of lignin was evaluated, and a potential inhibitory mechanism was proposed. Condensation of lignin was observed during DES treatment, and these condensed aromatic structures had an increased tendency to adsorb enzymes through hydrophobic interactions. As well as hydrophobic interactions mediated by lignin condensation, an increase in phenolic hydroxyl groups resulted in a greater amount of hydrogen bonds between cellulases and lignin that appeared to inhibit enzymatic hydrolysis yields of cellulose (39.96-42.86% to 31.96-32.68%). Although large amounts of -COOHs were generated, the elevated electrostatic repulsion as a result of ionic groups was insufficient to decrease non-productive adsorption.

Abstract:The high viscosity of deep eutectic solvent (DES) limits its application in industrial production due to reduced mass transfer efficiency within the solvent system. Incorporating water into the solvent system can effectively reduce the viscosity of DES. In this study, DES based on glycerol and choline chloride were prepared, and the physicochemical properties of DES-water mixtures and the impact of various water contents in the solvent system on the pretreatment efficiency of poplar was investigated. At a 35% water content in the solvent system, the solvent's viscosity drops by 95%. The separation results from poplar experiments demonstrated that adding 10%−15% water to the solvent system promoted lignin dissolution and increased its extraction rate. Following pretreatment, the solid product cellulose exhibited a purity level exceeding 80%, with an enzymatic hydrolysis rate exceeding 95%. Structural characterization analysis revealed that the total phenolic hydroxyl content of lignin increased, along with an increase in cellulose crystallinity. Adding water to the solvent system improved both the purity of lignin and the content of active hydroxyl groups. Additionally, there was a more uniform distribution in molecular weight. This study comprehensively discussed the feasibility of using water as an auxiliary solvent for DES-based lignocellulose pretreatment. This approach is expected to facilitate industrialization and enhance preferential lignin extraction during pretreatment.

Abstract:Ternary deep eutectic solvents (DESs) were developed to enable rapid and high-solid biomass pretreatment as well as concentrated sugar hydrolysate production. Six ternary DESs constituted choline chloride (ChCl) or guanidine hydrochloride (GH) as a hydrogen bond acceptor (HBA), ethylene glycol (EG) or propylene glycol (PG) or glycerin (GLY) as a polyol-based hydrogen bond donor (HBD), and p-toluenesulfonic acid (PTSA) as an acidic HBD. GH-EG-PTSA was the most effective, evidenced by 79% xylan and 82% lignin removal in only 6 min at 120 degrees C and 10 wt% solid loading. Even at 35 wt% solid loading, both GH-EG-PTSA and ChCl-EG-PTSA still removed more than 60% xylan and lignin in 30 min. Using a 20% solid loading and a low enzyme loading of 5 mg protein/g solid, 128 g/L glucose and 20 g/L xylose was obtained, with a glucose yield of 78.4%. Overall, this study demonstrated novel and high-performance ternary DESs for effective lignocellulose deconstruction.
