Abstract:Ocean deoxygenation, warming, and acidification resulting from global change and increasing nutrient inputs are major threats to marine ecosystems. Despite this, spatial and temporal patterns of oxygen availability and their impacts on marine life are understudied compared to warming and acidification, particularly in coastal tropical ecosystems. We measured the abundance of major groups of zooplankton in the context of five covarying environmental parameters [temperature, salinity, dissolved oxygen (DO), dissolved organic matter (DOM), and chlorophyll concentration] in a tropical estuary for one year. Partial least squares demonstrated that environmental conditions explained 20% of the variation in the community and found that temperature, salinity, DO, and DOM were most important (Variable Importance in Projection [VIP] > 0.8). A generalized linear model identified depth, DO, salinity, and chlorophyll as significant main effects, and temperature and DOM were also significant via two-way interactions (p < 0.05). When examined separately, the abundance of each zooplankton group was explained by a slightly different combination of environmental factors, but in all cases DO had large, significant effects, and in most cases DOM or its interactions were also significant. These results demonstrate that the seasonal cycle of hypoxia in this system significantly impacts the abundance of major zooplankton groups and likely also recruitment of benthic fauna through impacts on meroplankton and benthic-pelagic food webs.

Abstract:Hypoxia, triggered in large part by eutrophication, exerts widespread and expanding stress on coastal ecosystems. Hypoxia is often specifically defined as water having dissolved oxygen (DO) concentrations < 2 mg L−1. However, DO concentration alone is insufficient to categorize hypoxic stress or predict impacts of hypoxia on zooplankton and fish. Hypoxic stress depends on the oxygen supply relative to metabolic demand. Water temperature controls both oxygen solubility and the metabolic demand of aquatic ectotherms. Accordingly, to assess impacts of hypoxia requires consideration of effects of temperature on both oxygen availability and animal metabolism. Temperature differences across ecosystems or across seasons or years within an ecosystem can dramatically impact the severity of hypoxia even at similar DO concentrations. Living under sub-optimum DO can reduce temperature-dependent metabolic efficiencies, prey capture efficiency, growth and reproductive potential, thus impacting production and individual zooplankton and fish fitness. Avoidance of hypoxic bottom water can reduce or eliminate low-temperature thermal refuges for organisms and increase energy demands and respiration rates, and potentially reduce overall fitness if alternative habitats are sub-optimal. Moreover, differential habitat shifts among species can shift predator-prey abundance ratios or interactions and thus modify food webs. For example, more tolerant zooplankton prey may use hypoxic waters as a refuge from fish predation. In contrast, zooplankton avoidance of hypoxic bottom waters can result in prey aggregations at oxyclines sought out by fish predators. Hypoxic conditions that affect spatial ecology can drive taxonomic and size shifts in the zooplankton community, affecting foraging, consumption and growth of fish. Advances in understanding the ecological effects of low DO waters on pelagic zooplankton and fish and comparisons among ecosystems will require development of generic models that estimate the oxygen demand of organisms in relation to oxygen supply which depends on both DO and temperature. We provide preliminary analysis of a metric (Oxygen Stress Level) which integrates oxygen demand in relation to oxygen availability for a coastal copepod and compare the prediction of oxygen stress to actual copepod distributions in areas with hypoxic bottom waters.

A Dissolved Oxygen Threshold for Shifts in Bacterial Community Structure in a Seasonally Hypoxic Estuary

Abstract:Pelagic ecosystems can become depleted of dissolved oxygen as a result of both natural processes and anthropogenic effects. As dissolved oxygen concentration decreases, energy shifts from macrofauna to microorganisms, which persist in these hypoxic zones. Oxygen-limited regions are rapidly expanding globally; however, patterns of microbial communities associated with dissolved oxygen gradients are not yet well understood. To assess the effects of decreasing dissolved oxygen on bacteria, we examined shifts in bacterial community structure over space and time in Hood Canal, Washington, USA-a glacial fjord-like water body that experiences seasonal low dissolved oxygen levels known to be detrimental to fish and other marine organisms. We found a strong negative association between bacterial richness and dissolved oxygen. Bacterial community composition across all samples was also strongly associated with the dissolved oxygen gradient, and significant changes in bacterial community composition occurred at a dissolved oxygen concentration between 5.18 and 7.12 mg O2 L(-1). This threshold value of dissolved oxygen is higher than classic definitions of hypoxia (<2.0 mg O2 L(-1)), suggesting that changes in bacterial communities may precede the detrimental effects on ecologically and economically important macrofauna. Furthermore, bacterial taxa responsible for driving whole community changes across the oxygen gradient are commonly detected in other oxygen-stressed ecosystems, suggesting that the patterns we uncovered in Hood Canal may be relevant in other low oxygen ecosystems.

Abstract:The Bay of Bengal hosts persistent, measurable, but sub-micromolar, concentrations of oxygen in its oxygen-minimum zone (OMZ). Such low-oxygen conditions are not necessarily rare in the global ocean and seem also to characterize the OMZ of the Pescadero Basin in the Gulf of California, as well as the outer edges of otherwise anoxic OMZs, such as can be found, for example, in the Eastern Tropical North Pacific. We show here that biological controls on oxygen consumption are required to allow the semistable persistence of low-oxygen conditions in OMZ settings; otherwise, only small changes in physical mixing or rates of primary production would drive the OMZ between anoxic and oxic states with potentially large swings in oxygen concentration. We propose that two controls are active: an oxygen-dependent control on oxygen respiration and an oxygen inhibition of denitrification. These controls, working alone and together, can generate low-oxygen concentrations over a wide variability in ocean mixing parameters. More broadly, we discuss the oxygen regulation of organic matter cycling and N 2 production in OMZ settings. Modern biogeochemical models of nitrogen and oxygen cycling in OMZ settings do contain some of the parameterizations that we explore here. However, these models have not been applied to understanding the persistence of low, but measurable, concentrations of oxygen in settings like the Bay of Bengal, nor have they been applied to understanding what biological/physical processes control the transition from a weakly oxygenated state to a “functionally” anoxic state with implications for nitrogen cycling. Therefore, we believe that the approach here illuminates the relationship between oxygen and the biogeochemical cycling of carbon and nitrogen in settings like the Bay of Bengal. Furthermore, we believe that our results could further inform large-scale ocean models seeking to explore how global warming might influence the spread of low-oxygen waters, influencing the cycles of oxygen, carbon, and nitrogen in OMZ settings.
Oxygen limitations on marine animal distributions and the collapse of epibenthic community structure during shoaling hypoxia

Abstract:Deoxygenation in the global ocean is predicted to induce ecosystem-wide changes. Analysis of multidecadal oxygen time-series projects the northeast Pacific to be a current and future hot spot of oxygen loss. However, the response of marine communities to deoxygenation is unresolved due to the lack of applicable data on component species. We repeated the same benthic transect (n = 10, between 45 and 190 m depths) over 8 years in a seasonally hypoxic fjord using remotely operated vehicles equipped with oxygen sensors to establish the lower oxygen levels at which 26 common epibenthic species can occur in the wild. By timing our surveys to shoaling hypoxia events, we show that fish and crustacean populations persist even in severe hypoxia (<0.5 mL L(-1) ) with no mortality effects but that migration of mobile species occurs. Consequently, the immediate response to hypoxia expansion is the collapse of community structure; normally partitioned distributions of resident species coalesced and localized densities increased. After oxygen renewal and formation of steep oxygen gradients, former ranges re-established. High frequency data from the nearby VENUS subsea observatory show the average oxygen level at our site declined by ~0.05 mL L(-1) year(-1) over the period of our study. The increased annual duration of the hypoxic (<1.4 mL L(-1) ) and severely hypoxic periods appears to reflect the oxygen dynamics demonstrated in offshore source waters and the adjacent Strait of Georgia. Should the current trajectory of oxygen loss continue, community homogenization and reduced suitable habitat may become the dominant state of epibenthic systems in the northeast Pacific. In situ oxygen occurrences were not congruent with lethal and sublethal hypoxia thresholds calculated across the literature for major taxonomic groups indicating that research biases toward laboratory studies on Atlantic species are not globally applicable. Region-specific hypoxia thresholds are necessary to predict future impacts of deoxygenation on marine biodiversity.

Abstract:Oxygen minimum zones (OMZs) are permanent suboxic features of the oceanic water column that strongly influence zooplankton distributions and biogeochemical cycles. The lower interface of prominent OMZs is characterized by a subsurface zooplankton biomass peak and high biological activity. The calanoid copepod Lucicutia grandis is an indicator species for this habitat. Its ecology in the Arabian Sea was studied during the U.S. Joint Global Ocean Flux Study (JGOFS) program to understand planktonic distributional and developmental adaptations to oxygen gradients in suboxic environments and the role of the OMZ zooplankton in food webs, vertical flux processes, and carbon cycles. Zooplankton samples were obtained in vertically stratified multiple opening‐closing net and environmental sensing system (MOCNESS) tows to 1,000 m during four seasonal cruises. The vertical distribution of L. grandis was associated with the steep oxygen gradient from 0.07 to 0.15 ml L−1 at the base of the OMZ about 600–1,000 m. There was a clear progression with age of the depths and oxygen levels inhabited by different developmental stages within this zone, a phenomenon attributed to both physiological constraints and ecological interactions. The seasonal and spatial pattern of reproduction and development was keyed in part to the seasonal monsoon cycle, with final maturation of young stages into reproducing adults probably triggered by the direct and indirect effects of the seasonal or episodic input to depth of sinking particles. Gut contents included surface flux material, deep‐sea detrital material, zooplankton remains, and deep‐sea aggregate material, indicating that L. grandis occupies at least four different trophic levels. This was an active, not a diapausing, population, since both adults and immature stages fed and reproduced during all seasons. In contrast to the pelagic fauna that are severely impacted by coastal episodic hypoxia, the animals of the oceanic OMZ are uniquely adapted to the very low oxygen and strong spatial and temporal patterns of this widespread suboxic environment.

Abstract:Significance The loss of ocean oxygen caused by climate warming is a serious environmental issue and could lead to major declines in animal habitats. However, current Earth system models struggle in explaining observed trends in global ocean oxygen. Using a model, we demonstrate that marine oxygen concentrations depend sensitively on a rarely considered quantity known as the respiration quotient to prescribe the oxygen consumed per mole of organic carbon respired. Using a combination of direct chemical measurements across major biomes in the eastern Pacific Ocean plus changes in the global distribution of oxygen and carbon, we show how the respiration quotient increases systematically with temperature. The findings imply that we can experience more intense declines in ocean oxygen with warming.

Abstract:In the Eastern Tropical North Pacific Oxygen Minimum Zone (ETNP‐OMZ), fish larvae undergo development amidst highly variable dissolved oxygen environments. As OMZs expand, understanding the implications of low‐oxygen environments on fish development becomes increasingly relevant for fisheries management and ecosystem modeling. Abstract:The Eastern Tropical North Pacific (ETNP) is a large, persistent, and intensifying oxygen minimum zone (OMZ) that accounts for almost half of the total area of global OMZs. Within the OMZ core (∼350–700 m depth), dissolved oxygen is typically near or below the analytical detection limit of modern sensors (∼10 nM). Steep oxygen gradients above and below the OMZ core lead to vertical structuring of microbial communities that also vary between particle-associated (PA) and free-living (FL) size fractions. Here, we use 16S amplicon sequencing (iTags) to analyze the diversity and distribution of prokaryotic populations between FL and PA size fractions and among the range of ambient redox conditions. The hydrographic conditions at our study area were distinct from those previously reported in the ETNP and other OMZs, such as the ETSP. Trace oxygen concentrations (∼0.35 μM) were present throughout the OMZ core at our sampling location. Consequently, nitrite accumulations typically reported for OMZ cores were absent as were sequences for anammox bacteria (Brocadiales genus Candidatus Scalindua), which are commonly found across oxic-anoxic boundaries in other systems. However, ammonia-oxidizing bacteria (AOB) and archaea (AOA) distributions and maximal autotrophic carbon assimilation rates (1.4 μM C d–1) coincided with a pronounced ammonium concentration maximum near the top of the OMZ core. In addition, members of the genus Nitrospina, a dominant nitrite-oxidizing bacterial (NOB) clade were present suggesting that both ammonia and nitrite oxidation occur at trace oxygen concentrations. Analysis of similarity test (ANOSIM) and Non-metric Dimensional Scaling (nMDS) revealed that bacterial and archaeal phylogenetic representations were significantly different between size fractions. Based on ANOSIM and iTag profiles, composition of PA assemblages was less influenced by the prevailing depth-dependent biogeochemical regime than the FL fraction. Based on the presence of AOA, NOB and trace oxygen in the OMZ core we suggest that nitrification is an active process in the nitrogen cycle of this region of the ETNP OMZ.

Abstract:Recent changes in climate and eutrophication have caused increases in oxygen depletion in both freshwater and marine ecosystems. However, the impact of oxygen stress on zooplankton, which is the major trophic link between primary producers and fish, remains largely unknown in lakes. Therefore, we studied 41 lakes with different trophic and oxygen conditions to assess the role of oxygen stress on zooplankton communities and carbon transfer between phytoplankton and zooplankton. Samples were collected from each lake at the peak of summer stratification from three depth layers (the epilimnion, metalimnion, and hypolimnion). Our results revealed that freshwater zooplankton were relatively tolerant to anoxic conditions and the greatest changes in community structure were found in lakes with the highest oxygen deficits. This caused a switch in dominance from large to small species and reduced the zooplankton biomass in lower, anoxic layers of water, but not in the upper layers of water where the oxygen deficits began. This upper anoxic layer could thus be a very important refuge for zooplankton to avoid predation during the day. However, the reduction of zooplankton in the lower water layers was the main factor that reduced the effectiveness of carbon transfer between the phytoplankton and zooplankton.

Abstract:Ongoing ocean deoxygenation is threatening marine organisms globally. In eastern boundary upwelling systems, planktonic copepods dominate the epipelagic zooplankton, being crucial in the marine food web. Yet, they must cope with severe hypoxia caused by shoaling of the oxygen minimum zone. Based on laboratory experiments during 2021, we found differential responses in the metabolic rate (MR) and critical oxygen partial pressure of three abundant copepods. Calanoides patagoniensis doubled its MR during the upwelling season, so better exploiting the spring phytoplankton bloom for feeding and reproduction while maintaining their critical oxygen partial pressure unchanged between seasons. Contrastingly, Paracalanus cf. indicus and Acartia tonsa, maintained their MRs throughout seasons, but significantly increased their critical oxygen partial pressure during the upwelling period, becoming less tolerant to hypoxia. Field observations showed that oxygen levels equal to or lower than the critical oxygen partial pressure is a common condition (70% of occurrence) that copepods encounter during the year in the upper 50 m layer. These findings suggest a species‐dependent trade‐off between the MR and the critical oxygen partial pressure, where some species can maintain the latter despite fluctuations in their MR (improved hypoxia tolerance) or maintain their MR at the expenses of a larger critical oxygen partial pressure (improved energy expenditures). These adaptive responses, under oxygen levels equal to or lower than the critical oxygen partial pressure, suggest that exacerbated hypoxia, driven by ocean deoxygenation and increased upwelling, will alter copepod distribution and cause higher copepod mortality, with potentially drastic consequences for marine food webs.

Abstract:Secular decreases in dissolved oxygen concentration have been observed within the tropical oxygen minimum zones (OMZs) and at mid- to high latitudes over the last approximately 50 years. Earth system model projections indicate that a reduction in the oxygen inventory of the global ocean, termed ocean deoxygenation, is a likely consequence of on-going anthropogenic warming. Current models are, however, unable to consistently reproduce the observed trends and variability of recent decades, particularly within the established tropical OMZs. Here, we conduct a series of targeted hindcast model simulations using a state-of-the-art global ocean biogeochemistry model in order to explore and review biases in model distributions of oceanic oxygen. We show that the largest magnitude of uncertainty is entrained into ocean oxygen response patterns due to model parametrization of p CO 2 -sensitive C : N ratios in carbon fixation and imposed atmospheric forcing data. Inclusion of a p CO 2 -sensitive C : N ratio drives historical oxygen depletion within the ocean interior due to increased organic carbon export and subsequent remineralization. Atmospheric forcing is shown to influence simulated interannual variability in ocean oxygen, particularly due to differences in imposed variability of wind stress and heat fluxes. Microbial respiration in contrasting ocean provinces via high-frequency optode assays

Abstract:Microbial respiration is a critical component of the marine carbon cycle, determining the proportion of fixed carbon that is subject to remineralization as opposed to being available for export to the ocean depths. Despite its importance, methodological constraints have led to an inadequate understanding of this process, especially in low-activity oligotrophic and mesopelagic regions. Here, we quantify respiration rates as low as 0.2 μ mol O 2 L -1 d -1 in contrasting ocean productivity provinces using oxygen optode sensors to identify size-fractionated respiration trends. In the low productivity region of the North Pacific Ocean at Station Papa, surface whole water microbial respiration was relatively stable at 1.2 μ mol O 2 L -1 d -1 . Below the surface, there was a decoupling between respiration and bacterial production that coincided with increased phytodetritus and small phytoplankton. Size-fractionated analysis revealed that cells <5 μ m were responsible for the majority of the respiration in the Pacific, both at the surface and below the mixed layer. At the North Atlantic Porcupine Abyssal Plain, surface whole water microbial respiration was higher (1.7 μ mol O 2 L -1 d -1 ) than in the Pacific and decreased by 3-fold below the euphotic zone. The Atlantic size-fraction contributions to total respiration shifted on the order of days during the evolution of a phytoplankton bloom with regular storm disturbances. The high-resolution optode method used in the Atlantic captured these significant shifts and is consistent with coinciding stain-based respiration methods and historical site estimates. This study highlights the dynamic nature of respiration across vertical, temporal, and size-fractionated factors, emphasizing the need for sensitive, high-throughput techniques to better understand ocean ecosystem metabolism.

Abstract:The gravitational biological pump is not large enough to account for microbial heterotrophic activity in the mesopelagic ocean. Migrating zooplankton may be a key source of organic matter transport to depth. Here we show signatures of zooplankton in the suspended organic matter at the zooplankton vertical migration depth in the Eastern Tropical North Pacific Oxygen Deficient Zone (ETNP ODZ). We examine the mesozooplankton community in metagenomic depth profiles using the mitochondrial cytochrome c oxidase (COI) gene as a marker in the ETNP and Eastern Tropical South Pacific (ETSP) ODZs and at the oxic Hawaii Ocean Timeseries (HOT). Additionally, eukaryotic transcripts (polyA-selected) were examined for zooplankton in the ETNP. While zooplankton eDNA increased in the ODZ, zooplankton eRNA decreased in the ODZ, similar to previous net-based data, implying that eDNA is better preserved under anoxia. At all stations, Cnidaria, often missed in net-based data, contributed greatly to the zooplankton eDNA/eRNA. SAR11 abundance, determined from the single-copy core gene (rpoB), significantly correlated with zooplankton eDNA, with R2 values >0.8 at all stations. Strong correlations between SAR11 and zooplankton have not been previously reported, but are logical as SAR11 bacteria consume and zooplankton excrete simple dissolved organic compounds. SAR11 bacteria possessed genes to utilize urea and taurine in the ODZ, both compounds known to be excreted by zooplankton. In ODZs, SAR11 bacteria preferentially used the taurine degradation pathway leading to C and N assimilation, not the pathway for organic S assimilation, probably due to additional sources of organic S in ODZs.
Microbial community structures and important taxa across oxygen gradients in the Andaman Sea and eastern Bay of Bengal epipelagic waters

Abstract:In oceanic oxygen minimum zones (OMZs), the abundances of aerobic organisms significantly decrease and energy shifts from higher trophic levels to microorganisms, while the microbial communities become critical drivers of marine biogeochemical cycling activities. However, little is known of the microbial ecology of the Andaman Sea and eastern Bay of Bengal (BoB) OMZs. In the present study, a total of 131 samples which from the Andaman Sea and eastern BoB epipelagic waters were analyzed. The microbial community distribution patterns across oxygen gradients, including oxygenic zones (OZs, dissolved oxygen [DO] ≥ 2 mg/L), oxygen limited zones (OLZs, 0.7 mg/L < DO < 2 mg/L), and OMZs (DO ≤ 0.7 mg/L), were investigated. Mantel tests and Spearman's correlation analysis revealed that DO was the most important driver of microbial community structures among several environmental factors. Microbial diversity, richness, and evenness were highest in the OLZs and lowest in the OZs. The microbial community compositions of OZ and OMZ waters were significantly different. Random forest analysis revealed 24 bioindicator taxa that differentiated OZ, OLZ, and OMZ water communities. These bioindicator taxa included Burkholderiaceae, HOC36, SAR11 Clade IV, Thioglobaceae, Nitrospinaceae, SAR86, and UBA10353. Further, co-occurrence network analysis revealed that SAR202, AEGEAN-169, UBA10353, SAR406, and Rhodobacteraceae were keystone taxa among the entire interaction network of the microbial communities. Functional prediction further indicated that the relative abundances of microbial populations involved in nitrogen and sulfur cycling were higher in OMZs. Several microbial taxa, including the Thioglobaceae, Nitrospinaceae, SAR202, SAR406, WPS-2, UBA10353, and Woeseiaceae, may be involved in nitrogen and/or sulfur cycling, while also contributing to oxygen consumption in these waters. This study consequently provides new insights into the microbial community structures and potentially important taxa that contribute to oxygen consumption in the Andaman Sea and eastern BoB OMZ.

Abstract:Decline of the dissolved oxygen in the ocean is a growing concern, as it may eventually lead to global anoxia, an elevated mortality of marine fauna and even a mass extinction. Deoxygenation of the ocean often results in the formation of oxygen minimum zones (OMZ): large domains where the abundance of oxygen is much lower than that in the surrounding ocean environment. Factors and processes resulting in the OMZ formation remain controversial. We consider a conceptual model of coupled plankton–oxygen dynamics that, apart from the plankton growth and the oxygen production by phytoplankton, also accounts for the difference in the timescales for phyto- and zooplankton (making it a "slow-fast system") and for the implicit effect of upper trophic levels resulting in density dependent (nonlinear) zooplankton mortality. The model is investigated using a combination of analytical techniques and numerical simulations. The slow-fast system is decomposed into its slow and fast subsystems. The critical manifold of the slow-fast system and its stability is then studied by analyzing the bifurcation structure of the fast subsystem. We obtain the canard cycles of the slow-fast system for a range of parameter values. However, the system does not allow for persistent relaxation oscillations; instead, the blowup of the canard cycle results in plankton extinction and oxygen depletion. For the spatially explicit model, the earlier works in this direction did not take into account the density dependent mortality rate of the zooplankton, and thus could exhibit Turing pattern. However, the inclusion of the density dependent mortality into the system can lead to stationary Turing patterns. The dynamics of the system is then studied near the Turing bifurcation threshold. We further consider the effect of the self-movement of the zooplankton along with the turbulent mixing. We show that an initial non-uniform perturbation can lead to the formation of an OMZ, which then grows in size and spreads over space. For a sufficiently large timescale separation, the spread of the OMZ can result in global anoxia.

Abstract:Oxygen minimum zones (OMZ) are characterized by a dissolved oxygen (DO) concentration <0.5 ml L −1 . In situ DO measurements suggest that the global OMZ upper limit is shallower than 60 years ago due to global warming. The benthic foraminifera assemblage, trace elements, and biological productivity and denitrification tracers have been used in paleoceanographic research, given that they indicate dysoxic and suboxic changing conditions of bottom water. Studies of benthic foraminifera assemblages in the southwestern margin of Baja California Sur are scarce. Thus, it is necessary to conduct a paleoecological analysis of the benthic foraminifera community on an approximately 1200-year timeline. The sediment core LONO09-MC02 (1200 years old) collected at 680 m depth was examined to analyze benthic foraminifera relative abundances and estimate the DO concentration in the OMZ of Baja California Sur's southwestern margin. The DO concentrations inferred from benthic foraminifera assemblages ranged between 0.09- and 0.17-ml l −1 (4.4 and 8.0 μM). This suggests that the OMZ has maintained dysoxic conditions over the past 1200 years. Moreover, the cluster and principal component analysis defined dysoxic, suboxic, and oxic assemblages. The decoupling between geochemical tracers was associated with biogeochemical cycles of the North Pacific subtropical gyre and varied in response to intermediate water circulation in the Pacific Ocean. The low estimate of dissolved oxygen concentration corresponded with minimal solar activity, favoring La Niña-like oceanographic conditions with high primary productivity.

Abstract:Global warming and local eutrophication simultaneously lower oxygen (O2) saturation and increase biological O2 demands to cause deoxygenation. Tropical shallow waters, and their coral reefs, are particularly vulnerable to extreme low O2 (hypoxia) events. These events can drive mass mortality of reef biota; however, they currently remain unaccounted for when considering coral reef persistence under local environmental alterations and global climatic change. In this Perspective, we integrate existing biological, ecological and geochemical evidence to consider how O2 availability and hypoxia affect reef biota, with particular focus on the ecosystem architects, reef-building corals, that operate as both O2 consumers and producers. We pinpoint fundamental knowledge gaps and highlight the need to understand sub-lethal hypoxia effects that are likely already in play.

Abstract:Hypoxia is one of the predominant water quality issues affecting estuaries and coastal ecosystems, and its impact is often monitored using benthic macroinvertebrates. The M-AMBI (Multivariate AZTI Marine Biotic Index) is an index that meets the needs of small and large-scale monitoring as it is scalable. However, gaps remain as to the sensitivity of M-AMBI to hypoxia as few studies are available. Using Pensacola Bay in the northern Gulf of Mexico (USA) as a case study, we sought to evaluate the time scales over which benthic macrofauna respond to dissolved oxygen conditions from May through September 2017. Combined continuous DO monitoring and benthic sampling identified important differences in DO exposure on benthic habitat condition based on both the duration and frequency of low oxygen. We identified periods of 7 to 31 days as critical windows of exposure prior to a measurable benthic response, and that both duration and exposure to varying low oxygen conditions as well as the recovery period of oxygen to > 5 mg L −1 are important to benthic habitat health. While the duration of exposure to DO from < 2 mg L −1 to near anoxia remains an important factor in benthic health, benthic organisms can better tolerate periods of low oxygen when reoxygenation occurs after a short time interval. More research is needed to better quantify the relationship between oxygen stress and recovery on benthic habitats, particularly in systems where low DO exposure and recovery can vary over timescales of hours to days.

Abstract:Oxygen minimum zones (OMZs) are hot spots for redox-sensitive nitrogen transformations fueled by sinking organic matter. In comparison, the regulating role of sulfur-cycling microbes in marine OMZs, their impact on carbon cycling in pelagic and benthic habitats, and activities below the seafloor remain poorly understood. Using 13C DNA stable isotope probing (SIP) and metatranscriptomics, we explored microbial guilds involved in sulfur and carbon cycling from the ocean surface to the subseafloor on the Namibian shelf. There was a clear separation in microbial community structure across the seawater-seafloor boundary, which coincided with a 100-fold-increased concentration of microbial biomass and unique gene expression profiles of the benthic communities. 13C-labeled 16S rRNA genes in SIP experiments revealed carbon-assimilating taxa and their distribution across the sediment-water interface. Most of the transcriptionally active taxa among water column communities that assimilated 13C from diatom exopolysaccharides (mostly Bacteroidetes, Actinobacteria, Alphaproteobacteria, and Planctomycetes) also assimilated 13C-bicarbonate under anoxic conditions in sediment incubations. Moreover, many transcriptionally active taxa from the seafloor community (mostly sulfate-reducing Deltaproteobacteria and sulfide-oxidizing Gammaproteobacteria) that assimilated 13C-bicarbonate under sediment anoxic conditions also assimilated 13C from diatom exopolysaccharides in the surface ocean and OMZ waters. Despite strong selection at the sediment-water interface, many taxa related to either planktonic or benthic communities were found to be present at low abundance and actively assimilating carbon under both sediment and water column conditions. In austral winter, mixing of shelf waters reduces stratification and suspends sediments from the seafloor into the water column, potentially spreading metabolically versatile microbes across niches. IMPORTANCE Microbial activities in oxygen minimum zones (OMZs) transform inorganic fixed nitrogen into greenhouse gases, impacting the Earth's climate and nutrient equilibrium. Coastal OMZs are predicted to expand with global change and increase carbon sedimentation to the seafloor. However, the role of sulfur-cycling microbes in assimilating carbon in marine OMZs and related seabed habitats remain poorly understood. Using 13C DNA stable isotope probing and metatranscriptomics, we explore microbial guilds involved in sulfur and carbon cycling from ocean surface to subseafloor on the Namibian shelf. Despite strong selection and differential activities across the sediment-water interface, many active taxa were identified in both planktonic and benthic communities, either fixing inorganic carbon or assimilating organic carbon from algal biomass. Our data show that many planktonic and benthic microbes linked to the sulfur cycle can cross redox boundaries when mixing of the shelf waters reduces stratification and suspends seafloor sediment particles into the water column.
