Abstract:Lipase is the key enzyme for catalyzing triglyceride hydrolysis in vivo, and lipase inhibitors have been used in the management of obesity. We present the first report on the use of lipase-adsorbed halloysite nanotubes as an efficient medium for the selective enrichment of lipase inhibitors from natural products. A simple and rapid approach was proposed to fabricate lipase-adsorbed nanotubes through electrostatic interaction. Results showed that more than 85% lipase was adsorbed into nanotubes in 90 min, and approximately 80% of the catalytic activity was maintained compared with free lipase. The specificity and reproducibility of the proposed approach were validated by screening a known lipase inhibitor (i.e., orlistat) from a mixture that contains active and inactive compounds. Moreover, we applied this approach with high performance liquid chromatography-mass spectrometry technique to screen lipase inhibitors from the Magnoliae cortex extract, a medicinal plant used for treating obesity. Two novel biphenyl-type natural lipase inhibitors magnotriol A and magnaldehyde B were identified, and their IC50 values were determined as 213.03 and 96.96 mu M, respectively. The ligand-enzyme interactions of magnaldehyde B were further investigated by molecular docking. Our findings proved that enzyme-adsorbed nanotube could be used as a feasible and selective affinity medium for the rapid screening of enzyme inhibitors from complex mixtures. (C) 2015 Elsevier B.V. Abstract:Fe 3 O 4 /poly(styrene-co-maleic anhydride) core-shell composite microspheres, suitable for binding enzymes, were prepared using magnetite particles as seeds by copolymerization of styrene and maleic anhydride. The magnetite particles were encapsulated by polyethylene glycol, which improved the affinity between the magnetite particles and the monomers, thus showing that the size of the microspheres, the amount of the surface anhydrides, and the magnetite content in the composite are highly dependent on magnetite particles, comonomer ratio, and dispersion medium used in the polymerization. The composite microspheres, having 0.08-0.8 μm diameter and containing 100-800 μg magnetite/g microspheres and 0-18 mmol surface-anhydride groups/g microsphere, were obtained. Free α-amylase was immobilized on the microspheres containing reactive surface-anhydride groups by covalent binding. The effects of immobilization on the properties of the immobilized α-amylase [magnetic immobilized enzyme (MIE)] were studied. The activity of MIE and protein binding capacity reached 113,800 U and 544.3 mg/g dry microspheres, respectively. The activity recovery was 47.2%. The MIE had higher optimum temperature and pH compared with those of free α-amylase and showed excellent thermal, storage, pH, and operational stability. Furthermore, it can be easily separated in a magnetic field and reused repeatedly.

Abstract:Abstract By the facile adhesion way, the novel composite complex by polydopamine (PDA) and magnetic graphite nanosheets (Fe3O4@GNSs) has been successfully synthesized. The resulting composite was characterized by means of scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectra, and Raman spectra, X-ray diffraction, X-ray photoelectron spectroscopy, and vibrating sample magnetometry. Meanwhile, the PDA functionalized Fe3O4@GNSs (Fe3O4@GNSs-PDA) was applied for Candida rugosa lipase (CRL) immobilization covalently without any toxic coupling agent. Combining the superior physical properties and chemical stability of Fe3O4@GNSs and the well biocompatibility, functional characteristics of PDA, the Fe3O4@GNSs-PDA composite displayed several advantages, including the high enzyme capacity, enzyme activity and stability and a decrease in enzyme loss. Our work demonstrated that the mussel-inspired Fe3O4@GNSs can be extended to many other applications such as biocatalytic, genetic and industrial.

Abstract:Metal-organic frameworks (MOFs) have shown great potential as supports for enzyme immobilization, but their application is limited by separation difficulties due to their nanoscale size. Herein, magnetic MOFs (Fe 3 O 4 @MIL-53-NH 2 (Al)) were prepared under mild conditions, and an ionic liquid (IL) was used as linker to covalently immobilize lipase in Fe 3 O 4 @MIL-53-NH 2 -IL/PPL. The IL was composed of imidazolium cations and bis(trifluoromethylsulfonyl)imide anions, which enhanced the hydrophobicity of the support. The synthesized biocatalyst Fe 3 O 4 @MIL-53-NH 2 -IL/PPL showed excellent activity, 2.4 times higher than free lipase, and its denaturation, thermal, and storage stability were all satisfactory. The activity of the biocatalyst remained at up to 88.4% after ten recycles. The excellent magnetic responsiveness facilitated the collection of the biocatalyst, which may be beneficial to its reusability, and the multiple interactions between lipase and support may enhance the structural stability of lipase, which was investigated by circular dichroism spectroscopy. Compared to free lipase, both the substrate affinity and catalytic efficiency of Fe 3 O 4 @MIL-53-NH 2 -IL/PPL were improved. The magnetic MOF composite materials prepared through the post-synthesis method exhibited good potential in the field of lipase immobilization. The preparation method of immobilized lipase in this work provides a meaningful reference for the application of other MOF composites in the field of enzyme immobilization.

Abstract:Various hydrophobic supports have been used for lipase immobilization since the active site of lipase can be opened in a hydrophobic environment. Nevertheless, the increase of lipase activity is still limited. This study demonstrates a hyperactivation-protection strategy of lipase after immobilization on poly(n-butyl acrylate)-polyaldehyde dextran (PBA-PAD) core-shell nanoparticles. The inner hydrophobic PBA domain helps to rearrange lipase conformation to a more active form after immobilization into the PAD shell. More importantly, the outer PAD shell with dense polysaccharide chains prevents the immobilized lipase from contact with outside aqueous medium and revert its conformation back to an inactive form. As a result, under optimal conditions the activity of lipase immobilized in PBA-PAD nanoparticles was enhanced 40 times over the free one, much higher than in any previous report. Furthermore, the immobilized lipase retained more than 80 % of its activity after 10 reaction cycles.
Calcium-based MOFs as scaffolds for shielding immobilized lipase and enhancing its stability

Abstract:The enzyme immobilization technology has become a key tool in the field of enzyme applications; however, improving the activity recovery and stability of the immobilized enzymes is still challenging. Herein, we employed a magnetic carboxymethyl cellulose (MCMC) nanocomposite modified with ionic liquids (ILs) for covalent immobilization of lipase, and used Ca-based metal-organic frameworks (MOFs) as the support skeleton and protective layer for immobilized enzymes. The ILs contained long side chains (eight CH2 units), which not only enhanced the hydrophobicity of the carrier and its hydrophobic interaction with the enzymes, but also provided a certain buffering effect when the enzyme molecules were subjected to compression. Compared to free lipase, the obtained CaBPDC@PPL-IL-MCMC exhibited higher specific activity and enhanced stability. In addition, the biocatalyst could be easily separated using a magnetic field, which is beneficial for its reusability. After 10 cycles, the residual activity of CaBPDC@PPL-IL-MCMC could reach up to 86.9%. These features highlight the good application prospects of the present immobilization method.

Abstract:<p>Compared to the traditional chemical reduction method, a green and efficient strategy was successfully applied to regulate the surface heterogeneity of graphene oxide (GO). In this work, different kinds of amino acids were used to reduce and modify GO. The reduction and adsorption disparities of amino acids contributed to the difference of structural heterogeneity in terms of density, thermal tolerance and disorder of the oxygen-containing groups of GO. The modification also resulted in discrepancies in surface properties of the GO samples, such as hydrophobicity and surface charge. The functionalized GO was employed as a carrier for the immobilization of lipase from <em>Thermomyces lanuginosus</em> (lipase TL). The storage stability, the temperature and pH sensitivity of the immobilized biocatalysts and the free lipase TL were studied. Lipase immobilized on functionalized GO demonstrates superior enzymatic loading and relative activity compared to GO, with a maximum increase of 70% and 47%, respectively. The relative activity of lipase immobilized on functionalized GO was 19.21 times more than that of commercial immobilized lipase TLIM. Through molecular dynamics simulations we concluded that the increase in the relative activity of lipase TL is mainly related to its adsorption orientation and slight conformational change. Thus, our research offered a green and effective strategy to regulate the heterogeneity of nanomaterials for a suitable microenvironment to enhance the performance of immobilized lipase.</p>

Tailoring a robust nanozyme formulation based on surfactant stabilized lipase immobilized onto newly fabricated magnetic silica anchored graphene nanocomposite: Aggrandized stability and application

Abstract:Candida rugosa lipase (CRL) was treated with surfactants and immobilized onto the novel formulated magnetic graphene anchored silica nanocomposite (Fe3O4/SiO2/Gr NC). For this purpose, the surface of lipase was initially coated with Triton-X 100 and cetyltrimethylammonium bromide surfactants, to stabilize enzyme in its open form and was then adsorbed onto aminated Fe3O4/SiO2/Gr NC. Glutaraldehyde (GA) was then utilized to cross-link the adsorbed lipase onto the NC. The fabricated NC and conjugated lipase was characterized by various techniques such as FT-IR, XRD, TGA, SEM, TEM, CLSM, CD and Fluorescence spectroscopy. The magnetic character of the as-synthesized NC was verified by AGM investigation. CD and fluorescence spectroscopic analysis demonstrated slight structural rearrangements in lipase upon conjugation. The surfactant stabilized immobilized lipase demonstrated significantly enhanced thermostability, tolerance to various metal ions and inhibitors. The immobilization yield obtained owing to lipase interfacial activation by Triton X 100 and CTAB was remarkably enhanced by 6-folds and 3-folds, respectively which were remarkably higher in comparison to free immobilized lipase. The fabricated nanobiocatalysts were employed to synthesise green apple flavour ester, ethyl valerate via esterification reaction. Triton X 100 stabilized immobilized lipase was a better performer in yielding green apple flavour ester, demonstrating about 90% ester yield as compared to 78% yield obtained by CTAB stabilized immobilized lipase preparation. The obtained outcomes suggested that enzyme structure was stabilized by the GA treatment if executed in the absence or in the presence of detergent, and that, in the company of detergent, a conformation of the lipase with the exposed active center to the medium provided an aggrandized catalytic performance.

Abstract:The work reported in this paper is aimed at exploring the feasibility of immobilizing alkali lipase from Penicillium expansum on a bimodal ceramic foam, which has both macro- and micro-pore structures. After being chemically modified with a silane coupling agent, the ceramic foam was used as a support for lipase immobilization. To determine the preferable immobilization conditions, the effects of the amount of enzyme for loading, immobilization time, temperature, and pH on enzyme activity were investigated. The results showed that the chemically modified ceramic foam has a high loading capacity and a strong binding ability for the lipase. Thanks to the low internal mass transfer resistance, the ceramic foam has greatly enhanced the rate of immobilization. As a comparison, the immobilized-lipase activity was much higher than that on many frequently used porous materials like diatomite, alumina and activated carbon.

Abstract:A nanocarrier Fe3O4@OA@DA, with the structure of a dopamine functionalized Fe3O4 nanoparticles coated with oleic acid (OA), was simply prepared by dipping the Fe3O4@OA magnetic nanoparticles in dopamine solution. Lipase from Thermomyces lanuginosus was immobilized onto the obtained nanocarrier via covalent coupling. A high immobilization efficiency of 88%, and an immobilization yield of 97% with enzyme loading of 13.09 mg/g-support were achieved under the optimal conditions: immobilization time 10 h, pH 7.0, and enzyme concentration 0.27 mg/mL. Compared with the free enzyme, thermostability of the immobilized lipase was 2.9-fold improved at 60 degrees C. More than 75% initial activity of the immobilized lipase was remained after ten repeated uses. The V-max and K-m of the free and immobilized lipase were determined as 2.51 mmol/min, 2.13 mmol/min and 8.86 mM, 10.50 mM, respectively. These results indicated that the prepared magnetic oleic acid nanoparticle with dopamine functionalization is a good carrier for lipase immobilization, and has the potential uses for immobilization of other enzymes.
Preparation and Characterization of Magnetic Metal–Organic Frameworks Functionalized by Ionic Liquid as Supports for Immobilization of Pancreatic Lipase

Abstract:Enzymes are difficult to recycle, which limits their large-scale industrial applications. In this work, an ionic liquid-modified magnetic metal–organic framework composite, IL-Fe3O4@UiO-66-NH2, was prepared and used as a support for enzyme immobilization. The properties of the support were characterized with X-ray powder diffraction (XRD), Fourier-transform infrared (FTIR) spectra, transmission electron microscopy (TEM), scanning electronic microscopy (SEM), and so on. The catalytic performance of the immobilized enzyme was also investigated in the hydrolysis reaction of glyceryl triacetate. Compared with soluble porcine pancreatic lipase (PPL), immobilized lipase (PPL-IL-Fe3O4@UiO-66-NH2) had greater catalytic activity under reaction conditions. It also showed better thermal stability and anti-denaturant properties. The specific activity of PPL-IL-Fe3O4@UiO-66-NH2 was 2.3 times higher than that of soluble PPL. After 10 repeated catalytic cycles, the residual activity of PPL-IL-Fe3O4@UiO-66-NH2 reached 74.4%, which was higher than that of PPL-Fe3O4@UiO-66-NH2 (62.3%). In addition, kinetic parameter tests revealed that PPL-IL-Fe3O4@UiO-66-NH2 had a stronger affinity to the substrate and, thus, exhibited higher catalytic efficiency. The results demonstrated that Fe3O4@UiO-66-NH2 modified by ionic liquids has great potential for immobilized enzymes.

Abstract:Surface chemistry of carriers plays a key role in enzyme loading capacity, structure rigidity, and thus catalyze activity of immobilized enzymes. In this work, the two model enzymes of horseradish peroxidase (HRP) and glucose oxidase (GOx) are co-immobilized on the lysozyme functionalized magnetic core-shell nanocomposites (LYZ@MCSNCs) to enhance their stability and activity. Briefly, the HRP and GOx aggregates are firstly formed under the crosslinker of trimesic acid, in which the loading amount and the rigidity of the enzyme can be further increased. Additionally, LYZ easily forms a robust anti-biofouling nanofilm on the surface of SiO 2 @Fe 3 O 4 magnetic nanoparticles with abundant functional groups, which facilitate chemical crosslinking of HRP and GOx aggregates with minimized inactivation. The immobilized enzyme of HRP-GOx@LYZ@MCSNCs exhibited excellent recovery activity (95.6 %) higher than that of the free enzyme (HRP&GOx). Specifically, 85 % of relative activity was retained after seven cycles, while 73.5 % of initial activity was also remained after storage for 33 days at 4 °C. The thermal stability and pH adaptability of HRP-GOx@LYZ@MCSNCs were better than those of free enzyme of HRP&GOx. This study provides a mild and ecofriendly strategy for multienzyme co-immobilization based on LYZ functionalized magnetic nanoparticles using HRP and GOx as model enzymes.

Abstract:Interactions of enzymes with supports significantly affect the activity and stability of immobilized enzymes. Herein, amino-functionalized ionic liquid (IL)-grafted magnetic zeolitic imidazolate framework-90 (MZIF-90) was prepared and used to immobilize porcine pancreatic lipase (PPL). The nanocomposites were fully characterized; meanwhile, the interactions between ILs and ZIF-90 were calculated based on density functional theory. The prepared biocatalyst (PPL-ILs/MZIF-90) had a lipase loading of 178.3 mg/g and hydrolysis activity up to 287.5 U/g. When the biocatalyst was used to synthesize isoamyl acetate, the reaction media, molar ratio of alcohol/acid, temperature, and reaction time were optimized. Under the optimized reaction conditions (in hexane, alcohol/acid = 3:1, under 45 °C, reacted for 9 h), the ester yield reached 85.5%. The results of the stability test showed that PPL-ILs/MZIF-90 retained 88.7% of the initial activity after storing for 35 days and 92.5% of the initial activity after reusing for seven cycles for synthesizing isoamyl acetate. Moreover, the secondary structure analysis showed that the synthesized supports protected the active conformation of immobilized lipase, which lead to the enhanced catalytic performance. Additionally, the biocatalyst can be easily separated with a magnet, which facilitated the reusability. This study provides insights regarding the application of metal organic framework composites in the field of enzyme catalysis.
In Situ Preparation of Magnetic Fe3O4-chitosan Nanoparticles for Lipase Immobilization by Cross-Linking and Oxidation in Aqueous Solution

Abstract:A new and simple method has been proposed to prepare magnetic Fe(3)O(4)-chitosan (CS) nanoparticles by cross-linking with sodium tripolyphosphate (TPP), precipitation with NaOH and oxidation with O(2) in hydrochloric acid aqueous phase containing CS and Fe(OH)(2), and these magnetic CS nanoparticles were used to immobilize lipase. The effects on the sequence of adding NaOH and TPP, the reaction temperature, and the ratio of CS/Fe(OH)(2) were studied. TEM showed that the diameter of composite nanoparticles was about 80 nm, and that the magnetic Fe(3)O(4) nanoparticles with a diameter of 20 nm were evenly dispersed in the CS materials. Magnetic measurement revealed that the saturated magnetisation of the Fe(3)O(4)-CS nanoparticles could reach 35.54 emicro/g. The adsorption capacity of lipase onto nanoparticles could reach 129 mg/g; and the maximal enzyme activity was 20.02 micromol min(-1)mg(-1) (protein), and activity retention was as high as 55.6% at a certain loading amount.

Abstract:BACKGROUND: Biocatalytic processes often require a full recycling of biocatalysts to optimize economic benefits and minimize waste disposal. Immobilization of biocatalysts onto particulate carriers has been widely explored as an option to meet these requirements. However, surface properties often affect the amount of biocatalysts immobilized, their bioactivity and stability, hampering their wide applications. The aim of this work is to explore how immobilization of lipases onto magnetite nanoparticles affects their biocatalytic performance under carefully controlled surface modification.METHODOLOGY/PRINCIPAL FINDINGS: Magnetite nanoparticles, prepared through a co-precipitation method, were coated with alkyl silanes of different alkyl chain lengths to modulate their surface hydrophobicity. Candida rugosa lipase was then directly immobilized onto the modified nanoparticles through hydrophobic interaction. Enzyme activity was assessed by catalytic hydrolysis of p-nitrophenyl acetate. The activity of immobilized lipases was found to increase with increasing chain length of the alkyl silane. Furthermore, the catalytic activities of lipases immobilized on trimethoxyl octadecyl silane (C18) modified Fe(3)O(4) were a factor of 2 or more than the values reported from other surface immobilized systems. After 7 recycles, the activities of the lipases immobilized on C18 modified nanoparticles retained 65%, indicating significant enhancement of stability as well through hydrophobic interaction. Lipase immobilized magnetic nanoparticles facilitated easy separation and recycling with high activity retaining.CONCLUSIONS/SIGNIFICANCE: The activity of immobilized lipases increased with increasing alkyl chain length of the alkyl trimethoxy silanes used in the surface modification of magnetite nanoparticles. Lipase stability was also improved through hydrophobic interaction. Alkyl silane modified magnetite nanoparticles are thus highly attractive carriers for enzyme immobilization enabling efficient enzyme recovery and recycling.

Abstract:A core/sheath nanofiber membrane with poly[bis(p-methylphenoxy)]phospha-zene (PMPPh) as the sheath and easily spinnable polyacrylonitrile (PAN) as the core was prepared via a coaxial electrospinning process. Field-emission scanning electron microscopy and transmission electron microscopy were used to characterize the morphology of the nanofiber membrane. It was found that the concentration of the PAN spinning solution and the ratio of the core/sheath solution flow rates played a decisive role in the coaxial electrospinning process. In addition, the stabilized core/sheath PMPPh nanofiber membrane was investigated as a support for enzyme immobilization because of its excellent biocompatibility, high surface/volume ratio, and large porosity. Lipase from Candida rugosa was immobilized on the nanofiber membrane by adsorption. The properties of the immobilized lipase on the polyphosphazene nanofiber membrane were studied and compared with those of a PAN nanofiber membrane. The results showed that the adsorption capacity (20.4 ± 2.7 mg/g) and activity retention (63.7%) of the immobilized lipase on the polyphosphazene nanofiber membrane were higher than those on the PAN membrane.

Abstract:Magnetic porous hydrophobic microspheres were prepared by modified suspension copolymerization of methacrylate (MMA) and divinylbenzene (DVB) in the presence of oleic acid coated magnetite (Fe 3 O 4 ), and the microspheres were used as biocarrier for the lipase immobilization. The results showed that the magnetic microspheres possessed spherical shape, core/shell structure, porous structure and high magnetic content, and the size and structure of magnetic microspheres had no significant changes after enzyme binding. The particle average size of microspheres was 66 μm, the magnetic content of microspheres was up to 31%, and the magnetization saturation values of the core/shell magnetic microspheres were measured at 300 K to be 11.02 emu g −1 . Lipase was immobilized on the magnetic porous carrier at up to 16.30 mg/g carrier. Activity and enantioselectivity of the immobilized lipase for the synthesis of R-HMPC acetate were investigated, indicating an interfacial activation of the enzyme after immobilization. Moreover, the pH dependency and operational stability of the immobilized lipase were studied, and they possess high stability and can be reused for ten cycles with loss 10% activity.
Lipase/tannic acid magnetic hydrogel microspheres and its continuous catalytic application

Abstract:The immobilization of enzyme by covalent bond usually uses toxic chemicals, which not only reduces the enzyme activity, but also is not conducive to its application in the food field. In this work, the surface of magnetic hydrogel microspheres was modified with tannic acid, which was oxidized into quinone, and then covalently linked to the lipase through Schiff base reaction. Further using the water absorption and water retention characteristics of hydrogel microspheres, a micro-water-oil interface can be provided for lipase in the organic phase reaction system, which provides favorable conditions for opening the α -helix "lid" of lipase active center and exposing the active center. Meanwhile, the synthesis of esterification catalyzed by lipase at the oil-water interface was promoted. In comparison to free lipase, the produced lipase/tannic acid magnetic hydrogel microspheres (L-PTAM2) exhibited superior temperature and pH stability and can basically sustain the activity of free lipase. After 20 consecutive cycles, L-PTAM2 still maintained about 57% of its original activity and could be quickly separated from the product. It was used for continuous catalytic production of benzyl acetate with a yield of 82%.
