Abstract:Feeding silkworms with functional materials as additives to produce naturally modified silk is a facile, diverse, controllable, and environmentally friendly method with a low cost of time and investment. Among various additives, carbon dots (CDs) show unique advantages due to their excellent biocompatibility and fluorescence stability. Here, a new type of green fluorescent carbon dots (G-CDs) is synthesized with a high oil-water partition ratio of 147, a low isoelectric point of 5.16, an absolute quantum yield of 71%, and critically controlled surface states. After feeding with G-CDs, the silkworms weave light yellow cocoons whose green fluorescence is visible to the naked eye under UV light. The luminous silk is sewn onto the cloth to create striking patterns with beautiful fluorescence. Such G-CDs have no adverse effect on the survival rate and the life cycle of silkworms and enable their whole bodies to glow under UV light. Based on the strong fluorescence, chemical stability, and biological safety, G-CDs are found in the digestive tracts, silk glands, feces, cocoons, and even moth bodies. G-CDs accumulate in the posterior silk glands where fibroin protein is secreted, indicating its stronger combination with fibroin than sericin, which meets the requirements for practical applications.
Silkworms Transformed with Chimeric Silkworm/spider Silk Genes Spin Composite Silk Fibers with Improved Mechanical Properties

Abstract:The development of a spider silk-manufacturing process is of great interest. However, there are serious problems with natural manufacturing through spider farming, and standard recombinant protein production platforms have provided limited progress due to their inability to assemble spider silk proteins into fibers. Thus, we used piggyBac vectors to create transgenic silkworms encoding chimeric silkworm/spider silk proteins. The silk fibers produced by these animals were composite materials that included chimeric silkworm/spider silk proteins integrated in an extremely stable manner. Furthermore, these composite fibers were, on average, tougher than the parental silkworm silk fibers and as tough as native dragline spider silk fibers. These results demonstrate that silkworms can be engineered to manufacture composite silk fibers containing stably integrated spider silk protein sequences, which significantly improve the overall mechanical properties of the parental silkworm silk fibers.

Abstract:Comprehensive SummaryFluorescent silk has potential application in many fields such as bioimaging, tissue engineering scaffolds, luminescent marks, and dazzling fabrics. Among the methods to endow natural silk with fluorescent properties, feeding silkworms with fluorescent additives is facile, low‐cost and environment friendly, which has the prospect of large‐scale production. In this paper, we reviewed the research progress for this aim in the past ten years, and summarized the unified characteristics for the substances that can enter the silk gland by digestive tract of silkworms. The advantages and disadvantages of various fluorescent materials for this application are compared in detail. And the future research directions are suggested to overcome the shortcomings of the present research.

Abstract:Functional fluorescent silkworm silk holds promise for many important applications in biomedical engineering, optics, and photonics. However, it remains a challenge to obtain fluorescent silk in scale-up with both good mechanical properties and highly stable fluorescence simultaneously. In this work, we report a highly efficient strategy to produce fluorescent silk through directly feeding silkworm larvae with graphene quantum dots or CdSe/ZnS core–shell quantum dots. The obtained quantum dots-reinforced luminescent silkworm silk has superior mechanical strength and toughness, stable fluorescence, and good biocompatibility in comparison with the normal or fluorescent dye-colored silk. The strategy proposed in this work is environmental and economical and, also importantly, can generate superior luminescent silks in large scale. This study also provides possible cues for fabricating durable, fluorescent microdevices, and fabrics.

Abstract:Abstract Fluorescent silk is fundamentally important for the development of future tissue engineering scaffolds. Despite great progress in the preparation of a variety of colored silks, fluorescent silk with enhanced mechanical properties has yet to be explored. In this study, we report on the fabrication of intrinsically super-strong fluorescent silk by feeding Bombyx mori silkworm carbon nanodots (CNDs). The CNDs were incorporated into silk fibroin, hindering the conformation transformation, confining crystallization, and inducing orientation of mesophase. The resultant silk exhibited super-strong mechanical properties with breaking strength of 521.9 ± 82.7 MPa and breaking elongation of 19.2 ± 4.3%, improvements of 55.1% and 53.6%, respectively, in comparison with regular silk. The CNDs-reinforced silk displayed intrinsic blue fluorescence when exposed to 405 nm laser and exhibited no cytotoxic effect on cells, suggesting that multi-functional silks would be potentially useful in bioimaging and other applications.

Abstract:Fluorescent polymer dots (Pdots) have the advantages of excellent optical properties, great biocompatibility and high photostability. Herein, we feed ultra-low doses Pdots to silkworms for the first time and aim to prepare dual-performance modified silks. After Pdots feeding, the fluorescence signal of cocoons and degummed silks increases significantly, which is more stable and more uniform than that of post-treatment silks. Moreover, Pdots hinder the conformation transformation of silk fibroin and improve the mechanical property of twisted silk strand. The highest elongation at break point is 20.75 ± 0.03% and breaking strength is 271.7 ± 3.8 MPa. With excellent fluorescence and mechanical properties, the optimized silks are successfully applied as a scaffold for cell culture and imaging. Furthermore, we investigate the metabolism of Pdots in the silkworms for understanding the behaviours of Pdots in the process of silks synthesis and secretion.

Abstract:Silkworm silk fibers have been woven into textiles for thousands of years, because of their attractive luster, good mechanical properties, excellent biocompatibility, and large-scale production. With the development of human society, preparation of silk fibers with modified or enhanced properties are highly desirable for potential applications in structural materials and smart textiles. Herein, we realized the reinforcement of multiple properties of silk fibers by feeding silkworms with Ag nanowire (Ag NW) modified diets. The obtained silk fibers show obviously enhanced comprehensive mechanical properties, including improved tensile strength, elongation at break, tensile modulus, and toughness, which are increased by 37.2%, 37.6%, 68.3%, and 69.8%, respectively. Furthermore, compared with unmodified silk, the electrical conductivity and thermal conductivity of modified silk fibers are improved by 246.4% and 32.1%, respectively. The analysis on the components and structure shows that the incorporated Ag NWs lead to increased content of random coil/α-helix, improved orientation of crystallites, and increased content of Ag compared to pristine silk fibers, which may contribute to the enhanced mechanical, electrical, and thermal properties.Graphical Abstract

Abstract:Silkworm silk is gaining significant attention from both the textile industry and research society because of its outstanding mechanical properties and lustrous appearance. The possibility of creating tougher silks attracts particular research interest. Carbon nanotubes and graphene are widely studied for their use as reinforcement. In this work, we report mechanically enhanced silk directly collected by feeding Bombyx mori larval silkworms with single-walled carbon nanotubes (SWNTs) and graphene. We found that parts of the fed carbon nanomaterials were incorporated into the as-spun silk fibers, whereas the others went into the excrement of silkworms. Spectroscopy study indicated that nanocarbon additions hindered the conformation transition of silk fibroin from random coil and alpha-helix to beta-sheet, which may contribute to increased elongation at break and toughness modules. We further investigated the pyrolysis of modified silk, and a highly developed graphitic structure with obviously enhanced electrical conductivity was obtained through the introduction of SWNTs and graphene. The successful generation of these SWNT- or graphene-embedded silks by in vivo feeding is expected to open up possibilities for the large-scale production of high-strength silk fibers.

Abstract:Carbon dots (CDs) are widely studied for years due to their unique luminescent properties and potential applications in many fields. However, aggregation-caused quenching, monotonous emission modes, and unsustainable preparation impose restrictions on their performance and practical applications. Here, this work reports the facile synthesis of sustainable silk-derived multimode emitting CDs with dispersed-state fluorescence (DSF), aggregation-induced fluorescence (AIF), and aggregation-induced room temperature phosphorescence (AIRTP) through radiating sericin proteins in a household microwave oven (800 W, 2.5 min). The structure, luminescent properties, and the mechanism are investigated and discussed. The sericin-derived CDs have graphitized cores and heteroatom-cluster-rich surfaces. The DSF corresponds to the graphitized cores and the AIF origins from the aggregation-induced abundant orbital energy levels on the heteroatom-cluster-rich surfaces. The presence of abundant hydrogen bonds and small gap between the lowest singlet and triplet excited states induces AIRTP. Finally, based on the unique multimode emission of the prepared CDs, their applications in high-performance white-light-emitting diode, information encryption, anti-counterfeiting, and visual humidity sensors are demonstrated.

Naturally Derived Luminescent Material in Engineered Silk and Its Application as a Fluorescent Dye with a Large Stokes Shift and Sensing Capability

Abstract:Silkworms have provided valuable byproducts (spanning from high-quality textiles to health supplements) to humans for millennia. Despite their importance in sericultural economy and biotechnology, manifold possibilities inherent in the myriad natural or artificially generated silk varieties have been underestimated. In this paper, we report that the Yeonnokjam silk strain, which shows light-green color, contains quercetin fluorochrome (QueF) in sericin, and QueF can be used as a fluorescence dye with a large Stokes shift and high sensitivity to environmental temperature and pH, thus functioning as an environmental sensing material. A Stokes shift exceeding 180 nm, a quantum efficiency of 1.28%, and a rapid fluorescence decay of 0.67 ns are obtained, which are influenced by solvent polarities. Moreover, QueF can be used as a UV blocker as well, and its low cytotoxicity and biocompatibility further suggest promising prospects for diverse application in cosmetics and medical materials in the future.

Abstract:<p>Fluorescent silk fibroin materials have wide applications in tissue engineering and bioassay system, but the inhomogeneity of fluorescence and high amount of additive hinder their development. In this study, we reported a super-strong composite silk with excellent blue fluorescence intensity and uniformity by feeding silkworm trace amount of hexaphenylsilole (HPS) (0.0012‰ in the diet), an important aggregation-induced emission (AIE) molecule. The incorporation of HPS nanoparticles in silk altered the second structures of silk fibroin, reduced crystallinity and crystallite size and induced orientation of crystalline region. In addition, the obtained composite silk exhibited strong mechanical properties with breaking elongation of 11.9 ± 1.7% and breaking strength of 503.0 ± 44.4 MPa, improvement of 17.8% and 54.0%, respectively, in comparison with regular silk. These super-strong and uniform fluorescent silks may pave a route to prevent the quenching effect of conventional fluorescent silk fibroin materials and be potentially applied in biomedical field.</p>

Abstract:The fate of xenobiotics in living organisms is determined by their in vivo absorption, distribution, metabolism and excretion. A convenient and scalable animal model of these biological processes is thus highly beneficial in understanding the effects of xenobiotics. Here we present a silkworm model to investigate the molecular properties-directed absorption, distribution and excretion of fluorescent compounds as model xenobiotics through introducing the compounds into the silkworm’s diet and monitoring the resulting color and fluorescence in the silkworm’s body. The efficient uptake of xenobiotics into silk has been further studied through quantitative analysis of the intrinsically colored and highly luminescent silk secreted by silkworm. Our findings provide first-hand insights to better understand the molecular properties that allow specific materials to be incorporated into silk while it is being produced in the silk gland. The use of resulting luminescent silk as scaffold for tissue engineering application has been demonstrated to clearly reveal the interaction of silk with cells. Furthermore, this new development also paves a way to produce various functional silk embedded with stimuli-sensitive dyes or drugs as novel biomaterials for in vivo applications.
Gamma-ray Irradiation Synthesis of Silk Fibroin-Coated CdS Quantum Dots

Abstract:Silk fibroin coated CdS quantum dots(SF-CdS QDs) were successfully synthesized in an aqueous system at room temperature under 60Co γ-ray irradiation.The prepared QDs were characterized by transmission electron microscopy(TEM),energy dispersion spectrometry(EDS),Fourier transform infrared spectrometer(FT-IR),X-ray diffraction(XRD),ultraviolet-visible spectroscopy(UV-vis) and photoluminescence(PL) spectrometry.The synthesized QDs were about 5 nm in diameter with excellent water-solubility.The QDs showed strong visible yellow-green luminescence under UV excitation,at an emission peak of around 538 nm.This method could be conveniently extended to fabricate other nanoparticles coated with silk fibroin.

Abstract:The development of red emission carbon dots with bright solid-state fluorescence would significantly broaden their application in optoelectronic devices and sensors. Herein, a red-emissive carbon dot-based nanocomposite has been synthesized through chemical bonding with cellulose films. The red emission originating from the surface states of carbon dots was maintained in the cellulose films. Due to the stable chemical bonding, the photoluminescence intensity and emission wavelength remained unchanged for 12 months, and the quantum yield of the composite was enhanced over 4 times. It also showed outstanding stability in water or weak acid-base environments under pHs ranging from 2 to 11. Therefore, the mechanism of chemical bonding that eliminated the defects and preserved the efficient radiative process through surface states was proposed.

Improving the mechanical property of silk by feeding silkworm with chiral carbon dots

Abstract:The influence of chiral materials on organisms is crucial. However, there is little research on the impact of chiral carbon dots (CDs), a kind of typical chiral materials, on biology. Herein, chiral CDs (L-/D-CDs) were synthesized using the thermal polymerization method from citric acid and chiral cysteine. The effect of chiral CDs on silkworms was explored through feeding silkworms with chiral CDs. The breaking strength of silk fibers (667.9 MPa) in D-CDs group exhibit a 71.4 % increase compared with control-silk (389.5 MPa), while the breaking strength of silk fibers in L-CDs group increases by 51.6 %. In addition, Fourier transform infrared spectra display CDs can prevent the transformation from random coil/α-helix structures to β-sheet structures. Furthermore, D-CDs group exhibit the highest percentage of four primary amino acids (glycine, alanine, serine, and tyrosine) relative to the total amino acids in silkworm hemolymph. This percentage is elevated by 70.5 % compared to the control group, thereby furnishing an ample supply of raw materials for the synthesis of silk proteins. In contrast, L-CDs group exhibit increase by 39.3 %. Our work provides new ideas and approaches for studying the effects of chiral materials on living organisms.

Abstract:Nitrogen-doped carbon dots (CDs) have attracted great interest due to their extraordinary properties, especially their enhanced emission efficiency, and thus a facile synthesis of nitrogen-doped CDs with high emission efficiency is critical for practical applications. To improve the emission efficiency of CDs, herein we employed Bombyx mori silk, which has high nitrogen content, as a raw material to prepare photoluminescent nitrogen-doped carbon dots through one-pot hydrothermal synthesis, and found that the as-prepared CDs have a photoluminescence (PL) quantum yield of 13.9%, and display amphoteric properties depending on the pH, are highly photostable, have low toxicity and are suitable for bioimaging.

Abstract:Carbon dots (CDs) have gained tremendous attention over a wide range from fluorescence sensors to optoelectronic devices. In this context, N, S-codoped carbon dots (MO-CDs) of 2.7 nm were synthesized via a facile hydrothermal method using a single precursor Methyl Orange (MO). Upon excitation at 430 nm, the MO-CDs exhibit a yellow emission with an absolute quantum yield (QY) of 15.3%. In addition, the as-prepared MO-CDs possess excellent resistance to high ion strength, high H 2 O 2 concentration, and UV exposure. Because of their good fluorescence properties, a light-emitting diode (LED) device that emits yellow light was fabricated by mixing MO-CDs and siloxane. Moreover, the electrospun nanofibers with yellow fluorescence were prepared by electrospinning technology. This work shows that the CDs derived from MO possess special fluorescent characteristics, which can be utilized as a multifunctional carbon material.

Abstract:In this work, we report a simple and thoroughly green approach to prepare photoluminescent carbon nanodots (C-dots) using soya bean grounds as a carbon source without any chemical additives. The obtained C-dots with an average diameter of 3.0 nm or so exhibited excellent biocompatibility and typically multicolor fluorescence. The practicality of C-dots for NIH3T3 cells imaging was validated, and NIH3T3 cells treated with C-dots became bright and showed different colors under different excitation wavelength.

Abstract:This novel materials assembly technology endows the designated materials with additional/enhanced performance by fixing "functional components" into the materials. Such functional components are molecularly recognized and accommodated by the designated materials. In this regard, two-photon fluorescence (TPF) organic molecules and CdTe quantum dots (QDs) are adopted as functional components to functionalize silk fibers and films. TPF organic molecules, such as, 2,7-bis[2-(4-nitrophenyl) ethenyl]-9,9-dibutylfluorene (NM), exhibit TPF emission quenching because of the molecular stacking that leads to aggregation in the solid form. The specific recognition between -NO2 in the annealed fluorescent molecules and the -NH groups in the silk fibroin molecules decouples the aggregated molecules. This gives rise to a significant increase in the TPF quantum yields of the silk fibers. Similarly, as another type of functional components, CdTe quantum dots (QDs) with different sizes were also adopted in the silk functionalization method. Compared to QDs in solution the fluorescence properties of functionalized silk materials display a long stability at room temperature. As the functional materials are well dispersed at high quantum yields in the biocompatible silk a TPF microscope can be used to pursue 3D high-resolution imaging in real time of the TPF-silk scaffold.
